Seed collection strategies for plant restoration with the aid of neutral genetic diversity

  • One key step in the plant restoration process is the collection of seeds from the field. For the selection of source populations of target plant species for translocation purposes (reintroduction or reinforcements), several approaches are possible. A practical method involves the use of data from reciprocal transplant studies. If no direct data are available, knowledge of population genetics and the phylogeography of the target species can serve as an alternative. In this short review, we briefly propose guidelines for those collecting seeds for plant species restoration based on population genetics theory, focusing on two main questions: Where does the plant material come from and how are sources designated, and how are seeds efficiently collected from local populations? While genetic data on a larger scale (phylogeography and population genetics) are needed to form a reply to the first question, similar data on a smaller scale (fine-scale genetic structures within populations) are necessary to shed light on the second issue.

  • The genus Iksookimia (Actinopterygii: Cypriniformes: Cobitidae) is a bottom-dwelling freshwater loaches, which are well-known as their endemism and high geographic variation. However, population genetic relationships among Iksookimia spp. have remained unclear due to a shortage of genetic markers that can be applied generally in the genus. Here, we developed high-resolving microsatellite markers using I. koreensis and I. longicorpa as representatives of Iksookimia species because of their wide distribution range and phylogenetic position. Ten of polymorphic microsatellite loci were isolated from Iksookimia koreensis and were successfully cross-amplified in I. longicorpa. The mean number of observed alleles per locus was about 10.4 (range, 2-17) for I. koreensis and about 13.2 (range, 2-24) for I. longicorpa. The loci, IK03 and IK08, deviated from the Hardy-Weinberg equilibrium in I. koreensis, after applying the Bonferroni correction. The microsatellite markers obtained in the present study will be useful to evaluate population genetic structure and to establish conservation strategies for I. koreensis and related Iksookimia species.

Genetic diversity of spotted scat (Scatophagus argus) in Vietnam based on COI genes

  • A spotted scat, Scatophagus argus, has a high nutritional value and is among Asia's most widely consumed fish species. Thua Thien Hue's consumption market considers this species to be of high economic value and requires protection and conservation of the population. However, the studies on the identification and genetic diversity of S. argus distributed in Vietnam are still lacking. Therefore, mitochondrial cytochrome c oxidase subunit I (COI) gene was utilized to distinguish different populations and investigate the genetic diversity of two populations of S. argus from Tam Giang lagoon, Thua Thien Hue province (n = 31) and Ca Mau province (n = 14). The sequencing results indicated 13 distinct haplotypes among 45 sequences. Five single nucleotide polymorphisms were observed to distinguish Hue spotted scat population. The S. argus population in Ca Mau province was higher haplotype diversity (Hd) and nucleotide diversity (π) than those of Thua Thien Hue province, which demonstrates that there are minor differences between haplotypes. There were genetic distances ranging from 0%-4% within the populations and 6.67% between the two populations. In addition to the sequencing, the comparison of morphology, biology, culture, and the growth rate was sufficient to distinguish the spotted scat S. argus in Thua Thien Hue from Ca Mau.

Genetic variation of the endangered species Halenia coreana (Gentianaceae)

  • Halenia coreana is an endangered, endemic species that is distributed in only a few locations in Korea, such as Mts. Hwaaksan and Daeamsan. It has been recently segregated from H. corniculata, broadly distributed in cold temperate regions that include northern Japan, the Russian Far East, northeastern China, Mongolia, and eastern Europe, where population sizes are usually large. To examine the genetic diversity of H. coreana and evaluate the level of genetic differentiation of the species compared with that of H. corniculata, we surveyed 183 candidate simple sequence repeats (SSR) motif markers for H. coreana and H. corniculata from sequence data of amplified fragments of a specific length in the genome. A total of 17 genomic-SSR markers were selected to examine the levels of genetic diversity and differentiation using 17 samples of H. coreana and 60 samples of three populations of H. corniculata. The results here suggest that the genetic diversity of H. coreana is very low with a high frequency of inbreeding within its population. We found that H. coreana is genetically differentiated from H. corniculata, supporting the recognition of the geographically isolated H. coreana as a distinct species.

A survey of the genome-wide genetic variation of Hibiscus hamabo (Malvaceae)

  • Hibiscus hamabo (Malvaceae) is a deciduous shrub mainly found in northeast Asia, including China, Japan, and Korea. Due to its limited distribution on Jejudo Island and at several sites in Jeollanam-do in Korea, H. hamabo has been designated as an endangered species by the Ministry of the Environment and has been the subject of several restoration programs. In this study, we quantified genetic variations using double-digestion restriction-associated DNA sequencing technology in 96 individuals of H. hamabo from 13 distinct populations in Korea. We determined 3,352 genome-wide single nucleotide polymorphism loci after stringent filtering processes and analyzed the level of genetic variation within and among populations as well as the population differentiation and genetic ancestry with various assumptions pertaining to the population origin. Our results indicated weak differentiations among populations surveyed in this study but clearly suggested that most of the H. hamabo populations maintain a relatively high level of genetic diversity as evidence of frequent genetic exchanges among populations via outcrossing or sequential gene flows. For a more detailed analysis of the origin of Korean H. hamabo and its demographic history, it will be necessary to expand sampling in China and Japan.

