Inhibitory Effect of Kochujang Extracts on Chemically Induced Mutagenesis

  • Antimutagenic effects of 5 kinds of kochujang(Korean red pepper soybean paste) samples compared with doenjang(Korean soy paste) were studied using the Ames test with Salmonella typhimurium TA100 and the SOS chromotest, with E. Coli PQ37. Th eantimutatenic effects of methanol extracts from red pepper powder and meju(fermented soybean) powder, the major ingredients of the kochujang,were also evaluated for the mutagenicity of aflatoxin B1 (AFB1) in the Ames assay. The methanol extracts from the kochujang samples showed lower antimutagenicities than those of doenjang against AFB1 and N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in the Ames assay. Traditional kochujang I and II exhibited strong antimutagenic activity against AFB1 and MNNG. The traditional kochujang samples against MNNG were aslo observed in the SOS chromotest system with the same fashions as shown in the Ames mutagenicity test. The methanol extracts from meju powder had the strongest inhibitory effects on mutabenicity induced by AFB1, however, those form red pepper powder showed lower inhibition rate than kochujang. These results suggest that traditional kochujang exhibit higher antimutagenic acitivity than the commercial variety, and that meju powder seems to be one of the major antimutagenic components in kochujang.

Prevention of Swelling and Quality Improvement of Sunchang Traditional Kochujang by Natural Additives (천연첨가물을 이용한 전통고추장의 유통중 팽창억제 및 품질개선)

  • To suppress the gas forming caused by some yeast in the pack of traditional kochujang prepared at Sunchang area, which is the most severe problem during distribution of commercial products, mustard or horseradish powder as natural preservatives was mixed to the ingredients of kochujang and left for fermentation in clay pot as commercial scale for 180 day. The composition changes including quality and gas formaing in the kochujang containing those additives were monitored physicochemically and organoleptically. The fermented kochujang containing those additives were monitored physicohemically and organoleptically. The fermented kochujang containing 0.6% of horseradish showed lower organic acid content than that of the control but amino type nitrogen content, which is one of the important quality reference of kochujang, was higher in the one containing 0.6% natural preservatives. No color changed in the kochujang containing mustard but L and a value were increased in the kochujang containing horseradish. No difference of free reducing sugars was showed by the addition of those natural preservatives and the addition of 0.6% horseradish to kochujang had completely stopped gas forming during fermentation. The overall quality of the kochujang containing 0.6% horseradish was superior than that of the other treatments.

Sensory Characteristics and Preference of Various Chinese Foods added Kochujang by Chinese Focus Group (고추장 첨가 중국 음식에 대한 중국인의 관능적 특성 및 기호도 분석)

  • The purpose of this study was to analyze the characteristics of Chinese foods in main ingredients, sauces, and cooking methods, and to assess the applicability of kochujang in Chinese foods. Twenty Chinese foods and commercial kochujang were selected by a Chinese head cook. The main ingredients were 24 kinds such as pork, chicken, shrimp, bok choy, bean curd, Chinese noodles etc., which were generally used in Korean foods. The main Chinese sauces were 11 kinds, and soy sauce was used in 12 foods, shang loo tau soy sauce in 2, wine in 8, oyster sauce in 7, rice wine in 6. The classification of cooking units in all Chinese foods was performed and the cooking frequency was deep frying>pan frying>boiling>stir frying>steaming>roasting. The proper amount of kochujang (weight ratio) was decided by pre-test of the Chinese head cook and Chinese food added kochujang was assessed by a Chinese focus group. The overall preference of Chinese food added kochujang was assessed highly and was related to the kinds of sauces, and to the kinds of main ingredients. The foods which used soy sauce or shang loa tau soy sauce were preferred. Sweetness by using kochujang wasn't preferred in several foods. Saltiness and aftertaste by using kochujang didn't affect the food preference, and the intensity of color and pungency were evaluated as a moderate level. An increased amount of kochujang would be possible in several foods. However, the increase in the amount of kochujang for intensive color and pungency in Chinese foods was directly related to the increase of sweetness, which had been discussed as the main problem in lowering the preference. Therefore, the screening of various Chinese foods harmonized with kochujang and the determination of the proper mixing ratio with Chinese sauces are very important. The results of this study could be used as basic data for the promotion of kochujang consumption in the Chinese market.

