Evaluation of Antioxidant, Anti-cholinesterase, and Anti-inflammatory Effects of Culinary Mushroom Pleurotus pulmonarius

  • Culinary mushroom Pleurotus pulmonarius has been popular in Asian countries. In this study, the anti-oxidant, cholinesterase, and inflammation inhibitory activities of methanol extract (ME) of fruiting bodies of P. pulmonarius were evaluted. The 1,1-diphenyl-2-picryl-hydrazy free radical scavenging activity of ME at 2.0 mg/mL was comparable to that of butylated hydroxytoluene, the standard reference. The ME exhibited significantly higher hydroxyl radical scavenging activity than butylated hydroxytoluene. ME showed slightly lower but moderate inhibitory activity against acetylcholinesterase (AChE) and butyrylcholinesterase than galantamine, a standard AChE inhibitor. It also exhibited protective effect against cytotoxicity to PC-12 cells induced by glutamate ($10{\sim}100{\mu}g/mL$), inhibitory effect on nitric oxide (NO) production and inducible nitric oxide synthase protein expression in lipopolysaccharide-stimulated RAW 264.7 macrophages, and carrageenan-induced paw edema in a rat model. High-performance liquid chromatography analysis revealed the ME of P. pulmonarius contained at least 10 phenolic compounds and some of them were identified by the comparison with known standard phenolics. Taken together, our results demonstrate that fruiting bodies of P. pulmonarius possess antioxidant, anti-cholinesterase, and inflammation inhibitory activities.

An In Vitro and In Vivo Cholinesterase Inhibitory Activity of Pistacia khinjuk and Allium sativum Essential Oils

  • Objectives: Alzheimer's disease (AD), an overwhelming neurodegenerative disease, has deleterious effects on the brain that consequently causes memory loss and language impairment. This study was intended to investigate the neuroprotective activity of the two essential oils (EOs) from Iranian Pistacia khinjuk (PK) leaves and Allium sativum (AS) cloves against β-Amyloid 25-35 (Aβ25-35) induced elevation of cholinesterase enzymes in AD. Methods: The EOs of PK (PKEO) and AS (ASEO) were prepared and analyzed in terms of extraction yield, phenolic content, and cholinergic markers in vitro. Moreover, both were administered orally to adult male Wistar rats at concentrations of 1, 2, and 3%. The inhibitory potential of PKEO and ASEO was compared with Donepezil (0.75 mg/kg) against the high activities of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzymes. Results: PKEO reached an inhibition rate of 83.6% and 81.4% against AChE and BChE, respectively. ASEO had lower anti-cholinesterase activity (65.4% and 31.5% for the inhibition AChE and BChE). PKEO was found to have more phenolic content than ASEO. A significantly positive correlation was observed between the total phenolics and anti-cholinesterase potential. In rats, both EOs decreased the enzyme activity in a concentration-dependent manner. As compared with Donepezil, the significant difference in the AChE and BChE inhibition occurred as rats were treated with PKEO 3% (p < 0.05). Conclusion: It could be concluded that PKEO and ASEO are potent inhibitors of AChE and BChE in rats that hold promise to be used for the treatment of AD.

Antioxidant and Cholinesterase Inhibitory Activities of the By-products of Three Pandalid Shrimps

  • The antioxidant and cholinesterase inhibitory activities of the acetone and dichloromethane ($CH_2Cl_2$) extracts of the by-products (heads, shells, and tails) of Pandalus borealis, Pandalus hypsinotus, and Pandalopsis japonica belonging to the family Pandalidae were investigated and their bioactivities were compared. The antioxidant and cholinesterase inhibitory activities of the organic solvent extracts of three shrimp by-products were evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-Azino-bis[3-ethylbenzothiazoline-6-sulfonic acid] ($ABTS^+$) radical scavenging activities, reducing power and xanthine oxidase (XO) inhibitory activity assays and Ellman's colorimetric method. The extracts of P. hypsinotus exhibited the highest antioxidant and cholinesterase inhibitory activities. The acetone extracts showed more potent activities toward antioxidant and cholinesterase inhibition compared with the $CH_2Cl_2$ extracts. Furthermore, the total carotenoid contents of the acetone extracts were higher than those of the $CH_2Cl_2$ extracts. Thus, the carotenoid contents may affect antioxidant and cholinesterase inhibition. Our results suggest that the shrimp by-products could act as a nutraceutical agent to prevent oxidative stress and Alzheimer's disease.

