Our previous report has suggested that the decrease of fibronectin level during mvogenesis is due to the decreased Bvailabilitv of receptor (matrix assembly receptor) for 29-kDa fragment of fibronectin. In the present study, we demonstrate that G protein and adenvlate cvclase system are involved in the regulation of fibronectin matrix assembly and that when fibronectin level in extracellular matrix decreases, the postmitotic fusion-capable cells emerge more frequently from the proliferative population. This proposal is based on the following observations. (1) Cholers toxin, which increases intracellular CAMP, caused a decrease in the ability of mvoblasts to incorporate fibronectin into extracellular matrix. (2) Cholera toxin decreased the proliferation of mvoblasts and Induced the precocious fusion. (3) decAMP, which was found to induce the precocious fusion and decrease the proliferation of myoblasts, decreased the fibronectin level in extracellular matrix and matrix assembly receptor for fibronectin- (4) RGOS, whlh inhibits the incorporation of fibronectin into extracellular matrix, induced the precocious fusion and reduced the proliferaton of mvoblasts. These results suggest that CAMP regulates the fibronectin levels in extracellular matrix and that the alteration of fibronectin level is involved in regulation of the proliferation and differentiation of chick embryonic mvoblasts.
In the present study, the effect of intrathecal (i.t.) or intracerebroventricular (i.c.v.) administration with cholera toxin (CTX) on the blood glucose level was examined in ICR mice. The i.t. treatment with CTX alone for 24 h dose-dependently increased the blood glucose level. However, i.c.v. treatment with CTX for 24 h did not affect the blood glucose level. When mice were orally fed with D-glucose (2 g/kg), the blood glucose level reached to a maximum level at 30 min and almost returned to the control level at 120 min after D-glucose feeding. I.c.v. pretreatment with CTX increased the blood glucose level in a potentiative manner, whereas i.t. pretreatment with CTX increased the blood glucose level in an additive manner in a D-glucose fed group. In addition, the blood glucose level was increased in formalin-induced pain animal model. I.c.v. pretreatment with CTX enhanced the blood glucose level in a potentiative manner in formalin-induced pain animal model. On the other hand, i.t. pretreatment with CTX increased the blood glucose level in an additive manner in formalin-induced pain animal model. Our results suggest that CTX administered supraspinally or spinally differentially modulates the regulation of the blood glucose level in D-glucose fed model as well as in formalin-induced pain model.
The PI-oduction of virulence factors such as cholera toxin, heinolysin and hemagglutinin in V cliolerae non-01 and non-0139 were examined. Among 65 strains isolated from environmental and clinical blood sources, 29 (14.6%) strains produced hemolysin only, 35(53.9%) sh.ains produced both hemolysin and hemagglutinin. From one 037 slrain isolated from environmenl, cholera toxin, ctx gene, hemolysin, and hemagglutinin were detected. All of the strains isolated from clinical and environmental sources showed hemolytic activity against human 0 group e~ythrocytes. In inhibition patterns of heinagglotination, 5 of 18 clinical strains (27.8%) were inhibited by less than 1% mannose and galactose, while, among the 47 environmental isolates. hose paltems by less than 1% mannose and galactose 55.4% wel-e inhibited. Thel-ehre, exohamagglutinin positive rate was high in clinical blood isolates but in environnlental sources, the rate was almost similar lo ihe rate or endohemagglutinin positive. These results indicaled that V cholerae non-01 and non-0139 produced various virnlence factors such as cholera toxin, hemolysin, and hemagglutinin but not a single factor. Further studies are need for epidemiological or bacteriological shtdies of V cholerae 037 isolated from environment.