Decreasing Effect of Kochujang on Body Weight and Lipid Levels of Adipose Tissues and Serum in Rats Fed a High-Fat Diet (고지방식이를 섭취한 흰쥐에서 고추장의 체중 및 지방조직과 혈청내의 지질감소효과)

  • Effect of kochujang (Korean red pepper soybean paste) on the reduction of body weight and lipid levels of adipose tissues and serum were investigated in the rats fed a high-fat diet. Male Sprague-Dawley rats were fed for four weeks with diets containing either a normal diet (ND, based on the AIN-76TM diet), high fat diet (HFD, supplemented with 20% corn oil in ND), CK diet (10% commercial kochujang in HFD), TK I diet (10% 0 day (non-) fermented traditional kochujang in HFD), TKII diet (10% 6-month fermented traditional kochujang in HFD), or RPP diet (3% red pepper powder in HFD). CK, TK I and TKII in the diet significantly reduced the body weight gain compared to the control (HFD), whereas red pepper powder had no such effect. TKII supplementation in the high-fat diet reduced epididymal and perirenal fat pad weight significantly (p<0.05). Liver total lipid was lower in TKII diet group than other kochujang and RPP diet groups. TKII and CK decreased the content of triglyceride in epididymal and perirenal fat pad that increased by high-fat diet. The cholesterol content of the perirenal fat pad was the lowest in the TKII diet group. TKII was also effective in lowering serum cholesterol level. These results indicated that properly ripened (fermented for 6-month) traditional kochujang exhibited more suppressive effects on body fat gain and lipid levels of adipose tissue and serum than RPP, commercial kochujang and the kochujang without fermentation.

Inhibitory Effects of Kochujang Extracts on the Tumor Formation and Lung Metastasis in Mice

  • Effects of kochujang (Korean red pepper soybean paste) extracts on tumor formation, natural killer (NK) cell activity in spleen and glutathione S-transferase (GST) activity in liver were investigated in the sarcoma-180 cell transplanted mice. Inhibitory effects of these samples on lung metastasis of colon 26-M3.1 cells were also evaluated in the Balb/c mice. The injection of methanol extracts from traditional kochujang I (TK I, 0-day fermented), II (TKII, 6-month fermented), commercial kochujang (CK, 1-month fermented) and red pepper powder (RPP) significantly reduced tumor formation in Balb/c mice (p<0.05), TKII decreased tumor growth by 46% compared with control, resulting in the smallest tumor weight. The transplantation of sarcoma-180 cells increased the spleen/body weight ratio of Balb/c mice, while TKI and TKll significantly decreased this index (p<0.05). The effect of TKll and CK, fermented kochujang, on the NK cell activity of splenocytes was higher than that of sarcoma-180 cells transplanted control group. TK II recovered the activity of hepatic GST that was decreased by the transplantation of sarcoma- 180 cells in to the mice. All kochujang-treated mice had significantly fewer lung metastatic colonies than control mice. TKII was the most effective in inhibiting lung metastasis of colon 26-M3.1 cells. These results indicated that optimally ripened (6-month) TK had more suppressive effects on tumor formation and lung metastasis than RPP and kochujang without fermentation and commercially prepared kochujang in mice.

Analysis of the quality characteristics of Kochujang prepared using pastes from different peach varieties (복숭아 품종별 페이스트를 이용한 고추장의 품질 특성)

  • In this study, we aimed to increase the utilization of non-commercializable peach fruits collected during harvesting and distribution and to promote the transition of a high value-added industry. After producing pastes from the flesh of different peach varieties, we used these pastes as ingredients in the production of Kochujang. We then performed a comparative analysis on the quality characteristics of the peach-added Kochujang and commercially available Kochujang products. Peach-added Kochujang exhibited a lower pH and reduced sugar, salinity, reducing sugar, ash, and calorie content than those of commercially available Kochujang products. However, the Kochujang had slightly higher pH and moisture content than commercially available products due to high organic acid and moisture contents of peach flesh. In terms of content of inorganic elements, commercial Kochujang products exhibited higher sodium and magnesium content, whereas peach-added Kochujang had higher potassium content. In particular, the lower sodium and higher potassium content of peach-added Kochujang indicates that this product could be used as a functional food to prevent various diseases caused by excessive sodium intake among Koreans. After comparing the products in terms of phenol and flavonoid content, we concluded that ethanol extracts of peach-added Kochujang showed higher phenol content than those of the commercially available Kochujang products. In contrast, the distilled water extracts of peach Kochujang showed higher DPPH free radical scavenging activity than that of commercially available Kochujang products.

