Analysis of Aroma Compounds of Cinnamon by Solid Phase Microextraction (Solid Phase Microextraction을 이용한 계피의 향기성분 분석)

  • The volatile components of cinnamon bark were extracted by using different isolation methods, simultaneous distillation extraction (SDE) and solid phase microextraction (SPME). Then the volatile components were analyzed by gas chromatography(GC) and mass selective detector(MSD). 30 compounds were identified in cinnamon bark. In SPME technique, several factors influencing the equilibrium of the aroma compounds between sample and SPME fiber was taken into account, including the kind of SPME fiber, extraction temperature and extraction time. Four different SPME fibers were tested, namely polydimethylsiloxane (PDMS), poly acrylate(PA), divinyl- benzene-carboxen-polydimethylsiloxane (DVB/CAR/PDMS) and carbowax/divinylbenzene(CW/DVB). Among these SPME fiber, PDMS coating fiber showed the best results. The profile of volatile compounds of cinnamon bark at different extraction temperature and extraction time were investigated by 100$\mu\textrm{m}$ PDMS fiber.

Bioconversion Constituents of Galgeun-tang Fermented by Lactobacillus plantarum (Lactobacillus plantarum 발효에 의한 갈근탕의 생물 전환 성분 연구)

  • Galgeun-tang (GGT) is a traditional medicinal formula that is widely prescribed to treat cold, asthma, and hives in Korea. Fermented herbal medicines can be made more effective than normal herbal medicines by increasing the absorption and bioavailability of the active compounds. In this study, we fermented Galgeun-tang to produce bioconversion constituents using Lactobacillus plantarum (GGT144), and found that four peaks were decreased, three peaks were increased and two new peaks appeared in the HPLC-DAD chromatogram. After HPLC-DAD-guided fractionation of the newly-appearing compounds (1 and 5) and the increased (6, 7, and 9) compounds, the structure of the compounds was determined using NMR and MS. Using this approach the compounds were identified to be pyrogallol (1), daidzein (5), liquiritigenin (6), cinnamyl alcohol (7), and formononetin (9), respectively. In addition, the decreased compounds were identified to be daidzin (2), liquiritin (3), ononin (4), and cinnam aldehyde (8) using HPLC-DAD analysis with standard compounds. The high performance liquid chromatography method was used to quantify the nine constituents in GGT and GGT144. All calibration curves of the standard compounds displayed excellent linearity with a $R^2$ > 0.9968.

Effect of Burkholderia contaminans on Postharvest Diseases and Induced Resistance of Strawberry Fruits

  • This study takes strawberry-fruits as the test material and discusses the effect of Burkholderia contaminans B-1 on preventing postharvest diseases and inducing resistance-related substances in strawberry-fruits. Soaking and wound inoculating is performed to analyze the inhibitory effects of different treatment solutions on the gray mold of postharvest strawberry-fruits. The count of antagonistic bacteria colonies in the wound is found, and the dynamic growth of antagonistic bacteria and the pathogenic fungus is observed by electron microscopy. The results indicated that, either by soaking/wound-inoculating, the fermentation and suspension of antagonistic bacteria significantly reduced the incidence of postharvest diseases of strawberry-fruits. With wound inoculation, the inhibition rate of antagonist fermentation and suspension ($1{\times}10^{10}cfu/ml$) respectively reached 77.4% and 66.7%. It also led to a significant increase in the activity of resistance-related enzymes, i.e., phenylalanine ammonia lyase (PAL), 4-coumarate coenzyme A ligase (4CL), cinnamate-4-hydroxylase (C4H) and chalcone isomerase (CHI). On 1 d and 2 d post-treatment, the activity of 4CL was respectively 3.78 and 6.1 times of the control, and on 5 d, the activity of PAL was increased by 4.47 times the control. The treatment of antagonistic bacteria delayed the peaking of cinnamyl-alcohol dehydrogenase (CAD) activity and promoted the accumulation of lignin and total phenols. The antagonistic bacteria could be well colonized in the wounds. On 4-5 d post-inoculation, the count of colonies was $10^8$ times of that upon inoculation. Electronmicroscopy indicated that the antagonistic bacteria delayed the germination of pathogenic spores in the wounds, and inhibited further elongations of the mycelia.

