  • Normal tensile strength in collagen fibrils is due to intermolecular and intramolecular crosslinks which are known to be altered in aging. Pyridinoline, a mature crosslink which is stable and nonreducible, is derived from two hydroxyallysine and one hydroxylysine residues of collagen fibrils. The excess formation of pyridinoline in collagen is associated with making the tissue stiffer, less soluble and less digestible by enzymes. Lysyl oxidase is the enzyme that initiates the biosynthesis or crosslinks in collagen by catalyzing the oxidative deamination of the lysyl and hydroxylysyl residues in these molecules, and its activity is inhibited by $\beta$-aminopropionitrile(BAPN). Our previous work demonstrated that the pyridinoline content of bone collagen significantly was increased during incubation for 5 weeks at 37$^{\circ}C$ invitro, but it was diecrased by the addition of ascorbic acdi(AsA). In this study, we clarified the specific action of AsA in aging process in vitro enzymatic reaction.

The Effects of Rosiglitazone on in vivo Synthesis of Bone Collagen in Mice (Rosiglitazone이 마우스의 골조직 Collagen생성에 미치는 영향)

  • This study was performed to investigate the effect of rosiglitazone, a new antidiabetic agent, on in vivo synthesis of bone collagen. The mice were divided into low-fat diet group (LF), high-fat diet group (HF), and high-fat diet with rosiglitazone (6.3 $\mu\textrm{g}$/kcal diet) group (HF-Rosi), The synthesis of bone collagen was measured by stable isotope-mass spectrometric technique using $^2$$H_2O$ as a tracer. The $^2$$H_2O$ labeling protocol consisted of an initial intraperitoneal injection of 99.9% $^2$$H_2O$, to achieve approximately 2.5% body water enrichment followed by administration of 4% $^2$$H_2O$ in drinking water for 3 weeks. Although body weight gain and daily diet intake were not significantly different between groups, HF-Rosi had slightly higher body weight gain and daily diet intake than LF and HF. In addition, HF-Rosi showed significantly higher body fat content than LF and HF. Bone collagen synthesis was reduced in HF than LF and further decreased by the treatment of rosiglitazone. These results suggest rosiglitazone affect body fat content and bone turnover in mice.

The Effects of Artemisia Princeps var. Orientalis Extracts on Serum Lipids and Connective Tissues Collagen in Ovariectomized Rats (쑥이 갱년기 장애 유도 흰쥐의 혈중 지질 및 결합조직 중 Collagen 함량에 미치는 영향)

  • The aim of this study was to evaluate the effects of Artemisia Princeps Var. Orientalis (AP) on serum lipids and the collagen content of the connective tissues in ovariectomized rats. From day 2 until day 37 after the ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups: sham-operated rats (Sham), ovariectomized control rats (OVX-control), and ovariectomized rats supplemented with the AP 50 mg/kg bw/day (OVX-AP). The AP ethanol extracts were orally administrated 1 mL per day. The OVX rats were significantly heavier than the sham-operated rats at all time points, but supplementation with the AP extracts tended to gain weight less than OVX-control. Although total-cholesterol was increased at OVX-control, supplementation with the AP extracts tended to result in less than OVX-control. Triglyceride was significantly decreased after supplemented with the AP extracts (p<0.05). HDL-cholesterol is appeared higher AP extracts group than OVX-control. According to the results, we could know the fact that AP extracts were effective on serum lipids content throughout decreasing total-cholesterol, triglyceride and increasing HDL-cholesterol in ovariectomized rats. Supplementation with the AP extracts prevented a decrease in the collagen level in bone and cartilage tissues. These results are consistent with the conclusions based on the estrogenic activities of AP. Therefore, it may be used to possibly improve the quality of life in menopausal women.

Antifibrotic Effects of Oriental Herbs Extraction on Liver Cirrhosis (한약재(자하거, 백출, 차전자) 추출물의 간경화 억제효능에 관한 연구)

  • Hepatic cirrhosis is an important feature of chronic liver disease. Liver cirrhosis is characterized by hyperaccumulation of fibrous tissue components and is commonly observed in latter or terminal states of chronic hepatic disease. The antifibrotic effects on liver cirrhosis by oriental herbs extraction material were examined in bile duct ligated rats. Oriental herbs extraction (0.99 mg/kg rat weight/day) was administrated to cirrohotic rats for 4 weeks. Liver collagen content of bile duct ligated rats was significantly increased. And liver histology showed collagen fiber deposition was increased as well as the normal architecture was lost with large zone of necrosis being observed. Herbs extraction administrated rats showed significantly decreased liver collagen content, accumulation of collagen fiber in histological analysis, and biochemical markers of hepatic diseases. Those results demonstrate the usefulness of herbs extraction materials as an antifibrotic agent for liver cirrhosis.

Physicochemical and histopathological parameters of broilers with dorsal cranial myopathy

  • Objective: This study aimed to investigate the effect of dorsal cranial myopathy (DCM) on chicken meat quality. Methods: Sixty-six Ross 308 AP broilers, 47 days old, of both sexes, weighing about 3.51 kg, were slaughtered according to standard industrial practices, and evaluated for meat color, pH, chemical composition, collagen content, fatty acid profile, and histopathological parameters. Comparisons between normal and DCM-affected meat were performed using Student's t-test at the 5% significance level. Results: Histological analysis of muscle tissues affected by DCM showed myofiber degeneration, proliferation of inflammatory cells, fibroplasia, and necrosis with fibrosis. DCM samples had lower protein content and higher moisture, ash, insoluble collagen, total collagen, and pH. DCM-affected meat was redder and more yellowish. There were no differences in lipid or soluble collagen contents between groups. DCM-affected meat had higher percentages of arachidonic acid (C20:4n-6) and eicosapentaenoic acid (C20:5n-3). Conclusion: This study revealed that DCM-affected meat exhibits considerable changes in quality parameters.

