Development of 'Damogy' Cultivar of Gomchwi with Disease Resistant, High Quality, and Yield (고품질 내병 다수성 곰취 신품종 '다목이' 육성)

  • A new Gomchwi cultivar 'Damogy ' was bred by crossing between Gomchwi (Ligularia fischeri (Ledeb.) Turcz.) and Handaeri-gomchwi (Ligularia fischeri var. spiciformis Nakai). The selection and investigation of growth and yield characteristics were conducted from 2006 to 2011 in field and greenhouse of Highland Agriculture Research Institute, NICS, Rural Development Administration, Korea. On a newly developed cultivar 'Damogy', Color of petiole ear was purple, petiole trichome was exist, light of leaf back was not exist, and density of leaf vein was degree 4. Plant height, leaf length, leaf width and petiole length were 45.9, 16.9, 21.1 and 29.0 cm, respectively in the $3^{rd}$ year in growth characteristics. Plant size was also higher than that of Gondalbi. Bolting and flowering time were Aug. $23^{th}$ and Sept. $10^{th}$, respectively. In contrast, Gondalbi showed quite earlier bolting time more than 38 days compared with 'Damogy' and consequently earlier flowering time more than 26 day. 'Damogy' showed similar leaf number (129) per plant compared to 'Gondalbi' (130). Furthermore, yield was higher 'Damogy' (1,889 g/plant) than in 'Gondalbi' (798 g/plant). 'Damogy' showed higher leaf thickness (0.71 mm) than 'Gonalbi' (0.46 mm), and consequently showed more hardness in leaf characteristics ($28.3kg/cm^2$) compared with 'Gondalbi' ($23.0kg/cm^2$). 'Damogy' showed higher resistance in the susceptibility of powdery mildew disease compared to 'Gondalbi'. 'Damogy' variety was registered plant variety protection right as a No. 89 on December 2015.

Analysis of Epidemiological Characteristics, PFGE Typing and Antibiotic Resistance of Pathogenic Escherichia coli Strains Isolated from Gyeonggi-do (경기도에서 분리한 병원성대장균의 역학적 특성 및 PFGE, 항생제 내성 연구)

  • This study was conducted to survey the epidemiological characteristics and the isolated strains for pathogenic E. coli which was the major causative organisms for food poisoning occurred at school food services in the Gyeonggi-do area during the past three years. We investigated 19 accidents of food-borne disease outbreaks by pathogenic E. coli at school food services from 2010 to 2012. Food-borne disease outbreaks by pathogenic E. coli were usually occurred at direct management type (18 accidents, 95%) and high schools. For the seasonal factors, 13 accidents (65%) were occurred in June to September, especially the end of August and September after the summer holidays. The first patients were occurred on Wednesday (7 accidents, 37%) and Thursday (7 accidents, 37%), and they were mainly reported on Thursday (7 accidents, 37%) and Friday (5 accidents, 26%). The exposure of risk was estimated in Monday (4 accidents, 21%), Tuesday (7 accidents, 37%) and Wednesday (4 accidents, 21%), and kimchi (5 accidents, 50%) was estimated as the food of the high risk responsible for the outbreaks. 98 isolates of pathogenic E. coli consisted of PEC (50%), ETEC (34%), EAEC (15%), and EHEC (1%). The antibiotic resistance of pathogenic E. coli showed in the descending order of ampicilline (40%), nalidixic acid (37%), trimethoprim/sulfamethoxazole (24%), and tetracycline (19%). The antibiotics of second and third generation cephalosporins, cabarpenem, aminoglycosides, and second generation quinolones had antimicrobial susceptibilities and cefalotin, ampicillin/sulbactam and chloramphenicol showed medium resistance at 29%, 25%, and 6% respectively, and 70% of isolates were resistant to more than one antibiotic. By the PFGE analysis, they were classified into nine major groups and 31 profiles with 57% pattern similarity. It was very difficult to find the correlation of antimicrobial susceptibilities and genotype in the small scale-food poisoning, but the similarity of antimicrobial resistance and PFGE patterns in the large scale-food poisoning enabled the outbreaks to estimate the same pathotype of E. coli derived from identical origins.

Resistance Degree of Radish Cultivars to Fusarium oxysporum f. sp. raphani according to Several Conditions (발병조건에 따른 무 품종들의 시들음병에 대한 저항성 차이)

  • This study was conducted to establish the efficient screening system for resistant radish to Fusarium oxysporum f. sp. raphani. Five radish cultivars ('Myoungsan', 'Chungdu', 'Jangsaeng', 'Hannongyeorm', and 'Chungsukungjung') showing different degree of resistance to the fungus were selected. And the development of Fusarium wilt of the cultivars according to several conditions such as root wounding, dipping period of roots in spore suspension, inoculum concentration, and incubation temperature to develop the disease was tested. In distinguishing the resistance degree of the radish cultivars to the disease, non-cut roots were more effective than cut roots. And occurrence of Fusarium wilt of the radish plants increased in the proportion to increase of root-dipping period and spore concentration of the fungus. Thus, optimum conditions to differentiate susceptible and resistant cultivars to the disease were root-dipping period of 0.5 hour and spore concentration of $1{\times}10^7\;conidia{\cdot}mL^{-1}$. Disease severity of Fusarium wilt on the cultivars was changed with incubation temperature and the radish seedlings incubated at $25^{\circ}C$ represented the most difference of resistance and susceptibility to Fusarium wilt. From the above results, we suggest that the efficient screening method for resistant radish to Fusarium oxysporum f. sp. raphani would be to dip non-cut roots of fourteen-day-old radish seedlings in spore suspension of $1{\times}10^7\;conidia{\cdot}mL^{-1}$ for 0.5 hour and to transplant the inoculated plants to plastic pots with fertilized soil, and then to incubate the radish plants at a temperature of $25^{\circ}C$ for development of Fusarium wilt.

