Buffer capacities for two Bo horizon soils or Andisols developed from different parent materials have been investigated. The titration curves from column leaching experiment show that buffering occurred at pH 4.0 and 6.0. The buffer intensity or soil developed from pyroclastic materials (P-soil) is higher than that from basalts (B-soil). From batch test we have found that proto-imogolite and/or imogolite may control Al solubility as well as $Al(OH) _3$in the moderate acid condition. The buffer intensities ($\beta$) of P-soils were plotted on the theoretical buffering curve of $Al(OH)_3$, while $\beta$ of B-soils approached to that of proto-imogolite, which shows the solubility of short-range-order materials in P-soil control the buffer capacity. Buffering at pH 6.0 is thought to be the result of dissolution of some silicate clays and exchange reactions between $H^{+ }$and base-forming cations. Considering the amount of annual acid precipitation, aluminum solubility of Andisols, and the low BS (Base Saturation percentage), it can be predicted that prolonged acid precipitation will reduce the buffer capacity of soils and lead to soil acidification.

Computational simulations of adsorption columns were carried out to investigate the removal characteristics of VOCs from a laundry shop. n-Decane was selected as the representative component among the VOCs emitted, and the activity of the adsorbents, such as activated carbon, was evaluated using commercial CFD code. The mathematical framework was composed of continuity and Navier-stokes equations, and the simulation was performed using the Matlab program. The adsorption isotherms of LDF, Freundlich, and Langmuir were evaluated, and the adsorption amount of the adsorption isotherms with the adsorption parameter was compared. The simulation was carried out using a particle porosity, dispersion coefficient, particle density, bed diameter, and bed length of 0.79, 42.4 ㎠/min, 485 g/L, 2.0 cm, and 2.5 cm, respectively. The effect of the gas velocity, dispersion coefficient, and voidage on the adsorption amount was compared in the Langmuir adsorption isotherm. The simulation was carried out in the velocity range of 50 to 200 cm/min, dispersion coefficient range of 100 to 400 ㎠/min, and particle porosity range of 0.66 to 0.79. The simulation results of activated carbon with benzene coincided with the Langmuir isotherm. Three types of adsorption isotherm were compared under similar conditions, and the simulation results showed the efficient adsorption condition for hydrocarbons.

This paper presents an investigation on the ultimate behavior of steel cable-stayed bridges using nonlinear finite element analysis method. Cable-stayed bridges exhibit various geometric nonlinearities as well as material nonlinearities, so rational nonlinear finite element analysis should be performed for investigation of the ultimate behavior. In this study, ultimate behavior of steel cable-stayed bridges was studied using rational ultimate analysis method. Nonlinear equivalent truss element and nonlinear frame element were used for modeling the cable, girder and mast. Moreover, refined plastic hinge method was adopted for considering the material nonlinearity of steel members. In this study, the 2-step analysis method was used. Before live load analysis, initial shape analysis was performed in order to consider the dead load condition. For investigation of the ultimate behavior of steel cable-stayed bridges, analysis models which span length is 920.0 m were used. Radiating type and fan type were considered as the cable-arrangement types. With various quantitative evidences such as load-displacement curves, deformed shapes, locations of the yield point or region, bending moment distribution and so on, the ultimate behavior of steel cable-stayed bridges was investigated and described in this paper.

Responses of plankton populations to nutrient enrichment in mesocosm experiments in Shihwa lake were simulated using FVCOM. Dissoloved oxygen module was added to the FVCOM to simulate impacts of its decreased levels. The ecological model included the major components of the pelagic ecosystem including nutrients, phytoplankton (pico-, nano-, micro-), zooplankton (two groups of protozoa, mesozooplankton), particulate organic matter, dissolved organic matter and bacteria, and was calibrated using trophodynamic data collected from Gyeonggi Bay and Shihwa Lake. The model was able to reproduce major responses of plankton populations to nutrient enrichment, including phytoplankton of different size groups, change of dominance of protozoa from < 20 ${\mu}m$ oligotrichs to scuticociliates, and reponses to bacteria and low levels of dissolved oxygen in water column of the mesocosms.

