갓에서 Chlorophyll 유도체 및 $\beta$-Carotene의 분리와 이들의 항산화 작용 (Isolation of Chlorophyll Derivatives and $\beta$-Carotene from Mustard Leaf and Their Antioxidative Activities on the Lipid Autoxidation)

  • Chlorophyll a and b, pheophytin a and b and $\beta$-carotene, crude chlorophylls and carotenoids (CCC) extracts of mustard leaf kimchi were isolated by DEAE-sepharose CL-6B and Sepharose CL-6B colume and TLC. The effects of chlorophyll a and b, pheophytin a and b and $\beta$-carotene on linoleic acid autoxidation were examined by the determination of peroxide value and conjugated dienoic acid content. Among them, chlorophyll a showed greater antioxidative activity than others, followed by chlorophyll b, pheophytin a, pheophytin b and $\beta$-carotene. Degradation of pheophytin b was observed to be slower than others and $\beta$-carotene showed highest degradation level during the autoxidation reaction of linoleic acid.

  • 크롬에 의한 무와 배추 유식물의 생리학적 반응을 조사하기 위하여 수경재배 조건하에서 크롬용액의 농도에 따른 무와 배추종자의 발아율 세포신장, ${\alpha}$-amylase활성도, 엽록소, 단백질함량의 차이를 비교하였다. 크롬처리농도가 증가함에 따라 농도의존적으로 무와 배추종자의 발아율, 세포신장, ${\alpha}$-amylase 활성도가 감소하였는데 배추에서 보다 무에서 그 저해정도가 심하게 나타났다. 무와 배추에서 크롬처리시 약간의 chlorophyll 함량 감소가 있었으며 chlorophyll a가 b보다 더 저해를 받았다. 크롬농도 증가에 따른 ${\alpha}$-amylase 활성도, chlorophyll a와 b, 단백질함량과의 관계를 다중회귀분석한 결과 단백질> ${\alpha}$-amylase>chlorophyll a>chlorophyll b의 순서로 저해를 받았다.

Nondestructive and Rapid Estimation of Chlorophyll Content in Rye Leaf Using Digital Camera

  • We have developed and tested a new method for nondestructive estimation of chlorophyll- and nitrogen-contents in rye leaf. It was found that the relation-ships among nitrogen, chlorophyll content and fresh weight were significantly positive correlated. Nitrogen and chlorophyll content were positively correlated whereas correlation coefficients among R, G, R-B and G-B on the basis of photo-numerical values were negative. We have found that R/(R-B) obtained from data of digital camera is the best criterion to estimate the chlorophyll content of leaves. The regression curves of the relation between R/(R-B) and chlorophyll content were also calculated from the data collected on cloudy days. The coefficients of determination ($\textrm{r}^2$) were ranged from 0.33 to 0.99. In this study, the accuracy in estimating chlorophyll content from the color data of digital camera image could be improved by correcting with R, G, and B values. It is suggested that, for practical purposes, the image values estimated with sufficient accuracy using a portable digital camera can be applied for determining chlorophyll content and nitrogen status in plant leaves.

Physiological and Biochemical Responses of Fifteen Rice Cultivars to UV-B Radiation

  • This study was conducted to examine the physiological and biochemical responses against UV-B radiation in the seedling of 15 different rice cultivars, having the different physiological sensitivities. Out of 15 rice cultivars tested, moderate and susceptible groups showed significant decreases in biomass and RGR (relative growth rate). Contents of total chlorophyll were reduced remarkedly by irradiation of UV-B. In all rice cultivars tested, the content of chlorophyll a was strongly decreased, while the contents of chlorophyll b were slightly reduced without showing clear different among three groups and 15 cultivars. Carotenoid content was largely reduced by UV-B radiation, whereas polyamine content was moderately increased. The contents of MDA (malondialdehyde) that reflect the level of lipid peroxidation of cell membranes were clearly increased by UV-B stress, showing higher content in susceptible cultivars than moderate and torelant cultivars. The physiological important parameters highly related to visible injury were leaf color, chlorophyll, carotenoid, and lipid peroxidation, whereas biomass and polyamines were not closely correlated. Based on this results, it was concluded that changes of visible injury and the contents of chlorophyll and MDA could be adequately applied and utilized as physiological indicators to UV-B radiation.

