Compound 48/80로 유발된 Allergy에 대한 생혈윤부음(生血潤膚飮)의 효과 (Inhibitory Effect of Saingheylyunbooem on Compound 48/80 Stimulated Allergic Reaction)

  • The present study was conducted to investigate the anti-allergic activity of Saingheylyunbooem(SHU)). We investigated the anti-allergic effects of SHU in RBL-2H3 basophilic leukemia cells by compound 48/80, a mast cell degranulator in mice. SHU inhibited ${\beta}$-hexosaminidase and histamine release from compound 48/80 stimulated RBL-2H3 cells. The in vitro anti-inflammatory activities of SHU in LPS-stimulated RAW 264.7 cells were investigated. SHU inhibited NO production effectively dowregulated the expression of iNOS mRNA and iNOS protein expression in LPS-stimulated RAW 264.7 cells. These result provide evidences that SHU may be beneficial in the treatment of allergic inflammtory disease.

갈근해기탕(葛根解肌湯)의 항 Allergy 및 항염증 효과 (Inhibitory Effect of Galgeunhaegi-Tang on Compound48/80 Stimulated Allergic Reaction)

  • The present study was conducted to investigate the anti-allergic activity of Galgeunhaegi-Tang(GHT). We investigated the anti-allergic effects of GHT in RBL-2H3 basophilic leukemia cells by compound48/80, a mast cell degranulator and compound 48/80 induced anaphylactic shock in mice. GHT significantly inhibited ${\beta}$-hexosaminidase and histamine release from compound 48/80 stimulated RBL-2H3 cells. In addition, GHT effectively inhibited anaphylactic shock in mice by 40% at a dose 100 mg/mouse versus PBS treated control after the l.p injection(8 mg/kg) of compound 48/80. The in vitro anti-inflammatory activities of GHT in LPS-stimulated RAW 264.7 cells were investigated. GHT inhibited NO production in LPS-stimulated RAW 264.7 cells and effectively dowregulated the expression of iNOS mRNA and iNOS protein expression in LPS-stimulated RAW 264.7 cells. These result provide evidences that GHT may be beneficial in the treatment of allergic inflammtory disease.

궁귀향소산(芎歸香蘇散)의 항Allergy 및 항염증 효과 (Inhibitory Effect of Allergic Reaction by Gunggwihyangso-San)

  • The present study was conducted to investigate the anti-allergic activity of Gunggwihyangso-San(GHS). We investigated the anti-allergic effects of GHS in RBL-2H3 basophilic leukemia cells by compound 48/80, a mast cell degranulator and compound 48/80 induced anaphylactic shock in mice. Gunggwihyangso-San significantly inhibited ${\beta}$-hexosaminidase and histamine release from compound 48/80 stimulated RBL-2H3 cells. In addition, GHS effectively inhibited anaphylactic shock in mice by 50% at a dose 80 mg/mouse versus PBS treated control after the I.p injection(8 mg/kg) of compound 48/80. 한다열소탕(寒多熱少湯)의 항Allergy 및 항염증 효과 (Anti-Allergic Effect of Handayeolso-Tang)

  • Handayeolso-Tang(HDT) has been used as traditional medicine for the treatment of Taeumin TaeYang-Hanguel. The present study was conducted to investigate the anti-allergic activity of Handayeolso-Tang(HDT). We investigated the anti-allergic effects of HDT in RBL-2H3 basophilic leukemia cells by compound48/80, a mast cell degranulator and compound 48/80 induced anaphylactic shock in mice. HDT significantly inhibited ${\beta}-hexosaminidase$ and histamine release from compound 48/80 stimulated RBL-2H3 cells. In addition, HDT effectively inhibited anaphylactic shock in mice by 45% at a dose 120 mg/mouse versus PBS treated control after the I.p injection(8 mg/kg) of compound 48/80. The in vitro anti-inflammatory activities of HDT in LPS-stimulated RAW 264.7 cells were investigated. HDT inhibited NO production in LPS-stimulated RAW 264.7 cells and effectively dowregulated the expression of iNOS mRNA and iNOS protein expression in LPS -stimulated RAW 264.7 cells. These result provide evidences that HDT may be beneficial in the treatment of allergic inflammtory disease.

고장초의 항알레르기 효과 (Anti-allergic Effect of Zizania latifolia Turcz Extracts)

  • 고장초 메탄올 추출물의 항알레르기 활성을 조사하기 위해, 비만세포주인 RBL-2H3 세포를 이용하였으며, 고장초 추출물을 처리한 후 IgE-항체, A23187 및 PMA를 이용하여 세포를 활성화시켜 분비되는 $\beta$-hexosaminidase, TNF-$\alpha$의 양을 측정하였다. 그 결과 고장초 추출물이 활성화된 비만세포에서 분비되는 $\beta$-hexosaminidase, TNF-$\alpha$를 농도 의존적으로 억제하는 것을 확인할 수 있었다. 또한, 비만세포에 탈과립이 발생할 경우 나타나는 형태학적 변화에 대한 시료의 영향을 검색하기 위해 고장초 추출물을 처리한 세포에 compound 48/80을 처리하였으며, 그 결과 고장초 추출물을 처리한 경우 대조군에 비해 탈과립이 현저히 억제되는 것을 관찰하였다. 따라서 고장초 추출물은 다양한 자극으로 인한 비만세포의 탈과립 저해를 통해 알레르기 반응을 억제시키는 것으로 판단되며, 본 연구결과는 고장초의 항알레르기 기능성 소재로의 이용 가능성을 시사해준다.

