Growth Inhibition of Microcystis aeruginosa by a Glycolipid-Type Compound from Bacillus subtilis C1

  • We attempted to identify the compound responsible for the growth inhibition of Microcystis aeruginosa occurring when a culture broth of Bacillus subtilis C1 was added to the medium. The active compound was purified from B. subtilis C1 culture broth by adsorption chromatography and HPLC, and was identified as a type of glycolipid based on $^1H$ NMR and MS analyses. The purified active compound completely inhibited the growth of M. aeruginosa at a concentration of 10 ${\mu}g/ml$. This is the first report of a glycolipid produced by a Bacillus strain that has potential as an agent for the selective control of bloom-forming M. aeruginosa.

  • We designed the compound CVT (Continuously Variable Transmissions) by combining power circulation mode CVT and power split mode CVT, which have been proposed for connecting 2K-H II differential gear to the V- belt type CVU (Continuously Variable Unit), as an input coupled type. With the designed compound CVT, we carried out theoretical analysis and performance experiments. We proved that the compound CVT had a better performance than either of the power circulation mode or power split mode.

비만세포 매개 즉시형 알레르기 반응에 대한 연명초의 억제 효과 (Inhibitory Effect of Isodon japonicus Hara on Mast Cell-Mediated Immediate-Type Allergic Reactions)

  • The effect of aqueous extract of Isodon japonicus Hara (Labiatae) (IJAE) on mast cell-mediated immediate-type allergic reactions was investigated. IJAE inhibited compound 48/80-induced systemic anaphylaxis and immunoglobulin E (IgE)-mediated local anaphylaxis. When IJAE was pretreated at the same concentration with systemic anaphylaxis, serum histamine levels were reduced in a dose-dependent manner. IJAE dose-dependently inhibited histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80. The level of cAMP in human mast cell line (HMC-1) cells, when IJAE was added, significantly was increased, compared with that of normal control. These results indicate that IJAE will beneficial in the treatment of immediate-type allergic reaction.

  • 최근 국내에서 도심지 홍수로 인한 피해 발생이 증가하면서 차집된 다량의 강우를 일시에 배출할 수 있는 와류 유입구에 대한 관심이 증가하고 있다. 이 중 접선식 유입구는 구조가 단순하고 크기가 작아 적용성이 높으나 설계 유량보다 작은 유량에 대해서 와류가 형성되지 못하는 단점이 있다. 본 연구에서는 이를 보완하여 새롭게 제안된 2가지 종류의 복단면형 접선식 와류 유입구의 수리 특성을 3차원 수치모의를 통해서 분석하였다. 기존에 제시된 한계수심 이론과 자유 배수 조건 이론을 이용하여 복단면 형상의 접선식 와류 유입구의 특성을 흐름 상태, 수면형, 수심-유량 관계, 공기 기둥 면적비 측면에서 분석하였다. 종합적인 수리 특성 분석 결과 완경사형 복단면 접선식 유입구가 최적안으로 판단되나 설계 유량이 작은 경우에는 급경사형 복단면도 가용한 것으로 나타났다.

가미보중익기탕이 알레르기반응에 미치는 영향에 대한 실험적 연구 (Experimental Study on the Anti-allergic Effects of Gamibojungikgi-tang)

  • This experimental research has been done to study the effects of Gmibojungikgi-tang(GBJIKT) on the anti-allergic reaction. We found the several important results from the research which has been performed by two experiments toward immediately type and delayed type in order to study the effects of GBJIKT on hypersensitivity response to mice. The results obtained from our research are as following: The survival rate of one group to which we injected only the compound 48/80 is almost 0% according to its density and timing test. In the other hand, the survival rates of the other group to which we injected both of the compound 48/80 and GBJIKT are 10%, 10%, 20%, 30%, 20%, and 40% according to 0.025, 0.05, 0.1, 0.25 0.5 and 1(mg/g) of compound 48/80. Time dependency test also shows the 0% survival rates in 5 and 10 minutes. GBJIKT revealed the significantly inhibitory effect on Compound 48/80 induced Mast cell degranulation. GBJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to picry1 chloride GBJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to sheep red blood cell. Our research provides the important evidence that GBJIKT is benificial to the prevention and treatment of allergy related diseases.