한국산 녹차 5종의 물의 온도 및 우린 횟수별에 따른 Chlorophyll 함량의 변화 (The Changes of Chlorophyll Contents in 5 Korean Green Teas by the Extraction Temperatures and the Number of Soaking)

  • This study was carried out to examine the changes of chlorophyll a and b contents by the extraction temperatures and the number of soaking using five kinds of Korean green teas(3 kinds of green tea and 2 kinds of tea bag) which were obtained from a local market in Daegu city, Korea. The chlorophylls were extracted with 80% acetone, and determined by a visible spectrophotometer. The concentration of chlorophyll in Sulrok(tea bag) were the highest among all kinds of teas when they were extracted at 60, and decreased with the number of soaking increased. On the other hand, the chlorophyll content of green teas was the highest at 80, and decreased with the increase of the number of soaking as the tea bag did. It was found that the Korean tea bag contained more chlorophyll than green teas did, and green teas contained significantly more chlorophyll b than chlorophyll a.

Greening에 따른 유채 자엽의 엽록소-단백질 복합체 형성 (Formation of Chlorophyll-Protein Complexes in Greening Rape Cotyledons)

  • The formation of chlorophyll-protein complexes (CP-complexes) during the greening of rape cotyledons (Brassica napus cv. Yongdang) was investigated by the SDS-polyacrylamide gel electrophoresis. The total chlorophyll content and Chl a/b ratio were also determined. In addition, the effects of dark treatment on the CP-complex patterns during greening have been examined with respect to their photosynthetic electron transport activity. Greening has brought about the increasein total chlorophyll content and the decrease in Chl a/b ratio, but there have been no changes in Chl a/b ratio after 24 hrs of greening. The light-harvesting chlorophyll a/b-protein complex (LHCP-complex0 was predominant during the initial greening period. Thereafter, the amout of chlorophyll a-protein complex (CP I-complex) was gradually increased. Twenty-four-hr dark treatment immediately after illumination for 6 hrs and 12 hrs resulted in the increase of the Chl a/b ration and the CP I complex, otherwise the decrease of the LHCP-complex. The LHCP/CP I ratio was gradually decreased with further greening, and appeared no change after 48 hrs illumination. The investigation of the photosynthetic electron transport activity indicated that photosystem (PS) II activity (H2Olongrightarrowp-PD*+FeCy**) did not change, but the activity of PS I was increased suddenly due to the dark treatment. The data suggests that the increase of CP I-complex may result in that of P-700, that is, the increase of PS I activity.

N,N-Dimethylformamide와 Dimethylsulfoxide를 이용한 감귤 잎의 엽록소 측정방법 확립 (Establishment of Analytical Method for Chlorophyll Using the N,N-Dimethylformamide and Dimethylsulfoxide in Citrus Leaves)

  • BACKGROUND: Purpose of the study was to establish the extinction coefficients of chlorophyll a and b in N,N-dimethylformamide(DMF) and Dimethylsulfoxide(DMSO) solvents and to find out the conditions of optimal extraction temperature and time in citrus leaves. METHODS AND RESULTS: Chlorophyll a and b standards were dissolved in DMF and DMSO. Extinction coefficients of chlorophyll pigments were determined and their contents were quantified using spectrophotometer. Chlorophyllous pigments of citrus(Citrus unshiu Marc. cv. Okitsu wase) leaves were extracted at 25, 40, 60 and $80^{\circ}C$ for 4, 6, 8, 24 and 48 hours to determine the optimal extraction condition. CONCLUSION: The extinction coefficients of chlorophyll a(Chl a) and chlorophyll b(Chl b) of DMF extracts for high extinction wavelength were 663.8 and 647.2 nm. Similarly, the high extinction wavelength of DMSO extracts were 665.8 and 649.0 nm for chl a and b respectively. Chl a, Chl b and total chlorophyll content of DMF extracts were Chl a = $12.10A_{663.8}-2.74A_{647.2}$, Chl b = $21.94A_{647.2}-5.06A_{663.8}$ and total $Chl=19.193A_{647.2}+7.04A_{663.8}$. Similarly, Chl a, Chl b and total Chl of DMSO extracts were Chl a = $14.53A_{665.8}-5.40A_{649.0}$, Chl b = $26.98A_{649.0}-7.11A_{665.8}$ and total $Chl=21.58A_{649.0}+7.43A_{665.8}$. The chlorophyll extracts of DMF and DMSO were very stable in dark. High chlorophyll contents of citrus leaves were found at $80^{\circ}C$ extraction for 6 hours in DMF and at $80^{\circ}C$ extraction for 24 hours in DMSO. However, the chlorophyll content was decreased significantly after 8 hours in DMF extraction while it was remained up to 30 hours in DMSO extraction.

