Optimal Extraction and Characteristics of Lignocellulytic Enzymes from Various Spent Mushroom Composts (다양한 버섯 수확 후 배지로부터 목질섬유소 분해효소의 최적 추출 및 특성)

  • Recovery of ${\alpha}$-amylase (EC 3.2.1.1), lignin peroxidase (EC 1.11.1.14), laccase (EC 1.10.3.2), xylanase (EC 3.2.1.8), ${\beta}$-xylosidase (EC 3.2.1.37), ${\beta}$-glucosidase (EC 3.2.1.21) and cellulase (EC 3.2.1.4) from spent mushroom composts (SMCs) of Pleurotus cornucopiae, Pleurotus ostreatus, Pleurotus eryngii, Hericium erinaceum, Lyophyllum ulmarium, Agrocybe cylindracea, Lentinus lepideus, and Flammulina velvtipes were investigated using different extraction buffers. The maximum recovery of the enzymes was mostly detected in SMC extracts with tap water and 0.25% Triton X-100 by shaking incubation (200 rpm) for 2 h at $4^{\circ}C$. The xylanase (152 U/g) and laccase (8.1 U/g) activities were the highest in SMC extracts from F. velvtipes and P. eryngii. In addition, high enzymatic activities of ${\alpha}$-amylase (3.6 U/g) and cellulase (3.4 U/g) was detected in SMC extract of A. cylindracea. Futhermore, cellulase and laccase activities of SMCE from P. eryngii were compared to commercial enzymes.

The Soil Removal of Artificially Soiled Fabrics with Commercial Detergents at Various Washing Conditions (시판 세제를 사용하여 세척 조건에 따른 인공오염포의 세척성)

  • Formulations of compact and supercompact heavy-duty detergents, which are being used widely around the world, differ from those of conventional heavy-duty detergents. Significant differences in composition exist between the compact detergents and the conventional detergents. The compact detergents have a higher content of surfactants, oxygen bleach and enzymes than the conventional detergents. We have studied to find the most effective washing conditions of artificially soiled cloths with a commercial, supercompact, heavy-duty detergent and a drum type washing machine which is becoming the preferred type in Korea. Moreover, we have studied the washing performance with an impeller type washing machine, which has heretofore been the most popular type in East Asia. With the drum-type washing machine, washing performances improved as the washing temperatures went up and the washing times were lengthened. Though the rate of soil removal with a double recommended dosage was higher than with the recommended amount at $20^{\circ}C$, the effects of the higher dosage decreased as the washing temperatures increased. Finally, the washing performances with the two different dosages were the same at $60^{\circ}C$. The washing performances at $40^{\circ}C$ with the recommended dosage for 90 minutes were the same as with the double recommended dosage for 45 minutes. The soil removal efficiencies with the impeller-type washing machine were much lower than those of the drum-type washing machine. The reasons for this were the higher bath ratio that led to the lower concentration of wash liquor, the shorter washing time, and the lower washing temperature.

  • Defatted rice polishings (DRP) was subjected to chemical treatments i.e., 0.4 N HCl, and 6% $H_2O_2$, with or without physical treatment i.e. extrusion cooking. The treated DRP was evaluated chemically and biologically using male broiler chicks (108) of approximately uniform weight, selected out of 220 chicks, previously fed on commercial diets for 7 days as a settlement period. The chicks were then divided into 36 experimental units of 3 chicks each. Each experimental diet was randomly allotted to three experimental units and fed for 10 days to broiler chicks. The experimental diets were designated as A (Commercial), B (10% HCl treated DRP), C (20% HCl treated DRP), D (10% HCl plus extruded DRP), E (20% HCl plus extruded DRP), F (10% $H_2O_2$ DRP) and G (20% $H_2O_2$ DRP), H (10% $H_2O_2$ plus extrusion DRP) and I (20% $H_2O_2$ plus extrusion DRP), J (10% untreated DRP), K (20% untreated DRP) and L (Protein free). The birds fed on diet L were used to measure the endogenous nitrogen loss. The biological evaluations of diets containing differently treated DRP were compared with a commercial feed and feeds containing untreated defatted rice polishings. It was observed that these treatments liberated bound nutrients, making them more accessible to the normal digestive enzymes and increased their apparent nutrient availability. This process probably also detoxified the anti-nutritive factors i.e. phytates, lectin, trypsin inhibitor present in DRP. The results of the feeding trials revealed that diets containing 6% $H_2O_2$ treated DRP showed better weight gain, feed consumption and utilization, protein efficiency and digestibility, biological value and net protein utilization than all other treatments.

  • This review was written to introduce updated data on the structure and function of the major milk proteins identified as allergens, the characterization of their epitopes in each allergenic milk proteins, and the reduction of milk protein allergenicity. Most mammalian milk protein, even protein present at low concentration, are potential allergens. Epitopes identified in milk proteins are both conformational(structured epitope) and sequential epitopes(linear epitope), throughout the protein molecules. Epitopes on casein and whey proteins are reported to be sequential epitope and conformational epitopes, respectively. Conformational epitopes on whey protein are changed into sequential epitope by heat denaturation during heat treatment. Several methods have been proposed to reduce allergenicity of milk proteins. Most ideal and acceptable method to make hypoallergenic milk or formula, so far, is the hydrolysis of allergenic milk proteins by enzymes that has substrate specificity, such as pepsin, trypsin, or chymotrypsin. Commercial formulas based on milk protein hydrolysate are available for therapeutic purpose, hypoantigenic formula for infants from families with a history of milk allergy and hypoallergenic formula for infants with existing allergic symptoms.

