Chlorella protothecoides의 밀킹 전후 연속 배양 시스템을 통한 유용물질 분석 (Analysis of High-Value Materials through Continuous Cultivation System from Pre-and Post-Milking of Chlorella protothecoides)

  • Chlorella are source of valuable compounds as lipids, proteins, carbohydrates, bioactive compounds. To continuous obtain the high-value materials of Chlorella protothecoides, we performed continuous cultivation after applying milking techniques to C. protothecoides grown with culture for 7 days in optical panel bioreactor (OPBR) system. Fatty acid and lutein in extracts from pre- and post-milking of C. protothecoides were analyzed using gas chromatography and high performance liquid chromatography, respectively. C. protothecoides were rich in unsaturated fatty acids with a high content of oleic acid(C18:1), which is suitable as a biofuel feedstock. The fatty acid content in pre- and post-milking of C. protothecoides was decreased from 126.424mg/g d.w. to 119.341mg/g d.w, and the lutein content decreased from 0.258mg/g d.w. to 0.178mg/g d.w. The results of this study demonstrate the feasibility of milking C. protothecoides for production of lipids for biofuels production. It was confirmed that microalgae can continuously obtain lutein present in a trace amount through a continuous culture from milking.

제주지역 문화행정과 예술경영의 영역설정과 체계정립에 관한 연구 (A Study on the Evaluation of Equality and the Direction of Location Selection for the Concert Hall in Jeju)

  • 본 연구의 목적은 문화행정과 예술경영 두 분야의 영역이 중복 적용되는 현장에서 그 추구하는 가치와 역할체계의 정립을 전문가조사를 통하여 시도하고, 공공부문과 민간경영부문에서 두 영역이 맥락의 차이를 이해하게 하며 예술의 발전에 기여할 방향을 제안하는 것에 있다. 두 분야의 공통점과 차이점, 선행연구 고찰을 통하여 7개의 가치를 선정하고 각 가치의 하위지표를 정하였다. 이를 제주지역 전문가를 대상을 설문조사를 하여 AHP 기법을 이용하여 우선순위를 분석 하였다. 분석결과를 바탕으로 가치 간 우선순위로 영역의 범위를 설정하고, 하위지표 간 우선순위로 체계를 정립하였다. 문화행정은 공공성, 형평성, 투명성의 영역을 기본으로 하며 예술경영은 생산성, 공공성, 대응성을 기본영역으로 한다. 이를 바탕으로 문화행정은 지역문화의 보존과 활용, 문화의 공진화, 문화예술교육의 확대에 대한 체계에 집중할 필요가 있으며, 예술경영은 목표달성의 정도, 투입대비 산출의 정도, 지역문화의 보존과 활용에 대한 체계에 집중할 필요가 있다. 또한 제주지역에서는 문화의 공진화와 지역문화의 보존과 활용에 대한 공통적인 지향점을 갖고 체계를 정립할 필요성이 있음을 알 수 있었다.

살수방식에 따른 재배용기내 Gas 조성 및 콩나물의 생육 변화 (Growth of Soybean Sprouts and Concentration of $CO_2$ Produced in Culture Vessel Affected by Watering Methods)

