P(3HB) Accumulation in Alcaligenes eutrophus H16(ATCC 17699) under Nutrient-Rich Condition and Its Induced Production from Saccharides and Their Derivatives

  • Poly(3-hydroxybutyrate)(P(3HB)) accumulation under nutrient-rich condition with various amounts of $(NH_4)_2 SO_4$ was systematically investigated. The results of the electron-microscopy and the solvent extraction showed that the P(3HB) accumulation is unavoidable even under nutrient-rich condition. This indicates that in a two-step culture of Alcaligenes eutrophus H16, the researches should be careful in interpreting the data of polyhydroxyalkanoates(PHAs) accumulation in terms of the carbon-source fed in the second step because the two-step culture product contains the P(3HB) produced under nutrient-rich condition. The polyester production capability in a two-step batch culture of A. eutrophus H16(ATCC 17699) was also investigated using various saccharides and their derivatives such as glucose, fructose, gluconic acid, glucaric acid, sorbitol, lactose, galactose, and mannose. The polyesters synthesized were characterized by 500 MHz$^{1}H-NMR$ spectroscopy, intrinsic viscosity$[\eta]$ measurement in chloroform and differential scanning calorimetry(DSC). 500 MHz $^{1}H-NMR$ analysis showed that all polyesters synthesized generally contained 1~2 mol% of 3HV. Another finding is that the glucose utilization can be increased by changing the autoclaving procedure of the substrate to enhance the P(3HB) production yield up to 46 wt% of P(3HB) in dry cells.

고온, 고습조건이 저장중 가공 잎담배의 pH, 색상 및 화학성분에 미치는 영향 (Effects of High Temperature and Humidity on the pH, Color and Some Chemical Constituents of Processed Leaf Tobacco During Storage)

  • This study was carried out to investigate the effects of high temperature and humidity on the ageing of processed leaf tobacco. Four(1993) or six(1994) grades (Both flued-cured and burley) of processed leaf packed in carton box were stored under the natural and artificial conditions. When the Processed leaf was stored in the controlled mom at 40℃ with 75% R.H.(1993) for 40 days, the leaf pH was decreased. The decreasing rate of leaf pH was similar to that of leaf stored under the natural warehouse condition for 15 months. The degree of lightness and yellowness of leaf also decreased, while the decaying or darkening of the leaf was observed. When the processed leaf was stored in the controlled room at 35℃ with 65% R.H.(1994) for 90 days, the pH of flue-cured was decreased 0.22, which was similar to that of the leaf stored under the natural warehouse condition for 15 months, and the lightness, and redness of the leaf were higher than those of the control. As compared with the leaf stored under the natural condition for 2 years, the smoking quality of leaf stored under this condition was similar or somewhat better. The pH of burley tobacco changed little compared to that of flue-cured during storage in this study.

영농기 필지논에서의 인 (P) 농도와 산화환원전위 (Eh)의 변화 특성 (Variation of Phosphorus Concentration and Redox Potential in a Paddy Field Plot During Growing Season)

  • The purpose of this study is to investigate characteristics of total phosphorus (TP) and phosphate phosphorous ($PO_4$-P) concentrations in ponded water and redox potential (Eh) in paddy soil during the growing season. The TP and $PO_4$-P concentrations showed twice peak values after basal dressing and tillering fertilization. The ratio of $PO_4$-P to TP showed low values (0.07~0.18), indicating that most of phosphorus is particlulate. The $PO_4$-P concentrations significantly decreased with dissolved oxygen (DO) concentrations. The Eh showed high values (179~636 mV) under non-ponded aerobic condition, but low values (74~112 mV) under ponded anaerobic condition The TP and $PO_4$-P concentrations in ponded water increased shortly after tillering fertilization even if phosphorus was not applied. This may be due to the release of dissolved phosphorus from the bottom sediment and its associated algal and water flea blooms under anaerobic condition. Therefore, proper water management should be needed shortly after tillering fertilization.

