한강에서 분리한 장구균의 항생제 내성과 분자생물학적 분석 (Antibiotics Resistance and Molecular Analysis of Enterococcus Isolated from the Han-river in Korea)

  • 2008년 3월, 서울의 상수원으로 이용되는 한강에서 분리된 Enterococcus 76주를 동정하고 이 균주들에 대한 항생제 감수성과 항생제 내성유전자 구조분석, 교차접합, PFGE 및 MLST를 실시하였다. 분리 균주 중 E. casseliflavus는 25주이었으며, E. faecalis 및 E. hirae가 각각 4주 및 1주이었다. 항생제 감수성을 조사한 결과 15주가 vancomycin에 대해 내성을 나타내었으며 E. faecium 11주와 E. casseliflavus 4주가 VRE인 것을 알 수 있었다. VRE를 대상으로 vanA 유전자 검출을 통해 6주의 E. faecium가 vanA를 가지는 VREF임을 밝혔으며, vanA가 포함된 transposon인 Tn1546 구조를 분석한 결과 Km36과 Km37은 Tn1 type, Km20과 Km38은 Tn2 type 그리고 Km 39와 Km40은 Tn3 type으로 나타났다. PFGE 결과 6주의 VREF 중 Km36과 Km37은 동일한 subtype을 나타내었으며, 나머지 4주는 각기 다른 subtype을 나타냈다. VREF 6주를 대상으로 MLST를 실시한 결과 3주는 ST78, 나머지 3주는 각각 ST18, ST192 및 ST230로 나타났다. 이들의 clonal complex는 모두 병원에서 분리되는 CC17로부터 파생된 것으로 파악되었으며 4주는 CC78에, 나머지 2주는 각각 CC18과 CC192에 포함된 것을 확인할 수 있었다.

  • The aim of this study was to investigate the susceptibility of mutans streptococci (S. mutans and S. sobrinus) and Streptococcus anginosus, for seven antibiotics, penicillin G, amoxicillin, ciprofloxacin, cefuroxime, erythromycin, bacitracin, and vancomycin. The minimum inhibitory concentration (MIC) of seven antibiotics against 3 species (type strains) of mutans streptococci and S. anginosus, 10 strains (wild type) of S. mutans, 7 strains (wild type) of S. sobrinus, and 11 strains (wild type) of S. anginosus, were measured by broth dilution method. All of the type strains of mutans streptococci and S. anginosus had the same susceptibility for penicillin G, amoxicillin, cefuroxime and bacitracin. Type strain of S. anginosus was sensitive in ciprofloxacin, but those of mutans streptococci were not. All of the clinical isolates of mutans streptococci and S. anginosus had the same susceptibility for the seven antibiotics. Our data reveal that mutans streptococci and S. anginosus have similar antibiotic-resistant character. In addition. these results may offer the basic data to verify the antibiotic-resistant mechanism of mutans streptococci and S. anginosus.

Neisseria gonorrhoeae분리균주의 특성 (Characterization of Isolated strains of Neisseria gonorrhoea)

  • N. gonorrhoeae 81균주를 320개의 가검물에서 분리동정한 후에 증식에 대한 VCN의 영향과 lsovitalex의 양에 따른 영향, 당분해능에 관한 실험과 ${\beta}-lactamase$생산균의 검출 및 plasmid분리에 대하여 연구하였다. 전체 320개의 가검물중에서 81균주의 N.gonorrhoeae가 분리동정되었고, chocolate agar에서는 81균주 중 72균주가,1% Isovitalex와 1 % VCN을 함유한 Thayer- Martin배지에서는 80균주가, 2 % Isovitalex와 1 % VCN이 든 Thayer-Martin 배지는 55균주가 N. gonorrhoeae의 특성을 나타냈다. Coagglutination시험에 양성인 81균주 중에서 67균주는 48시간 배양후에,10균주는 72시간 배양후에 glucose 배지에서 산을 생산하였으나, 나머지 4균주는 96시간 배양후에도 산을 생산하지 않았다. 인천지역 특수직업여성에서 분리된 81균주의 N.gonorrhoeae 중에서 41 균주가 ${\beta}-lactamase$를 생산하였고, ${\beta}-lactamase$ 생산 균주인 N. gonorrhoeae PL-118은 2.6. 4.5 및 24.5Mdal의 plasmid DNA를 포함하였다.

Genotypes of Clinical and Environmental Isolates of Cryptococcus neoformans and Cryptococcus gattii in Korea

  • Multilocus sequence typing analysis was applied to determine the genotypes of 147 (137 clinical and 10 environmental) Cryptococcus neoformans and three clinical Cryptococcus gattii isolates from 1993 to 2014 in Korea. Among the 137 clinical isolates of C. neoformans, the most prevalent genotype was ST5 (n = 131), followed by ST31 (n = 5) and ST127 (n = 1). Three C. gattii strains were identified as ST57, ST7, and ST113. All environmental isolates were identified as C. neoformans with two genotypes, ST5 (n = 7) and ST31 (n = 3). Our results show that C. neoformans isolates in Korea are genetically homogeneous, and represent a close genetic relationship between clinical and environmental isolates.

