Chronic Treatment of Ethanol Inhibits Proliferation of Normal Fibroblasts, but Not Oncogenic ras-Transformed Cells

  • The adverse effects of ethanol on cell proliferation have been described for a variety of tissues and cells. In the present study, we investigated whether chronic ethanol intoxication impairs the cell proliferation and DNA synthesis induced by oncogenic $H-ras^{V12}$ - and $v-K-ras^{V12}$-transformed cells. Ethanol treatment inhibited the cell proliferation and the DNA synthesis of control parental fibroblasts in a time- and dose-dependent manner. In contrast, ethanol did not suppress the proliferation of either oncogenic $H-ras^{V12}$ - or $v-K-ras^{V12}$ -transformed fibroblasts. Microinjection of oncogenic $H-Ras^{V12}$ protein induces DNA synthesis and ethanol treatment did not interfere with the DNA synthesis. The antiproliferative toxicity of ethanol was rescued by antioxidants, such as N-acetylcysteine and 4-methlpyrazole. These results indicate that the antiproliferative action site of ethanol toxicity lies upstream or is independent of Ras and ethanol exerts its toxicity through a free radical formation.

Chironomus riparius의 급성 및 만성영향에 의한 농약의 퇴적토 독성평가 (Sediment Toxicity Assessment of Pesticides using Chironomus riparius Acute and Chronic Effect)

  • BACKGROUND: Pesticides is exposed in an aquatic environment and effected to benthic animals. Especially, sediment-associated pesticides is required for determination of sediment toxicity on aquatic organisms. This study was conducted to evaluate the impact of six pesticides (chlorfluazuron, difenoconazole, dithianon, flufenoxuron, flutianil, pendimethalin) on Chironomus riparius in aquatic ecosystems. METHODS AND RESULTS: Chlorfluazuron, difenoconazole, dithianon, flufenoxuron, flutianil and pendimethalin were used as a model compounds, which have a sediment-associated potential ($K_{oc}$>3). Acute and chronic toxicity tests on Chironomus riparius were performed at six concentrations of each pesticide with four replicates of each based on OECD test guideline 235 and 218. The calculated 48-h $EC_{50}$ values of chlorfluazuron, flutianil, pendimethalin, difenoconazole, dithianon and flufenoxuron were 6.72, 2.55, 2.27, 0.77, 0.30 and 0.11 mg/L, respectively. Flufenoxuron was the lowest 48-h $EC_{50}$ value in this study. The No Observed Effective Concentration (NOEC) and the Lowest Observed Effect Concentration (LOEC) of flufenoxuron for Chironomus riparius in 28-days test were 30 and $60{\mu}g/kg$, respectively. CONCLUSION: Pesticides of the sediment-associated have the potential effect for Chironomus riparius in aquatic ecosystems. Therefore, sediment toxicity assessment of these pesticides should be further investigated to evaluate the impact to benthic organisms.

Gasoline 취급 사업장의 작업환경 측정 및 위해성 평가 (Working Environment and Risk Assessment of Gasoline in Workplace)

  • 화학물질로부터 근로자의 건강보호를 위해 사용량이 많으나 작업환경 및 유해성 평가 자료가 부족한 gasoline에 대해 유해성(hazards)을 조사하고 국내 취급실태 및 일부 취급 사업장의 작업환경 측정을 통해 위해성(risk assessment)을 평가하였다. 연구결과 gasoline은 생식독성 추정 및 생식세포 변이원성 물질인 1B, 그리고 IARC Group 2B, ACGIH A3 물질이었다. 작업환경 측정결과는 TLV-TWA 기준 $900mg/m^3$미만이었으며, 유해성 평가결과 발암 $RfC_{(Worker)}$$0.3mg/m^3$, 만성흡입독성 $RfC_{(Worker)}$$2.7mg/m^3$, 발달독성 $RfC_{(Worker)}$$2.7mg/m^3$이었다. 그리고 위험도 평가에서 발암성은 459, 만성흡입독성은 51, 발달독성은 51이었으며, 이를 토대로 한 gasoline의 위험도는 1을 초과하는 물질로 평가되었다.