고려인삼 Chlorophyll a/b Binding Protein(Cab) 유전자의 동정 및 분자적인 특성분석 (Molecular Characterization of a cDNA Encoding Chlorophyll a/b Binding Protein (Cab) from Panax ginseng C. A. Meyer)

  • 광계II(PSII)는 고등식물의 chloroplast에서 두 개의 광합성 반응중심 중의 하나이다. Chlorophyll a/b 광수확 복합체는 광계II를 위한 안테나 역할을 수행한다. 본 연구에서는 인삼의 잎조직을 제작한 cDNA library로부터 chlorophyll a/b-binding protein (Cab) 유전자를 분리하였다. 인삼 Cab유전자는 935 bp의 염기와 265개의 아미노산 잔기(pI 5.63)로 구성된 한 개의 ORF를 포함하고 있으며, 단백질의 분자량은 28.6 kDa으로 추정되었다. 인삼에서 분리한 Cab 유전자는 기존에 식물에서 보고된 유전자들과 유사성을 나타내었으며, 유사도는 $68-92\%$로 나타났다. 아미노산 서열을 비교하여 유연관계를 분석한 결과 인삼의 Cab 유전자는 비교된 P. persica (AAC34983), A.thaliana (AAD28771), G. hirsutum (CAA38025), G. max (AAL29886), V. radiata (AAF89205) 등과 동일한 그룹으로 분리되었다.

오존에 노출된 덩굴식물류, 쇠뜨기, 쑥의 광색소 함량과 SOD 활성 변화 (Photosynthetic Pigment Concentrations and Changes of SOD Activities on Liana, Equisetum Arvense and Artemisia Princeps Exposured to Ozone)

  • 덩굴식물 개머루, 계요등, 까마귀머루, 담쟁이덩굴, 댕댕이덩굴, 인동덩굴, 하늘타리 등 7수종과 덩굴식물이 아닌 쇠뜨기, 쑥 등 2수종을 이용하여 광색소 함량과 SOD활성에 대하여 분석한 결과는 다음과 같다. 100 ppb 처리구에서 볼 때 엽록소 a, 엽록소 b, 총 엽록소 함량, 카로테노이드에서 담쟁이덩굴이 가장 많이 증가하였으며, 엽록소 b와 a의 비에서는 쇠뜨기, 카로테노이드와 총 엽록소의 비에서는 계요 등에서 가장 많이 증가하였다. 덩굴식물과 덩굴식물이 아닌 쇠뜨기, 쑥의 비교에서는 엽록소 b와 엽록소 a의 비에서 확실한 차이를 보였다. 엽록소 a와 b의 비에서보면 엽록소 b보다 엽록소 a가 더 민감한 차이를 보였으며, 수종에서는 담쟁이덩굴, 덩굴식물과 덩굴식물이 아닌 쇠뜨기, 쑥의 비교에서는 덩굴식물이 민감한 것으로 나타났다. SOD 활성에서는 100 ppb 처리구에서 덩굴식물류 7수종과 쇠뜨기, 쑥의 처리전과 처리후의 차이는 쑥에서 3535.7 unit/g로 매우 크게 증가하였으며, 계요 등이 109.3 unit/g로 가장 적게 증가하였고, 개머루 한 수종에서 131.7 unit/g로 감소했다.

비료와 가뭄 스트레스에 의한 부추의 엽록소 변화 (Changes in the Chlorophyll of Garlic Chives (Allium tuberosum) Resulting fromFertilizer and Drought Stress)

  • 부추는 아시아를 포함한 전세계적으로 분포하는 초본이다. 한국에서 부추는 양념용 채소와 국수용 고명에 사용된다. 질소비료, 인산비료, 칼리비료와 수분 스트레스가 부추(Allium tuberosum Rotter) 잎에 미치는 영향에 대해 조사하였다. 엽록소 a 함량은 10 mg/l 질소 비료에서 0.386였고, 50 mg/l에서는 0.584였다. 엽록소 a, b, 전체 엽록소량은 대조군에 비해 유의한 차이를 나타내었다(p<0.05). 엽록소 a, b, 전체 엽록소에 대한 피어슨 상관계수(Pearson's r)는 각각 0.940, 0.966, 0.971였다. 잎의 엽록소 a, b, 전체 엽록소는 인산비료인 경우 40 mg/l가 50 mg/l 시비보다 높았다. 인산 비료인 경우 10 mg/l에서 전체 엽록소량은 0.312였고 50 mg/l 일 때 0.589였다. 엽록소 효율과 습도와의 관계를 산출하였다. 광합성 효율의 척도로 기저 수준의 형광산율(Fo), 최대 형광산율(Fm), 전자소멸 상태(Fv), 최대 PSII 광계비율(Fv/Fm)의 기울기는 각각 -0.931, 0.972, 972, 0.950였다. 질소, 인산, 칼륨 비료와 가뭄스트레스는 부추의 엽록소함량과 효율성에 영향을 주었다.