Effect of Gene actA on the Invasion Efficiency of Listeria monocytogenes, as Observed in Healthy and Senescent Intestinal Epithelial Cells

  • Listeria monocytogenes can asymptomatically inhabit the human intestine as a commensal bacterium. However, the mechanism by which L. monocytogenes is able to inhabit the intestine without pathogenic symptoms remains unclear. We compared the invasion efficiency of L. monocytogenes strains with the 268- and 385-bp-long actA gene. Clinical strains SMFM-CI-3 and SMFM-CI-6 with 268-bp actA isolated from patients with listeriosis, and strains SMFM-SI-1 and SMFM-SI-2 with the 385-bp gene isolated from carcasses, were used for inoculum preparation. The invasion efficiency of these strains was evaluated using Caco-2 cells (intestinal epithelial cell line), prepared as normal and healthy cells with tightened tight junctions and senescent cells with loose tight junctions that were loosened by adriamycin treatment. The invasion efficiency of L. monocytogenes strains with the 268-bp-long actA gene was 1.1-2.6-times lower than that of the strains with the 385-bp-long gene in normal and healthy cells. However, the invasion efficiency of both types of strains did not differ in senescent cells. Thus, L. monocytogenes strains with the 268-bp-long actA gene can inhabit the intestine asymptomatically as a commensal bacterium, but they may invade the intestinal epithelial cells and cause listeriosis in senescent cells.

대구지방 소아 및 토양에서 분리한 Microsporum gypseum의 교배형에 대한 연구 (Study on Mating Type of Microsporum gypseum Isolated from Children Patients and Soils in Taegu Area)

  • Microsporum gypseum complex의 교배형 및 감염원을 조사하기 위하여 대구에 거주하는 소아에서 분리한 M.gypseum 17주와 환자주변의 초등학교 운동장, 화단 및 어린이 놀이터의 토양에서 분리한 60주를 표준균주 A.gypsea 및 A.incurvata와 교배 실험을 실시하였다. 소아에서 분리된 M.gypseum 17주의 성별 완전형의 분포는 남아에서 A.incurvata가 14주 이었고, A.gypsea는 3주이었다. 여아에서는A.incurvata만 4주 분리되었다. 병형별 분리빈도는 Tinea facial가 A.gypsea 2주, A.incurvata 10주로 다른 부위보다 많이 분리되었다. 교배형의 분포는 환자에서 분리한 17주중 A.gypsea는 3주중 "+"형 2주, "-"형이 1주이었고, A.incuwata는 "+"형이 6주, "-"형이 8주이었다. 토양에서 분리한 60주의 교배형의 분포는 A.gypsea는 31주중 "+"형 16주, "-"형이 15주이었고, A.incurvata는 "+"형이 15주, "-"형이 14주이었고 특히 소아에서 많이 분리된 A.incurvata가 어린이 놀이터에서 많이 분리된 것을 보면 소아의 M.gypsen 감염은 놀이터와 관련성이 있을 것으로 사료된다. 분리된 A.incurvata가 어린이 놀이터에서 많이 분리된 것을 보면 소아의 M.gypsen 감염은 놀이터와 관련성이 있을 것으로 사료된다.

Comparison of Antibiotic Resistance of Blood Culture Strains and Saprophytic Isolates in the Presence of Biofilms, Formed by the Intercellular Adhesion (ica) Gene Cluster in Staphylococcus epidermidis

  • To elucidate the question of whether biofilm formed by the intercellular adhesion (ica) gene cluster has influences on antibiotic resistance in Staphylococcus epidermidis, we compared 124 skin strains with strains isolated from 50 blood cultures that cause septicemic diseases. The results revealed that the blood culture isolates were more resistant to the antibiotics tested than the saprophytic isolates. Moreover, antibiotic multiresistance was more prevalent in the clinical isolates. In the blood culture isolates, $46\%$ of the strains were resistant to three or more antibiotics, whereas only $12\%$ of the saprophytic isolates were resistant to three or more antibiotics. Interestingly, these characteristics were highly correlated with the biofilm formed by the ica gene cluster. In biofilm-producing strains, $84\%$ of the blood culture isolates and $44\%$ of the saprophytic isolates were antibiotic multiresistant, whereas only $22\%=;and\;9\%$, respectively, were antibiotic multiresistant in biofilm-nonproducing strains. Additionally, in the biofilm-producing ica-positive strains, $89\%$ of the blood culture isolates and $57\%$ of the saprophytic isolates were antibiotic multiresistant. However, the rate of the antibiotic multiresistance in the ica-negative strains was very low, thus indicating that the biofim formed by the lea gene cluster in S. epidermidis is an important pathogenic factor in association with the antibiotic multiresistance.