클로르피리포스 제조업장의 기중 농도와 작업자의 적혈구 콜린에스테라제 활성도 (Airborne Chlorpyrifos Concentrations and RBC Cholinesterase Activity of Workers in Its Formulation Workplace)

  • Airborne chlorpyrifos concentrations in formulation workplaces have not been determined in Korea. The aim of the study was to evaluate chlorpyrifos concentrations of air in a formulation workplace and recognize the RBC cholinesterase activities for the formulation workers. 30 air samples (personal or area sampling) were collected and bood samples from 10 workers were collected for RBC chlorinesterase (RBC AChE) activity in a factory on May 2008. Air samples were collected by the National Institute for Occupational Safety and Health (NIOSH) method and were analyzed by GC-MS. Airborne chlorpyrifos concentrations ranged from 0.003 to $12.90mg/m^3$ and geometric mean (GM) was $0.15\;mg/m^3.$ Compared to Korean Occupational Exposure Limit (KOEL) of 0.1 (2011) or $0.2\;mg/m^3$ (2008), at 95% confidence, airborne concentrations exceeded the KOEL 69.7% or 56.1% of the time or less, indicating that this concentration level was unacceptable according to exposure assessment using a LogNorm2$^{(R)}$. Since the workers were continually at work on the organophosphate or carbamate pesticides formulation, individual baseline for RBC AChE activity was not determined. As the results of comparison with reference average value of RBC AChE activity, it was found that a worker was below 70% RBC AChE activity and five workers were abnormal.

수은 폭로 근로자의 혈중 Zinc-Protoporphyrin 및 혈청 Cholinesterase Activity에 관한 조사 연구 (A Study on the Blood Zinc-Protoporphrin and Serum Cholinesterase Activity of Workers Exposed to Mercury Vapor)

  • This study aims at investigating the relationships between the urinary mercury concentration and blood zinc-protoportphyrin, serum cholinestrase activity, making 149 workers exposed to mercury vapor and 68 workers who were not exposed to mercury among the workers in a flurorescent lamp manufactureing factory an object of this investigation. The results are as follows ; 1. In an exposed group the number of those whose urinary mercury concentration showed over $100{\mu}g/l$ was 21 persons (14.3%) among 147 workers. The average urinary mercury concentration was $52.1{\pm}46.1{\mu}g/l$($1.8-361.2{\mu}g/l$), which proved to be higher than the average concentration in a control group. 2. In an exposed group, the average concentration of blood zinc-protoporphyrin was $27.8{\pm}12.5{\mu}g/dl$($12.2-101.5{\mu}g/dl$), which proved to be somewhat higher than the average concentration in a control group. But it did not show a significant difference. 3. In an exposed group, the average concentration of serum cholinesterase activity showed $1936.7{\pm}341.0IU/l$(1,120,0-2,8750IU/l), which proved to be lower than the average concentration in a control group. 4. The relational coefficient between urinary mercury concentration and blool zinc-protoporphyrin, serum cholinesterase activity of the whole workers exposed to mercury showed little difference. While the relational coefficient between the urinary mercury concentration and blood zinc-protoporphyrin of the workers whose urinary mercury concentration showed over $100{\mu}g/l$ was relatively high, which was 0.62.

유기린계 살충제가 흰쥐의 Cholinesterase활성에 미치는 영향 (Effects of Organophosphorous Insecticides on Mice Cholinesterase Activity)

  • Changes in cholinesterase (ChE) activity, electrophoretic pattern of ChE and histopathologic state on the mice serum, brain and liver by administration of organophos-phorous insecticides were studied. The mice ChE activities on serum, brain and liver decreased by increasing of concentration and time both administration of malathion and DDVP, whereas on serum and brain the activities of the 7 days after administration decreased, and then presented the gradually slight recovery in course of time. The ChE on serum and liver showed many isozyme bands by polyacrylamide gel electrophoresis but several on brain. And isozyme bands disappeared and diffused by administration of organophosphorous insecticides and development of time. The mice liver with administration group of malathion on histopathologic test showed midzonal necrosis between central vein and portal area, and with administration group of DDVP mainly presented portal necrosis on location of potal area.