This study investigated the effect of surfactants (nonionic surfactant (Tween 20), biosurfactant) on enhancing transport of bacteria (Bacillus subtilis ATCC 6633) in geological materials. Column experiments were performed under various surfactant conditions with columns packed with quartz sand (particle size distribution: 0.5~2.0 mm, mean diameter: 1.0 mm). Bacterial mass recovery, sticking efficiency, and other parameters were quantified from breakthrough curves. Results indicate that bacterial attachment to sand surfaces increased considerably in the presence of mineral salt medium (MSM), especially at the inlet, which was due to the increase of ionic strength by MSM. It was observed that bacterial transport in sand columns was enhanced in the presence of surfactant. Results also show that simultaneous injection of both surfactant and MSM or pre-injection of surfactant was more effective in bacterial transport enhancement than after-injection of surfactant. This study suggests that transport of bacteria in geological materials could be influenced by surfactants and their injection methods.
Full-length cDNAs are essential for the correct annotation of genomic sequences and for the functional analysis of genes and their products. To elucidate the functions of a large population of Chinese cabbage (Brassica rapa) genes and to search efficiently for agriculturally useful genes, we have been taking advantage of the full-length cDNA Over-eXpresser (FOX) gene hunting system. With oligo dT column it purify the each mRNA from the flower organs, leaf and stem tissue. And about 120,000 cDNAs from the library were transformed into $\lambda$-pFLCIII-F vector. Of which 115,000 cDNAs from the library were transformed into T-DNA binary vector, pBigs for transformation study. We used normalized full-length cDNA and introduced each cDNA into Arabidopsis by in planta transformation. Full-length Chinese cabbage cDNAs were expressed independently under the CaMV 35S promoter in Arabidopsis. Selfed seeds were harvested from transgenic Arabidopsis. We had selected 2,500 transgenic plants by hygromycin antibiotic tolerant test, and obtained a number of transgenic mutants. Each transgenic Arabidopsis was investigated in morphological changes, fertility and leaf colour. As a result, 285 possible morphological mutants were identified. Introduced cDNA was isolated by PCR amplification of the genomic DNA from the transgenic mutants. Sequencing result and BLAST analysis showed that most of the introduced cDNA were complete cDNAs and functional genes. Also, we examined the effect of Bromelain on enhancing resistance to soft rot in transgenic Chinese cabbage 'Osome'. The bromelain gene identified from FOX hunting system was transformed into Chinese cabbage using Agrobacterium methods. Transformants were screened by PCR, then RT-PCR and real time PCR were performed to analyze gene expression of cysteine protease in the T1 and T2 generations. The anti-bacterial activity of bromelain was tested in Chinese cabbages infected with soft rot bacteria. The results showed that the over-expressed bromelain gene from pineapple conferred enhanced resistance to soft rot in Chinese cabbage.
Residual solvents in foods are defined as organic volatile chemicals used or produced in manufacturing of extracts or additives, or functional foods. The solvents are not completely eliminated by practical manufacturing techniques and they also may become contaminated by solvents from packing, transportation or storage in warehouses. Because residual solvents have no nutritional value but may be hazardous to human health, there is a need to remove them from the final products or reduce their amounts to below acceptable levels. The purpose of this study was to develop and evaluate an analytical method for the screening of residual solvents in health functional foods. Furthermore, the aim of this study was to constitute a reasonable management system based on the current state of the market and case studies of foreign countries. Eleven volatile solvents such as MeOH, EtOH, trichloroethylene and hexane were separated depending on their column properties, temp. and time using Gas Chromatography (GC). After determining the GC conditions, a sample preparation method using HSS (Head Space Sampling) was developed. From the results, a method for analyzing residual solvents in health functional foods was developed considering matrix effect and interference from the sample obtained from the solution of solvents-free health functional foods spiked with 11 standards solutions. Validation test using the developed GC/HSS/MS (Mass Spectrometry) method was followed by tests for precision, accuracy, recovery, linearity and adequate sensitivity. Finally, examination of 104 samples grouped in suits was performed by the developed HSS/GC/MS for screening the solvents. The 11 solvents were isolated from health functional foods based on vapor pressure difference, and followed by separation within 15 minutes in a single run. The limt of detection (LOD), limit of quantification (LOQ), recovery and coefficient of variation (C.V.) of these compounds determined by the HSS/GC/MS were found to be 0.1 pg/mL, 0.1-125 pg/g, 51.0-104.6%, and less than 15%, respectively. Using the developed HSS/GC/MS method, residual solvent from 16 out of 104 health functional products were detected as a EtOH. This method therefore seems t o be a valuable extension ofanalytical method for the identification of residual solvents in health functional food.

