Fusobacterium nucleatum is a gram-negative bacteria associated with diverse infections like appendicitis and colorectal cancer. It mainly attacks the epithelial cells in the oral cavity and throat of the infected individual. It has a single circular genome of 2.7 Mb. Many proteins in F. nucleatum genome are listed as "Uncharacterized." Annotation of these proteins is crucial for obtaining new facts about the pathogen and deciphering the gene regulation, functions, and pathways along with discovery of novel target proteins. In the light of new genomic information, an armoury of bioinformatic tools were used for predicting the physicochemical parameters, domain and motif search, pattern search, and localization of the uncharacterized proteins. The programs such as receiver operating characteristics determine the efficacy of the databases that have been employed for prediction of different parameters at 83.6%. Functions were successfully assigned to 46 uncharacterized proteins which included enzymes, transporter proteins, membrane proteins, binding proteins, etc. Apart from the function prediction, the proteins were also subjected to string analysis to reveal the interacting partners. The annotated proteins were also put through homology-based structure prediction and modeling using Swiss PDB and Phyre2 servers. Two probable virulent factors were also identified which could be investigated further for potential drug-related studies. The assigning of functions to uncharacterized proteins has shown that some of these proteins are important for cell survival inside the host and can act as effective drug targets.

This research aims to assess the behavior of reinforced concrete (RC) residential buildings when moment-resisting frames (MRFs) are used as the lateral resisting system. This investigation was conducted using MIDAS Gen v.19.0. Buildings with various plan footprints (Square, Rectangular, Circular, Triangular, and Plus-Shaped), and different heights (15 m, 30 m, 45 m, and 60 m) are investigated. The defined load cases, the equivalent static lateral load pattern, and the response spectrum function were defined as stated by the American Standard (ASCE 7-16), the 1997 Uniform Building Code (UBC97), the Egyptian Code for Loads (ECP-201), and the European Standard (EC8). Extensive comparisons of the results obtained by the different codes (including the story displacement, the story drift, and the base shear) were undertaken; to assess the response of moment-resisting multi-story framed buildings under lateral loads. The results revealed that, for all study cases under consideration, both ECP-201 and EC8 gave smaller base shear, displacement, and drift by one third to one fourth, around one fourth, around one fifth, respectively for both the ELF and RSA methods if compared to ASCE 7-16 and UBC97.
In the genome annotation of Escherichia coli MG1655, the orf382 (1,149 bp) is designated as a gene encoding an alcohol dehydrogenase that may be Fe-dependent. In this study, the gene was amplified from the genome by PCR and overexpressed in Escherichia coli BL21(DE3). The recombinant $6{\times}$His-tag protein was then purified and characterized. In an enzymatic assay using different hydroxyl-containing substrates (n-butanol, $\small{L}$-threonine, ethanol, isopropanol, glucose, glycerol, $\small{L}$-serine, lactic acid, citric acid, methanol, or $\small{D}$-threonine), the enzyme showed the highest activity on $\small{L}$-threonine. Characterization of the mutant constructed using gene knockout of the orf382 also implied the function of the enzyme in the metabolism of $\small{L}$-threonine into glycine. Considering the presence of tested substrates in living E. coli cel ls and previous literature, we believed that the suitable nomenclature for the enzyme should be an $\small{L}$-threonine dehydrogenase (LTDH). When using $\small{L}$-threonine as the substrate, the enzyme exhibited the best catalytic performance at $39^{\circ}C$ and pH 9.8 with $NAD^+$ as the cofactor. The determination of the Km values towards $\small{L}$-threonine (Km = $11.29{\mu}M$), ethanol ($222.5{\mu}M$), and n-butanol ($8.02{\mu}M$) also confirmed the enzyme as an LTDH. Furthermore, the LTDH was shown to be an ion-containing protein based on inductively coupled plasma-atomic emission spectrometry with an isoelectronic point of pH 5.4. Moreover, a circular dichroism analysis revealed that the metal ion was structurally and enzymatically essential, as its deprivation remarkably changed the ${\alpha}$-helix percentage (from 12.6% to 6.3%).
Acoustic characteristics of water sediment were experimentally studied in laboratory. Water saturated sand sediment less than the grain size of 0.5 mm diameter is uniformly distributed in an acryl box (100 mm×100mm×42mm) with material thickness 1 mm. Pores in the acryl box are modeled as the structure of cylindrical pore tubes (diameter 3 mm and length 42 mm) filled with water. Cylindrical pore tubes have porosities 0%, 5%, 11%, 18% and 26 % controlled by the tube numbers. Transmitted acoustic waves through sand sediment specimen are analyzed as the functions of porosity and frequency from 0.3 MHz to 4 MHz. Transmitted acoustic waves are mixed with the first-kind wave from whole specimen and the second-kind wane from cylindrical pore tubes. For the center frequency 1 MHz, the first kind wave is dominant but for the center frequency 2.25 MHz, the second kind wave is dominant. In the case of the first-kind wave, as the porosity increases, the transmission coefficient decreases and the sound speed decreases to the sound speed of water. As the frequency increases, the transmission coefficient decreases but the sound speed is almost constant. In the case of the second-kind wave, as the porosity increases, the transmission coefficient increases but the sound speed is almost constant. The transmission coefficient and the sound speed are almost constant as a function of frequency.