Study on the quality assessment of Liquidambaris Storax (소합향(蘇合香)의 규격 설정을 위한 연구)

  • Objective : Liquidambaris Storax, the processed resin of Liquidambar orientalis Mill., has been therapeutically used as orifice-opening medicinals in Korean Medicine. However, unauthenticated resinous drugs from various origins were sold as Liquidambaris Storax. This study aimed to establish the quality assessment of genuine Liquidambaris Storax and to provide quantitative differences of Liquidambaris Storax in herbal markets. Methods : Bibliographic research on ancient Liquidambaris Storax was performed using classic books of herbology. The quality assessment was approached by searching modern books of herbal medicines. The marker compounds in Liquidambaris Storax from various origins were quantitatively analyzed using high-performance liquid chromatography. Results : L. orientalis was botanically characterized by palmately 5-lobed shape of leaves. Genuine Liquidambaris Storax was recognized as resinous or semi-solid form (ancient―Song dynasty) to liquid form (Myeong/Cheong dynasty) and two processed forms were available in current markets. Genuine liquid Liquidambaris Storax should be lengthened as a thread when it was lifted by a stick and be sunken under the water. The crystals of cinnamic acid was seen under microscopic observation and the scent of benzaldehyde occurred after the treatment with potassium permanganate. Cinnamyl alcohol, cinnamic acid, and cinnamaldehyde were quantified in two liquid samples of Liquidambaris Storax, while cinnamaldehyde was not quantified in resinous (semi-solid) sample. Conclusion : This study presents the detailed methods of quality assessment for genuine Liquidambaris Storax as well as the status in the herbal markets. Further quantitative study with diverse samples is required to establish the quality standard of Liquidambaris Storax.

Selection of Low Lignin-high Biomass Whole Crop Silage Rice Elite Line for the Improvements of Forage Digestibility and Fermentation

  • Lignin modification has been a breeding target for the improvements of forage digestibility and fermentation in whole crop silage(WCS) rice. In rice, gold hull and internode 2 (gh2) was identified as a lignin-deficient mutant. gh2 exhibits a reddish-brown pigmentation in the hull and the internode is located on the short arm of chromosome 2 and codes for cinnamyl-alcohol dehydrogenase (CAD). To develop WCS rice variety improved digestibility and fermentation, we measured acid detergent fiber (ADF), lignin and total digestible nutrient (TDN) calculated from ADF (TDN=88.9-(0.79% × ADF) and performed marker-assisted selection using CAD(Os2g0187800) gene first intron region specific marker with 55 Jungmo1038/J.collection lines. Those lines had lignin content range from 0.82 to 6.61%, ADF from 15.8 to 45.8%, TDN from 52.7 to 78.8 compared to 'Jungmo1038'(1.53,20.7,72.6), 'J.collection'(0.98,12.8,78.8%) and gh2 were introgressed into 44 lines. Considering on these genotype and low-lignin phenotype, we finally selected 2 elite lines(Suweon668, Suweon669). Suweon668 and Suweon669 line are high biomass-low lignin lines that the ADF content is relatively low, even though the dry matter weight is high. Also they have lodging and shattering resistance and glabrous leaf and hull important to improve cattle palatability. Our results will provide that rice can be improved for forage digestibility and fermentation with low lignin concentration.

The overexpression of Arachis hypogaea resveratrol synthase 3 (AhRS3) modified the expression pattern of phenylpropanoid pathway genes in developing rice seeds

