고혈압 흰쥐에서 cumambrin A의 혈압 강하 효과 (Effect of cumambrin A Treatment on Blood Pressure in Spontaneously Hypertensive Rats)

  • We have isolated a sesquiterpene lactone, cumambrin A from the dried flowers of Chrysanthemum boreale Makino and reported its chemical structure. The aim of the present study was to investigate whether the exogenous administration of cumambrin A has a pharmacological effect on normalization of blood pressure in the spontaneously hypertensive rats (SHR). In vitro studies: Relaxative response induced by cumambrin A was increased with dose-dependent manner and showed maximizing response at a concentration of 5?0-4 M. Further, this relaxative response was significantly increased at a condition of endothelium present than that of endothelium denuded. in vivo studies: The normalizing effect of cumambrin A on blood pressure was also increased with time-dependent manner and then gradually recovered to normal condition at approximately 4 hrs after cumambrin A treatment.

자생식물 Essential Oil 5 종의 항 아토피피부염 활성 연구 (A Study on the Activities of Five Natural Plant Essential Oils on Atopic Dermatitis)

  • 본 연구는 개똥쑥(Artemisia annua L., AA), 유자(Citrus junos Sieb. ex TANAKA, CJ), 산국화(Chrysanthemum boreale Makino, CB), 잣나무(Pinus koraiensis, PK), 금강송(Pinus densiflora for. erecta, PD) 총 5 종의 자생식물 essential oil의 항 아토피 효능을 확인하기 위한 실험이다. 항균 효과를 확인하기 위해 자생식물 essential oil 5 종을 Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans 총 4 종류의 균에 처리했다. 항염증 효과를 확인하기 위해 TNF-α와 IFN-γ (TI)를 처리한 HaCaT 세포에 5 종의 자생식물 essential oil을 처리했다. AA, CJ, CB, PK, PD은 10 mg/mL 농도에서 Candida albicans에 대한 항균 효과를 나타냈다. AA (1 ㎍/mL), CB (1 ㎍/mL), PK (0.1, 1 ㎍/mL)에서 thymus and activation-regulated chemokine (TARC) 생성량이 억제되었고, AA 및 PK (1 ㎍/mL)에서 macrophage-derived chemokine (MDC) 생성량이 억제되었으며, AA (0.1, 1 ㎍/mL), PK (0.1, 1 ㎍/mL)에서 IL-6 생성량이 억제되었다. AA, CB, PK essential oil의 항균 및 항염증 효과가 확인되었으며, 이에 아토피 피부염 완화에 기여할 수 있음을 기대한다.

흰쥐에서 Cumambrin A의 대동맥 이완작용 (Effect of Cumambrin A on the Relaxation of Rat Aorta)

  • We previously reported that the exogenous administration of cumambrin A, a sesquiterpene lactone from the dried flowers of Chrysanthemum boreale Makino has a pharmacological effect on normalization of blood pressure in the spontaneously hypertensive rats (SHR). In the present study, we further investigated the effect of cumambrin A on the relaxation of phenylephrine-induced precontracted rat aortic artery rings. The potency of cumambrin A was than compared to verapamil, a well known $Ca^{2+}-channel$ blocker. The results demonstrate that the isolated rat aortic arteries are relaxed to basal tension at a concentration of $5{\times}10^{-5}\;M$ cumambrin A treatment. The results also show that the phenylephrine-induced contraction is inhibited by a pretreatment of cumambrin A. Co-treatment of cumambrin A and verapamil showed a strong synergetic effect on the relaxation of rat aortic artery rings. Thus, these data demonstrate that cumambrin A is a potent relaxant of rat aortic smooth muscle and suggest that cumambrin A modulates intracellular or extracellular $Ca^{2+}$ mobilization.

감국, 산국 및 구절초꽃 캔디의 항산화활성과 품질특성 (Quality Characteristics and Antioxidant Activity of Chrysanthemum indicum L., C. boreale M., and C. zawadskii K. Flowers Candies)

  • 감국, 산국 및 구절초 열수추출물의 첨가량을 달리하여 캔디를 제조하고 이들의 품질특성과 항산화특성 및 관능특성을 조사하였다. 색도의 경우 추출물의 첨가량의 증가와 함께 L값(명도)은 감소하였고, a값(적색도)과 b값(황색도)은 증가하였다. pH의 경우 감국, 산국 및 구절초 캔디는 각각 5.81$\sim$5.92, 5.77$\sim$5.85 및 5.83$\sim$5.94로 추출물 첨가량이 증가할수록 감소하였으며, 산도는 추출물의 첨가량이 증가함에 따라 증가하였다. 수분함량도 추출물의 첨가량이 증가함에 따라 증가하였다. 용해도는 초기에 급격하게 증가하였으며, 경도는 추출물의 첨가량이 증가할수록 감소하였다. 총 폴리페놀 및 플라보노이드 함량, 전자공여능, 총 항산화력은 추출물 첨가량의 증가에 따라 증가하였으며, 구절초 캔디가 각각 0.815$\sim$2.706 mg/g, 0.147$\sim$0.856 mg/g, 15.217$\sim$76.236% 및 0.514$\sim$3.167 mg AA eq/g으로 감국 및 산국 캔디보다 높았다. 관능평가 결과 색은 추출물 첨가량이 증가함에 따라 감소하였으며, 향의 경우 감국 캔디는 15% 첨가구, 구절초 캔디는 10% 첨가구에서 각각 5.20과 5.35로 높게 평가되었다. 맛의 경우 산국 캔디는 추출물 첨가량의 증가에 따라 감소하였으며, 감국과 구절초 캔디의 경우는 10% 첨가구에서 각각 5.30과 5.60으로 높게 나타났다. 조직감은 경도가 낮을수록 낮은 점수를 얻었다. 종합적인 기호도에서 감국과 구절초 캔디의 경우 10% 첨가구에서 각각 5.75와 6.00으로 높은 평가를 받았으나, 추출물의 첨가량이 증가할수록 낮은 점수를 받았다.