무전해 Ni 도금에 의한 선택적 CONTACT HOLE 충전 (Selective Contact Hole Filling by electroless Ni Plating)

  • The effect of activation and electroless nickel plating conditions on contact properties was investi-gated for selective electroless nickel plating of Si wafers in order to obtain an optimum condition of con-tact hole filling. According to RCA prosess, p-type silicon (100) surface was cleaned out and activated. The effects of temperature, DMAB concentration, time, and strirring were investigated for activation of p-type Si(100) surface. The optimal activation condition was 0.2M HF, 1mM PdCl2, 2mM EDTA,$ 70^{\circ}C$, and 90sec under ultrasonic vibration. In electroless nickel plating, the effect of temperature, DMAB concentra-tion, pH, and plating time were studied. The optimal plating condition found was 0.10M NiSO4.H2O, 0.11M Citrate, pH 6.8, $60^{\circ}C$, 30minutes. The contact resistance of films was comparatively low. It took 30minutes to obtain 1$\mu\textrm{m}$ thick film with 8mM DMAB concentration. The film surface roughness was improved with decreasing temperature and decreasing pH of the plating solution. The best quality of the film was obtained at the condition of temperature $60^{\circ}C$ and pH 6.0. The micro-vickers hardness of film was about 800Hv. Plating rate of nickel on the hole pattern was slower than that of nickel on the line pattern.

간호대학생의 삶의 질에 미치는 영향에 대한 융합연구 (A Convergence Study on Factors Influencing Quality of Life of Nursing Students)

  • 본 연구의 목적은 간호대학생의 삶의 질에 미치는 영향을 파악하기 위한 경로모형 연구이다. 연구대상은 B시와 G시에 소재하고 있는 간호학과 재학중인 학생 209명을 대상으로 하였다. 수집된 자료는 SPSS 22.0과 Amos 22.0 프로그램을 이용하여 분석하였다. 연구결과 간호대학생의 삶의 질에 직접적으로 영향을 미치는 변수로는 건강상태, 스트레스였으며, 스트레스에 직접적으로 영향을 미치는 변수는 경제상태, 건강상태로 확인되었다. 즉, 경제상태가 낮을수록(${\beta}=-,067$ p=.030), 건강상태가 나쁠수록(${\beta}=-.109$, p=.003) 스트레스를 많이 받는 것으로 나타났으며, 건강상태가 좋을수록(${\beta}=.083$, p=.013) 스트레스가 적을수록(${\beta}=.-213$, p=.001) 삶의 질이 높은 것으로 나타났다. 본 연구결과를 토대로 간호대학생들의 삶의 질을 향상시키기 위해 정보적 지지와 정서적 지지를 고려한 교육 프로그램 개발이 이루어져야 하겠다.

맞춤형 주거환경수정을 이용한 노인의 건강상태 차이비교 분석: 커뮤니티케어 사업을 중심으로 (Comparative Analysis of Differences in Health Condition with Elderly Age Using the Customized Home Modification : Focusing on Community Care Projects)

  • 본 연구는 65세 이상 노인 36명을 대상으로 커뮤니티 케어 사업에서의 주거환경수정이 낙상 위험환경과 대상자의 건강상태에 미치는 영향에 대해 알아보고자 하였다. 주거환경 및 대상자의 특성에 대한 평가를 위해 HOME FAST와 WHODAS-K 2.0을 사용하였으며, 집단 사전·사후 실험연구로 진행되었다. 평가를 바탕으로 주거환경수정 서비스를 시행한 결과, 낙상 위험수준은 통계적으로 유의하게 감소되는 변화를 나타내었으며(p<.01), 건강상태의 경우에는 이동성(p<.01)과 사회참여(p<.05)영역에서 통계적으로 유의하게 향상되어 전반적 건강상태(p<.05)의 변화에도 긍정적인 변화를 보이는 것으로 확인되었다. 향후 더 많은 연구 대상자 확보하고, 포괄적인 주거환경 평가를 활용한 연구가 필요하며, 주거환경수정의 질적 서비스 제공을 위해 대상자 우선순위 선별과정과 전문인력 인프라 확충이 요구된다.

여성의 생리적인 현상에 따른 피부상태와의 상관성 관련 요인에 관한 연구 (The Research for Skin Condition Related with Physiological Phenomenon)

  • 본 연구는 경남 창원시내에 거주하는 20~40대 여성 315명을 조사대상으로 하여 단순무작위추출(simple landom sampling)하여 설문지조사 방법에 의해 자기 기록식으로 기록하였다. 본 연구는 피부상태에 따른 피부의 유형을 세안 후 30분간 아무것도 바르지 않은 상태에서 뺨부위를 검진법을 통하여 체크하여 지성, 중성, 건성, 지루성으로 구분하여 각 조건들과의 상호작용을 알아보았다. 각 요인들간의 관련성을 알아보기 위해 X2-검정을 실시하였으며, 자료처리는 PC/SAS를 이용하였다. 대상자의 일반적 특성과 피부상태에서 연령, 직장 및 결혼 여부는 통계적으로 유의성이 있었다(p<0.01). 연령이 많을수록 건성으로 진행되는 경향이 있고 직장여성과 기혼여성일 경우 건성의 비율이 높은 편이었다. 스트레스 후 피부반응과 일반적인 특성 및 생리적 현상과의 관련성에서 여드름이 나는 부위와 배변횟수 사이에는 통계적으로 유의한 차이가 있었고, 각 X2-검정의 값은 42.961(P<0.01), 7.345(P<0.05)였다. 피부트러블반응과 여드름 나는 시기와의 사이에는 유의성이 인정되었다(P<0.01).