From the present study, the basis for the application of flow induced vibration to the heat recovery steam generator tube array would be prepared. For the previous mentioned purpose, the present CFD analysis introduced a single fin tube and calculated with the unsteady laminar flow over the single fin tube. The characteristics of vortex shedding and lift and drag fluctuation over the fin tube was investigated. The derived nondimensional lift PSD was compared with the results of the previous experimental studies and the characteristics of lift and drag PSD over a single fin tube was established from the present CFD study.

The present CFD analysis introduced circular cylinder tube array and calculated using unsteady laminar flow for the tube array. The characteristics of lift and drag fluctuations over the cylinder tube array was investigated. The derived nondimensional lift and drag PSD was compared with the results of the previous experimental research and the characteristics of lift and drag PSD for a circular cylinder tube array was established from the present CFD study.

The heat transfer characteristics of the drag reducing polymer solutions are investigated experimentally in the thermal entrance region of circular tube flows. Fluids used in experiments are the aqueous solutions of high molecular polymer, polyacrylamide Separan AP-273 and the range of polymer concentrations is from 20 to 1000 wppm. Two stainless steel tubes with inside diameter 8.5mm(L/D=712) and 10.3mm(L/D=1160) are used for the heat transfer flow loops. The flow loop is set up to measure friction factors and heat transfer coefficients of test sections in two different modes; the recirculating flow system and once-through flow system. The test tubes are heated directly by electricity to apply the constant heat flux boundary conditions to the wall. Three different types of adaptors are used to observe the effects of the upstream flow conditions of the heat transfer test sections. The viscosity and characteristic relaxation time of the test fluids circulating in the flow system are measured by the capillary tube viscometer and falling ball viscometer at regular time intervals. The installed adaptors exhibit slight effect on the entrance heat transfer of Newtonian fluid. However, no noticeable effects are observed for the entrance heat transfer of the drag reducing fluids. The order of magnitude of the thermal entrance lengths of the drag reducing fluids which follow the minimum friction asymptote is much longer than that of Newtonian fluids in turbulent flows. A new dimensionless parameter, the viscoelastic Graetz number, is defined and all the experimental data are recasted in terms of the viscoelastic Graetz number. The local Nusselt number of the viscoelastic fluids is represented as a function of flow behavior index n and the viscoelastic Graetz number. As degradation continues the viscosity and the characteristic relaxation time of the testing fluids decrease. Weissenberg number defined by the relaxation time and D/V appears to be a proper dimensionless parameter in describing degradation effects on heat transfer of the viscoelastic fluids.
In this paper, we propose the development of deep learning structure to improve quality of polygonal containers. The deep learning structure consists of a convolution layer, a bottleneck layer, a fully connect layer, and a softmax layer. The convolution layer is a layer that obtains a feature image by performing a convolution 3x3 operation on the input image or the feature image of the previous layer with several feature filters. The bottleneck layer selects only the optimal features among the features on the feature image extracted through the convolution layer, reduces the channel to a convolution 1x1 ReLU, and performs a convolution 3x3 ReLU. The global average pooling operation performed after going through the bottleneck layer reduces the size of the feature image by selecting only the optimal features among the features of the feature image extracted through the convolution layer. The fully connect layer outputs the output data through 6 fully connect layers. The softmax layer multiplies and multiplies the value between the value of the input layer node and the target node to be calculated, and converts it into a value between 0 and 1 through an activation function. After the learning is completed, the recognition process classifies non-circular glass bottles by performing image acquisition using a camera, measuring position detection, and non-circular glass bottle classification using deep learning as in the learning process. In order to evaluate the performance of the deep learning structure to improve quality of polygonal containers, as a result of an experiment at an authorized testing institute, it was calculated to be at the same level as the world's highest level with 99% good/defective discrimination accuracy. Inspection time averaged 1.7 seconds, which was calculated within the operating time standards of production processes using non-circular machine vision systems. Therefore, the effectiveness of the performance of the deep learning structure to improve quality of polygonal containers proposed in this paper was proven.
The purpose of this paper was to develop size & shape optimization programming algorithm of plane trusses. The optimum techniques applied in this study were extended penalty method of Sequential Unconstrained Minimization Techniques(SUMT) and direct search method with multi-variables proposed by Hooke & Jeeves. Upper mentioned two methods were used iteratively at each level of size and shape optimization routines. The design variables of size optimization were circular steel tube(structural member) diameter and thickness, those of shape optimization were joint coordinates, and the objective function was represented as total weight of truss. During the optimum design, two level procedures of size and shape optimization were interacted iteratively until the final optimum values were attained. At the previous studies about shape optimization of truss, the member sectional areas and coordinates were applied as design variables. So that they could not apply the buckling effect of compression member. In this paper, actual sizes of member and nodal coordinates are used as design variables to consider the buckling effect of compression member properly.
