Histopathological changes in fingerlings of Japanese flounder, Paralichthys olivaceus, with severe scuticociliatosis (스쿠지카섬모충에 중감염된 넙치 (Paralichthys olivaceus) 치어에 대한 병리조직학적 관찰)

  • In order to elucidate the patterns of tissue damage evoked by the scuticociliatids, eighteen fingerlings of Japanese flounder, Paralichthys olivaceus, heavily infected with an unidentified scuticociliatid were histopathologically examined. Skin layer with the underlying musculature were severely necrotized due to the infestation of the ciliates. However in the early lesions, both dermis and myofibres remained relatively intact compared with other surrounding loose connective tissues. Mild damages were found in more dense tissues. One or more scuticociliatids were recognized in the blood and lymph vessels of the loose connective tissues with or without destructive changes. Many of nerve trunks or ganglia were also parasitized with less marked histological damage in the parenchyma. Dura and its adjacent tissues in the spinal cord were severely necrotized with massive accumulation of the ciliates in subdural space. The parasitic invasion in the central nervous system was usually confined to the cortical region. In the gill, variable degenerative changes were occurred due to the invasion of the ciliates recognized in the blood vessels of branchial arches or primary filaments. From these results, it was strongly suggested that the scuticociliatids are very actively penetrated into the deep tissues mainly through the severe destruction of the loose connective tissue components and that the vascular system could play a role in the rapid distribution of the ciliates to the remote tissues or organs.

An Experimental Study on the Effect of Tissue Conditioner on the Oral Mucosa (Tissue Conditioner가 구강조직에 미치는 영향에 관한 실험적 연구)

  • An experimental study was performed to study the tissue reactions to tissue conditioners in rabbits. Thirty rabbits were divided into three groups, 10, Hydro-cast group, 10, Coe-comfort group, and 10, heat-cured resin group. Tissue conditioners and heat-cured resin were embedded in the oral mucosa of rabbits. The tissue specimens were removed on 3rd, 7th, 14h, 21st and 28th day after embedding and examined under microscope after staining them with H-E stain, Van Gieson's stain, Masson's trichrome stain and PAS reaction. The results were as follows : 1. Tissue reactions to tissue conditioners were somewhat different from each other in the early stage, but, with the increase of the embedding period, the fibrous capsule was thickened in both. These tissue reactions were similar to those to heat-cured resin. 2. Newly formd fibrous components were stained deep-red with Van Gieson's stain and dark-green with Masson's trichrome stain. But their stainability was decreased as collagenous fibers became matured. 3. Newly formed fibrous components showed intense PAS reactivity, but PAS reactivity was reduced as the connective tissue capsule became completed.

INFLUENCE OF CO-CULTURED FIBROBLASTS ON THE DIFFERENTIATION OF MOUSE CALVARIA-DERIVED UNDIFFERENTIATED MESENCHYMAL CELLS IN VITRO (복합 및 유격배양한 섬유모세포가 마우스 두개관 미분화간엽세포의 골세포 분화에 미치는 영향)

  • This study was designed to evaluate the influence of fibroblasts or connective tissue from mouse oral mucosa on differentiation of neonatal mouse calvaria-derived osteoblasts and mineralization of bone nodules. Primary cell cultures from mouse calvarial osteoblasts and 2-4 passaged fibroblasts from oral mucosa were co-cultured in monolayer cultures, devided into 6 experimental group according to cell density or cell confluency. Osteoblasts were also co-cultured with fibroblasts in $Transwell^{(R)}$ culture plate with different co-cultured period according to osteoblast differentiation. The alkaline phosphatase activity were measured in monolayer cultures and cultures using $Transwell^{(R)}$. The mineralized bone nodules were presented by Von Kossa staining and density of mineralized nodules was measured by image analysis. The connective tissues with or without osteoblast seeding were cultured and examined histologically by Von Kossa and Trichrome Goldner staining. The results were as follows; 1. Prolonged maturation of matrix and delayed mineralization of bone nodules were resulted in monolayer cultures. 2. Co-culture of fibroblast with osteoblast using $Transwell^{(R)}$ during osteoblast proliferation stage stimulated proliferation of osteoblasts and increased alkaline phosphatase activity and mineralization of bone nodules. 3. Co-culture of fibroblast with osteoblast using $Transwell^{(R)}$ during matrix mineralization stage decreased and delayed mineralization of bone nodules. 4. In vitro cultured connective tissue with osteoblast seeding resulted in proliferation of osteoblasts and matrix formation with mineralization.

