Design of In-situ Self-diagnosable Smart Controller for Integrated Algae Monitoring System

  • The rapid growth of algae occurs can induce the algae bloom when nutrients are supplied from anthropogenic sources such as fertilizer, animal waste or sewage in runoff the water currents or upwelling naturally. The algae blooms creates the human health problem in the environment as well as in the water resource managers including hypoxic dead zones and harmful toxins and pose challenges to water treatment systems. The algal blooms in the source water in water treatment systems affects the drinking water taste & odor while clogging or damaging filtration systems and putting a strain on the systems designed to remove algal toxins from the source water. This paper propose the emerging In-Situ self-diagnosable smart algae sensing device with wireless connectivity for smart remote monitoring and control. In this research, we developed the In-Site Algae diagnosable sensing device with wireless sensor network (WSN) connectivity with Optical Biological Sensor and environmental sensor to monitor the water treatment systems. The proposed system emulated in real-time on the water treatment plant and functional evaluation parameters are presented as part of the conceptual proof to the proposed research.

  • Human hepatocytes, with complete hepatic metabolizing enzymes, transporters and cofactors, represent the gold standard for in vitro evaluation of drug metabolism, drug-drug interactions, and hepatotoxicity. Successful cryopreservation of human hepatocytes enables this experimental system to be used routinely. The use of human hepatocytes to evaluate two major adverse drug properties: drug-drug interactions and hepatotoxicity, are summarized in this review. The application of human hepatocytes in metabolism-based drug-drug interaction includes metabolite profiling, pathway identification, P450 inhibition, P450 induction, and uptake and efflux transporter inhibition. The application of human hepatocytes in toxicity evaluation includes in vitro hepatotoxicity and metabolism-based drug toxicity determination. A novel system, the Integrated Discrete Multiple Organ Co-culture (IdMOC) which allows the evaluation of nonhepatic toxicity in the presence of hepatic metabolism, is described.

Effects of Cell Status of Bovine Oviduct Epithelial Cell (BOEC) on the Development of Bovine IVM/IVF Embryos and Gene Expression in the BOEC Used or Not Used for the Embryo Culture

  • The objective of this study was to investigate the effects of cell status of BOEC on development of bovine IVM/IVF embryos and gene expression in BOEC before or after culturing of embryos. The developmental rates beyond morula stage in the BOEC co-culture group was significantly higher than in the control group (p<0.05). In particular, blastocyst production in the BOEC co-culture group (28.3%) was dramatically increased compared with the control group (7.2%). In the in vitro development of bovine IVM/IVF embryos according to cell status, the developmental rates beyond morula stage in the primary culture cell (PCC) co-culture group were the highest of all experimental groups. Expression of genes related to growth (TGF-${\beta}$ EGF and IGFBP), apoptosis (Bax, Caspase-3 and p53) and antioxidation (CuZnSOD, MnSOD, Catalase and GPx) in different status cells of BOEC for embryo culture was detected by RT-PCR. While EGF gene was detected in isolated fresh cells (IFC) and PCC, TGF-${\beta}$ and IGFBP were found in IFC or PCC after use in the embryo culture, respectively. Caspase-3 and Bax genes were detected in all experimental groups regardless of whether the BOEC was used or not used in the embryo culture. However, p53 gene was found in IFC of both conditions for embryo culture and in frozen/thawed culture cells (FPCC) after use in the embryo culture. Although antioxidant genes examined were detected in all experimental groups before using for the embryo culture, these genes were not detected after use. This study indicated that the BOEC co-culture system used for in vitro culture of bovine IVF embryos can increase the developmental rates, and cell generations and status of BOEC might affect the in vitro development of bovine embryos. The BOEC monolayer used in the embryo culture did not express the growth factors (TGF-${\beta}$ and EGF) and enzymatic antioxidant genes, thereby improving embryo development in vitro.

  • This experiment was investigated the effect of presence of granulosa cells from follicles of different size on bovine oocyte maturation, cleavage and development to late stage. The nuclear and cytoplasmic maturation of oocytes in the IVM-IVF system are critical for subsequent embryo development. Granulosa cells when the co-cultured with oocytes may interact with cumulus-oocytes complexes and influence the development competence of the oocytes. Granulosa cells from medium (2~6 mm) and large(>1O mm) size follicles were recovered by aspiration, washed 3 times by centrifugation at 500 x g for 5 min. and used for co-culture at a concentration of 2~3 x 106 cells/mi. The oocytes were matured in vitro (IVM) for 24 hrs. in TCM-199 supplemented with 35 $\mu$g/ml FSH, 10 $\mu$g/ml LH, 1 $\mu$g/ml estradiol-17$\beta$ and granulosa cells at 39$^{\circ}C$ under 5% $CO_2$ in air. They were fertilized in vitro (IVF) by epididymal spermatozoa treated with heparin for 24 hrs., and then the zygotes were co-cultured in vitro (I VC) with bovine oviductal epithelial cells for 7 to 9 days.   • Bovine embryos at 2-to 8-cell produced by in vitro matured/in vitro fertilized(IVM/IVF) were cultured in TCM 199 or Synthetic oviduct fluid(SOF) with 10% fetal bovine serum(FBS) or cocultured with cumulus or bovine oviduct epithelial cell(BOEC) in TCM-199 or SOF medium. In experiment 1, the proportions of embryos developed to morula and blastocysts stages in TCM 199 medium were higher when they were co-cultured with cumulus cell(29%) or BOEC(33%) than that of TCM 199 with 10% FBS(12%, P<0.01). In experiment 2, embryos deived from IVM/IVF were cultured in SOF with 10% FBS or cocultured with cumulus cell or BOEC in same medium. The higher development rates of IVM/IVF embryos developed beyond morula stages were obtained in cumulus cell co-culture group(39%) than those of BOEC group(26%) and SOF with 10% FBS group(17%, P<0.01). The present results indicated that the early development of IVM/IVF embryos can be maintained efficienty in SOF with cumulus cell co-culture.

