Searching of Possible Target Enzymes for Herbicide Development using Commercial Plant-Specific Inhibitors (식물 특정효소저해제의 생물활성 조사에 의한 신규제초제 작용점 탐색)

  • This study was conducted to search new target enzymes of novel herbicide candidate. Total of 107 biochemical inhibitors reported to inhibit over than 100 different plant enzymes were purchased from commercial chemical companies. 15 inhibitors and 34 enzymes were selected by germination assay, seedling assay, wheat leaf disc assay, and whole plant assay. Among them, seven compounds of purine, phehyl-hydrazine, o-phenanthroline, oleylamine, dicyclohexylcarbodiimide, 7,8-benzoquinoline, and aminooxyacetic acid showed high herbicidal activity in the whole plant assay under greenhouse while 7,8-benzoquinone, 8-hydroxyquinoline, 2,2'-dipyridyl, and o-phenanthroline inhibited seed germination of barnyardgrass, rice, and tomato at concentrations of 1.25 to $5{\mu}M$. The compounds of 7,8-benzoquinoline, chlorpromazine, cyanuric fluoride, 4-methylpyrazole, oleylamine, tranylcypromine, and trifluoperazine inhibited the growth of cyanobacteria at 30 to $100{\mu}M$. The compounds of dicyclohexylcarbodiimide and chlorpromazine exhibited whitening effect on tile wheat leaf disc at $100{\mu}M$. These results suggest that the plant-specific enzyme inhibitors which have biological activities may supply the target enzyme for developing new herbicide candidate.

Optimum Potting Medium and Nitrogen and Phosphorus Levels in the Soil for Root Nodule Formation in Black Locust (Robinia pseudoacacia L.) Seedlings (아까시나무(Robinia pseudoacacia L.) 유묘의 뿌리혹 형성에 적절한 배양토, 질소, 인 수준 구명에 관한 연구)

  • The objectives of this study were to find out proper potting medium and nutrient levels in the soil to promote the root nodule formation in black locust (Robinia pseudoacacia L.) seedlings. Commercial potting medium, compost, organic fertilizer, molded forest fertilizer, and compound fertilizer were used at different mixing rates to bring in various levels of mineral nutrients in the soil. Seedlings were grown in pots in a greenhouse for three months. Commercial potting medium containing peatmoss, vermiculite, and geolite was not suited for early nodule formation due to lack of nutrients, even though it produced good total dry weight. Compost was the best medium to promote both high total dry weight production and nodule formation with providing the proper levels of nitrogen and phosphorus in the soil. Molded forest fertilizer was acceptable for nodule formation. Compound fertilizer and organic fertilizer was not suited for nodule formation. The potting medium should contain optimum levels of nitrogen (0.05-0.2%) and phosphorus (100-600ppm) to promote early nodule formation in black locust seedlings.

  • Faced with global agenda of greenhouse abatement program including regulations and $CO_2$ emission trading scheme, shipping companies are enforced to a high level of efficiency in fuel consumption. Accordingly shipbuilding companies worldwide are required to develop fuel-efficient ships which otherwise traditionally consume a great amount of fossil fuels. In this dissertation, relevant to the improvement of fuel efficiency for commercial ships, design methodology through the numerical simulations are intensively described. This work consists of derivation of effective hydrodynamic design practice based on the application of longitudinal grooves to effectively improve the pressure distribution around ship hull. The primary objective of the present study is to improve ship resistance performance using longitudinal grooves which originate from long strips on the abdomen of humpback whale. Several groove shapes have been extensively investigated and the proposed shape efficiently controlled the variation of pressure distributions acting on the hull surface.

  • By releasing predatory mite, Amblyseius swirskii Athias-Henriot, with 25 individuals per cucumber plant in a commercial greenhouse (1,600 $m^2$), the population of western flower thrips, Frankliniella occidentalis (Pergande), was kept under the economic threshold level. The initial thrips population of 18.3 individuals per leaf decreased to 2.1 individuals per leaf after four weeks, while the number of A. swirskii reached to 16.6 individuals per leaf. It was also observed that at a point where the thrips occurred in high density of about 34.3 individuals per leaf, A. swirskii population built up on the lower part of cucumber plant first, but as time passed the predatory mites moved upward to the top of the plant and aggregated.

Isolation and In vitro and In vivo Antifungal Activity of Phenylacetic acid Produced by Micromonospora aurantiaca Strain JK-1