The Histopathology of Hyeungbangjihwang-tang Extracts on the Liver Cirrhosis of Fisher 344 Rats Induced by Long Time Carbon Tetrachloride Injection. (사염화탄소의 장기(長期) 투여(投與)로 유발된 백서(白鼠) 간경변(肝硬變)에 대한 형방지황탕(荊防地黃湯)의 조직병리학적 영향)

  • Objective : This study was carried out to investigate the effects of oral administration of Hyeungbangjihwang-tang (HBJHT) on the liver cirrhosis of rats induced by Carbon tetrachloride for 10 weeks. Method : The histopathological changes were observed. The HBJHT extracts were daily dosed at 50, 100 and 200mg/kg for 12 weeks. Results : Severe hepatocellular necrosis and ballooning, hyperplasia of connective tissue, subdued reduction of hepatic lobule, and hyperplasia of bile duct in portal triad were dramatically decreased in the HBJHT-treated group compared to that of the Carbon tetrachloride-treated control group in histopathological observation. The diameter of hepatic lobules was significantly enlarged in the HBJHT-treated group compared to that of the Carbon tetrachloride-treated control group, and the amounts of connective tissue, degenerative cells and bile ducts were significantly and dose-dependently decreased. Conclusion : It is concluded that HBJHT has a significant recovering effect on the liver of rats induced by carbon tetrachloride.

  • A depressed alvelolar ridge and a collapsed interdental papilla in the anterior region may interfere with esthetic prosthodontic repair. The resultant sequelas will cause overcontoured prosthesis or dark shadow under pontic which may not suffice our patients' esthetic demands. So, surgical periodontal treatment has been needed for esthetic repair in deformed ridges and extensive interproximal spacing. Our treatment option alters Dr.Abram's roll technique to make palatally extended connective tissue secondary pedicle flap to obtain additonal connective tissue. Blood supply maintenance, least trauma, and no secondary wound is advantages in this modification.

Histopathological Studies on Experimental Nitrate Poisoning in Rabbits (질산염중독(窒酸鹽中毒)에 관한 병리조직학적연구(病理組織學的硏究))

  • In order to clarify the histopathological changes resulting from nitrate poisoning, rabbits were experimentally poisoned by the oral administration of $KNO_3$ or $NaNO_2$ and examined clinically and histopathologically. In addition, the quantitative changes of glycogen level in hepatic cells were histochemically observed. The results obtained were summarized as follows: 1. Clinical symptoms observed from the acute cases which died within 2 hours after the administration were severe cyanosis of visible mucosa, frequent urination, and dyspnea. However, in chronic cases administrated daily with $KNO_3$ for 43, 50 and 74 days respectively, no marked symptoms were observed. 2. Macroscopic changes observed in acute cases were severe methemoglobinemia, cloudy swelling of hepatic cells, hemorrhage and hyperemia of gastric mucosa, and hyperemia of other organs. In chronic cases there were marked hyperemia, dark-red coloring and increasing of consistency in liver and kidney, and swelling of spleen. 3. Microscopic changes observed in acute cases were hemorrhage and hyperemia of various organs, cloudy swelling and centrilobular necrosis of hepatic cells and necrosis of convoluted tubular epithelium in kidney. In chronic cases there were round cell infiltration of the interlobular connective tissue and epithelial proliferation of interlobular bile ducts in the liver, and necrosis of the convoluted tubular epithelium and proliferation of interstitial connective tissue in kidney, thickening of alveolar septa of lungs, activated hemopoiesis of bone marrow, and myeloid metaplasia of sqlenic pulp. 4. Glycogen storage in liver cells was decreased in acute cases, on the contrary, increased in chronic cases.

