Rheological Properties of Concentrated Dandelion Leaf Extracts by Hot Water or Ethanol

  • Basic rheological data of dandelion leaf concentrates were determined to predict processing aptitude and usefulness of dandelion leaf concentrates as functional food materials. Hot water and 70% ethanol extracts of dandelion leaves were concentrated at 5, 20, and 50 Brix, and their static and dynamic viscosities, and Arrhenius plots were investigated. Most concentrated dandelion leaves extracted with hot water and 70% ethanol showed flow behaviors close to Newtonian fluid based on power law model evaluation. Apparent viscosity of concentrated dandelion leaves extracted with hot water and 70% ethanol decreased with increasing temperature. Yield stresses of concentrated dandelion leaves extracted with hot water and 70% ethanol by Herschel-Bulkley model application were 0.020-0.641 and 0.017-0.079 Pa, respectively. Activation energies of concentrated dandelion leaves extracted with hot water and 70% ethanol were $2.102-32.669{\times}10^3$ and $1.657-5.382{\times}10^3\;J/mol{\cdot}kg$ with increasing concentration, respectively. Loss modulus (G") predominated over storage modulus (G') at all applied frequencies, showing typical flow behavior of low molecular solution. G' and G" of concentrated dandelion leaves extracted with hot water slowly increased with increasing frequency compared to those of concentrated dandelion leaves extracted with 70% ethanol.

수수 식물체 추출물의 제초활성 및 작물 선택성 (Herbicidal activity and crop injury of aqueous extracts of sorghum leaves)

  • Herbicidal effects and crop selectivity of aqueous leaf extracts of sorghum (Sorghum bicolor L.) were evaluated against several weed species for developing sustainable weed management in organic farming. Aqueous sorghum leaf extracts were highly phytotoxic to different weed species. No broadleaf weeds were germinated in the concentration of 5 fold or higher concentrated sorghum leaf extracts and 90% of seed germination was inhibited within that range in grass species. Sorghum leaf extracts strongly inhibited the growth of different weeds by pre-emergence and foliar applications in greenhouse condition. Foliar application of sorghum leaf extracts had a higher inhibitory effect than the pre-emergence application. Broadleaf weed species were more susceptible than grasses to the application of sorghum leaf extract in foliar applications than grasses. Galium spurium, Erigeron candensis, and Rumex japonicus were completely killed at the highest concentrated sorghum leaf extract both in pre-emergence and foliar application. Most broadleaf weed species were inhibited more than 80% at pre-emergence application at 50 fold concentrated sorghum leaf extract. G. spurium, E. candensis, R. japonicus, Eclipta alba, Plantago asiatica and Portulaca oleraeea were most susceptible to sorghum leaf extract in foliar application. Growth of most broad leaf weed species was suppressed by greater than 90% at 50 fold concentrated sorghum leaf extract. Most crop species were tolerant to sorghum leaf extract but shoot growth was slightly reduced by the application of 40~50 fold concentrated extracts, Sorghum leaf extract may used to control weeds in organic fanning without affecting the growth of crop.

천마추출액 농축 비율을 달리하여 제조한 천마음료의 품질특성 (Quality Characteristics of Chun Ma (Gastrodiae rhizoma) Beverage Prepared Using Concentrated Extracts)

  • 천마음료를 제조시 첨가물로 천마 추출액을 달리하여 제조한 pH, 당도, 색도, 총페놀 함량, DPPH radical 소거 활성능, 유리당, 유기산 및 유리아미노산 함량, 무기질 함량, 관능 검사를 분석하여 품질 특성을 검토하였다. 천마음료 제조 시 GCE 5($5^{\circ}$Brix 천마 농축 추출액), GCE 10($10^{\circ}$Brix 천마 농축 추출액), GCE 15($15^{\circ}$Brix 천마 농축 추출액), GCE 20($20^{\circ}$Brix 천마 농축 추출액)으로 나누어서 제조하였다. pH는 GCE 20이 가장 높은 값(4.68)을 GCE 5가 가장 낮은 값(4.37)을 나타내었으며, 당도는 GCE 20가 $19.6^{\circ}$Brix으로 가장 높은 값을 나타내었다. b값과 총페놀함량은 추출농축액의 첨가농도가 높아짐에 따라 조금씩 증가하는 경향을 보이고 있으며, 총페놀 함량은 GCE 20이 232.23 mg%으로 가장 높은 값을 나타내었다. DPPH radical 소거 활성능에서는 모든 처리구에서 75.07~76.00%로 큰 차이가 나타나지 않았다. 유리당 함량은 fructose, glucose, sucrose, maltose는 GCE 20이 각각 340.34 mg%, 199.55 mg%, 93.96 mg%, 73.84 mg%로 가장 높은 값을 나타내었다. 유기산 중 GCE 15의 경우 tartaric acid, malic acid, lactic acid, succinic acid가 각각 89.89 mg%, 88.32 mg%, 339.52 mg%, 324.96 mg%으로 가장 높은 값을 나타냈다. 유리아미노산과 무기질 함량은 첨가되는 추출농축액의 농도가 높아짐에 따라 유리아미노산과 무기질의 함량은 높아지는 것으로 나타났다. 유리 아미노산은 Glu>Gly>Ser>Arg>Hylys의 순으로 많이 함유하고 있는 것으로 나타났다. 천마 음료 중 가장 많이 함유하고 있는 Glu의 경우 GCE 20이 249.15 ${\mu}g/100g$로 가장 높았고, GCE 5가 61.5 ${\mu}g/100g$로 가장 낮은 값을 나타냈다. 무기질 함량은 모든 처리구에서 K>Na>Mg>Ca 순으로 많이 함유되어 있는 것으로 나타났으며, 관능 평가 결과 GCE 5는 색, 향, 전체적인 기호도에서는 가장 높은 값을 나타냈다. 이상의 결과 천마를 소재로 한 음료 제조시 천마 고유의 맛과 향에 대한 거부감이 심한 것을 고려하여 상업적인 생산이 되어야 할 것으로 사료된다.