십전대보탕이 알레르기반응에 미치는 영향 (The Experimental Study on the Anti-allergic Effects of Sipjeondaebo-tang)

  • This experimental research has been done to study the effects of Sipjeondaebo-tang(SOT) on the anti-allergic reaction. 麻黃辛芎散의 卽時型 알레르기 反應 抑制 效果에 關한 硏究 (Studies on Inhibitory Effect of Immediate-Type Allergic Reaction of Mahwang-Shin-Gung-San)

  • Mahwang-Shin-Gung-San has been found inhibiting the mast cell-mediated anaphylactic reaction. This report describes an inhibitory effect of Mahwang-Shin-Gung-San (MSGS) on the immediate-type cutaneous allergic reactions. MSGS has concentration -dependently inhibited the ear swelling response induced by compound 48/80 in mouse by intradermal injection. The mast cells in mouse ear tissue undergone ear-swelling response by compound 48/80 were stained by alcian blue/nuclear fast red. MSGS significantly inhibited the compound 48/80-induced degranulation from mast cells in ear tissue. MSGS concentration-dependently inhibited the histamine release from the rat peritoneal mast cells (RPMC) by compound 48/80. We also studied the effect of MSGS on mast cell-dependent passive cutaneous anaphylaxis (PCA) activated by dinitrophenyl IgE antibody. MSGS showed potent inhibition of PCA by oral administration. These results indicate that MSGS inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

辛夷花에 의한 卽時型 알레르기의 反應 抑制 效果 (Inhibitory Effect of Immediate-Type Allergic Reaction by Magnoliac flos)

  • The mast cell plays a pivotal role in initiating allergic response by secreting intracytoplasmic granular mediators such as histamine. Magnoliae flos has been used for treatment of allergic disease in Korea. However, its effect in experimental models remains unknown. The present report describes an inhibitory effect of Magnoliae flos on mast cell-mediated immediate-type allergic reactions. Topical application of compound 48/80 can induce an ear swelling response in normal($WBB6F_1-+/+$) mice but not in the congenic mast cell-deficient $WBB6F_1-W/W^v$ mice. Magnoliae flos inhibited concentration-dependently mast cell-dependent ear swelling response induced by compound 48/80 by topical application. Magnoliae flos inhibited concentration-dependently passive cutaneous anaphylaxis induced by anti-dinitrophenyl (DNP) IgE in rats by topical application. Magnoliae flos also inhibited concentration-dependently the histamine release from the rat peritoneal mast cells by compound 48/80 or anti-DNP IgE. Moreover, Magnoliae flos had a significant inhibitory effect on compound 48/80-induced systemic anaphylactic reaction. These results indicate that Magnoliae flos inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

보중익기탕이 알레르기반응에 미치는 영향에 대한 실험적연구 (The Experimental Study on the Anti-allergic Effects of Bojungikgi-tang)

  • 보중익기탕이 즉시형 및 지연형 알레르기에 대한 면역 반응에 미치는 영향을 알아보고자 항 알레르기에 대한 실험을 즉시형과 지연형으로 구분하여 시행하여 다음과 같은 결론을 얻었다 Compound 48/80 유발 아나필락시스 반응에 미치는 영향은 농도별 실험에서 보중익기탕 투여군은 생존율이 비순차적이었으나 생존율이 증가하였다. 또한 시간의존적 실험에서 보중익기탕 투여군은 다소의 생존율 변화를 보였으며, Compound 48/80 유발 비만세포 탈 과립 율의 측정을 통하여 대조군에 비해 보중익기탕 투여군은 현저하게 유의한 (p<0.005) 감소효과가 인정되었다. Picryl chloride에 의한 접촉성 피부염증 반응에 미치는 효과에 관한 실험과 면양적혈구에 의한 지연형 족부종 반응에 미치는 효과에 관한 실험에서 대조군에 비하여 보중익기탕 투여군은 유의한 억제효과가 인정되었다. 이러한 실험결과에서 보중익기탕이 즉시형 및 지연형 알레르기반응에 있어 모두 효과를 발휘하는 것으로 나타났으며 보중익기탕의 항 알레르기 효과를 질병의 치료에 응용할 수 있을 것으로 생각된다. 또한 향후 지속적인 실험과 연구를 통해 임상적 치료와 활용에 발전이 있을 것이라 사료된다.

Phenolic and Furan Type Compounds Isolated from Gastrodia elata and their Anti-Platelet Effects

  • Nine phenolic ($1\~9$) and two furan type (10, 11) compounds, were isolated from the methanolic extract of the tuber of Gastrodia elata Blume (Orchidaceae) in the course of continuing search for platelet anit-aggregating plant components. Compound 1 was identified as 4,4'-dihy-droxybenzyl sulfone, a novel compound for the best of our knowledge. Compound 10, 5-hydroxymethyl-2-furancarboxaldehyde, was isolated for the first time from this plant. Compound 1 ($IC_{50};\;83{\mu}M$) was about four times more inhibitory to U46619 induced aggregation than ASA ($IC_{50};\340{\mu}M$). Compound 9, 4,4'-dihydroxy-dibenzylether, ($IC_{50};\;5{\mu}M$, $3{\mu}M\;and\;33{\mu}M$, respectively) was $10\~}80$ fold more potent than ASA ($IC_{50};\;420\;{\mu}M,\;53\;{\mu}M\;and\;340\;{\mu}M$ respectively) to collagen, epinephrine and U46619 induced aggregation, although it is less active than ASA to AA induced aggregation.
