Insecticidal activity of the crude extract and its fractions of Custard apple (Annona reticulata L.) (커스타드애플(Annona reticulata L.)씨 추출물과 그 분획물의 살충활성)

  • In recent years, many agricultural scientists are studying on eco-friendly farming methods. Among of lots of the methods, the natural insecticides are highly motivated and interested due to their safety and biodegradable issues, and readily available source of bioinsecticides. In this study, the crude extract of custard apple (Annona reticulata L.) seed and its three fractions which were separated based on polarity indexes were examined for their insecticidal activities against Myzus persicae Sulzer and Nilaparvata lugens S.. The crude extract (Aceton/MeOH) showed strong insecticidal activities against both insects at 3.00 mg/mL ($LD_{50}$=0.45mg/mL for M. persicae S. and 1.42 mg/mL for N. lugens S.). Furthermore, simple fractionation with hexane, chloroform, and water lead to increase three-folds insecticidal activity on chloroform fraction ($LD_{50}$=0.13mg/mL for M. persicae S. and 1.14 mg/mL for N. lugens S.). The results suggest that A. reticulata extracts might be used to control for M. persicae effectively.

Antioxidative Effects Oil Pueraria Root Extracts (갈근 추출물의 항산화효과)

  • In order to investigate possible utilization as a naturall antioxidant, antioxidative fraction from pueraria root powder was obtained by solvent extraction. PO,CO and TBA values were measured at $60^{\circ}C$, $100^{\circ}C$, $145^{\circ}C$, respectively, after adding the pueraria root extract to the final concentration of 0.1% to the lard, beef tallow, pallm and soybean oil. The results of antioxidative effect of pueraria root extract to edible oils and fats were as follows : 1. Antioxidative fractions in pueraria root were extracted by sequential solvent systems using ethanol, methanol : isopropyl alcohol (1: 4) and chloroform : methanol(95 : 5). 2. Antioxidative activity of the extracts to edible oils and fats was more effective than that of adding 100 ppm $\alpha$-tocopherol at $60^{\circ}C$. 3. At $60^{\circ}C$ and $100^{\circ}C$, antioxidative effect was beef tallow>palm oil>lard>soybean oil in its orders. 4. At $145^{\circ}C$, the pueraria root extract showed antioxidative activity in beef tallow, lard and palm oil, but not in soybean oil. 5. The content of saturated fatty acids by the heat treatment was remained smalll change, but that of unsaturated fatty acids was noticeable decreased.

Antimicrobial Effect of Citrus unshiu Markovich Extracts on Food-Borne Pathogens (청피 추출물이 식중독 유발 미생물의 증식에 미치는 영향)

  • This study was performed to investigate the antimicrobial effects of the Citrus unshiu Markovich extracts against food-borne pathogens. First, the Citrus unshiu Markovich was extracted with methanol at room temperature, and fractionation of the methanol extracts from Citrus unshiu Markovich was carried out by using petroleum ether, chloroform, ethyl acetate, and methanol. The antimicrobial activity of the Citrus unshiu Markovich extracts was determined using the paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of Citrus unshiu Markovich showed the highest antimicrobial activity against Bacillus cereus and Shigella sonnei. A synergistic effect was found in combined extracts of Citrus unshiu Markovich and Hedyotis diffusa Willd as compared to each extract alone. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Citrus unshiu Markovich against Bacillus cereus and Shigella sonnei. The ethyl acetate extract of Citrus unshiu Markovich showed strong antimicrobial activity against Bacillus cereus at the concentration of 5,000 ppm. The 5,000 ppm of ethyl acetate extract from Citrus unshiu Markovich retarded the growth of Bacillus cereus more than 24 hours and Shigella sonnei up to 24 hours. The ethyl acetate extracts of Citrus unshiu Markovich have shown an antimicrobial effect against Bacillus cereus and Shigella sonnei.