Vibrio cholerae is a very important pathogenic bacterium that has to be monitored in seafood and ships' ballast water. Various methods have been developed to identify this bacterium, yet these methods are time-consuming and have limitations for their sensitivity to detect contamination. The purpose of the present study was to develop a robust and reliable method for identifying V. cholerae. Peptide nucleic acid (PNA) probes were developed to use for PNA-based asymmetrical real-time PCR techniques. The toxigenic Cholera enterotoxin subunit B (ctxB) gene was selected as a target for detecting V. cholerae and the gene was synthesized as a positive template for conventional and real-time PCR. Real-time PCR primers and PNA probes were designed and standard curves were produced for the quantitative analysis. The selected PNA probes reacted specifically to V. cholerae without any ambiguity, even among closely related species, and the detection limit was 0.1 cfu/100 mL. Taken together, the PNA probes and asymmetrical qPCR methods developed in this present study could contribute to the rapid, accurate monitoring of V. cholerae in marine environments, and as well as in seafood and ships' ballast waters.

Red pine (Pinus densiflora) is widely used traditional medicine, pharmacological and nutritional values from which the phytochemical compounds are derived. The present study was aimed to examine the antibacterial effects in the absence and presence of a immature red pine cone extract against 13 microorganisms. The components in the aqueous extract from immature red pine cone were identified by GC-MS. About 1.4% of total polyphenolic compound was measured in aqueous extract collected from immature red pine cone. Also, the high concentration of ${\beta}$-phellenandrene, ${\alpha}$-pinene, limonene, bornyl acetate and aldehyde was detected in total ion chromatograms. Of total 13 microorganisms, 4 microorganisms including Pseudomonas aeruginosa, Vibrio cholera, Listeria monocytogenes, Klebsiella pneumonia were effectively killed by aqueous extract of immature red pine cone. The highest anti-bacterial effect was detected in P. aeruginosa, followed by V. cholera, L. monocytogenes and K. pneumonia. In case of P. aeruginosa, the largest diameter of inhibition zone was maintained to 1/2 solution treated cells and slightly decreased at 1/4 and 1/8 solution treated cells. Also, in test used V. cholera and L. monocytogenes, the inhibition zone was strongly formed in only 1 and 1/2 solution treated cells, while K. pneumonia showed the very small diameter of inhibition zone in all concentrations. Therefore, these results suggested that the aqueous extracts of immature red pine cone should be considered as a new and potentially important anti-bacterial substrate to effectively prevent the microbial infection and penetration.
Vibrio cholerae, cause of the life-threatening diarrheal disease cholera, can be divided into different serogroups based on the structure of its lipopolysaccharide (LPS), which consists of lipid-A, core-polysaccharide and O-antigen polysaccharide (O-PS). The O1 serogroup, the predominant cause of cholera, includes two major serotypes, Inaba and Ogawa. These serotypes are differentiated by the presence of a single 2-O-methyl group in the upstream terminal perosamine of the Ogawa O-PS, which is absent in the Inaba O-PS. To ensure the consistent quality and efficacy of the current cholera vaccines, accurate measurement and characterization of each of these two serotypes is highly important. In this study, we efficiently screened a phage-displayed human synthetic Fab library by bio-panning against Ogawa LPS and finally selected three unique mAbs (D9, E11, and F7) that specifically react with Ogawa LPS. The mAbs bound to Vibrio cholerae vaccine in a dose-dependent fashion. Sequence and structure analyses of antibody paratopes suggest that IgG D9 might have the same fine specificity as that of the murine mAbs, which were shown to bind to the upstream terminal perosamine of Ogawa O-PS, whereas IgGs F7 and E11 showed some different characteristics in the paratopes. To our knowledge, this study is the first to demonstrate the generation of Ogawa-specific mAbs using phage display technology. The mAbs will be useful for identification and quantification of Ogawa LPS in multivalent V. cholerae vaccines.
Of mushrooms found in Japan, approximately 35 species are classified as poisonous mushrooms. The mushroom poisonings caused by these mushrooms are classified as three types accodring to the symptoms and their toxic constitutents. The first type is the cholera-like symptom which is induced mainly by the toxic constitutents such as cyclopeptide and gyromitrin. The second type is the neurological mainfestations induced by muscarine (cholinergic), muscimol (delirium), psilocybin (hallucinogenic) and coprine (antabuse-like).
