The influence of (-0-Hydroxycitrate(HCA), shown to be a competitive inhibitor of adenosine 5-triphosphate(ATP) citrate lyase, on food intake and body weight, serum triglyceride and cholosterol level, in vivo rates of fatty acid and cholesterol synthesis, and fat cell number and size was investigated. 3 groups of female, 5 weeks old Sprague Dawley rats, 8 animals each, were ad libitum meal-fed or pair-fed(3 hours from 10 : 00 to 13 : 00) AIN based high glucose diet for a total period of 8 weeks. Providing normolipidemic rats orally with 400mg of HCA formula containing approximately 20mg of HCA 1 hour prior to daily feeding schedule significantly depressed in vivo hepatic rates of fatty acid and cholesterol synthesis in the liver and adipose tissue. Serum triglyceride and cholesterol levels were significantly reduced by HCA. At the end of treatment period, the rats administered with HCA resulted in a significantly reduction in body weight gain. The reduction in weights was attributable to a significant decrease in fat cell size with a smaller extent, but not significant, reduction in fat cell number. Rats receiving HCA demonstrated less food intake than the controls ; however, this decreased caloric intake was not fully responsible for the HCA induced depression of hepatic and adipocytic lipogenesis, since experiment using pair-fed cojntrol rats showed, less magnitude but similar results. Both a anorectic and an antilipogenic properties of HCA seem to be responsible for this weight reduction activity of HCA. The outcome of this study suggest that metabolic regulation may be a feasible approach to the control of obesity and hyperlipidemia.

To study effects of wild garlic on components of lipid and protein, blood glucose level, and cholinesterase activity in rat's serum, Sprague-Dawley strain of 40 male rats are divided into 5 groups which are Normal, Control, A, B, and C group. Normal group is fed only Basal diet, and Control group is fed Basal diet and 0.5ml/day of 2.5% cholesterol solution, In addition to a diet of control group, A group is fed 25% wild garlic flour, B group 0.5ml/2day of concentratded wild garlic juice, C group 0.5ml/2day of concentrated ethanol extract of wild garlic, After 8 weesk the rats were fasted for 12 hours, and then decapitated to collect blood. The results of analysis of the rat's serum were summarized as follows, 1. Wild garlic diet, specially ethanol extract has an influence on decreasing the level of total cholesterol and blood glucose in rat's serum, 2. HDL-cholesterol and phospholipid content of rat's serum are increased by wild garlic diet. Therefore I think that wild garlic is good food for preventing development of atherosclerosis and diabetes.

This study was designed to investigate the effect of dietary vitamin E levels on lipid metabolism in caffeine-fed rats. Male Sprague-Dawley rats were fed on diet containing one of three levels of vitamin E(37.5, 750, or 1,500mg/kg diet0 which was with or without 0.3% caffeine. The rats were sacrificed after 5 and 10 weeks of the feed periods. Results obtained from this study were as follows ; Net weight gain, feed intake and FER in the caffeine added groups were significantly decreased as compared to those of only vitamin E fed group. Liver and kidney weights tended to increase in the caffeine added groups, but spleen and heart weights were not affected. Total lipid contents in serum and liver tended to decrease as dietary vitamin E became increasing and caffeine diet adding. Serum total cholesterol content tended to increase in the caffeine added group. But, liver cholesterol content were decreased. And the higher the dietary vitamin E level became, the lower serum and liver cholesterol contents were. Serum and liver triglyceride contents were significantly lower in the caffeine added groups than in the only vitamin E fed groups. The degree of decrease was more evident as dietary vitamin E level became more increasing. Serum aminotransferase activity was not affected in all experimental groups.

Effect of potato polyphenolic compound on the concentrations of serum and liver lipids, serum glucose, and urine protein was investigated in Sprague Dawley rats by feeding a diet containing 0.5% of cholesterol for 2 weeks. Polyphenolic compound extracted from potato (Solanum tuberosum variety Dejima) was supplemented at a 0.5% level in the basal diet or the cholesterol diet. The supplementation of potato polyphenolic compounds decreased slightly the concentrations of total cholesterol and VLDL+LDL-cholesterol in serum, and atherosclerotic index in rats fed the cholesterol diet, while those measurements were not altered by the supplementation of potato polyphenolic compounds in rats fed the basal diet. In the rats fed the basal diet or the cholesterol diet containing potato polyphenolic compounds, urine protein increased by 12% and 27% at 1st week, respectively, but this change was not seen at 2nd week. The concentration of serum glucose, however, was not significantly different in the dietary groups.