  • Toxic effects of chronic adminstration of methamphetamine (MA) to SD rats were studied in respect to histopathological changes induced in each organ. In experimental groups liver weight decreased and brain weights increased markedly compared with controls in the 12th month after subcutaneous injection of 0.5 mg and 5mg/kg/BW MA. Serum alkaline phosphotase levels increased, but marked decrease of cholesterol, triglyceride, and BUN levels were checked depending on both the dose of MA and duration of treatment.

Glucose-6-phosphate dehydrogenase를 이용한 Moina macrocopa의 중금속 독성 검정 (Heave Metal Toxicity Test in Moina macrocopa with Glucose-6-phosphate Dehydrogenase Activity)

  • A rapid, inexpensive enzymatic method is proposed for indirect water quality testing in terms of heavy metal toxicity. The activity of glucose-6-phosphate dehydrogenase was applied for heavy metal toxicity test as an effective criterion in water quality. The toxicity of Pb (lead) and Cd (cadmium) for water flea, Moina macrocopa, were evaluated for 2-8 days with variables of mobilization ability. And the reproduction impairment of Moina macrocopa were investigated as the parameter of chronic toxicity test for Pb and Cd. As a result, the EC$_{50}$ for immobilization of Moina macrocopa were Pb and Cd were 1.6749 and 0.4683, respectively. The values of reproductive impairment to Moina macrocopa for Pb and Cd were 9.5938 and 8.3264 in EC$_{50}$ A significant alteration of G6PDH (Glucose-6-phosphate dehydrogenase) activity of Moina macrocopa was observed when Cd and Pb were treated in media. The results obtained indicate that G6PDH activity of Moina macrocopa can be used as an indicative parameter in aquatic toxicity tests for heavy metals.als.

  • Arsenic is a unique element with distinct physical characteristics and toxicity whose importance in public health is well recognized. The toxicity of arsenic varies across its different forms. While the carcinogenicity of arsenic has been confirmed, the mechanisms behind the diseases occurring after acute or chronic exposure to arsenic are not well understood. Inorganic arsenic has been confirmed as a human carcinogen that can induce skin, lung, and bladder cancer. There are also reports of its significant association to liver, prostate, and bladder cancer. Recent studies have also suggested a relationship with diabetes, neurological effects, cardiac disorders, and reproductive organs, but further studies are required to confirm these associations. The majority of research to date has examined cancer incidence after a high exposure to high concentrations of arsenic. However, numerous studies have reported various health effects caused by chronic exposure to low concentrations of arsenic. An assessment of the health effects to arsenic exposure has never been performed in the South Korean population; thus, objective estimates of exposure levels are needed. Data should be collected on the biological exposure level for the total arsenic concentration, and individual arsenic concentration by species. In South Korea, we believe that biological exposure assessment should be the first step, followed by regular health effect assessments.

Toxicity of disulfiram, a therapeutic agent for chronic alcoholism, to mouse spleen cells

  • Disulfiram is a drug used to treat alcohol dependence. Recent studies have shown that disulfiram also has anti-cancer effects. Considering that many anti-cancer agents have side effects, including immunosuppression, it is important to check if disulfiram has some cytotoxicity to immune cells. In this study, mouse spleen cells were treated with disulfiram and the metabolic activity was measured. Disulfiram increased the cell death of spleen cells according to annexin V-FITC/PI staining analysis. In addition, disulfiram decreased the mitochondrial membrane potential of spleen cells. The toxicity of disulfiram was concentration dependent. Interestingly, disulfiram affected the population of lymphocytes and the subset of spleen cells was altered. This study provides clinicians and researchers with valuable information regarding the toxicity of disulfiram to mouse spleen cells, particularly lymphocytes.