Characteristics of Volatile Flavor Compounds in Kochujang Prepared with Commercial Enzyme During Fermentation (효소제를 사용한 개량식고추장의 숙성과정 중 휘발성 향기성분의 특성)

  • Kochujang was prepared for this study with raw material inoculated by commercial enzyme of amylase and protease. Volati1e compounds of Kochujang were analyzed using a purge and trap method during fermentation and identified with GC-MSD. Total 54 kinds of volatile flavor components like 16 kinds of alcohol, 16 kinds of ester, 7 kinds of acid, 4 kinds of aldehyde, 2 kinds of alkane, 1 kind of benzene, 3 kinds of ketone, 1 kind of alkene, 2 kind of amine, 1 kind of phenol, other 1 were found. Total number of volatile flavor detected right after manufacturing were 23 kinds like 3 kinds of alcohol, 6 kinds of ester, 3 kinds of aldehyde. After 30 days storage, total number of volatile flavor went up to 31 kinds with addition of 4 kinds of alcohol, 1 kind of ester. The total number of volatile flavor after 120 days storage were increased to 49 kinds. Volatile flavor compounds detected during the storage period were total 20 kinds like 6 kinds of alcohol such as 2-methyl-1-propanol, ethanol, 3-methyl-1-butanol, 5 kinds of ester such as ethyl acetate, isoamyl acetate, ethyl butyrate, 3 kinds of aldehyde such as butanal, acetaldehyde and 6 kinds of others. Even though peak area % of flavor compound varied depends on fermentation period, ethanol, ethyl acetate, ethyl butyrate, ethenone, 2-methyl-1-propanol, 3-methyl-1-butanol were the main compounds that consisted of flavor from Kochujang which was made with enzyme treatment. Ethly acetate showed the highest result in the treatment of right after manufacturing, 3-methyl-1-butanol had up to 90th day and ether were the other days.

Yield Stress of Kochujang with Vane Method (Vane 방법을 이용한 고추장의 항복응력 측정)

  • The vane method was used to measure yield stresses of five commercial kochujang samples under a controlled shear-rate operating condition. Magnitudes of vane yield stress were higher than those of yield stress using Casson model with a concentric cylinder viscometer. Magnitudes of vane yield stresses showed great differences between the static $({\sigma}_s)$ and dynamic yield stresses $({\sigma}_d)$ of kochujang samples with undisturbed structure (UDS) and with broken down structure (BDS). A dimensionless yield number $(N{\sigma}_o)$ was determined from the ratio of ${\sigma}s$ to ${\sigma}d$ in order to describe the existence of temporary structure of kochujang.

  • This study was carried out to investigate the bacterial microflora of commercial and traditional Kochujang. Computer aided idetification systems were used to identify the bacteria in Kochujang. Seven genera such as Bacillus, Corynebacterium, Enterococcus, pasteurella, Pseudomonas, Staphylococcus and Streptococcus in commercial process and 10 genera such as Bacillus, Corynebacterium, Enterococcus, Flavimonas, Flavobacterium, Gemella, Pasteurella, Pseudomonas, Staphylococcus and Streptococcus in traditional process were identified. Distribution of genus Bacillus was $56{\sim}70%$ during fermentation in the commercial process and $38{\sim}50%$ in the traditional process. B. lichenoformis strains identified in this study were classified into three types by their physiological characteristics.

Investigation of quality characteristics and alcohol content in commercial Korean fermented sources (시판 장류의 알코올 함유량 및 품질특성 조사)

  • The purpose of this study was to investigate alcohol content of commercial Korean fermented sources and to analyze the quality parameters such as titratable acidity, pH, salinity, reducing sugar, alcohol and yeast cell count. Eighty-seven samples of Doenjang, 68 samples of Kanjang and 57 samples of Kochujang were collected and divided into conventional and improved products depending on manufacturing method. As a result, the titratable acidities of conventional Doenjang, Kanjang and Kochujang were 1.72, 1.23 and 1.25%, and those of the improved products were 1.87, 1.80, and 1.00%, respectively. The pH values of conventional Doenjang and Kochujang were lower than those of the corresponding improved products and the pH of conventional Kanjang was higher than improved products. The salinities of the conventional products were higher than the improved products, and an the reducing sugar and alcohol content of the conventional products were lower than the improved products. The detection rate of alcohol (above 1%) in conventional Doenjang, Kanjang, and Kochujang were 6, 10 and 39%, respectively while the corresponding improved products showed much higher rates of 100, 76 and 100%, respectively. There was no significant difference in the yeast cell count between the alcohol-containing products and the alcohol-free products. Accordingly, further study is necessary to develop a manufacturing method using non-degradable sugar and salt control to reduce the production of alcohol in Korean source.