In order to investigate the changes in bioactive materials induced in goldfish brains by $^{18}F$ irradiation, the variations in the neurotransmitter levels in the whole brain were studied. The distance between the goldfish and 580 mCi of $^{18}F$ was about 4 cm, and the exposure lasted for 4 hrs. The absorption level calculated based on the distance, exposure time, and half-life of $^{18}F$ was approximately 2 Gy. After sacrifice by $^{18}F$ irradiation or untreated conditions, ten brains were dissected or immediately frozen, respectively. The tissues were extracted in acetic acid. After lyophilization, the samples were dissolved in distilled water and were further purified on a reverse-phase HPLC column. There were no differences in the intensities of the bioactive materials between $^{18}F$-exposed goldfish and control goldfish, while the only peak corresponded to 13 min, which indicated a significant increase in the irradiated brains. Our analysis has found that this compound is tryptophan. This result suggests that $^{18}F$ leads to changes in a classical neurotransmitter, tryptophan, in both the brains of control goldfish and goldfish contaminated by irradiation.
The prostate cancer is the critical health problem, increasing of its related death in worldwide. Unfortunately present surgery and chemotherapeutic choices seem to be impossible in curing or controlling prostate cancer, because metastasis occasionally advances even after these potentially curative therapies. Therefore, there is immediate need to alternative chemoprevention and chemotherapeutic agents. Over one hundred species of dried medicinal herbs were tested for proliferation inhibitory effects on prostate cancer cell line, PC-3. One of them, Anthriscus sylvestris was selected because of potent anti-proliferation effect. The dried root of A. sylvestris was extracted with 100% methanol for 2-3 days and its extract was fractionated by using ethyl acetate. And ethyl acetate layer was subjected to column chromatographies on silica gel, reverse phase-18 (RP-18) and Sephadex LH-20, in turn. Finally, the pure compound was obtained by crystallization in methanol at $4^{\circ}C$ for overnight and identified as deoxypodophyllotoxin by NMR spedorscopic and physico-chemical analyses. In addition, it was confirmed that deoxypodophyllotoxin clearly inhibits the proliferation of PC-3 cells in a dose and time-dependent manner.

Ochratoxin A and aflatoxin may be detected from naturally fermented foods due to the contamination of the mycotoxin-producing molds or un-prudential use of the mycotoxin producing starter strains during the fermentation. This study was carried out to analyze the production of ochratoxin A and aflatoxin under the various environmental conditions. For the experiment, the effects of different temperature, culture media, and fermentation time on the production of ochratoxin A by Aspergillus usamii KFRI 999 and A. awamori KFRI 983 were analyzed. Additionally, the production of aflatoxin was assessed under the various temperature, initial pH, fermentation time and culture media during fermentation by A. flavus KACC 41403 and A. oryzae KACC 46471. The levels of ochratoxin A and aflatoxin were analyzed by HPLC. The result showed that the production of mycotoxin was greatly affected by the fermentation temperature. A. oryzae KACC 46471 did not produce aflatoxin. All of the mycotoxin producing strains showed the highest level of mycotoxin at $30^{\circ}C$. A. awamori KFRI 983 showed the lowest level of ochratoxin A in PDA media among the experimental medium. The results of the present study may be useful for the reduction of ochratoxin A and aflatoxin in various foods.