흡입물질이 흰쥐 Lactate Dehydrogenase와 Cholinesterase 활성변화에 미치는 영향 (Effects of Volatile Substances on Rat Lactate Dehydrogenase and Cholinesterase)

  • The effects of volatile substances inhalation on lactate dehydrogenase and cholinesterase in rats were investigated. Male Sprague-Dawley rats were exposed to marketed odorant, ethyl acetate and ethyl ether for 15 days. Enzyme activities were measured in serum and several tissues such as liver, lung, brain, heart, kidney and muscle to find differences of effects according to the organ. Cholinesterase activity in serum and most of tissues revealed time-dependent decrease in the case of marketed odorant inhalation. Especially in heart and kidney significant decrease was observed. Ethyl acetate exposure to rats revealed also decrease in serum and all tissues by 40% to 60%. Ethyl ether inhalation showed significant decrease by 30% to 50%. Lactate dehydrogenase activity was markedly increased in serum and similarly in heart, brain and kidney by exposure to marketed odorant. No changes were observed in liver. Ethyl acetate exposure to rats revealed increase in serum by about 200%, compared to normal group and in other tissues by 40% to 70% except in liver and muscle. Ethyl ether inhalation showed significant increase in serum by about 100%. There was no change in 'liver and slight increase in muscle.

Cholinesterase Inhibitory Activities of Alkaloids from Corydalis Tuber

  • Several isoquinoline alkaloids (1 - 18), which have basic chemical structures as protoberberine and aporphine skeletones, were evaluated for their inhibitory activities on AChE and BuChE. Among them, compounds 3, 4, 6, 8 and 12 showed the potent AchE activity with the $IC_{50}$ values ranging from $10.2{\pm}0.5\;{\mu}M$ to $24.5{\pm}1.6\;{\mu}M$, meanwhile, compound 14 - 17 exhibited strong inhibitory activity with $IC_{50}$ values from $2.1{\pm}0.2$ to $5.5{\pm}0.3\;{\mu}M$. Compounds 14 - 17 exhibited selective inhibition for AChE compared with BuChE. The isoquinoline alkaloid possesses aromatic methylenedioxy groups and quaternary nitrogen atoms are crucial for the anti-cholinesterase inhibitory activity.

말뚝버섯 자실체의 항산화, 항콜린에스테라제 및 염증 저해 활성 (Antioxidant, anti-cholinesterase, and inflammation inhibitory activities of fruiting bodies of Phallus impudicus var. impudicus L.)

