Studies on natural products are of great interest, due to the limits in development of synthesized medicine and its side effects. Deer antler is the most popular cure-all type drug among Asian folk medicines. In this study, we newly isolated the biologically active components from chloroform extract and 70% ethanol extract of deer antler, and analyzed their structures. First, the structure of monoacetyldiglyceride in deer antler was identified. To investigate the structure-activity relationship of monoacetyldiglycerides, we synthesized diverse substituted glycerides from glycerol, and confirmed their structures by spectroscopic methods. Among seven structurally-interesting compounds tested in this study, compound 1,2,3,5, and 6 showed activity toward [Ca$\^$2+/]$\_$i/ increase in fura-2 loaded rat pancreatic acinar cells. Second, 70% ethanol extract of deer antler stimulated insulin release from rat pancreatic islets. We found the most effective fraction was CN-Es-8 in 70% ethanol extract, and it increased intracellular Ca$\^$2+/ concentration in pancreatic ${\beta}$-cell.

This study was performed to investigate the taste composition and antioxidant properties of cheonggukjang containing Korean red ginseng (RGC), as compared to either general cheonggukjang (GC) or non-fermented boiled soybeans (BS). Amylase activity was the highest (576.7 unit/g) in RGC, whereas protease activity was the highest (326.0 unit/g) in GC. The total soluble sugar contents of BS, GC, and RGC were 2,027.5, 905.5, and 837.5 mg/100 g, respectively. RGC had the highest amount of total amino acids (2,127.4 mg/100 g) and essential amino acid (50.9%) among the samples. The ratio of sweet to bitter components was higher in RGC than in GC. Although the extracts of RGC had higher radical scavenging activity for 1,1-diphenyl-2-picrylhydrazyl (DPPH) than BS or GC, regardless of the extract concentration, the ethanol extract of RGC showed the highest scavenging ability (92.4%) at 2.0 mg/mL. The chloroform extracts from GC and RGC showed their greatest superoxide dimutase-like activities at 17.2 and 19.7% at a concentration of 2 mg/mL, respectively. Regardless of the samples, the nitrite scavenging ability was positively correlated to the extract concentration, and RGC had highest ability among samples under the same extract concentrations.
This study was carried out to investigate whether Chrysanthermum zawadskii var. latilobum Kitamura (C. zawadskii) extracts has an inhibitory effect against the mutagenicity by cigarette smoke condensates (CSC). C. zawadskii was extracted with 70% ethanol and the yield was 18.5%. We further fractioned 70% ethanol extract sequentially to diethylether, chloroform, dichloromethane, and aqueous water, and gained the yield of 17.5%, 5.6%, 5.8%, 32.8% and 35.5%, respectively. In the Ames test, there was no mutagenic effect of crude extract and its solvent fractions up to 2 mg/plate toward Salmonella typhimurium TA 98 with or without S-9 mix metabolic activations. On the contrary, the crude extract showed an inhibitory activity against the mutagenicity of CSC in the presence of S-9 mix metabolic activation. Diethyl ether layer among five solvent fractions showed the highest inhibitory activity. The inhibitory activity of diethyl ether fraction was also increased in a dose-dependent manner and the inhibitory rate was about 97.7% at the concentration of 1 mg/plate. In this study, we conclude that crude extract of C. zawadskii itself is potentially safe for mutagenicity, and the diethyl ether fraction has an inhibitory effect against the mutagenicity of CSC.