The Effect of Oriental Herbs on Growth and Aflatoxin Production by Aspergillus parasititus R-176 (한약재가 Aspergillus parasiticus R-716의 생육과 aflatoxin 생성에 미치는 영향)

  • The possible effeets of some oriental herbs, whleh have been used to treat eaneerllke dllleue In Korea, on growth and afIatonn production by Aspergillus parasiticus R-716 were investigated. Zedoaria rhizoma, Curcuma longa, Cyperus rotundus, Angelica gigas, Paeonia albiflora, Paeonia mountan, Atractylis ovata, and Pulsatilla koreana were extracted with chloroform. Among them the extract of paeonia mountan was remarkably effective on the growth inhibition, and Curcuma longa, Zedoaria rhizoma, Cyperus rotundus, Paeonia albiflora, Atractylis ovata also inhibited the growth. The ertraet of Atractylis ovata and Curcuma longa, also Inhibited the afIatoxin production but the others showed no effect at all or sometimes stimulated effect. With the addition of O.2ml extract of Atractylis ovata in 30ml SLS medium, the growth was delayed for about 2 days, and after 9 days, mycellum weight was 0.953g, and total afatoxin was reduced 5O%($792\mu\textrm{g}$)of that produced in the control($1547\mu\textrm{g}$). Aflatoxin per mycelium weight was decreased 32%($992\mu\textrm{g}$) of that produced in the control($1467\mu\textrm{g}$), but NADPH oxidase was higher as compared to the control. The extract of Atractylis ovata appeared to have a inhibitory effect on the growth and the aflatoxin production of Aspergillus parasiticus R-716.

Synergistic Antimicrobial Effect of Patrinia scabiosaefolia and Forsythiae fructus Extracts on Food-borne Pathogens (식중독 유발 세균의 증식에 미치는 패장과 연교 추출물의 상승 효과)

  • To investigate the antimicrobial effect of the Patrinia scabiosaefolia extracts against food-borne pathogens, we extracted the P. scabiosaefolia with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, ethyl acetate, and methanol, respectively. The antimicrobial activity of the P. scabiosaefolia extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of P. scabiosaefolia showed the highest antimicrobial activity against Escherichia coli and Shigella sonnei. Synergistic effect in inhibition was observed when P. scabiosaefolia extract was mixed Forsythiae fructus extract as compared to each extracts alone. Finally, the growth inhibition curves were determined by using ethyl acetate extracts of P. scabiosaefolia against Staphylococcus epidermidis and Shigella sonnei. The ethyl acetate extract of P. scabiosaefolia had strong antimicrobial activity against S. sonnei at the concentration of 4,000 ppm. At this concentration, the growth of S. Sonnei was retarded more than 72 hours and up to 48 hours for S. epidermidis. These results suggest that the ethyl acetate extracts of P. scabiosaefolia can be used for the efficient material against the growth of S. epidermidis and S. sonnei.

Chemical Components and Physiological Activities of Young Mulberry(Morus alba) Stem (뽕나무 어린줄기의 화학성분 및 생리활성)

  • In order to exploit as a new functional decocted beverage, chemical components, antioxidative and antimicrobial activities young stems of mulberry(Morus alba) were surveyed. The proximate composition was composed of crude fiber 51.12%, ash 13.46%, total sugar 10.38%, crude fat 9.10% and crude protein 5.01%. The P(295.9 mg%) was the highest mineral found in young stem of mulberry and Ca 289.6 mg%, K 209.6 mg%, Na 58.3 mg%, Mg 45.0 mg% and Fe 4.6 mg% in that order. Free sugars was composed of glucose 1.08%, galactose 022%, sucrose 0.20% and fructose 0.16%. Eight fatty acids in stem of mulberry were identified and the major fatty acids were linoleic acid(46.10%), palmitic acid(27.84%) and linolenic acid(10.85%). Among the 17 amino acids detected, total amino acid was 2,450.5 mg% and proline(313.7 mg%) was the most predominant. Methanol extract and ethyl acetate fraction showed stronger activity of the hydrogen donating activities, each of 77.24% and 80.08%, respectively. The methanol extract from young stem of mulberry showed the strongest antimicrobial activities to Bacillus subtitis and Bacitus cereus. Chloroform and ethyl acetate fractions from methanol extract of young stem showed a inhibition zone of 9.0∼19.0 ㎜ in diameter against pathogen bacteria.

Antioxidation and Antigenotoxic Effects of Buckwheat Sprout Extracts (메밀싹 추출물의 항산화 효과 및 유전독성억제 효과)

  • This study was carried out to determine the antioxidative and antigenotoxic effects of buckwheat (Fagopyrum esculentum Moench) sprout using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical donating method and micronucleus test. Buckwheat sprout were extracted with 70% ethanol and then further fractionated to n-hexane, chloroform, ethyl acetate (EtOAc), butanol and water. Among the five fractions, the EtOAc fraction showed the highest electron donating activity ($RC_{50}$ 26.1 ${\mu}g/mL$). The effects of buckwheat sprout extracts on the frequencies of micronucleated polychromatic erythrocytes (MNPCEs) induced by MNNG (N-methyl-N'-nitro-N-nitrosoguanidine) were investigated in the bone marrow. 10, 20, 40 and 80 mg/kg of each extract were administered to animals immediately after injection of MNNG and the exposure time was 36 hrs. Inhibition effects of buckwheat sprout ethanol extract were 23.4%, 40.6%, 56.3% and 73.4%, respectively. When the fraction of hexane, chloroform, ethyl acetate, butanol and water from 70% ethanol extract were treated with concentration of 80 mg/kg, the suppression rates of the MNPCE were 64.1, 67.9, 75.8, 74.2 and 63.3%, respectively.