  • 콩나물 재배에서의 살수방법과 그에 따른 콩나물 생육형태와 수율변화를 비교해본 결과 다음과 같다. 1. 살수방식에 따른 콩나물의 생육에 큰 차이를 나타내었으며, 왕복살수형, 용기담수형, 고정살수형 중 고정살수형 방식(재배통 상부에 하부타공된 고정형 물통설치에 의해 샤워형태로 살수)은 살수시 일시에 물을 샤워형식으로 쏟아내려 재배통 내부의 품온저하($22^{\circ}$ 유지), 내부 gas 배출, 유기물 제거 등의 효과가 탁월하여 콩나물 생육에 있어 몸통길이 13.3 cm로 다른 형식(15.2 cm, 15.9 cm)에 비해 짧았으나, 몸통두께(2.19 mm), 수율(535%)은 높게 나타나, 외관이 양호하고 부패가 억제되어 선도가 가장 잘 유지되었으며, 또한, 콩나물 재배시 필요한 용수량의 경우는 1회 살수량이 250∼300 L/통(원료콩 투입량 50 kg/통 기준)가 가장 양호한 것으로 나타났다. 2. 고정살수형 방식은 내부 gas($\textrm{CO}_2$ 5% 이하)배출이 탁월하여 재배완료 후 콩나물의 색택향상 및 부분부패 방지효과 등을 나타내는 장점이 있었으며, 재배 6일차에 재배통 하부(바닥깔판으로부터 약 30 cm이내)의 $\textrm{CO}_2$ 농도는 4-6%, $\textrm{O}_2$ 농도는 13-14%이었다. 따라서 재배시 재배통내 적정 gas조건은 농도 10% 이상, $\textrm{CO}_2$ 농도 5% 이하로 유지되어야 할 것으로 판단되었다. 3. 살수방식에 따른 재배통내의 품온 변화에서도 고정살수형 방식(4일차, $20^{\circ}$)이 용기담수형 방식(4일차, $22^{\circ}$)보다 품온 억제가 양호하여 콩나물의 생육향상 및 부패 등을 방지하므로 건강한 콩나물 생산의 중요한 요인임을 알 수 있었다.되지 않은 부분에 대해서 안전성 확보를 위하여 더 자세한 연구가 필요하다..6%이었다.program should be prepared through the long-period of monitoring and experiment.   • 정보통신 기술이 빠르게 발전하면서 사회와 문화 전반에 많은 영향을 미치고 있음에도 전통문화에 대한 체계적인 분류 및 최신 검색 기능을 제공하는 시스템은 아직까지 제공되고 있지 않다. 따라서 본 논문은 전통문화 콘텐츠 분류체계를 수립하여 3D 연표를 자동으로 생성하고 시각화함으로써 사용자에게 체계적인 분류 및 검색 기능을 제공함과 동시에 융복합서비스가 가능한 전통문화 검색 시스템을 개발하였다. 본 시스템은 전통문화 콘텐츠를 수집하는 기능, 수집된 전통문화 콘텐츠를 분류 및 저장하는 기능, 저장된 전통문화 콘텐츠에 기반을 두어 3D 디지털 연표를 자동으로 생성하는 기능을 제공한다. 또한, 시스템의 사용성 평가를 위해 시스템 만족도 설문지를 개발하였고 대학생 19명이 참여하여 시스템 사용성을 검증하였다. 실험 결과, 시스템 만족도에서는 모든 문항이 평균적으로 '만족한다'는 결과를 보였다.

  • The acute toxicity effect of triorganotin of trioganotin on the growth of microalgae and shellfish was investigated through flask culture. The value of 120 hr-LC$_{50}$ that is the median lethal concentration of TBTO on the shellfish (R. philipinarum) was found to be 6 $\mu$g/L. The acute toxicity effect of TBTO on T. suecica was obviously shown even at the concentration of 0.5 $\mu$g/L, and the effect diminished as the initial cell density increased. The effect also diminished less in the experiment done under aeration than in that done under non-aeration. To design a chemostat system for the test of chronic toxicity, the culture of T. suecica was executed in photobioreactor. In batch culture, the profiles of chlorophyII a and D.C.W. showed the growth of T. suecica very well, and the maximum specific growth rate was estimated to be 0.54 d$^{-1}$. with this value, as a dilution rate in contimuous culture, pH was nicely maintained between 7 and 9 when air was supplied with 3% CO$_{2}$. From all results and the natural environment of clam, a novel chemostat system was invented. Through this system, we can observe each independent toxicity effect of TBTO and plankton and combined toxicity effect as well.

Primary Culture of Endothelial Cells from Murine Brain Microvessels

  • It is important to coordinated interaction among neurons, astrocytes and endothelial cells to maintain the function of brain. To study their regulatory mechanisms in vitro system, the co-culture system among the isolated cells from brain may be needed. However, the method for purifying brain microvascular endothelial cells (BMEC) far culture have not established yet. In this study, the proper culture methods of mice cells using two different strains, CD1 and C57BL6, to obtain the pure and plentiful endothelial cells were described. The flatted-round forms of CD1 endothelial cells grew on the collagen-IV coating plates, while the purified cells from C57 mice preferred type collagen-I dishes for their growth. Both cells displayed anti-PECAM-1 (CD31) and von Willebrand Factor immune-reactivity. These results indicated that different coating materials not only improve attachment of isolated cells but also promoting growth of cells, suggesting that this method of purifying murine Brain microvascular endothelial cells (BMEC) provides a suitable model to investigate blood-brain-barrier (BBB) properties within neurovascular unit in vitro.