BACKGROUND/OBJECTIVE: The aim of this study was to examine the effect of high dietary methionine (Met) consumption on plasma and hepatic oxidative stress and dyslipidemia in chronic ethanol fed rats. MATERIALS/METHODS: Male Wistar rats were fed control or ethanol-containing liquid diets supplemented without (E group) or with DL-Met at 0.6% (EM1 group) or 0.8% (EM2 group) for five weeks. Plasma aminothiols, lipids, malondialdehyde (MDA), alanine aminotransferase (ALT), and aspartate aminotransferase were measured. Hepatic folate, S-adenosylmethionine (SAM), and S-adenosylhomocysteine (SAH) were measured. RESULTS: DL-Met supplementation was found to increase plasma levels of homocysteine (Hcy), triglyceride (TG), total cholesterol (TC), and MDA compared to rats fed ethanol alone and decrease plasma ALT. However, DL-Met supplementation did not significantly change plasma levels of HDL-cholesterol, cysteine, cysteinylglycine, and glutathione. In addition, DL-Met supplementation increased hepatic levels of folate, SAM, SAH, and SAM:SAH ratio. Our data showed that DL-Met supplementation can increase plasma oxidative stress and atherogenic effects by elevating plasma Hcy, TG, and TC in ethanol-fed rats. CONCLUSION: The present results demonstrate that Met supplementation increases plasma oxidative stress and atherogenic effects by inducing dyslipidemia and hyperhomocysteinemia in ethanol-fed rats.
This study was performed to investigate the increase of S-allylcysteine content, a biomarker of black garlic, during its aging period, as well as the serum lipid-lowering effects of black garlic extract in high fat diet fed rats. The highest content of S-allylcysteine was observed on the 15th day of the aging period. Sensory evaluation was also estimated to be extremely good on the 15th day of the aging period. High fat diet rats with induced hyperlipidemia were fed diets containing black garlic extract of low, medium, and high doses for 6 weeks. No significant difference in body weight gain and food efficiency was observed between normal, placebo and black garlic fed groups. Liver weight was significantly higher in black garlic fed groups than in the normal group. Total serum cholesterol and triglyceride were significantly lower in low, medium, and high dose groups than in the normal group. Also, HDL-cholesterol was significantly higher and LDL-cholesterol was significantly lower in black garlic diet fed groups than in the normal group. Hepatic levels including total lipid and cholesterol were especially decreased in the black garlic diet fed group than in the placebo group. These results suggest that black garlic intake reduces the levels of serum and hepatic cholesterol in high fat diet fed rats. In conclusion, black garlic has a potential to be used as a functional health food ingredient with beneficial effects on lowering cholesterol and triglyceride levels.
The purpose of this study was to investigate the effect of Lycii fructus on the serum lipid in rats fed high fat diet. We compared the effects of L. fructus and L.fructus water extract both adminstered with high fat diets on rats that had previously been on high fat or standard diets. Two separate experiments were conducted for 6 weeks. respectively. In experiment I, 4 groups of rats were fed experimental diets consisting of either \circled1 6 weeks of a standard diet(control), \circled2 6 weeks of a high-fat diet(HHC), \circled3 3 weeks of a high-fat diet followed by 3 weeks of a high-fat diet containing L. fructus(HHL) or \circled4 6 weeks of a high-fat diet with L. fructus extract in place of water for the last 3 weeks (HHT). In the second set of experiments, a high-fat diet (SHC), high-fat diet containing L.fructus(SHL) or high-fat diet with L. fructus extract in place of water (SHT) were fed for 3 weeks after 3 weeks of standard diet feeding. Rats fed L. fructus diet consumed more diets than high-fat diets. THe results of experiment I showed significant decreases(p<0.05) in serum triglyceride(TB) and low density lipoprotein-cholesterol (LDL-C) levels with L. fructus feedings, but did not show andy changes in total cholesterol (TC) level. High density lipoprotein-cholesterol (HDL-C) level was increased significantly(p<0.05) with L. fructus feedings. Therefore, the ration of LDL-C to HDL-C(LDL-C/HDL-C) which is used as an atherosclerosis index was significantly (p<0.05) low, while the HDL-c/TC ration was significantly(p<0.05) high with L.fructus intake. However, no significant were found in serum cholesterols and TG levels in experimentII. The results of these experiments indicate that , regardless of the feeding from, L. fructus can be beneficial in lowering serum TG and LDL-C levels for habitual high-fat diet intakers. L.fructus also seems to be effective in elevating serum HDL-C level, theregy having beneficial effects on atherosclerosis by influencing the serum lipoprotein profile.