자울약침액(紫菀藥鍼液)의 폐유(肺兪) 처치(處置)가 Type I Hypersensitivity에 미치는 영향 (Effect of Radix Asteris Herbal Acupuncture at $BL_{13}$ on the Type I Hypersensitivity)

  • Objectives : We studied the effects of Radix Asteris herbal acupuncture solution (RAHAS) on the type I hypersensitivity. Methods : In vivo, we measured compound 48/80 induced active systemic anaphylactic shock, anti-DNP IgE induced passive cutaneous anaphylaxis (PCA) and acetic acid induced microvascular permeability using ICR mice. In vitro, we showed effects on cytotoxicity and ${\beta}$-hexosaminidase release from RBL-2H3 cells. Results : In vivo, RAHAS pretreatments at $BL_{13}$ and optional points inhibited active systemic anaphylactic shock induced by compound 48/80 and microvascular permeability increased by acetic acid. PCA was only inhibited by RAHAS pretreatments at $BL_{13}$. In vitro, RAHAS treatments inhibited ${\beta}$-hexosaminidase release. Conclusion : These results suggest that RAHAS may be beneficial in the prevention of type I hypersensitive inflammatory response.

황금약침(黃芩藥鍼)이 Type 1 Hypersensitivity에 미치는 영향 (Effect of scutellariae radix pharmacopuncture on the type 1 hypersensitivity)

  • Objectives : We studied the effects of Scutellariae Radix pharmacopuncture solution (SRHAS) on the type 1 hypersensitivity. Methods : In vivo, we measured compound 48/80-induced active systemic anaphylactic shock using ICR mice and anti-DNP IgE-induced passive cutaneous anaphylaxis (PCA) using Sprague Dawley rats. In vitro, we showed effects on cytotoxicity and ${\beta}-hexosaminidase$ release from RBL-2H3 cells. Results : In vivo, SRHAS pretreatments (100% or 50%) at BL13 inhibited active systemic anaphylactic shock induced by compound 48/80. PCA was only inhibited by pretreatments of SRHAS at optional points. In vitro, $0.1{\sim}2%$ SRHAS treatments did not affect cell viability while ${\beta}$-hexosaminidase release was significantly inhibited. Conclusions : These results suggest that SRHAS may be beneficial in the inhibition of type I hypersensitive inflammatory response.

장수풍뎅이 유충 열수 추출물에 의한 항알레르기와 항염증 효과 (The Anti-inflammatory and Antiallergic Effects of Allomyrina dichotoma Larva Hot-water Extract)

  • 본 연구에서는 장수풍뎅이 유충 추출물을 이용하여 알레르기 및 염증 반응에 미치는 효과를 확인하였다. 장수풍뎅이 유충 추출물의 항알레르기와 항염증 효능은 Compound 48/80에 의해 활성화된 비만세포(RBL-2H3)와 lipopolysaccharide(LPS)로 활성화된 대식세포(Raw 264.7)로부터 분비 또는 발현되는 ${\beta}-hexosaminidase$, $TNF-{\alpha}$, IL-4, IL-6, COX-2, nitric oxide, 및 iNOS를 측정하여 관찰하였다. 비만세포에 장수풍뎅이 유충 추출물과 Compound 48/80을 함께 처리한 경우 비만세포의 탈과립에 의해서 분비되는 ${\beta}-hexosaminidase$의 양이 현저히 감소하였으며 $TNF-{\alpha}$, IL-4, COX-2의 발현 또한 효과적으로 감소시킴을 확인할 수 있었다. 또한 LPS에 의해 활성화된 대식세포(Raw 264.7)는 장수풍뎅이 유충 추출물 처리에 의해 염증반응인자인 IL-6와 nitric oxide의 분비량이 현저히 감소함을 확인할 수 있었다. 이상의 결과는 장수풍뎅이 유충 추출물이 알레르기와 염증반응의 원인 물질인 ${\beta}-hexosaminidase$ 억제 효과뿐만 아니라, 염증 cytokine의 발현을 저해하는 것으로 보아 알레르기와 염증 질환의 예방과 치료에 효과적으로 이용될 수 있음을 확인할 수 있었다.

  • Objectives : We studied anti-allergic effects of Cinnamomi Ramulus(CR) herbal acupuncture and Cinnamomi Ramulus extract. Methods : In vivo, animals were herbal-acupunctured with CR at both ST36s three times for 5 days. Then, we induced active systemic anaphylatic shock using compound 48/80 in ICR mice, and passive cutaneous anaphylaxis using anti-DNP IgE in Sprague Dawley rat. In vitro, we measured cell viability, ${\beta}$ -hexosaminidase release and the expressions of IL-4, TNF-${\alpha}$ and COX-2 mRNA in RBL-2H3 cells after treatment of various concentrations of CR extract. Results : In vivo, CR herbal acupuncture pretreatments at both ST36s inhibited compound 48/80-induced active systemic anaphylatic shock. Passive cutaneous anaphylaxis was inhibited by CR herbal acupuncture pretreatments at both ST36s and optional points. In vitro, CR extract treatments did not affect on cell viability and inhibited ${\beta}$-hexosaminidase release. CR extract treatments also decreased the expressions of IL-4, TNF-${\alpha}$ and COX-2 mRNA in RBL-2H3 cells. Conclusions : These results suggest that CR herbal acupuncture and CR extract should be beneficial in the inhibition of allergic inflammatory response.
