Comparison of Parametric and Bootstrap Method in Bioequivalence Test

  • The estimation of 90% parametric confidence intervals (CIs) of mean AUC and Cmax ratios in bioequivalence (BE) tests are based upon the assumption that formulation effects in log-transformed data are normally distributed. To compare the parametric CIs with those obtained from nonparametric methods we performed repeated estimation of bootstrap-resampled datasets. The AUC and Cmax values from 3 archived datasets were used. BE tests on 1,000 resampled data sets from each archived dataset were performed using SAS (Enterprise Guide Ver.3). Bootstrap nonparametric 90% CIs of formulation effects were then compared with the parametric 90% CIs of the original datasets. The 90% CIs of formulation effects estimated from the 3 archived datasets were slightly different from nonparametric 90% CIs obtained from BE tests on resampled datasets. Histograms and density curves of formulation effects obtained from resampled datasets were similar to those of normal distribution. However, in 2 of 3 resampled log (AUC) datasets, the estimates of formulation effects did not follow the Gaussian distribution. Bias-corrected and accelerated (BCa) CIs, one of the nonparametric CIs of formulation effects, shifted outside the parametric 90% CIs of the archived datasets in these 2 non-normally distributed resampled log (AUC) datasets. Currently, the 80~125% rule based upon the parametric 90% CIs is widely accepted under the assumption of normally distributed formulation effects in log-transformed data. However, nonparametric CIs may be a better choice when data do not follow this assumption.

  • The Genus Cis Latreille known as fungivorous ciids is taxonomically reviewed for the first time from Korea. Six unrecorded species - Cis boleti (Scopoli, 1763); Cis seriatopilosus Motschulsky, 1861; Cis mikagensis Nobuchi & Wada, 1955; Cis jezoensis Nobuchi, 1960; Cis sasajii Kawanabe, 2001; Cis hieroglyphicus Reitter, 1877- are present. All of Cis members are associated with the fruiting body of higher fungi. The author provides the taxonomical information and the ecological information on the host fungi of the Korean Cis species.

Production of cis, cis-Muconic Acid from Benzoic Acid via Microbial Transformation

  • For the production of cis, cis-muconic acid via biocatalytic conversion reactions from a toxic cosubstrate, benzoic acid, a fed-batch process using computer-controlled DO-stat feeding was developed. The mutant strain of Pseudomonas putida BM014 produced cis, cis-muconic acid from benzoic acid with high conversion yield. More than 32 g/L of cis, cis-muconic acid was accumulated in 42h and a productivity of 1.4g/(L.h)was achieved.

Preparation of a Large Quantity of CIS-9, trans-11 and trans-10, cis-12 Conjugated Linoleic Acid(CLA) Isomers from SYnthetic CLA

  • Conjugated linoleic acid(CLA) refers to a collective term of positional and geometric isomers of linoleic acid, which are different in their biological activities. The predominant isomer of CLA in animal tissues is cis-9, trans-11; smaller amounts of trans-10, cis-12 CLA isomers, CLA methyl ester (CLA-ME) was chemically syn-thesized from linoleic acid by the alkaline isomerization method. The synthetic CLA-ME, mainly composed of cis-9, trans-11 CLA and trans-10, cis-12 CLA, was dissolved in acetone, stored at 68$^{\circ}C$ for 1 day, and the supernatant(cis-9, trans-11 CLA-Me) was separated from the precipitate (trans-10, cis-12 CLA-Me). After the processes were repeated three times at -68$^{\circ}C$, the whole processes were repeated three times at -71$^{\circ}C$ in order to increase the purity of these two isomers. The cis-9, trans-11 CLA-Me and trans-10, cis-12 CLA isomers were further purified by the urea adduct. Purities of the cis-9, trans-11 CLA-Me and trans-10, cis-12 CLA-Me were 90.3 and 99.9%, respec-tively. This method could be employed for the preparation of a large quantity of highly purified cis-9, trans-11 CLA-Me or trans-10, cis-12 CLA-Me from synthetic CLA-Me.

Enhancement of cis,cis-Muconate Productivity by Overexpression of Catechol 1,2-Dioxygenase in Pseudomonas putida BCM114

  • For enhancement of cis,cis-muconate productivity from benzoate, catechol 1,2-dioxygenase (C12O) which catalyzes the rate-limiting step (catechol conversion to cis,cis-muconate) was cloned and expressed in recombinant Pseudomonas putida BCM114. At higher benzoate concentrations (more than 15 mM), cis,cis-muconate productivity gradually decreased and unconverted catechol was accumulated up to 10 mM in the cae of wild-type P. putida BM014, whereas cis,cis-muconate productivity continuously increased and catechol was completely transformed to cis,cis-muconate for P. putida BCM114. Specific C12O activity of P. putida BCM114 was about three times higher than that of P. putida BM014, and productivity was enhanced more than two times.