  • The actinomycete strain JK-1 that showed strong inhibitory activity against some plant pathogenic fungi and oomycetes was isolated from Jung-bal Mountain in Ko-yang, Korea. The strain JK-1 produced spores singly borne on sporophores and the spores were spherical and 0.9-1.2 11m in diameter. The cell wall of the strain JK-1 contained meso-diaminopimelic acid. The actinomycete strain JK-1 was identified as the genus Micromonospora based on the morphological, physiological, biochemical and chemotaxonomic characteristics. From the 168 rDNA analysis, the strain JK-1 was assigned to M aurantiaca. The antibiotic MA-1 was purified from the culture broth of M aurantiaca JK-1 using various purification procedures, such as Diaion HP20 chromatography, C18 flash column chromatography, silica gel flash column chromatography and Sephadex LH-20 column chromatography. $^{1}H-$, $^{13}C-NMR$ and EI mass spectral analysis of the antibiotic MA-1 revealed that the antibiotic MA-1 is identical to phenylacetic acid. Phenylacetic acid showed in vitro inhibitory effects against fungal and oomycete pathogens Alternaria mali, Botrytis cinerea, Magnaporthe grisea, Phytophthora capsici and yeast Saccharomyces cerevisiae at < 100 $\mug$ $ml^{-1}$. In addition, phenylacetic, acid completely inhibited the growth of Sclerotinia sclerotiorum, Bacillus subtilis, Candida albicans, Xanthomonas campestris pv. vesicatoria at < $\mug$ $ml^{-1}$. Phenylacetic acid strongly inhibited conidial germination and hyphal growth of M grisea and C. orbiculare. Phenylacetic acid showed significantly high levels of inhibitory' effect against rice blast and cucumber anthracnose diseases at 250 $\mug$ $ml^{-1}$. The control efficacies of phenylacetic acid against the two diseases were similar to those of commercial compounds tricyclazole, iprobenfos and chlorothalonil .n the greenhouse.

  • High oil price and global warming problem are being continued all over the world. For this reason, fuel economy and emission of greenhouse gas are regulated by law in many countries. Therefore many companies are researching and producing hybrid electric vehicles (HEVs) which substitute conventional internal combustion engine vehicle. However, these researches and productions are restricted to mainly passenger cars. Because of cost and physical problems, commercial vehicles are difficult to evaluate fuel economy. So simulations are important and it is necessary to know how sensitive parameters that enter into simulation affect. In this paper, forward simulations using AVL Cruise were conducted for analysis of fuel economy for parallel hybrid bus and were repeated by changing each parameter. Based on these results, root mean square errors (RMSE) are calculated for analysis of fuel economy sensitivity. The number of target parameters are 15. These parameters were classified with high and low sensitivity parameter relatively.

Evaluating the Influence of Liquid Organic Polymer on Soil Aggregation and Growth of Perennial Ryegrass (유기중합물이 토양의 입단화와 페레니얼 라이그래스의 성장에 미치는 영향)

  • Soil aggregate is a vigorous procedure including soil physical, chemical, and biological processes. Pore space created by binding these particles together improves retention and exchange of air and water. Various researches have reported that the benefits of organic polymers that may increase aggregate stability. The purpose of the study was to determine if a liquid organic polymer mixture has any influence on perennial ryegrass quality or soil aggregation. $Turf2Max^{(R)}$ was applied to two soils as a source of liquid organic polymer. Fine-loamy soil from local Iowa topsoil with 4.0% organic matter was screened and dried. Commercial baseball infield clay, $QuickDry^{(R)}$, was used as the second soil There were three rates of liquid organic polymer (0, 2, and 4%). there was no visual improvement in turf grass color, quality, or growth by using organic polymer. It is possible that aggregate stability increases with use of organic polymer. The aggregate stability study needs to be repeated in the greenhouse and then substantiated under field conditions for these preliminary observations.

  • For environmental reasons and because of our limited energy resources, high-efficiency power generation technology will be necessary in the future. Ultra-supercritical (USC) power generation technology is the key to managing the greenhouse gas problems and energy resource problems discussed in the Kyoto Protocol to the United Nations Framework Convention on Climate Change. Other countries and manufacturers are trying to build commercial power plants. In this paper, an efficient method of achieving near-zero emission operation of a high-efficiency fossil power plant using USC power generation is discussed. Development of USC power generation in Korea has been supported by the Korean government in two phases: Phase I was USC key technology development from 2002 to 2008, and Phase II is USC development and technology optimization from 2010 to 2017.

  • In recent years, eco-friendliness and high fuel economy have become important issues for commercial tractors. Electric tractors are often required for operation in a greenhouse. However, the battery capacity limits the available operation time. To overcome this problem, a plug-in hybrid electric tractor is considered a reasonable alternative. This tractor has a basic driving ability and can operate in various working modes such as mower, rotary, loader, and trailing. This study focuses on the energy management strategy by considering various working modes.

Study on Integrated Control against Root-knot Nematode of Fruit Vegetables (Oriental Melon and Cucumber) in Vinyl House (시설과채류(오이, 참외)의 뿌리혹선충 종합방제에 관한 연구)

  • To reduce crop damage from root-knot nematode, Meloidogyne spp. chemical, cultural, physical control methods were compared in commercial greenhouse for 3 years from 1992 to 1994. Timing of sol exchange for economic losses due to the nematodes is important: every 3 years for coarse sand (Masatto) and every 4 years for red-yellow soil. Control effects on Meloidogyne spp. in cucumber were 78.2% in Dazomet DP, 72.1% Carbo G+Soil reverse+submergence, and 66.3% in Carbo G+submergence. Pesticide effects were temporally different after treatment: 77.7~80.6% in 20 days and 33.7~49.5% in 60 days. Cropping system in oriental melon gave an excellent control effect of 81.1% at oriental melon/rice culture. All methods controled root-knot nematodes at the time of the treatment but the number of the nematodes increased at the end of the season. However, soil exchange was effective for 3 years. The most economic control practice is rotation with rice for every three years.