Induction of Quinone Reductase, an Anticarcinogenic Marker Enzyme, by Medicinal Herb Extracts

  • To search for novel cancer preventive agents, we assessed the quinone reductase (QR)-inducing activities of medicinal herb extracts in cultured murine hepatoma cells (hepalclc7 cells). Among 216 herb extracts tested in this study, 8 kinds of herbal extracts were found to induce QR activity in hepalclc7 cells by more than 2-fold when used at the concentration of 25 $\mu\textrm{g}$/$m\ell$. The methanol extracts of Aster koraiensis NK and Pulsatilla koreana Nakai induced QR by 252 and 223 % , respectively, at the concentration of 25 $\mu\textrm{g}$/$m\ell$. Most of the herbal extracts with QR inducing-activity increased the enzyme activity in a typical dose-dependent manner. The QR activity in BP$^{r}$ cl cells was induced move than 50 % by the extracts of Pulsatilla koreana Nakai, Inula helenium, Physalis alkekengi var, francheti (Masters) Makino, Chrysanthemum zawadskii var. latilobum Kitamuva, Auemisia keiskeana Miquel, Chfsanthemum boreale Makino. In conclusion, hlsatilla koreana Nakai, Aster koraiensis N.K, and Chfsanthemum zawadskii var. iatilobum Kitamura, which showed relatively high QR induction, merit further animal study to evaluate their potential as cancer preventive agents.

Three Different Viruses Isolated from Typical Weed Plants that Grown Adjacent to Common Crop Fields

  • Weeds are widely grown in the field and are infected by many viruses. A survey was conducted to identify viruses infecting weeds in Korea. Virus-infected weed samples including Rorippa indica (L.) Hiern, R. islandica (Oed.) Bord, Crepidiastrum denticulatum (Houtt.) Pak & Kawanno, Achyranthes japonica (Miq.) Nakai, and Chrysanthemum boreale (Makino) Makino were collected in Kyonggi Province. These weeds were grown in the greenhouse and were isolated on 10 test plants. Several virus isolates were isolated fron infected tissues and were further studied by host range assay, serological test, electron microscopy (EM), reverse transcription-polymerase chain reaction (RT-PCR) and sequencing. Each isolated virus strain was mechanically transmitted to weeds and various hosts including Nicotiana spp., Brassica spp., Vigna unguiculata, Capsicum annuum, and Cucumis sativus and showed systemic mosaic, vein clearing, necrosis, mottle, malformation, chlorosis, and/or death of host plants in some cases. Each virus was then purified using infected leaves and observed by EM. From these results three viruses were isolated and identified as Turnip mosaic virus (TuMV), Broad bean wilt virus (BBWV), and Cucumber mosaic virus (CMV). RT-PCR using virus-specific oligonucleotide primers and the cloning were conducted to determine the nucleotide sequences of coat proteins of the three viruses their amino acid sequence were deduced. The amino acid sequence homologies were about 92.7 to 99.7%, 96.2 to 97.7%, and 93.9 to 98.6% to other reported TuMV, BBWV, and CMV strains, respectively. These results suggest that many weeds may serve as primary inoculum source of diseases caused by TuMV, BBWV, CMV and that the management of these viral diseases can be achieved through weed control.

수종의 국화과 식물에서 분리한 Sesquiterpene Lactone들의 생리활성(제3보) -Nitric oxide 방출 및 ACAT 저해활성- (Biological Activities of Sesquiterpene Lactones isolated from Several Compositae Plants Part 3 -Inhibitory Activity on Nitric Oxide Release and ACAT-)

  • Nine sesquiterpene lactones, which were isolated from Hemisteptia lyrata Bunge, Chrysanthemum zawadskii Herbich var. latilobum Kitamura and Chrysanthemum boreale Makino were evaluated for the inhibition upon the nitric oxide release from the Raw cell and for the ACAT (acyl Co A: cholesterol acyltransferase) inhibitory assay. In the nitric oxide release inhibitory experiment, hemistepsin B $(IC_{50}\;=\;0.05\;{\mu}g/ml,\;0.15\;{\mu}M),$ cumambrin B, tulipinolide and costunolide exhibited strong inhibition with $IC_{50}$ values $0.05\;{\mu}g/ml\;(0.15\;{\mu}M),\;0.1\;{\mu}g/ml\;(0.38\;{\mu}M),\;0.2\;{\mu}g/ml\;(0.69\;{\mu}M)$ and $0.2\;{\mu}g/ml(0.86\;{\mu}M),$ respectively. In the ACAT inhibitory assay, angeloylcumambrin B, tigloylcumambrin B, tulipinolide and costunolide exhibited strong inhibition with $IC_{50}$ values $33\;{\mu}g/ml\;(95.4\;{\mu}M),\;22\;{\mu}g/ml\;(63.6\;{\mu}M),\;38\;{\mu}g/ml\;(151\;{\mu}M)$ and $17\;{\mu}g/ml(73.3\;{\mu}M),$ respectively, whereas other compounds did not exhibit significant activity against ACAT above $100\; {\mu}g/ml.$.