Nutritional Factors Affecting Abdominal Fat Deposition in Poultry: A Review

  • The major goals of the poultry industry are to increase the carcass yield and to reduce carcass fatness, mainly the abdominal fat pad. The increase in poultry meat consumption has guided the selection process toward fast-growing broilers with a reduced feed conversion ratio. Intensive selection has led to great improvements in economic traits such as body weight gain, feed efficiency, and breast yield to meet the demands of consumers, but modern commercial chickens exhibit excessive fat accumulation in the abdomen area. However, dietary composition and feeding strategies may offer practical and efficient solutions for reducing body fat deposition in modern poultry strains. Thus, the regulation of lipid metabolism to reduce the abdominal fat content based on dietary composition and feeding strategy, as well as elucidating their effects on the key enzymes associated with lipid metabolism, could facilitate the production of lean meat and help to understand the fat-lowering effects of diet and different feeding strategies.

Chitooligosaccharide Prolongs Vase Life of Cut Roses by Decreasing Reactive Oxygen Species

  • Chitooligosaccharide (COS), as antioxidant, extensively applied to food and juice preservation. In the present study, influences of COS on vase life and ornamental value of cut roses were investigated. Results showed that vase life of cut roses treated by COS was longer 6.4 days than one of control and ornamental character of cut roses was improved effectively by COS. The increase of vase life and ornamental value were chiefly governed by that COS improved water absorption capacity of cut roses. Besides that, COS decreased the contents of superoxide anion and hydrogen peroxide and lowered the levels of malondialdehyde in turn during the senescence process of cut roses. That was because that COS not only enhanced activities of antioxidant enzymes glutathione reductase, but also improved reduced glutathione contents in petals of cut rose. Therefore, COS could be used in commercial preservatives to improve the longevity of cut roses.

Characteristics of White Water from Enzyme Deinking process for ONP at Low Alkalinity

  • Old newspaper was deinked using commercial cellulolytic enzymes and a surfactant complex at low alkalinity. The properties of the deinked pulp(DIP) were evaluated and the suspended solids content, cationic demand, turbidity, and chemical oxygen demand(COD) of the process water were measured. The results can summarized as follows, 1. The brightness and yield of the DIP were improved using enzymatic surfactant complex deinking. 2. The amount of foaming during deinking with the enzyme surfactant complex was higher than that with synthetic surfactant deinking. However, it was not sufficient to cause process problem. 3. The pH and turbidity of the white water from deinking with the enzyme surfactant complex were similar to those of the white water from surfactant deinking. 4. The suspended solids content, cationic demand, and COD of the white water from deinking with the enzyme surfactant complex were improved compared to those of the white water from surfactant deinking.

  • Optimum preparation conditions of Korean traditional sweet rice drink 'malt-Sikye' were 1hour of rice soaking time 30min of malt extraction time 60$^{\circ}C$ of malt extraction tem-perature 60$^{\circ}C$ of saccharification temperature 1hour of agitation interval 5hour of saccharification time 5.5 of pH 4% of malt concentration and 20% of rice content. The malt-Sikhy contained to 17.1% of total sugar 11.2% of reducing sugar o.34mg/ml of protein and 4.7${\mu}$mol/ml of amino acid. The most abundant sugar found in malt-Sikhye was maltose. The commercial amylolytic enzymes were not effec-tive in preparation malt-Sikye.

  • Effects of aqueous ammonia soaking to three annual plants (hemp woody core, tobacco stalk and corn stover) awere investigated to focus on the enzymatic saccharification characteristics change by this treatment. At two different levels of treatment ($90^{\circ}C$-16 h and $45^{\circ}C$-6 days), higher temperature treatment led to more enzymatic saccharification of cellulose to glucose by commercial cellulase mixtures (Celluclast 1.5L and Novozym 342 from Novozyme Korea). Difference among annual plants were significant. corn stover was the best response to enzymatic saccharification of cellulose and xylan by comercial enzymes all treatment conditions but tobacco stalk was the worst response to all of them. chemical composition or physical structure difference may brought this difference.

  • In this study, the production of total reducing sugar from macro green-algae Enteromorpha intestinalis by enzymatic hydrolysis was investigated. As a result of enzymatic hydrolysis using 13 kind commercial enzymes, the highest yield of 8.75% was obtained from Viscozyme L, which is multi-enzyme complex such as cellulase, arabanase, beta-glucanase, hemicellulase and xylanase. As a control, only 0.33% and 0.27% yield were obtained from 1% sulfuric acid and 0.05 M citrate buffer (pH 4.8), respectively. In the case of enzyme mixture, the mixture of $Viscozyme^{(R)}$ L and $Cellic^{(R)}$ CTec2 (1:1) was presented the highest yield of 10.67%. Finally, the 14.99% yield was obtained at 36 hr under the condition of 10% biomass and 30% enzyme mixture.