희귀 및 멸종위기식물인 광릉요강꽃의 개체군 구조 및 지속성 (Stage Structure and Population Persistence of Cypripedium japonicum Thunb., a Rare and Endangered Plants)

  • 광릉요강꽃(Cypripedium japonicum Thunb.)은 한국, 중국, 일본에 분포하는 동아시아 특산식물로, 지구 수준의 IUCN Red List "위기(Endangered, EN)"로 평가되며, 한국의 국가 Red List에서는 "멸종위기(Critically Endangered, CR)"에 포함된다. 본 연구는 광릉요강꽃 자생지에서 7년 동안 수행된 개체통계학적(demographic) 모니터링 자료를 바탕으로 개체군 구조 및 변화양상을 분석하고, 개체군의 지속성에 대하여 예측하였다. 광릉요강꽃은 국내 7개 지역(포천, 가평, 화천, 춘천, 영동, 무주, 광양)에서 자생지가 관찰되었고, 15개 아개체군에 4,356개체가 생육하는 것으로 확인되었다. 개체군 크기 및 구조는 지역별로 상이하였으며, 인위적 관리가 개체군 크기 및 구조 변화에 중요한 영향을 미치고 있었다. 7년 동안 개체 수 변화를 모니터링한 결과 광릉요강꽃의 개체군 생존력(Population Viability Analysis, PVA)은 지역별로 다양한 경향을 보였다: 향후 1세기 안에 멸절할 가능성은 포천 0.00%, 광양 10.90%, 춘천 24.05%, 화천 79.50%로 예측되었다. 모니터링이 수행된 위 연구지역은 현지 내 보호시설로 인위적 출입을 제한하고 있어 광릉요강꽃의 최대 위협요인인 인위적 남획 변수가 개체군 생존력에 반영되지 않았다. 즉, 실질적인 멸절 위험도는 본 연구에서 추정된 값보다 현저하게 클 것으로 예상된다. 국내 광릉요강꽃 개체군의 멸절위험도의 객관적 판단을 위해서는 향후 다양한 위협요인이 있는 여러 지역의 개체군 정보가 반영될 필요가 있으며, 국내·외로 광릉요강꽃 개체군에 대한 개체통계학적 모니터링을 확대해 나가야 할 것이다.

  • 식물 종은 주어진 환경조건과의 상호작용의 결과로 현재와 같은 특성을 나타낸다. 제비꽃속의 식물은 적응전략으로 개방화와 더불어 왕성한 영양번식체계를 선택하였거나 또는 폐쇄화를 발달시키는 전략을 선택하였다. 왕제비꽃은 제비꽃 속의 식물로 세계적으로 한반도와 중국 지린(길림)성의 동부에만 분포한다. 분포의 중심과 가장자리는 서로 다른 개체군 동태를 나타낼 것으로 예상된다. 왕제비꽃이 복합적인종자생산전략(Mixed-mating strategy)을 소유함에도 현재와 같은 제한적인 분포를 나타내는 원인을 알아볼 필요가 있다. 먼저 우리는 분포지의 식생환경을 조사하여 그 특성을 평가하였다. 식물계절학을 통해 생장특성을 알아보았다. 또한 개체군의 구조, 개방화의 결실특성, 폐쇄화의 생산성을 평가하였다. 그리고 2014년과 2018년 사이의 개체군크기 변화를 비교하였다. 분포지의 대부분은 계곡에 인접한 낙엽활엽수혼효림의 식생하부에 위치하였다. 4~5월에 자가불화합성의 개방화를 생산하였고 6~9월까지 폐쇄화를 생산하였다. 폐쇄화의 생산은 불확실한 환경변화에서 종자의 생산을 보증하기 위한 전략으로 평가되며 개방화에 비해 약 2배의 생산량을 나타내었다. 개체군의 구조는 안정적인 지역과 매우 불안정한 지역이 구분되었다. 2014년에 비해 2018년의 조사에서 개체군의 급격한 감소를 경험한 지역이 존재하였다. 이러한 큰 폭의 감소는 가장자리 개체군에서 크게 나타나는 것으로 평가되었다. 왕제비꽃 분포지는 가뭄이라는 자연적인 교란 요인과 분포지를 구성하는 기질에 따른 다양한 미소입지의 존재로 다양한 식물을 부양하였다. 이러한 불확실한 조건에서 폐쇄화를 통해 종자의 생산을 보증하고 그에 따라 자연적인 교란요인이 제거되었을 때 빠르게 유묘의 재정착과 개체군의 보충이 가능한 것으로 판단되었다. 그리고 환경요인은 모든 개체군 및 개체군 내의 모든 개체에 동일하게 작용하지 않는 것으로 추정되었다. 따라서 지역적으로 급격한 개체수의 감소에도 불구하고 완전한 소멸에 이르지 않을 것이고 환경의 변화에 따라 국지적인 소멸과 재정착이 반복될 것으로 예상되었다. 우리는 본 연구를 통해 수집된 종의 특성에 대한 정보를 바탕으로 추가적인 분포지의 조사가 필요함을 제안하였다. 또한 효과적인 보전 전략의 수립과 시행을 위해서는 보다 장기적인 개체군 동태와 개방화와 폐쇄화의 유전적 특성을 평가하는 것이 필요함을 제안하였다.