This study was carried out to investigate the antioxidant activities, total phenol, and flavonoid contents of 70% methanol, 70% ethanol, and chloroform-methanol (CM, 2:1, v/v) extracts from garlic (Allium sativum L.) peels. Antioxidant activity was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) radical and ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, ${\beta}$-carotene bleaching assay, and ferric reducing antioxidant power. Total phenol and flavonoid were in a range of 32.59-63.05 mg CAE/g, 5.26-9.22 mg QE/g, respectively. Total phenol and flavonoid were predominant with the values of $63.05{\pm}0.20mgCAE/g$ and $9.22{\pm}0.17mgQE/g$ in 70% ethanol extract. Additionally, 70% ethanol extract also displayed the strongest antioxidant activity followed by 70% methanol extract and CM extract in all assays. These results showed that garlic peel can be used as natural antioxidant in nutraceutical preparations to prevent human diseases.
This study was performed to investigate the antimicrobial effect of the Viscum album var. coloratum extracts against food-borne pathogens. First, the Viscum album var. coloratum was extracted with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol, respectively. The antimicrobial activity of the Viscum album var. coloratum extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The petroleum ether extracts of Viscum album var. coloratum showed the highest antimicrobial activity against Bacillus cereus and Shigella dysenteriae. Synergistic effect in inhibition was observed when Viscum album var. coloratum extract was mixed with Perillae folium extract as compared with each extract alone. Finally, the growth inhibition curves were determined by using petroleum ether extracts of Viscum album var. coloratum against Bacillus cereus and Shigella dysenteriae. The petroleum ether extract of Viscum album var. coloratum had strong antimicrobial activity against Bacillus cereus at the concentration of 5,000 ppm. At this concentration, the growth of Bacillus cereus was retarded more than 24 hours and up to 12 hours for Shigella dysenteriae. In conclusion, the petroleum ether extracts of Viscum album var. coloratum inhibit efficiently Bacillus cereus and Shigella dysenteriae.
In order to examine the biofunctions of glycosylceramide which is representative of sphingolipid, monoglycosylceramide (cerebroside) was isolated from rice bran extract. Crude glycosylceramides were isolated in large quantities and promptly by flash system column chromatography from rice bran extract, and purified by normal-phase HPLC using an evaporative light-scattering detector. One major cerebroside was obtained by HPLC used as an eluent consisting of chloroform, methanol and water (99:11:1, v/v/v), and the constituents were analyzed by MALDI/TOF-MS, FAB-MS, GC and 600 MHz $^1$H-NMR. The component sugar was estimated to be glucose. In the ceramide, the fatty acid component consist was 2-hydroxy arachidic acid. The long-chain base component was sphinga-4,8-dienine.
Objectives: The quorum-sensing-inhibitory and anti-biofilm activities of the methanol extract of E. globulus leaves were determined against clinically isolated multidrug-resistant Pseudomonas aeruginosa. Methods: The preliminary anti-quorum-sensing (AQS) activity of eucalyptus was investigated against a biosensor strain Chromobacterium violaceum ATCC 12472 (CV12472) by using the agar well diffusion method. The effect of sub-minimum inhibitory concentrations (sub-MICs) of the methanol extract of eucalyptus on different quorum-sensing-regulated virulence factors, such as swarming motility, pyocyanin pigment, exopolysaccharide (EPS), and biofilm formation, against clinical isolates (CIs 2, 3, and 4) and reference PA01 of Pseudomonas aeruginosa were determined using the swarm diameter (mm)-measurement method, chloroform extraction method, phenol (5%)-sulphuric acid (concentrated) method, and the microtiter plate assay respectively, and the inhibition (%) in formation were calculated. Results: The preliminary AQS activity (violacein pigment inhibition) of eucalyptus was confirmed against Chromobacterium violaceum ATCC 12472 (CV12472). The eucalyptus extract also showed concentration-dependent inhibition (%) of swarming motility, pyocyanin pigment, EPS, and biofilm formation in different CIs and PA01 of P. aeruginosa. Conclusion: Our results revealed the effectiveness of the E. globulus extract for the regulation of quorum-sensing-dependent virulence factors and biofilm formation at a reduced dose (sub-MICs) and suggest that E. globulus may be a therapeutic agent for curing and controlling bacterial infection and thereby reducing the possibility of resistance development in pathogenic strains.
This study was performed to investigate the inhibitory effect of Aloe vera on the growth and aflatoxin production of Aspergillus parasiticus. Spore suspension of A. parasiticus ATCC 15517 was inoculated on the yeast-extract sucrose broth containing 0.1%, 0.5%, 1.0% and 10.0% of chloroform extract of Aloe vera and then incubated at 30$\circ$C for 7 days. Mycelial weight was 160.7 mg/5ml in control group and decreased by the addition of the extract with no significance. The mold caused decrease in pH of the media with and without the extract. pH in the group contained 10.0% of the extract showed significantly higher value of 5.10 than that of 4.90 in control group (p<0.05). Fluorescence spots of four aflatoxins were observed under the 365 nm of UV light after extraction of the media and TLC. In the result of separation and determination by HPLC, the aflatoxins were produced in the order of $B_1, G_1, B_2$ and $G_2$ in all groups. Production of aflatoxins $B_1, B_2$ and $G_1$ was reduced by the addition of the extract and decreased as amount of the extract increased. The production of aflatoxins $B_1$ and $B_2$ significantly reduced when the media contained more than 1.0% of the extract, and $G_1$ more than 0.5%, respectively(p<0.05). No reduction and no significant difference among groups were observed in case of aflatoxin $G_2$. With the above result, the extract of Aloe vera reduced the production of aflatoxin by A. parasiticus though it did not inhibit mycelial growth.

The purpose of this study was to measure the antioxidant and physiological activities of seed from Gardenia jasminoides Ellis fructus (GJE) in Namhae Korea. We determined proanthocyanidin. GJE seed were extracted by 70% ethanol, chloroform:methanol, 2:1 volume ratio (CM) and n-butanol of three solvents. To investigate by the solvent extract of flavonoid content and value as a functional food ingredient of GJE seed through various antioxidant activities were performed. Solvent extract antioxidant activity of increasing concentrations (0.2, 0.4, 0.6 mg/mL) were significantly increased (p<0.05). The content of proanthocyanidin in GJE seed was $70.035{\pm}0.772mg$ catechin eqivalents/g dry weight. As a result of bioactivity assay, by a solvent of seed were found that the relationship with the increase of flavonoid content increased bioactivities. The antioxidant activity of the extract from the other except for the n-butanol extract on seed was observed at a high level. The results suggest that Gardenia jasminoides Ellis fructus seed can be used as a natural antioxidant.

Several extracts of different parts and solvent fractions of Cornus kousa were obtained and their functional material contents, antioxidant activities and tyrosinase inhibition effects were determined. Content of total polyphenols and flavonoids contents in flower were 169.638 $mg{\cdot}g^{-1}$ and 25.418 $mg{\cdot}g^{-1}$, respectively, which were much higher than those of other parts. Also, flower extracts showed the strongest effects on DPPH and ABTS radicals scavenging and ferrous ion chelating. In flower, leaf, and stem extracts, inhibition effects on peroxidation of linoleic acid determined by ferric thiocyanate(FTC) method were higher than a synthetic antioxidant, BHT. Tyrosinase inhibition activities were shown only in flower extract. Flower and leaf extracts, showing high biological activities in various system, were successively reextracted with n-hexane, chloroform, ethylacetate and n-butanol. Total polyphenol contents of water fractions were higher than any other solvent fractions in both flower and leaf, 67.006 $mg{\cdot}g^{-1}$ and 67.739 $mg{\cdot}g^{-1}$, respectively. But total flavonoid contents were higher in ethyl acetate fraction for flower extract and butanol fraction for leaf extract. Among the solvent fractions, the highest efficiency of free radical scavenging activities was obtained in ethyl acetate fraction for flower extract and n-butanol fraction for leaf extract. Tyrosinase inhibition activities were higher in water fraction for both flower and leaf extracts, 49.24% and 31.8%, respectively.