Impacts of Various Solvent Extracts from Wild Haw(Crataegus pinnatifida Bunge) Pulpy on the Antioxidative Activities (야생산사(Crataegus pinnatifida Bunge)과육 용매별 추출물의 항산화 효과)

  • Haw(Crataegus pinnatifida Bunge) belongs to the rose family and has beneficial health effects such as anti-obesity and gastroprotective activities. This study compared the antioxidative activities of various solvent extracts from haw. After removing seeds, pulpy was extracted with 70% methanol, 70% ethanol, chloroform :methanol(CM, 2:1, v/v), n-butanol, and ethyl acetate(EA). Total phenol contents were 71.46, 70.41, 47.63, 47.49 and 45.95 mg/g in the 70% methanol, 70% ethanol, CM, n-butanol and EA extracts, respectively. Total flavonoid contents of CM and n-butanol extracts were higher compared to other extracts at 1.63 mg/g and 1.59 mg/g, respectively, whereas EA extract had the lowest flavonoid content at 1.12 mg/g. Corresponding to total phenol contents, among $NO_2$ radical scavenging activity, antioxidant activity by ${\beta}$-carotene bleaching assay, and superoxide dismutase(SOD)-like ability, 70% methanol and 70% ethanol extracts showed higher antioxidative activities than the other extracts. However, the lowest activities were n-butanol extract in $NO_2$ radical scavenging activity as well as EA extract in antioxidant activity by ${\beta}$-carotene bleaching assay and SOD-like ability. These results suggest that haw extracts are available as a new natural antioxidant, and its activities are attributed to antioxidant substances such as phenolic compounds and flavonoids.

Isolation and Identification of an Antioxidant Substance from Ethanol Extract of Wild Grape (Vitis coignetiea) Seed (머루종자 에탄올 추출물로부터 항산화활성물질 분리 및 동정)

  • Antioxidant compound(s) were identified from the ethanol extract of wild grape (Vitis coignetiea) seed. Organic solvent fractions of n-hexane, chloroform, ethyl acetate and butanol were obtained from the ethanol extract of wild grape seed, among which ethyl acetate fraction showed the strongest reducing power. Ethyl acetate fraction was further purified through ODS column chromatography and HPLC, and isolated antioxidative active compound was identified through $^1H-NMR$ as (+)-catechin (52.7 g/100 g). (+)-Catechin and ethyl acetate fraction both showed approximately 80% scavenging effect. These results indicated (+)-catechin in the ethyl acetate fraction synergetically interacts with unknown antioxidative compound(s).

Effects of Turmeric (Curcuma longa L.) Bioactivity Compounds and Lipid Peroxidation Inhibitory Action (울금(Curcuma longa L.)의 생리활성 및 지질과산화 저해능에 미치는 영향)

  • The aim was to determine the physiological activity and antioxidant activity by lipid peroxidation inhibitory action of turmeric (Curcuma longa L.). Bioactive compound of total carotenoid $1.581{\pm}0.005mg$ ${\beta}$-carotene equivalents (BCE)/g dry weight. Total phenol content was the highest in the ethyl acetate (EA) extract, followed by chloroform:methanol (CM, 2:1, v/v) and 70% methanol extracts. Antioxidant effects (nitrogen oxide radical scavenging activity, nitrite scavenging activity, ${\beta}$-carotene bleaching assay, and lipid peroxidation inhibition action) of 70% methanol, CM, and EA extract of turmeric. Turmeric extracts yield were 70% methanol 16.54%, CM 5.64%, and EA 4.14%, respectively. Antioxidant activity of the samples exhibited a dose-dependent increase. However, in the current study, none of the samples evaluated showed activity as strong as the BHA (butylated hydroxyanisole) and trolox. Further, nitrite scavenging activity was the highest for the EA extract. As a result of this experiment, indicating their commercial value and potential applications in food and nutraceuticals.