Effect of UV -B radiation on seedlings of two Solidago virgaurea populations from the Mt. Hakusan area of Japan

  • We collected seeds of Solidago virgaurea plants growing at different altitudes on the Mt Hakusan area in Japan and cultivated them in a naturally-lit green house. Three-week-old seedlings were irradiated with supplemental UV-B for 12 h each day for 1 and 2 weeks. After a week of itradiation the seedlings of the population collected from the higher altitude at Oh-nanjiho (ON) had accumulated more anthocyanins than those from the lower altitude at Bettoh-deai (BD). Levels of anthocyanins in the ON seedlings were highly correlated with the dose of UV-B radiation and the correlation was also observed after 2 weeks. The growth of the third leaves was retarded by UV-B radiation in both populations. The extent of growth retardation in the third leaves was correlated with the dose of UV -B radiation in both populations. However, no significant difference in the extent of leaf area growth was observed between the ON and BD populations. The increase in plant fresh weight was extensively inhibited in the ON seedlings after 1 week of UV-B radiation. The inhibition was recovered to those in the BD population by 2 weeks irradiation. These results indicate that these populations respond differentially to supplementary UV -B radiation during the first week. Because flavonoids such as anthocyanins play an important role in protection against UV-B radiation in many plants, populations growing at higher altitude may be better able to adapt to increased global levels of UV-B radiation.

Genetic diversity and structure of natural populations of Picea jezoensis in South Korea

  • Picea jezoensis (Sieb. et Zucc.) Carriere is one of the major and widespread components of the cold-temperate and boreal forests in Russian Far East, northeast China, Korea, and Japan. However, it is restricted to a highly fragmented range in South Korea with small populations. Mean expected heterozygosity $(H_e)$ based on 22 loci in 11 isozyme systems was 0.077 for four sampled populations that covered the whole distribution range of P. jezoensis in South Korea. This value is within the range reported for conifers, but it is very low compared to that of other spruce species as well as that of P. jezoensis populations in Russian Far East. Most populations had a slight excess of heterozygotes and the Wright's $F_{IS}$ (-0.019) was comparable to that previously reported for other spruce species. In all of the four populations, the Wilcoxon sign-rank test indicated no greater heterozygosity than that expected for populations at mutation-drift equilibrium, suggesting that the populations have not been bottlenecked recently. Despite a fragmented range and isolated populations, population differentiation was not high $(F_{ST}=0.047)$ and the number of migrants per generation was 5.09. Nei's genetic distances were also small $({\bar{D}}=0.005)$ but strongly related to geographic distances between populations, suggesting an Isolation by Distance. The northernmost isolate, Mt. Gyebang population was genetically distinct from the other three populations. Implications for the conservation of genetic variation of P. jezoensis in South Korea were discussed.

Genetic Diversity and Population Genetic Structure of Cephalotaxus koreana in South Korea

  • The Korean plum yew (Cephalotaxus koreana Nakai) is a shade-tolerant, coniferous shrub. The seeds have been used as a folk medicine in Korea, and an alkaloid extract (HTT) is known to have anticancer properties. We estimated the genetic diversity of 429 trees in 16 populations in South Korea using 194 polymorphic amplicons from seven combinations of AFLP primer-restriction enzymes. The average number of effective alleles and the percentage of polymorphic loci were 1.37 and 79.4%, respectively. Shannon's diversity index and the expected heterozygosity were 0.344 and 0.244, respectively. We divided 16 populations into four groups on the UPGMA dendrogram and the PCA biplot. The first two principal components explained 84% of the total genetic variation. Genetic differentiation between populations explained 14% of total genetic variation, and the remaining 86% came from difference between individuals within populations, as determined by an analysis of molecular variance (AMOVA). However, the genetic differentiation did not correlate with the geographic distance between populations from the Mantel test. The Bayesian statistics, which are comparable to Wright's $F_{ST}$ and Nei's $G_{ST}$, were ${\theta}^I=0.406$ and ${\theta}^{II}=0.172$, respectively. The population genetic diversity was slightly lower, and the strength of genetic differentiation was much weaker, than the average of those plants having similar life histories, as assessed using arbitrary marker systems. We discuss strategies for the genetic conservation of the plum yew in Korea.