  • This study was conducted to determine the inheritance of duvatrienediol(DVTs) and cis-abienol concentration from flesh tobacco leaves. NC744, which has a very low levels of DVTs and no cis-abienol was used as a parent in cross to TI 1068 producing a normal amount of DVTs and cis-abienol. Presence of DVTs and cis-abienol on the leaf surface was determined using thin layer chromatography(TLC). Segregation pattern from F$_2$ and BC$_2$[ (TI 1068 $\times$ NC744) $\times$ NC744J generations revealed that TI 1068 have a single dominant gene controlling DVTs and cis-abienol production. And DVTs production was inherited independently of the ability to produce cis-abienol.

  • To obtain the genetic information about DVT( $\alpha$- and $\beta$-4,8,13- duvatriene-1,3-diols) and cis-abienol contents, two burley(Burley 21, KB 108), three sun-cured cultivars(N. tabacum L. cv. Yonbyun 3, Jahengyon and Jaraehong), TI 1068, and thirty F$_1$ hybrids derived from them were sampled and the diterpenes were analysed using thin layer chromatography (TLC) procedures. DVT exudation from the leaf scerface could Ie detected in all coltivars and F$_1$ hybrids tested. TI 1068 and Yonbyun 3 had cis-abienol exdudates. Burley 21, KB 108, Jaheungyon, and Jaraehong had no spot of cis-abienol. It is considered that cis-abienol exdudation might be controlled by dominant gene(s). The cytoplasmic effect on the cis-abienol exdudation was not detected.

  • A cis-1,2-dichloroethylene (cis-DCE)-degrading anaerobic bacterium, Clostridium sp. strain KYT-1, was isolated from a sediment sample collected from a landfill site in Nanji-do, Seoul, Korea. The KYT-1 strain is a gram-positive, endospore-forming, motile, rod-shaped anaerobic bacterium, of approximately $2.5{\sim}3.0\;{\mu}m$ in length. The degradation of cis-DCE is closely related with the growth of the KYT-1 strain, and it was stopped when the growth of the KYT-1 strain became constant. Although the pathway of cis-DCE degradation by strain KYT-1 remains to be further elucidated, no accumulation of the harmful intermediate, vinyl chloride (VC), was observed during anaerobic cis-DCE degradation. Strain KYT-1 proved able to degrade a variety of volatile organic compounds, including VC, isomers of DCE (1,1-dichloroethylene, trans-1,2-dichloroethylene, and cis-DCE), trichloroethylene, tetrachloroethylene, 1,2-dichloroethane, 1,1,1-trichloroethane, and 1,1,2-trichloroethane. Strain KYT-1 degraded cis-DCE at a range of temperatures from $15\;to\;37^{\circ}C$, with an optimum at $30^{\circ}C$, and at a pH range of 5.5 to 8.5, with an optimum at 7.0.

  • The purpose of this study was to develop a gas chromatographic (GC) method to analyze fatty acids in milk fat with a single injection. The single-injection GC method we developed for analyzing fatty acid composition can separate a wide range of fatty acid methyl esters from butyric acid to docosahexaenoic acid. It separated 6 isomers of 18:1 (cis-6, cis-9, cis-11, trans-6, trans-9 and trans-11), 4 isomers of 18:2 (cis-9-cis-12, trans-9-trans-12, cis-9-trans-12 and trans-9-cis-12), and 4 isomers of conjugated 18:2 (cis-9- trans-11, trans-9-cis-11, cis-10-trans-12 and trans-10-cis-12).

  • Amino acid cobalt(Ⅲ) complexes of 1,3-diminopropane-N,N'-di-α-(β-methyl)-pentanoic acid (H2dpdmp), uns-cis-[Co(dpdmp)(aa)] (aa=glycine, S-alanine, R-aspartic acid, sarcosine) have been prepared from the reaction between the uns-cis-[Co(dpdmp)Cl2]- complex and the corresponding amino acid. In the reaction with the uns-cis-[Co(dpdmp)Cl2]- complex, glycine and S-alanine have yielded both merridional and facial isomers, while R-aspartic acid and sarcosine, only merridional isomers. The stereospecific substitution reaction of R-aspartic acid to racemic uns-cis-[Co(dpdmp)Cl2]- complex has yielded two merridional diastereomers; ΛR-uns-cis- and ΛR-uns-cis-[Co(dpdmp)(R-asp)]. It is of interest to note that this is one of the few CoⅢ(ONNO)(aa) type complex preparations, which gives only one isomer with stereospecificity. On the other hand, two merridional products obtained from the reaction of sarcosine with racemic uns-cis-[Co(dpdmp)Cl2]- are turned out to be mixtures of optical isomers.