인공수초재배섬에서 수생식물 지상부 절취주기별 수중영양염류 제거효율 (Effect of Cut-off Intervals on Nutrients Removal Efficiency in Hydrophytes at the Artificial Vegetation Island)

  • We investigated the most effective cutting interval for underwater nutrient removal through cut off the emergent part of hydrophytes at artificial vegetation island (AVR) which was installed for the purpose of water quality improvement in Lake Paldang. We divided the planting area of Phragmites japonica into three parts according to the cutting intervals. The shoot height and relative growth rate of P. japonica, nutrient contents and biomass of cut off P. japonica were measured at each cutting interval. The amount of nutrients which were removed through cut off at each cutting interval was calculated. P. japonica showed full growth, 80% and 60% of full growth before first cut off at three-months, two-months and one-month cutting interval condition respectively. Three-month cutting interval condition showed the largest biomass of cut off P. japonica and one-month cutting interval condition showed the least. However the cut off P. japonica showed the highest content of nutrients at one-month cutting interval condition and the least at three-month cutting interval condition. The amount of phosphorus and nitrogen removal at two-month cutting off condition is the largest among three cutting interval conditions indicating that cut off the emergent part of P. japonica every two months is the most effective to remove the nutrients from water at AVR in eutrophic lakes.

생리식염수 냉수 거즈와 일반 냉수 거즈의 수술 후 갈증, 구강상태, 타액 산성도에 미치는 효과 비교 (A Comparison of Effect between Wet Gauze with Cold Normal Saline and Wet Gauze with Cold Water on Postoperative Thirst, Oral Cavity Condition, and Saliva pH)

  • Purpose: This study was aimed to compare the effect between wet gauze with cold normal saline and wet gauze with cold water on thirst, oral cavity condition, and saliva pH among postoperative patients. Methods: This study was a quasi-experimental study with 56 participants. The experimental group received wet gauze with cold normal saline, and control group received wet gauze with cold water for three times at 15 minute intervals. Data were collected using visual analog scale for thirst, oral assessment guide for oral cavity condition and acidity tape for saliva pH. Measurement were made before applying the gauze and at 15min, 30min, and 45min after applying gauze. Data were analyzed using repeated measured ANOVA. Results: Thirst, oral cavity condition, and saliva pH improved in both groups with increasing number of gauze application. Thirst for the experimental group improved more than for the control group (F=4.29, p=.009), oral cavity condition except saliva, and saliva pH were not significantly different between the groups. Conclusion: This study results indicated that nurses can apply wet gauze with cold normal saline to reduce thirst and saliva acidity, and to improve the oral cavity condition for postoperative patients.

원주지역 초등학교 아동의 영양지식과 식생활 태도에 관한 연구 -급식학교와 비급식학교 아동의 비교- (A Comparative Study on Nutritional Knowledges and Dietary Behaviors of Children in Elementary School by School Lunch Program in Won-Ju Province)

  • This study was to investigate the children's understanding level of nutritional knowledge and the degree of knowledge application into the actual living not only in the with-lunch school but also in the without lunch school. Having expected their obtaining of nutritional knowledge and practical applicating, we compared the with-lunch school children's understanding level of nutritional knowledge, dietary attitude, and completeness of diet life with those of without-lunch school children. In addition to this, we surveyed healthiness, Physical condition, and BMI (Body Mass Index) and compared these factors. The results are as follows. 1. Almost all of the children were standard in physical condition, however they recognized themselves fatty than normal. Mealtime consumption(p<0.05), BMI(p<0.05), and diet attitude points(p<0.001) showed significant difference in the children who regarded the themselves healthy. 2. High correlation was observed between parent's physical shape and mealtime consumption (p<0.05), quantity of eating food (p<0.001), children's BMI(p<0.001) and diet attitude(p<0.05) respectively. 3. The points of itemised nutritional knowledge test was different in accordance with the children's year grade (p<0.05), children's understanding level of health and physical condition(p<0.05), parent's physical shape (p<0.05), and mother's education level(p<0.05) respectively. The points of nutritional knowledge test in both schools showed no difference except the item of vitamin.