Meat Tenderness Characteristics of Ten Major Muscles from Hanwoo Steers according to Quality Grades of Carcasses

  • The objective of this study was to determine the influence of quality grade (QG) on meat tenderness characteristics of ten major muscles from Hanwoo steers. A total of 25 Hanwoo carcasses ($5\;carcasses{\times}5\;QGs$) were selected. Intramuscular fat content, collagen content, sarcomere length, and Warner-Bratzler shear force (WBSF) of Longissimus thoracis (LT), Longissimus lumborum (LL), Psoas major (PM), Semisponals (SS), Triceps brachii (TB), Semimembranosus (SM), Gluteus medius (GM), Rectus Abdominis (RA), Superficialis flexor (SF), and Internal and external intercostal (IC) were determined. IC had the highest fat content, followed by LT, RA, LL, PM, GM, SS, SF, TB, and SM. High-fat muscles such as LT, LL, IC, RA, and PM had significantly (p<0.05) different fat contents among QGs. Collagen contents were significantly (p<0.05) different among QGs. With decreasing QG, increasing collagen content was found in muscles. There were significant (p<0.05) differences in sarcomere length among QGs of several muscles. However, no significant (p>0.05) difference in sarcomere length was found among QGs for LL, PM, or RA muscle. PM had the lowest WBSF, followed by LL, LT, RA, IC, GM, SM, SF, SS, and TB. WBSF of QG $1^{{+}{+}}$ was lower than that of QG 1 for SS, TB, and SM. All muscles of QG 1 showed lower WBSF than QG 3 except TB or IC. Results of this study suggested that differences in WBSF among these 10 muscles by QG were due to differences in collagen content and sarcomere length.

Nutritional Characteristics of Water Extract from Kangaroo Tail. (캥거루 꼬리의 영양학적 특성)

  • To get the nutritional data of hot water extract from imported kangaroo tails, research was done about contents of collagen, chondroitin sulfate, protein, fat, mineral ions, and conjugated linoleic acid (CLA) in tails. Collagen content of sulfuric acid digested sample was way higher at bones than meats in both kangaroo tail and cow tail. Comparing kangaroo tail and cow tail, meat of kangaroo tail have 1.7 times higher collagen content than that of cow tail. Content of collagen in bone parts of kangaroo tail was also higher 1.2 times than that of cow tail. Meat sample of kangaroo tail (liquid extraction) have 1.3 times higher content of muco-polysaccharide than that of cow tail. In the born part, kangaroo tail was 2.4 times higher than cow tail in its content of muco-polysaccharide. The CLA content of the kangaroo tail showed the content that was higher than 0.9% of the cow tail for 4.9% and showed about 5.3 times high ratios. Especially in kangaroo tail, a band with high content of CLA was found between C18:1 and C18:2.

Collagen and Texture Properties of Commonly Consumed Fish Species in Korea as Sliced Raw Fishes (국내 다소비 횟감의 콜라겐과 조직감 특성)

  • This study investigated the collagen and texture properties of commonly consumed sliced raw fish species (CC-SRF) [olive flounder (OF), red seabream (RS), Atlantic salmon (AS), coho salmon (CoS) and sockeye salmon (SS)] distributed in Korea as sliced raw fishes. The crude lipid contents of CC-SRF were 5.5% for OF, 6.8% for RS, 18.5% for AS, 16.1% for CoS, and 5.7% for SS. The collagen content and solubility from CC-SRF were 622 mg/100 g and 78.0%, respectively, in OF, 270 mg/100 g and 75.6%, respectively, in RS, 237 mg/100 g and 24.1%, respectively, in AS, 341 mg/100 g and 65.7%, respectively, in CoS, and 246 mg/100 g and 17.9%, respectively, in SS. The texture of CC-SRF was affected by the lipid content, collagen content, acid solubility, hydroxylation, and cross linkage degree. The highest hardness of CC-SRF was obtained from OF, followed by RS, SS, AS and CoS. There was, however, no difference (P>0.05) in hardness between OF and RS and between AS and CoS.

Quality Evaluation of Red Seabream, Pagrus major by Physicochemical Method (물리 화학적 방법에 의한 참돔, Pagrus major의 품질판정)

  • The quality of cultured fish was evaluated in this study by using physicochemical method. The speed of a current was taken a decision 0.5 m/s by a preliminary experiment. No-exercise trained group was not much significantly changed moisture content, lipid content, collagen content and breaking strength during cultured time (P>0.05). But moisture content of exercise trained group was 72.05$\pm$1.45% at the first time and increased to 3% after 20 days. Lipid content was initially 5.36$\pm$0.90% and reduced to 46% during the cultured time. The breaking strength increased during cultured time, on the contrary collagen content did not change significantly. Relation between each elements during the cultured time, moisture content and lipid content was significantly correlated with breaking strength (P<0.05, r=0.60 and P<0.05, 0.56). But relation between breaking strength and collagen content was not clear (P>0.05, r=0.16). These results were suggested that the increase of breaking strength after exercise was affected by lipid content not collagen content.