동충하초(冬蟲夏草)의 항돌연변이(抗突然變異) 활성(活性)에 관한 연구(硏究) (A Study on Antimutagenic Activity of the Extracts from Paecilomyces japonica)

  • The fruiting bodies of Paecilomyces japonica been used for an anticancer and immuno-stimulating agent as an oriental medicine, Antimutagenecity test with SOS chromotest and antioxidant test with NBT method were carried out using the concentrated culture broth, the extract of mycelia, and that of fruiting bodies. Among the sample extracts tested, the extract of fruiting bodies was most effective to antimutagenecity against the mutagens tested such as MNNG, ethidium bromide(EtBr), 2-aminofluorene(AF) and nitrofluorene(NF). Antimutagenic activity of all the extracts were very effective against mutagen, MNNG. When the extracts were added to certain concentration, antimutagenic activity was enhanced against mutagen, MNNG and NF. Antioxidant activity of the extract from fruiting bodies was highest. However, its activity was very low, compared to ascorbic acid.

Analysis of Flavonoids in Concentrated Pomegranate Extracts by HPLC with Diode Array Detection

  • Three flavonoid compounds - quercetin, luteolin, and kaempferol - were analyzed from two commercially available concentrated pomegranate extracts produced in Turkey and Italy, respectively. The samples were freeze-dried and hydrolyzed by 0.4 M hydrochloric acid in 50% ethanol at $80^{\circ}C$. HPLC (high-performance liquid chromatography) with DAD (diode array detection) at a wavelength of 260 nm was used for the detection of the three flavonoids. The detection limits of the three compounds were in hundreds of picograms and the signal-to-noise ratio ranged from 4 to 5: quercetin: >308 pg, s/n=4.0; luteolin: >262 pg, s/n=4.5; kaempferol: >688 pg, s/n=5.0. Quercetin, but not luteolin and kaempferol, was detected in the both pomegranate extracts. The concentrations of quercetin were $49.7\;{\mu}g/g$ and $27.7\;{\mu}g/g$ in the two pomegranate extracts made in Turkey and Italy, respectively.

  • Antioxidant effect of several antioxidative components on the high purity $\alpha$-linolenic acid(HALA; ALA, 78.1%) ethyl ester concentrated from perilla oil were investigated by measuring weight-gains and peroxide value(POV) during storage at 5$0^{\circ}C$, 23$^{\circ}C$ and 4$^{\circ}C$. Amounts of antioxidant components were 0.2g/kg HALA ethyl ester for sesamin, sesangolin and butylated hydroxytolune(BHT), and 0.1g/kg for sesamol and 100g/kg for ether extracts from perilla seed. The oxidative stability of HALA ethyl ester was particularly increased by adding sesamol, ether extracts and BHT, but sesamin and sesangolin scarcely showed an antioxidant effect. POV on the HALA ethyl ester added sesamol and ether extract was less than 15.0meq/kg by 9 weeks of storage at 23$^{\circ}C$. However, in the case of low temperature storage at 4$^{\circ}C$, all the samples estimated showed less than 7.0meq/kg in POV by 5 months. Consequently, sesamol and ether extracts were recognized as available antioxidant components on the HALA ethyl ester from perilla oil.

Bacillus sp. CS-17의 색소 생성조건 및 색소 농축액의 항균특성 (Conditions for the Pigment Production by Bacillus sp. CS-17 and Antibacterial Activity of Pigment Concentrated Extracts)