  • Our previous study for developing seeds of Iksan 526 (I.526), an inbred line of resveratrol-producing transgenic rice line, showed that, in 20 days after heading (DAH) seeds, resveratrol was almost saturated and accumulation of piceid was highest though the expression of Arachis hypogaea resveratrol synthase 3 (AhRS3, GenBank DQ124938) was highest in 31 DAH seeds. In this study, it was investigated how the overexpression of AhRS3 affects phenylpropanoid pathway genes. p-Coumaroyl-CoA is derived from phenylpropanoid pathway and used as a substrate of AhRS3 reaction for resveratrol production. In 6, 13, 20, 31 and 41 (45 for Dongjin) DAH seeds of I526 and Dongjin, a wild type of I.526, respectively, the expression pattern of phenylpropanoid pathway genes, including phenylalanine ammonia-lyase (PAL: LOC_Os02g41630.2, LOC_Os04g43760.1), cinnamate 4-hydroxylase (C4H: LOC_Os05g25640.1), 4-coumarate-CoA ligase (4CL: LOC_Os02g08100.1), cinnamoyl-CoA reductase (CCR: LOC_ Os09g25150.1, LOC_Os08g34280.1), hydroxycinnamoyl-CoA shikimate/quinate hydroxycinnamoyl transferase (HCT: LOC_Os04g42250.2, LOC_Os02g39850.1) and cinnamyl alcohol dehydrogenase (CAD: LOC_Os02g09490.1), was examined using real time (RT)-PCR. Compared to developing seeds of Dongjin, RT-PCR results showed that the expression pattern of phenylpropanoid pathway genes was modified in developing seeds of I.526. In most genes, except for CAD, of I.526 developing seeds, the gene expression was highest in 20 DAH corresponding to biosynthesis of resveratrol and piceid, i.e. the expression of phenylpropanoid pathway genes was gradually increased by 20 DAH and decreased as seeds develop. Especially, in Dongjin, the highest expression of PALs and 4CL was in 6 DAH and their expression was gradually decreased as seeds develop. These genes expression data also exhibited that, in developing seeds of I.526, phenylpropanoid pathway genes were slightly or significantly (in some genes) upregulated compared to Dongjin. Therefore, the overexpression of AhRS3 changed the expression pattern of phenylpropanoid pathway genes in I.526 developing seeds and this modification for gene expression is closely related to biosynthesis of resveratrol and piceid.

Insecticidal Activity of Cinnamon Essential Oils, Constituents, and (E)-Cinnamaldehyde Analogues against Metcalfa pruinosa Say (Hemiptera: Flatidae) Nymphs and Adults (미국선녀벌레(Metcalfa pruinosa Say)에 대한 계피 정유 유래 물질의 살충 활성)

  • The insecticidal activity of the constituents of cinnamon essential oils and structurally related compounds against both the nymphs and adults of the citrus flatid planthopper Metcalfa pruinosa was examined using a direct-contact application. The toxicity of the cinnamon oil constituents and 21 (E)-cinnamaldehyde related compounds regarding the nymphs of M. pruinosa was evaluated using a leaf-dipping bioassay. Based on 24 h $LC_{50}$ values, hydro-cinnamic acid ($1.55mg/cm^2$) is the most toxic compound, followed by geranic acid ($1.59mg/cm^2$). The $LC_{50}$ values of 11 of the compounds including cinnamaldehyde are between $1.60mg/cm^2$ and $4.94mg/cm^2$. Low toxicities and no toxicity were observed with the other 15 ($5.24mg/cm^2$ to $13.47mg/cm^2$) and two compounds, respectively. Also, the toxicities of the cinnamon oil constituents and 21 cinnamaldehyde related compounds regarding the M. pruinosa adults were evaluated using a direct-spray method. The toxicity of eugenol (10.81 mg) is the most toxic compound for the adults of M. pruinosa, followed by geranic acid (30.68 mg). The $LC_{50}$ values of nine of the compounds including cinnamaldehyde are between 59.16 mg and 96.70 mg. Low toxicities and no toxicity were observed with the other 15 (105.44 mg to 255.76 mg) and three compounds, respectively. The spray formulations that comprise cinnamon bark and cinnamon green leaf oils resulted in 82.3% and 82.9% mortalities, respectively, toward the M. pruinosa adults in a ginseng field. Global efforts to reduce the level of highly toxic synthetic insecticides in agricultural environments justify further studies on cinnamon oils to ascertain whether the corresponding active principles can act as insecticides, when they are applied as a direct spray with contact action, for the control of M. pruinosa populations.
