A method based on RNA-transcript conformation polymorphism (TCP) was tested for detection of two PVY isolates in a mixed infection. Differences in electrophoretic mobility of RNA transcripts copied from PCR products of each virus isolate enabled the distinction between the two virus isolates in a mixed infection. The identities of the RNA transcripts and hence of the infecting virus isolates were determined by annealing to reference oligonucleotides containing unique strain-specific sequences visualized by retardation of transcript mobility in gel. The ratio at which both virus isolates could be detected was as low as 1:10. The suitability of this procedure for the study of mixed virus infections is discussed.
Molecular mechanics and conformation search methods were carried oyt to investigate the relationship between conformations and thromboxane synthetase ingibitory activities of om ega-pyridylalkenoic acids. The initial geometries of ${\omega}$-pyridylalkenoic acids and heme part of cytochrome P-450 were obtained from MM+ geometry optimization. The bond lenths and angles were not varied by step during the conformation searching. Stable conformers of some ${\omega}$-pyridylalkenoic acids were obtained by comformational search method. The distances were 8.5~10.8 ${\AA}$- between N atom at 3-position of pyridine ring and C atom at carboxylic group of stable ${\omega}$-pyridylalkenoic acids. The conformations of ${\omega}$-pyridylalkenoic acids and heme part complex were determined by same method. In theses structures, benzene ring and ethylene group in ${\omega}$-pyridylalkenoic acids are making the structure more rigid and increase inhbitory activity. The electron donating groups in C atom shich is connected to pyridine ring also increase activity.
N-Hydroxy diketopiperazine derivatives from L- and D-forms of Ser and Cys were synthesized for the first time and the ring conformation of both analogues in water was determined to be close to flat via NMR and ab initio calculations. However, the side chain of the Ser analogue was oriented toward a pseudo-equatorial position while that of the Cys analogue was slightly oriented toward a pseudo-axial direction, with a slightly distorted boat-shaped ring conformation. Among them, the D-Cys analogue was found to be a weak inhibitor of α- glucosidase.
Seven derivatives of 2,3-alkyl substituted N-cyanoaziridine were stereoselectively prepared from alkyl substituted alkenes and cyanamide. NMR spectral property and conformation of N-cyanoaziridine were analysed. Protons(3.34 ppm) at 2,3 position of N-cyanoaziridines are significantly deshielded in case of cyclopentyl fused N-cyanoaziridine. These protons are getting more anisotropically shielded with increasing the fused carbocycle size of N-cyanoaziridine. Therefore chemical shift for these protons are upfield shifted to 2.80 ppm in case of cyclooctyl fused N-cyanoaziridine. Their ED$_{50}$ values aganist L$_{1210}$, cell in vitro were evaluated as 0.5~8.0 $\mu$g/ml. Conformation of carbocycle of fused N-cyanoaziridine is more important to their cytotoxicity than the increment of the strain energy of 3-membered ring.
A linear undecapeptide, Substance P (SP) is involved in a wide variety of physiological processes such as pain, inflammation, salivation, and hypertension. Tertiary structure of SP in dodecylphosphocholine (DPC) micelles has been investigated by CD, NMR spectroscopy, and DGII calculation. CD spectrum of SP in the presence of 7.5 mM DPC micelles does not show any favorable secondary structure. The tertiary structure determined by NMR spectroscopy and DGII calculation shows that the Phe7-Phr8-Gly9-Leu10 region adopts a turn structure, while the N-terminal region is quite flexible. Both prolines in SP exist preferentially as the trans isoforms and the aromatic ring of Phe7 protrudes outward. Conformation of SP may be restrained by the contact of the Phe7 aromatic ring with the hydrophobic side chains of the DPC micelles and this interaction induces a turn structure. Structure of SP in aqueous solution in the presence of DPC micelles can represent a good model to study the conformation recognized by the receptor near neutral membrane.
Taking advantage of the heat inducible HSP82 gene in yeast, chromatin structure after transcription cessation was investigated. Alteration of chromating conformation within the HSP82 gene transcription unit into an active state has been shown to correlate with its transcriptional induction. It was thus of interest to examine whether the active chromatin state within the HSP82 mRNA analysis, the gene ceased its transcription within a few hours of cultivation at a normal condition after heat induction. In this condition, an active chromatin conformation in the HSP82 gene body was changed into an inactie state which was revealed by DNase I resistance and by typical nucleosomal cutting periodicity in the corresponding chromatin. These results thus ruled out the possibility of a long-term maintenance of the DNase I sensitive chromatin after transcription cessation. DNA replication may be a critical event for the chromatin reprogramming.

It has been postulated that acetylcholine exhibits both nicotine and muscarinic activity because of its ability to present two patterns of essential atoms to the receptors. These two patterns arise from the ability of the molecule to exist in more than one preferred conformation. The molecule S(+)-acetyl-${\beta}$-methylacetylcholine exhibits only muscarinic activity. Calculations using molecular orbital theory predict that this molecule prefers only the muscarinic conformation. This is offered as an explanation for the exclusive role of the molecule and as evidence supporting the twoconformation, two-activities hypothesis.
The conformation of synthetic oligosaccharide can be elucidated by employing molecular modeling and highfield proton NMR (nuclear magnetic resonance) spectroscopy. Information with respect to the composition and configuration of saccharide residues and the sequence and linkage positions of the oligosaccharide can be obtained by employing a variety of one- and two-dimensional NMR techniques and molecular modeling. These techniques are also useful in establishing the solution conformation of the oligosaccharide moiety. This study is focused on the elucidation of linkage positions of synthetic trisaccharides, Gal(β1-4)Glc(β1-3)Glc, Gal(β1-4)Glc(β1-4)Glc and Gal(β1-4)Glc(β1-6)Glc.

By means of NMR spectrum analysis of the synthesized MBBA and EBBA, it has been found that benzene rings of p-n-butylaniline in both MBBA and EBBA molecules do not conjugate with the central double bond and the benzene ring is twisted from molecular plane of N-(p-methoxy or ethoxy benzylidene) group. And as a result of MO studies, the minimum energy conformation is found for the conformation of $30^{\circ}$ twisted angle. One sees reasonable agreement between theory and experiment.
Reaction of tetrahomodioxa p-phenylcalix[4]arene with ethyl bromoacetate and potassium carbonate in ace-tone leads to the title tetra ester derivative, 7,13,21,27-tetra-phenyl-29,30,31,32 -tetrakis(ethoxycarbonyl)meth-oxy-2,3,16,17-tetrahomo-3,17-dioxacalix[4]arene, its structure was determined by NMR spectra as 1,4-alternate conformation. The molecular structure has been solved by X-ray diffraction methods. The molecule has a conformation with pseudo center of symmetry. The benzene ring A is up, ring C is down, B and D rings are flat with respect to the plane of the macrocyclic ring.