  • 전통 대두발효식품으로부터 색소 생성능과 protease 활성이 가장 강한 균을 선별하여 Bacillus sp. CS-l7로 동정하였다. CS-17 균의 균체증식은 배양 24시간, protease 활성은 배양 48시간, 그리고 색소 생성은 배양 72시간 후 최대에 달하였다. Bacillus sp. CS-17은 대두분말을 1.0% 첨가한 색소 생성용 기본배지에서 가장 높은 색소 생성능을 가지는 것으로 나타났다. 배양조건이 Bacillus sp. CS-17의 색소 생성에 미치는 효과를 조사한 결과, pH 8.5에서 $37^{\circ}C$, 72시간 배양하였을 때 최적의 색소 생성을 보였다. NaCl의 첨가는 색소 생성능을 억제하는 것으로 나타났다. 이상의 결과로 Bacillus sp. CS-17의 색소 생성 최적조건은 $37^{\circ}C$, pH 8.5에서 72시간동안 배양하였을 때로 추정되었다. 그람 양성균 5주 및 그람 음성균 6주에 대하여 색소 농축액의 항균활성을 paper disc법으로 조사한 결과, B. subtilis, P. aeruginosa, S. typhimurium, E. aerogenes, B. cereus, A. hydrophila의 성장에 대한 항균효과가 인정되었으나 그 활성은 전반적으로 미약하였다.

메밀 싹 추출물 에멀젼의 제조 및 에멀젼의 열 안정 특성 (Production and Heat-Stable Characteristics of Emulsion Made from Buckwheat Sprout Extracts)

  • 메밀 성분 중 루틴의 추출 효율을 높이기 위해 메밀을 발아한 후 루틴 및 polyphenol 추출 효율이 높은 추출방법을 선택하였다. 메밀의 발아는 흙 재배가 효율적이었으며, 8일 후에는 polyphenol 함량이 10.66 mg/g으로 메밀 종자에 함유되어 있던 polyphenol 함량의 10배 이상 높은 양을 얻을 수 있었다. 발아 메밀에서 추출된 루틴을 포함한 polyphenol 성분은 에멀젼을 제조하여 열에 대한 안정성을 높이는 데 효율이 있는지를 확인하였다. 에멀젼은 homo mixer만을 통과시키는 것보다 유압식 균질기를 통과시켰을 때 포접 효율이 높을 뿐만 아니라 열처리 온도에 대하여 열 안정성이 우수한 것을 알 수 있었다.

양파추출농축액(오니웰TM)의 저장 중 휘발성 향기성분 변화 (Changes of Volatile Compounds in Concentrated Onion Extracts (ONIWELLTM) during Storage)

  • 양파추출농축액(오니웰$^{TM}$)의 저장 유통과정 중 품질유지를 위한 척도로서 관능적요소 중 가장 중요한 휘발성 향기성분을 저장기간($30^{\circ}C$, 150일) 동안 분석하였다. 총 23종의 휘발성화합물이 동정되었는데, 이는 함황 화합물류 9종, 카르보닐 화합물류 5종, 퓨란류 4종, 방향족 화합물류 2종 및 기타 화합물류 3종이었다. 함황 화합물류는 전체함량의 80.3~62.6%로 중요한 향기성분임을 알 수 있었다. 이 중 dimethyl trisulfide가 50.1~42.1% 범위로 함황 화합물류 중에서 가장 높은 함량을 차지하였다. 총 함황 화합물류의 함량은 저장기간과 함께 증가하였는데, 특히 신선한 양파향의 dimethyl disulfide와 마늘향의 methyl propyl disulfide 함량이 유의적으로 증가하였다(p<0.05). 가열반응에 의해 생성된 4종의 퓨란류(furfural, 2-acetylfuran, 5-methyl-2-furfural, furfurylalcohol)가 동정되었는데, 함량면에서 함황 화합물 다음으로 많았다(14.2~12.9%). 이 중 furfural이 전체 퓨란류의 61.9~49.4%를 차지하였고, 저장기간과 함께 유의적으로 증가하였다. 카르보닐 화합물에서는 알데히드류가 4종, 케톤 1종으로 총 5종이 검출되었다. 함량면에서는 6.2~4.8% 범위로 퓨란류 다음으로 많았으며, 가열에 의한 지방산화 분해에 의해 생성된 화합물로 동정되었다. 따라서 이들 가열반응에 의해 유도된 퓨란류나 알데히드류가 양파 특유의 함황 화합물과 함께 양파추출농축액(오니웰$^{TM}$)의 풍미에 기여할 것으로 추정되었다.

  • The regulation of plasma lipid level, particularly LDL cholesterol, represents the focus of current therapy for atherosclerosis. And $Lp-PLA_2$ is able to hydrolyse oxidized phosphatidylcholine within LDL into lyso-PC and oxidized fatty acids. $Lp-PLA_2$ is a potential biomarker of coronary heart disease and plays an important proinflammatory role in the progression of atherosclerosis. We investigated the inhibitory effects of methanol extracts of 224 natural plants on $Lp-PLA_2$ activity. Seven kinds of methanol extracts of tested plants showed above 50% inhibitory effect with the concentration of $100\;{\mu}g/ml$. The concentrated aqueous suspensions of each methanol extract were partitioned with n-hexane, $CHCl_3$, and EtOAc. Among them, EtOAc extracts of Astilbe chinensis var. davidii (root) and Pourthiaea villosa var. brunnea (leaf) significantly inhibited $Lp-PLA_2$ activity at the same concentration.