  • Objectives: To investigate the role of TGF (Transforming growth factor-$\beta$) involved in the paracrinic communication during decidualization between UEC (uterine epithelial cells) and USC (uterine stromal cells), we have employed a co-culture system composed of human endometrial epithelial and stromal cells in defined hormonal conditions. Design: In the co-culture, endometrial epithelial cells cultured in the matrigel-coated cell culture insert are seeded on top of the endometrial stromal cells cultured within a collagen gel. The co-culture was maintained for 48 hours under the following hormonal conditions: progesterone dominant condition (100 nM P4 and 1 nM E2) or estrogen-dominant condition (100 nM E2 and 1 nM P4). 10 ng/ ml HGF and/or 10 ng/ml TGF-$\beta$1 are added. Methods: RT-PCR is utilized to detect mRNAs quantitatively. Enzyme-linked immunosorbent assay (ELISA) and immunohistochemical staining are utilized to detect proteins in the tissue. Results: Prolactin mRNA is expressed in the co-cultured stromal cells under the progesterone dominant condition. TGF-$\beta$1 and its receptors are expressed in both the co-cultured epithelial and stromal cells irrespective of the steroid present, which is in contrast with no or negligible expression of TGF-$\beta$1 or its receptor in cells separately cultured. Both estrogen and progesterone significantly elevate the concentration of hepatocyte growth factor (HGF) in the conditioned medium of the co-culture with the value of 4, 325 pg/ml in E2-dominant and 2, 000 pg/ml in P4-dominant condition compare to 150 pg/ml in no hormone. In separately cultured stromal cells, administration of HGF induces the expression of TGF receptor 1 in both hormonal conditions, but induction of TGF receptor 2 is only manifest in the P4-dominant condition. Administration of TGF-$\beta$ and HGF directly induce the decidualization marker prolactin mRNA in separately cultured stromal cells. Conclusion: It is likely that steroid hormones induces prolactin mRNA indirectly by promoting the cell to cell communication between the stromal and the epithelial cells. TGF-$\beta$ and HGF are two possible paracrine mediators in the human endometrial decidualization.

  • The objective of this study was to produce calves derived from in vitro fertilization of in vitro matured follicular oocytes. Oocytes aspirated from small antral folicles of ovaries obtained at a local slaughter house were matured and fertilized in vitro. At l8hrs after insemination with Korean native cattle semen, oocytes were co-cultured for 6~7 days by utilizing co-culture system with bovine oviduct epithelial cell. After co-culture, good or excellent quality late morulae or early blastocysts were selected by morphological criteria under stereo microscope. Selected embryos were transferred to recipients on day 6 or 7 (estrus = day 0). Recipients were monitored by observation for estrus and rectal palpation after 60 days from embryo transfer. One of them went to term with the birth of a calf. This case is the first production of calf derived from in vitro fertilization in Korea.

Enhancement of BDNF Production by Co-cultivation of Human Neuroblastoma and Fibroblast Cells

  • It has been proved that co-cultivation of human neroblastoma cells and human fibroblast cells can enhance nerve cell growth and the production of BDNF in perfusion cultivation. In batch co-cultivation, maximum cell density was increased up to 1.76${\times}$106 viable cells/mL from 9${\times}$105 viable cells/mL of only neuroblastoma cell culture. The growth of neuroblastoma cells was greatly improved by culturing both nerve and fibroblast cells in a perfusion process, maintaining 1.5${\times}$106 viable cells/mL, which was much higher than that form fed-batch cultivation. The nerve cell growth was greatly enhance in both fed-batch and perfusion cultivations while the growth of fibroblast cells was not. It strongly implies that the factors secreted from human fibrobast cells and/or the environments of co-culture system can enhance both cell growth and BDNF secretion. Specific BDNF production rate was not enhanced in co-cultures; however, the production period was increased as the cell growth was lengthened in the co-culture case. Competitive growth between nerve cells and fibroblast cells was not observed in all cases, showing no changes of fibroblast cell growth and only enhancement of the neuroblastoma cell growth and overall BDNF production. It was also found that the perfusion cultivation was the most appropriate process for cultivating two cell lines simultaneously in a bioreactor.

Biological Fixation of $CO_2$ by Chlorella sp. HA-1 in a Semi-Continuous and Series Reactor System

  • Characteristics of biological $CO_2$ fixation by Chlorella sp. HA-1 were investigated in a semi-continuous and series reactor system using an internally illuminated photobioreactor to overcome shortcomings of physicochemical technologies such as adsorption and membrane separation. High $CO_2$ fixation rate was achieved in the semi-continuous reactor system, in which the dilution ratios of the culture medium were controlled. The average $CO_2$ fixation rate was maintained almost constantly when the dilution ratio increased by 0.1 increment from the initial value of 0.5. The total removal efficiency of $CO_2$ was enhanced by employing a series reactor system. The average $CO_2$ fixation rate increased until 4.013 g $CO_2\;day^{-1}$ in a series operation of four reactors, compared to 0.986 g $CO_2\;day^{-1}$ in a batch operation mode. The total $CO_2$ fixation rate was proportional to the number of reactors used in the series reactor system. In the series reactor system of semi-continuous operation, a large amount of $CO_2$ was removed continuously for 30 days. These results showed that the present reactor systems are efficient and economically feasible for a biological $CO_2$ fixation.