Reliability and validity of Korean version of the OHIP for edentulous subjects: A pilot study (무치악 환자들을 위한 한국어 버전의 구강건강영향지수 신뢰도와 타당성 평가를 위한 모의연구)

  • Purpose. The purpose of this pilot study is to evaluate the reliability and validity of the Korean version of the oral health impact profile (OHIP-EDENT K) for edentulous patients. Materials and methods. The study was conducted on 12 patients who fabricated overdenture in the Department of Prosthodontics, Korea University, Guro Hospital. All subjects completed the Korean version of Oral Health Impact Profile (OHIP K) questionnaire. Shorten version of the OHIP called OHIP-14 K and OHIP-EDENT K were derived from the datasets. Cronbach's alpha was used to measure internal consistency of the summary scores for OHIP-EDENT K. The Spearman's correlation coefficient between the summary scores for OHIP-EDENT K and OHIP K was calculated to evaluate concurrent validity. Results. The reliability of the summary scores for OHIP-EDENT K was acceptable (α=.736). The Spearman's correlation coefficient of the summary scores for OHIP-EDENT K and OHIP K was 0.966, which was statistically significant (P<.001). OHIP-EDENT K exhibited less susceptibility to floor effects than OHIP-14 K and appeared to measure change as effectively as OHIP K. In order to prove the reliability, responsiveness and validity of OHIP-EDENT K, further studies with more samples are needed. Conclusion. The OHIP-EDENT K, a questionnaire on oral health-related QOL comprising 19 items, has measurement properties comparable with the full 49-item version. This modified shortened version can be an alternative questionnaire to full version of OHIP K and OHIP-14 K in edentulous patients.

Cation Content of Salt-Tolerant and -Susceptible Cultivars and Its Inheritance in Rice (벼 내염성 및 비내염성 품종들의 양이온함량과 그의 유전)

  • This study was conducted to obtain the basic information on salt-tolerance of rice cultivars in relations with growth retardation and cation content. As the salt-level increased, less retardation in plant height and dry weight were shown in salt-tolerant than salt-susceptible rice cultivars. Salt tolerant cultivars showed lower N $a^{+}$ and higher $K^{+}$ content and lower N $a^{+}$ / $K^{+}$ ratio at each salt-levels than salt-susceptible ones, while there were no significant differences in $Ca^{++}$ and $Mg^{++}$ content. At the tillering stage, the plant height and dry weight of the salt-treated plots were significantly correlated with N $a^{+}$ and $K^{+}$ content and N $a^{+}$ / $K^{+}$ ratio, implying that N $a^{+}$ and $K^{+}$ content could be an indicator of salt-tolerance of a rice cultivar. There were no tiller-depending differences in cation content in all cultivars. N $a^{+}$ content and N $a^{+}$ / $K^{+}$ ratio in leaves were lower at the top and higher at the bottom. In three $F_1$ hybrids between salt tolerant parent Pokkali and three salt susceptible parents, plant height, dry weight, $K^{+}$ content and N $a^{+}$ / $K^{+}$ ratio were similar to those of Pokkali parent, while N $a^{+}$ content was intermediate of the parents. So, it seemed that salt tolerance is dominant over salt susceptibility. In $F_2$ of Pokkali/wx817 cross, genetic segregation of plant height, dry weight, $K^{+}$ content and N $a^{+}$ / $K^{+}$ ratio varied continuously and was biased onto Pokkali side, while that of N $a^{+}$ content showed normal distribution with intermediate mode. Broad-sense heritability of the characters ranged from 0. 604 to 0. 811. Suggestions of Stone Materials for Restoration of Gwanghwamun Woldae in Seoul Based on Lithological Study (암석학적 연구를 통한 서울 광화문 월대 복원용 석재 제안)

  • This study is to analyze the lithological similarities between railing stones of Gwanghwamun Woldae and stone blocks stored in Donggureung Royal Tombs, Guri, to interpret the provenance of the stones, and to suggest the most suitable quarry for a new stones supply among the rocks in the Seoul-Pocheon area in order to select stones for the restoration of the Gwanghwamun Woldae. The railing stones in Donggureung are medium to coarse-grained pink biotite granite, and their lithological characteristics, magnetic susceptibility(Average 5.20 ×10-3 SI unit), and gamma spectrometer data(K 5.00~6.38%, U 4.92~8.56 ppm, Th 27.60~36.44 ppm) show similarities with the remaining railing stones in Gwanghwamun Woldae(Average 5.38). Therefore, the railing stones in Donggureung can be reused for the restoration of Gwanghwamun Woldae. They have similar lithological and geochemical charateristics to the pink biotite granite found the Suraksan Mt. and Buramsan Mr. areas in Seoul, suggesting that these areas are the source of the stone. However, since the Suraksan Mt. and Buramsan Mt. areas are currently unavailable for quarrying, lithological and geochemical analyses on granite from the Yangju and Pocheon areas are conducted to determine the suitability of the new stone for restoration. As a result, a pink biotite granite similar to the Woldae stones was identified in the Pocheon area, and it is suggested that the stones similar in grain size and colour to the railing stones of Gwanghwamun Woldae should be selected and used for the restoration of Gwanghwamun Woldae.