화학물질 우선순위 선정 기법(CRS-Korea)의 개발과 적용 (Development of Korean Chemical Ranking and Scoring System (CRS-Korea) and its Application to Prioritizing National Toxic Chemicals)

  • A chemical ranking and scoring (CRS-Korea) system was developed and proposed to use as the first step to prioritize the toxic chemicals for the purpose of monitoring and detailed risk assessment that might follow as necessary. The CRS-Korea system takes a basic concept of risk assessment (both human health risk and ecological risk) in that risk score is determined by the product of toxicity score and exposure score. Included in the toxicity category are acute toxicity, chronic/sub -chronic toxicity, carcinogenicity, and other toxicity. The exposure category consists of quantity released to the environment, bioconcentration, and persistence. A consistent scheme and a comprehensive chemical data base are offered in the CRS-Korea system to calculate a score for the each component in the two categories by using specific physicochemical, fate, and toxic properties and the quantity of the chemical used. The toxicity score is obtained by adding up all the individual scores for the components in the toxicity category. The exposure score is determined by multiplication of the score of the quantity released with the sum of persistent score and bioconcentration score. Equal weight is given to the toxicity score and the exposure score. As the CRS-Korea system was applied to identify 50 national priority chemicals, it was found that significant data gap exists on toxicity and fate properties and that the uncertainty associated with estimating the quantify released to the environment is notably high. The proposed CRS system is only a screening tool in the first step toward the priority setting and should be used with expert judgement and other considerations necessary.

A 24-Weeks Toxicity Study of Eryngium foetidum Linn. Leaves in Mice

  • Eryngium foetidum Linn. leaves (EF) are widely used in Thailand and many countries throughout Asia as a culinary seasoning and a traditional medicine. However, adverse effect of high dose consumption in long duration has not been evaluated. The aim of this study was to investigate chronic toxicity of EF in mice. Thirty-two ICR male mice were divided into 4 groups of 8 mice each. The mice were fed AIN-76 rodent diet, or AIN-76 rodent diet supplemented with ground freeze-dried EF at 0.8%, 1.6% and 3.2% that is equivalent to approximately 35, 73 and 155 times that of human consumption, respectively, at 97.5 percentile for a period of 24 weeks. At the end of experiment, the mice were euthanized and blood samples were collected for hematological and biochemical evaluations. Necropsy was performed while visceral organs such as lung, liver, kidneys, spleen etc. were collected, weighed and histopathologically examined. Blood urea nitrogen (BUN) results of mice in 1.6% and 3.2% EF diet groups were significantly higher than the BUN of control group. No significant difference was noted in other biochemical and hematological properties between the treatment groups and control; all results were within normal range. Histopathology of almost all visceral organs showed no significant changes. However, tubulonephrosis and chronic interstitial nephritis were observed in the groups treated with 1.6% and 3.2% EF diet. Body weight was reduced significantly at week 12 to week 20 when compared to the control group while relative kidney weights were significantly increased. In conclusion, the consumption of EF in diet at high doses illustrated the adverse effect on some biochemical parameters and histopathology in mice. Our findings suggested that EF daily consumption for 24 weeks, at higher doses than the 0.8% EF diet (35 times of human consumption), might cause adverse effect on kidney function in mice.

  • 새로운 제초성 N-phenyl-3,4-dimethylphthalimide 유도체의 구조변화에 따른 물리-화학 파라미터와 다루어진 바 없는 TOPKAT 프로그램으로 계산된 랫트 및 마우스 등의 급만성 독성에 관한 판별점수(DS) 및 치사율과의 관계(QSTR)를 정량적으로 검토하였다. 그 결과, 발암성은 랫트보다 마우스가 그리고 수컷보다는 암컷이 높은 경향이었다. $R_2$-기만이 변화하는 조건에서 Hansch-Fujita 식을 유도한 결과, 발암성에서 랫트 암컷을 제외한 마우스(암, 수) 및 랫트 수컷은 공통적으로 LUMO 에너지가 영향을 미치는 주 요인이었으며 마우스 암컷과 수컷의 발암성에 관한 선택성 요소는 주로 $R_2$-치환기 길이의 적정값(약 $(L)_{opt.}=5.0{\AA}$)에 의존적이었다. 또한, Free-Wilson 식으로부터 $R_2$-기의 기여도는 랫트 수컷의 경우, 탄화수소로 구성된 치환체가 그리고 그 이외의 경우에는 불소 치환체들의 기여도가 우세한 경향이었다.