In this experiment, three different diets were produced to investigate the effects of Jeju native chicken meat, from chickens that were fed a tangerine by-product, on physiological activities in rats. The first diet did not contain any chicken (TS), the second diet contained 10% chicken that had not been fed the tangerine by-product (T0), and the third diet contained 10% that had been fed the by-products (T1). These diets were provided to 11-week-old male rats for four weeks. Weight gain, feed intake, feed efficiency, liver, kidney, and epididymis fat weights were not significantly different among the TS, T0, and T1 groups. Total lipid, triglycerides, and cholesterol levels in the liver were significantly lower in T0 and T1 than in TS (p<0.05). And total lipid, phospholipid, triglycerides, total cholesterol, HDL-cholesterol, DL+VLDL-cholesterol, HDL-cholesterol/total cholesterol, and atherogenic index levels in the blood serum were similar between T0 and T1, which did not present any significant differences. The feed containing the tangerine by-product did not cause any statistically significant differences in serum protein, glucose, or hemoglobin. Finally, T0 and T1 showed similar trends in terms of $\gamma$-GTP, ALT, AST, and ALP activities, which again did not present any statistically significant differences.
This study was conducted to investigate the effects of shellfish hydrolysate on lipid metabolism in rats fed high fat diet. Male Sprague-Dawley rate weighting approximately 110g were fed basal control diet, high fat diet and high fat diet plus 4 different shellfish hydrolyates for 4 weeks. The shellfish hydrolysates from the different sources, were oyster, hard-shelled mussel, little neck clam and march clam. After 4 weeks, serum GOT, GPT, ${\gamma}$-GTP, triglyceride and total cholesterol was significantly decreased in shellfish hydrolysates supplementation with high fat diet compared to basal control and high-fat group(p<0.05). The total lipid and cholesterol content in liver showed significant decrease(p, 0.05). There were no different in serum GPT, HDL-cholesterol, liver total cholesterol and lipid of rats between basal control diet and high-fat diet. The unsaturated fatty acids, specific components of shellfishes were a little components in shellfish hydrolysate as they were a low and not different among the groups and were most well reflected in liver and plasma. Considering digestive and absorptive process of in human body, it was assumed that the hypolipidemic effect of shellfish was not under the influence of unsaturated fatty acids but the other components, peptides, taurine and betaine and so on was detected in the process of hyperlipidemia induced by high-fat diet.
Cladosiphon okamuranus is edible brown algae cultured commercially and massively at Okinawa coast. Dietary fiber occupying most of C. okamuranus isn't digested and absorbed by digestive enzymes of human. But it is known to prevent and treat constipation as metabolic disease. This study was to investigate the effects of dietary fiber extracted from C. okamuranus on the damage of liver and the blood cholesterol level in the high fat diet-fed rats. The effects were measured by the levels of aspartate aminotransferase (ALT), alanine aminotransferase (AST), total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL) and high density lipoprotein (HDL) in sera. The levels of ALT, AST, TC, TG and LDL were significantly reduced in the C. okamuranus-treated group compared with the high fat diet group. But HDL level was markedly increased. The results showed that the dietary fiber extracts from C. okamuranus have the inhibitive effects of cholesterol biosynthesis in the high fat diet-fed rats.