Molecular Classification of Commercial Spirulina Strains and Identification of Their Sulfolipid Biosynthesis Genes

  • Cyanobacterial strains of the genus Spirulina have recently been identified as an excellent source of sulfolipids, some of which possess anti-HIV properties. Thus, to investigate the distribution of sufolipid biosynthesis pathways in Spirulina, a genetic screening/phylogentic study was performed. Five different strains of Spirulina [Spirulina (Jiangmen), Spirulina sp., S. platensis, S. maxima, and Spirulina seawater] sourced from different locations were initially classified via 16S rDNA sequencing, and then screened for the presence of the sulfolipid biosynthesis genes sqdB and sqdX via a PCR. To assess the suitability of these strains for human consumption and safe therapeutic use, the strains were also screened for the presence of genes encoding nonribosomal peptide synthases (NRPSs) and polyketide synthases (PKSs), which are often associated with toxin pathways in cyanobacteria. The results of the 16S rDNA analysis and phylogenetic study indicated that Spirulina sp. is closely related to Halospirulina, whereas the other four Spirulina strains are closely related to Arthrospira. Homologs of sqdB and sqdX were identified in Spirulina (Jiangmen), Spirulina sp., S. platensis, and the Spirulina seawater. None of the Spirulina strains screened in this study tested positive for NRPS or PKS genes, suggesting that these strains do not produce NRP or PK toxins.

Characterization of Drug-Resistant Salmonella enterica Serotype Typhimurium by Antibiograms, Plasmids, Integrons, Resistance Genes, and PFGE

  • Forty-seven Salmonella Typhimurium (33 zoonotic, 14 clinical) strains were tested for antimicrobial resistance using the standard disk diffusion method. The presence of relevant resistance genes and class 1 integrons were investigated by using PCR. Pulsed-field gel electrophoresis (PFGE) and plasmid profiling were carried out to determine the genomic diversity of Salmonella Typhimurium. Approximately 57.4% of the S. Typhimurium strains were multidrug resistant (MDR) and showed high resistance rates to tetracycline (70.2%), sulfonamides (57.4%), streptomycin (53.1%), ampicillin (29.7%), nalidixic acid (27.6%), kanamycin (23.4%), chloramphenicol (21.2%), and trimethoprim (19.1%). Resistance towards cephalosporins was noted for cephalothin (27.6%), cephradine (21.2%), amoxicillin clavulanic acid (17.0%), and cephalexin (17.0%). Resistance genes, $bla_{TEM}$, strA, aadA, sul1, sul2, tetA, tetB, and tetC, were detected among the drug-resistant strains. Thirtythree strains (70.2%) carried class 1 integrons, which were grouped in 9 different profiles. DNA sequencing identified sat, aadA, pse-1, and dfrA genes in variable regions on class 1 integrons. Thirty-five strains (74.4%) were subtyped to 22 different plasmid profiles, each with 1-6 plasmids (2.0 to 95 kb). PFGE subtyped the 47 strains into 39 profiles. In conclusion, high rates of multidrug resistance were found among the Malaysian Salmonella Typhimurium strains. The emergence of multidrug-resistant Salmonella Typhimurium to cephalosporin antibiotics was also observed. The strains were very diverse and no persistent clone was observed. The emergence of MDR Salmonella Typhimurium is a worldwide problem, and this report provides information for the better understanding of the prevalence and epidemiology of MDR S. Typhimurium in Malaysia.

Screening and Characterization of Lactic Acid Bacteria Strains with Anti-inflammatory Activities through in vitro and Caenorhabditis elegans Model Testing

  • The present study was conducted to screen candidate probiotic strains for anti-inflammatory activity. Initially, a nitric oxide (NO) assay was used to test selected candidate probiotic strains for anti-inflammatory activity in cultures of the murine macrophage cell line, RAW 264.7. Then, the in vitro probiotic properties of the strains, including bile tolerance, acid resistance, and growth in skim milk media, were investigated. We also performed an in vitro hydrophobicity test and an intestinal adhesion assay using Caenorhabditis elegans as a surrogate in vivo model. From our screening, we obtained 4 probiotic candidate lactic acid bacteria (LAB) strains based on their anti-inflammatory activity in lipopolysaccharide (LPS)-stimulated RAW 264.7 cell cultures and the results of the in vitro and in vivo probiotic property assessments. Molecular characterization using 16S rDNA sequencing analysis identified the 4 LAB strains as Lactobacillus plantarum. The selected L. plantarum strains (CAU1054, CAU1055, CAU1064, and CAU1106) were found to possess desirable in vitro and in vivo probiotic properties, and these strains are good candidates for further investigations in animal models and human clinical studies to elucidate the mechanisms underlying their anti-inflammatory activities.