  • 본 연구에서는 말뚝버섯의 자실체 메탄올 추출물의 항산화, cholinesterase 저해 및 항염증 효과를 탐색하였다. DPPH 라디칼 소거능, hydroxyl radical 소거능, 철 이온 제거능 및 환원력 등의 항산화 효과를 측정한 결과 DPPH 라디칼 소거능, hydroxyl radical 소거능 및 환원력은 양성대조군으로 사용한 BHT에 비해 낮았으나 실험에 시용한 2.0 mg/ml의 농도에서 50% 이상의 저해효과를 나타내었고 철 이온 제거능은 BHT에 비해 높게 나타나서 다른 종류의 식의약용 버섯에 비해 항산화 효과가 우수하였다. 치매환자의 기억력 감퇴와 관련된 acetylcholinesterase와 butyrylcholinesterase의 저해실험에서 말뚝버섯 자실체의 메탄올 추출물은 실험에 사용한 전 농도 범위에서 양성대조군인 galanthamine에 비해 유의하게 낮았지만 1.0 mg/ml의 농도에서 50% 이상의 저해 효과를 나타냈다. In vitro 항염증 실험에서 RAW 264.7 대식세포에 서로 다른 농도의 메탄올 추출물을 처리한 후 염증 유발물질인 LPS를 처리하여 RAW 264.7 세포가 생성한 NO의 양을 측정한 결과 추출물을 투여한 실험군의 NO 농도가 LPS만 단독으로 처리한 양성대조군에 비해 유의하게 낮았고 처리한 메탄올 추출물의 농도가 증가함에 따라 생성된 NO의 양은 유의하게 감소하는 경향을 나타났다. 또한 in vivo 항염증 실험에서 먼저 각기 다른 농도의 메탄올 추출물을 생쥐의 뒷발에 주사한 후 추가로 기염제인 carrageenan을 주사하여 흰쥐 뒷발에 유도된 부종 (edema)이 추출물에 의해 저해되는 정도와 염증 치료제로 처방되는 indomethacin을 양성대조군으로 하여 실험을 진행하였다. 실험 결과 흰쥐에 주사한 말뚝버섯 자실체의 메탄올 추출물의 농도가 증가함에 따라 흰쥐 뒷발에 유도된 부종의 용적도 농도 의존적으로 유의하게 감소하는 것이 관찰되어 말뚝버섯 자실체에는 염증을 저해하는 성분이 함유되어 있는 것으로 사료되었다. 따라서 말뚝버섯 자실체의 메탄올 추출물에는 항산화, acetylcholinesterase과 butyrylcholinesterase의 저해 및 항염증 효과를 나타내는 유용 성분이 함유되어 있어 앞으로 의약용 기초 소재로서의 이용 연구가 필요하다고 사료된다.

농업인에서의 체내 잔류농약 검출 (Serum cholinesterase activities and serum bromide concentrations in farmers)

  • 배경 : 농약의 사용이 보편화되면서 점차 그 사용량이 증가되고 있다. 저자들은 농한기의 농업인을 대상으로 국내에서 가장 많이 사용되는 맹독성 농약인 methyl bromide와 organophosphate compound의 농업인 체내 잔류 정도를 혈청 bromide 농도와 cholinesterase 활성도도 측정을 통해 조사하고자 하였다. 방법 : 농한기에 경상남도 산청군과 하동군에 거주하는 121명의 농업인을 대상으로 농번기의 농약사용에 관한 설문조사와 농약의 잔류정도를 측정하기 위한 혈액을 채취하였다. 대조군으로는 간 기능이 정상이며 B형 간염 항원이 없는 사람 중에서 연령과 성별에 맞게 무작위로 240명을 선별하였다. 농업인과 대조군에서 혈중 bromide는 혈청을 이용하여 gold chloride방법으로 구하고, cholinesterase는 colorimetric assay로 측정하였다. 대상 농업인중에서 간기능 검사가 이상 소견을 보이거나 B형 간염 항원 양성인 33명과 이에 해당하는 대조군 65명은 비교대상에서 제외하였다. 결과 : 정상인 240명을 대상으로 산정한 혈청 bromide 농도의 참고치는 72.9 ug/mL이하, 혈청 cholinesterase 활성도의 참고치는 1.6-15.9 U/mL 이었다. 정상인 175명의 혈청 bromide 농도는 $29.8{\pm}22.5\;{\mu}g/mL$, 혈청 cholinesterase 활성도는 $8.7{\pm}3.6$ U/mL이었다. 농업인의 혈청 bromide 농도는 $31.7{\pm}18.0\;{\mu}g/mL$로 대조군과 차이를 보이지 않았고, 혈청 cholinesterase도는 $10.9{\pm}4.2$ U/mL로 대조군에 비하여 유의하게 높았다. 농업인을 경작 면적, 농약 사용시 장갑 및 마스크 착용 유무에 따라 분휴류여 비교해 본 결과, 혈청 bromide 농도나 혈청 cholinesterase 활성도 차이가 없었다. 결론 : 농한기 농업이과 정상인을 비교해 보았을 때 methyl bromide 중독시 증가하는 혈청 bromide 농도는 차이가 없었으며, organophosphate compound 중독시 감소되는 cholinesterase는 농한기 농업인에서 대조군보다 높은 값을 보였다. 따라서 혈청 bromide 농도와 cholinesterase 활성도는 농한기 농업인에서 농약의 체내잔류 지표로 유용성이 낮다고 사료된다.