  • The Realization of Ubiquitous is achieved by magic mirror and it is required more concrete study to realize it's functionality. Especially its function of professional fashion co-ordinator for managing the appearance could be of further use. The Objective of this Study is to establish RFID chip data base to put into a computer for making use of the functionality of the magic mirror aiming at suggesting the available information on the total fashion co-ordination. I sought firstly the code with binary system determining the criteria of accessories to be input in a RFID chip. Secondly, as the image with cloth is an important element for the total fashion co-ordination, desired co-ordination among the emphasis, harmony, character, season and accent can be made selectable classifying into a limit element and a common element to extract the codes. Thirdly, necessary conditions were given to the generated codes using Visual C++ program of Microsoft and the extracted codes as per groups were compared and analyzed.

  • Despite the rapid development of assisted reproductive technologies (ART) in recent years, implantation rates after replacement of embryos into the uterine cavity remains low. Several techniques such as culture conditions based on formulations of human tubal fluid and various ART techniques as GIFT, ZIFT, TET have been adopted in recent years to improve embryo viability in vitro and implantation rates. Also, coculture of human IVF-derived embryos have been used in an effort to increase the number of viable embryos following IVF and to improve synchrony between the developing embryo and the uterine environment. The aim of this study was to evaluate whether the use of co culture with autologous cumulus cells has a significant beneficial effect on the development of embryos in vitro and its relation to the pregnancy rates in 120 patients with previous failed IVF-ET from September, 1995 to January 1998. We obtained the results from which significant improvement in the quality of viable embryos were observed using a coculture system with autologous cumulus cells, but pregnancy rates in this group of patients did not differ from the rate in the standard IVF group during the same period. Our study shows that a simplified short-term coculture system with autologous cumulus cells may help rescue moderate quality embryos to cleave regularly.

Effects of Protein Sources and Co-culture on In Vitro Culture of IVF-derived Porcine Embryos (단백질 공급원 및 체세포와의 공배양이 돼지 체외수정란의 체외발달에 미치는 영향)

  • This study was conducted to investigate whether various protein sources and co-culture affect in vitro development of porcine zygotes derived from In vitro maturation/fertilization (IVM/IVF). These results obtained in these experiments are summarized as follows 1. When porcine oocytes matured and fertilized In vitro were cultured in NCSU 23 medium supplemented with various BSA concentrations (0.4, 0.8 and 3.2%), In vitro developmental rates of porcine zygotes to blastocyst stage were 22.9, 18.4 and 14.6%, respectively. High concentration of BSA (3.2%) showed a smaller nuclei number (36.1$\pm$11.8) of blastocysts than 0.4 and 0.8% BSA groups (53.2$\pm$27.4 and 61.2$\pm$22.5, respectively) (P<0.05). This result indicates that high concentration of BSA is detrimental on preimplantation development of IVF-derived porcine embryos. 2. No differences were detected in the developmental rate and mean nuclei number of porcine embryos between 10 and 20% FBS concentrations in culture medium. 3. IVF-derived porcine embryos co-cultured with mouse or porcine embryonic fibroblast cells showed a lower development to the blastocyst stage than those without co-culture system. Consequently, the present study suggests that high concentration of BSA as a protein source in culture medium suppresses development potential of porcine embryos produced In vitro. In addition, co-culture with somatic cells is not effective on in vitro development of IVF-derived porcine embryos to blastocyst stage.

  • Chlorella are source of valuable compounds as lipids, proteins, carbohydrates, bioactive compounds. To continuous obtain the high-value materials of Chlorella protothecoides, we performed continuous cultivation after applying milking techniques to C. protothecoides grown with culture for 7 days in optical panel bioreactor (OPBR) system. Fatty acid and lutein in extracts from pre- and post-milking of C. protothecoides were analyzed using gas chromatography and high performance liquid chromatography, respectively. C. protothecoides were rich in unsaturated fatty acids with a high content of oleic acid(C18:1), which is suitable as a biofuel feedstock. The fatty acid content in pre- and post-milking of C. protothecoides was decreased from 126.424mg/g d.w. to 119.341mg/g d.w, and the lutein content decreased from 0.258mg/g d.w. to 0.178mg/g d.w. The results of this study demonstrate the feasibility of milking C. protothecoides for production of lipids for biofuels production. It was confirmed that microalgae can continuously obtain lutein present in a trace amount through a continuous culture from milking.

  • The purpose of this study is to examine the concepts of cultural administration and art management, their context, and elements that each concept pursues, and to establish the domains and systems of cultural administration and art management in Jeju. Seven values were selected through the consideration of commonalities and differences and previous studies, and sub-indicators of each value were determined. Factors were analyzed using the AHP method. Cultural administration is based on the areas of publicity, equity, and transparency, while art management is based on productivity, publicity, and responsiveness. Cultural administration needs to focus on a system for the preservation and utilization of local culture, co-evolution of culture, and expansion of culture and arts education. Art management needs to focus on the level of achievement of goals, the level of output versus input, and the system for preserving and utilizing local culture. In addition, it was found that there is a need to establish a system in Jeju with a common aim of the co-evolution of culture and preservation and the utilization of local culture.