Metastatic Malignant Mixed Tumor of Mammary Glands in an Irish Setter Dog : A Case Report (개의 악성유선혼합종의 전이 예)

  • An 11 years old Irish Setter bitch was euthanlzed and necropsied because of clinical findings such as severe purulent nasal discharge and formation of large tumor mass, 8 ${\times}$8cm in size, in the abdominal cavity. A complete unilateral mastectomy had been carried out twice 14 and 22 months before necropsy. The surgically removed masess of the mammary glands had been diagnosed as malignant mixed tumor in each time. Grossly, tumor masses were observed in nasal cavity, infralumbar lymph node, lung, abdominal cavityn and brain. Microscopic findings of the surgically removed masses consisted of tumor epithelial cells, tumor hyaline cartilage-like structures and abundant connective tissues. The mass of the lymph node had similar microscopic features to those of the original malignant mixed tumor of the mammary glands. The tumor osseous tissue and osteoid were observed in the abdominal cavity, lung, and brain. Myoepithelial cells were frequently found on association with the metastatic tumors. From the results, it was concluded that malignant mixed tumor of the mammary glands metastasized to the infralunbar lymph node, abdominal cavity, lung and brain. In addition, the observation in this study supported two theories at the same time that the bone in malignant mixed tumor arises by endochondral ossification of the cartilage formed by the myoepithelial cells and arises by intramembranous ossification of stromal connective tissue or transformed myeopithelial cells. Solid carcinoma of the nasal epielia and granulosa cell tumor were also diagnosed in a mass of the nasal cavity and of the ovaries respectively.

Comparative studies of histopathologic pulmonary lesion of cattle and deer tuberculosis by Mycobacterium bovis (Mycobacterium bovis에 의한 소 및 사슴 결핵의 폐 병리조직학적 소견 비교연구)

  • Comparative studies of histopathologic lesions from 23 purified protein derivative (PPD) positive cattle, 21 slaughter cattle found with tuberculosis, and 11 tuberculosis-positive elk (Cervus elaphus) were performed. PPD positive cattle did not show specific histopathologic lesions in all 23 heads that were no visible lesion reactor. Slaughter cattle found with tuberculosis revealed microscopically classical granulomatous lesion (tubercle) with central caseous necrosis surrounded by mantle of epithelioid cells and Langhan's giant cells capsuled by connective tissue in lung. Elk was noted with some different lesion patterns with classical granulomatous lesion and suppurative abscesses that was composed of fibrin, degenerated cells without having connective tissue. In addition, many Langhan's giant cells infiltration in alveoli at peripheral lesion were observed in some cases of classical granulomatous lesion and suppurative abscesses.

Studies on the Appearance and Function of the Central Cavity of Corpus Luteum in the Ovaries of the Korean Native Cattle (한우 난소의 중심강이 있는 황체의 출현과 기능에 관한 연구)

  • The appearance and function of corpora lutea(CL) with a central cavity in the ovaries of Korean Native Cattle (KNC) were investigated endocrinologically and histochemically. The CL were enucleated from KNC ovaries within 2~3 hrs local slaughter house and classified with central cavity CL or not. Enzymatically dispersed luteal cell (1$\times$106 live cell/ml of Dulbecco's Modified Eagle Media) with or without cavity of CL cultured at 37$^{\circ}C$ in a humidified incubation (5% CO2 : 95% air) for 72hr. A central cavity in the CL of KNC was found in 58.8% of CL-1, 34.9% of CL 2, 39.1% of CL-3, and 11.1% of CL-4, respectively. There were no significant difference between protein content of CL with and without a central cavity. Mean progesterone secretion after 36h of in vitro luteal cell culture were significantly (p<0.05) higher in CL with central cavity than without cavity. However, the luteal cavitic wall was composed of the connective tissue band of the reticular and collagen fibers and then these connective tissue band extended into the CL with cavity. These results suggest that the central cavity of CL may be caused infertility in KNC.

  • A study of pulmonary diseases in the slaughtered cattle (n = 125) of Mymensingh town was conducted to study pathological findings using the standard methods from September 2001 through April 2002. The pulmonary lesions observed in this study included congestion, emphysema, anthracosis, pleuritis abscess and hemorrhage. The histopathological findings of congestion were characterized by hemorrhage, a large number of leukocytes infiltration in the lumen of the alveoli, bronchitis, bronchiolitis, proliferation of fibrous connective tissue and hyperplasia of bronchiolar epithelia. Histopathologically, emphysema was identified by the distended alveoli with thin and atrophied alveolar walls. In anthracosis, carbon particles were found in stroma and alveolar lumen. In pleuritic lesions, there was a proliferation of fibrous connective tissue along with the infiltration of mononuclear reactive cells. Abscesses were characteristic of the accumulation of neutrophils surrounded by immature fibroblasts forming a capsule like structure.
