PREPARATION OF MULTIFUNCTIONAL LOW MOLECULAR WEIGHT CHITOSAN AND ITS APPLICATION IN COSMETICS.

  • The aim of this study is to elucidate the anti-microbial activity and anti-oxidative activity of water-soluble chitosan with a molecular weight of 5,000-200,000. Water-soluble chitosans have demonstrated a regular anti-microbial activity on the tested strians by the paper disk method. In the MIC (Minimum Inhibitory Concentration) test, CC-01 (MW=5,000) with the lower MW showed the higher MIC value than the higher MW chitosan. The MW of chitosan increase, the MIC decreases. MICs of 4 chitosans(CC-02∼CC-05) against S. aureusTCC 65389, E coli ATCC 8739, p. aeruginosa, ATCC 9027 and C. albicans ,ATCC 10231 were 7.0-39.O$\mu\textrm{m}$, whereas MICs of chitosans against A. niger were over 2.OmM. Formula containing chitosan showed higher anti-microbial activities than the formula made with the chemical preservatives(Methylparaben 0.2% and Imidazolidinyl Urea 0.3%). Among 5 water-soluble chitosans, CC-03(MW=92,163) showed the most potent anti-oxidative activity (IC$\sub$50/ : 0.2mM). In conclusion, the water-soluble low molecular weight chitosan could be served as natural preservatives and antioxidant in cosmetics.

Growth Factor Releasing Porous Poly (${\varepsilon}-caprolactone$)-Chitosan Matrices for Enhanced Bone Regenerative Therapy

  • Drug releasing porous poly($\varepsilon$-caprolactone) (PCL)-chitosan matrices were fabricated for bone regenerative therapy. Porous matrices made of biodegradable polymers have been playing a crucial role as bone substitutes and as tissue-engineered scaffolds in bone regenerative therapy. The matrices provided mechanical support for the developing tissue and enhanced tissue formation by releasing active agent in controlled manner. Chitosan was employed to enhance hydrophilicity and biocompatibility of the PCL matrices. PDGF-BB was incorporated into PCL-chitosan matrices to induce enhanced bone regeneration efficacy. PCL-chitosan matrices retained a porous structure with a 100-200 $\mu$m pore diameter that was suitable for cellular migration and osteoid ingrowth. $NaHCO_3$ as a porogen was incorporated 5% ratio to polymer weight to form highly porous scaffolds. PDGF-BB was released from PCL-chitosan matrices maintaining therapeutic concentration for 4 week. High osteoblasts attachment level and proliferation was observed from PCL-chitosan matrices. Scanning electron microscopic examination indicated that cultured osteoblasts showed round form and spread pseudopods after 1 day and showed broad cytoplasmic extension after 14 days. PCL-chitosan matrices promoted bone regeneration and PDGF-BB loaded matrices obtained enhanced bone formation in rat calvarial defect. These results suggested that the PDGF-BB releasing PCL-chitosan porous matrices may be potentially used as tissue engineering scaffolds or bone substitutes with high bone regenerative efficacy.

  • In electrospraying of polymer solutions, metal sample collectors are often ineffective in fully removing solvent from sprayed particles and recovering redispersable sprayed particles. Herein, a novel electro spraying system, where sprayed particles were dispersed into laminar flow of dispersant (coagulation liquid), was designed for the nano-encapsulation of protein drugs. Chitosan and polyacrylic acid were used as the encapsulation materials. Aggregation of particles could be prevented by using this new electrospraying system, and unimodal size distribution was observed at an applied voltage between 4~16 kV and a low flow rate. The effects of the applied voltage on mean particle size were not significant on the other hand.

  • Molecular recognition technology has attracted considerable attention for improving the selectivity of a specific molecule by imprinting it on a polymer matrix. In this study, adsorption and release characteristics of chitosan based drug delivery films imprinted with sulindac (SLD) were investigated in terms of the plasticizer, temperature and pH and the results were also interpreted by the related mathematical models. The adsorption characteristics of target molecules on SLD-imprinted polymer films were better explained by the Freundlich and Sips equation than that of the Langmuir equation. The binding site energy distribution function was also useful for understanding the adsorption relationship between target molecules and polymer films. The drug release of SLD-imprinted polymer films followed the Fickian diffusion mechanism, whereas the drug release using artificial skin followed the non-Fickian diffusion behavior.

  • Chitosan, the natural biodegradable polymer derived from chitin by de- acetylation, has been widely applied to the textile finishing processes for excellent anti-microbial characteristic and handle improvement of fabric. The purpose of this study is to investigate the change of handle when cotton fabric is treated with chitosan-polyurethane mixed solution. The viscosity values of chitosan solutions were 8cps and 50cps, and the wet-pick-up% was maintained at 90%. In case of mixing with water soluble polyurethane, the mixture ratio of chitosan and polyurethane was settled on the solid content ratio of 1:0, 1:0.5, 1:1, 1:2. Also the change of physical properties by neutralization in NaOH solution was studied. The results can be summarized up as follows : 1. Extensibility(EM) and tensile energy(WT) of cotton fabric treated with chitosan are decreased, but bending rigidity(B) is remarkably increased. With the addition of polyurethane, the decrease of EM and WT is weakened and the increase of B is weakened. The case of neutralization is similar to the case of polyurethane addition. 2. By treating fabric with chitosan, FUKURAMI(Fullness and softness) is decreased, but KOSHI(Stiffness), SHARI(Crispness), HARI(Anti-drape Stiff ness) are increased. With the addition of polyurethane, the decrease of FUKURAMI is diminished and the increase of KOSHI, SHARI, HARI are diminished. 3. As the viscosity of chitosan solution increased, the air permeability value increased. The addition of polyurethane decreased the air permeability.

A Study on Adsorption of Heavy Metal Ions Using Chitosan and Chitosan Derivative (Chitosan 및 Chitosan유도체를 이용한 중금속 이온 흡착에 관한 연구)

  • We have synthesized the water-insoluble chitosan derivative, N-dithiocarboxy chitosan sodium salt, through the reaction of chitosan with carbon disulfide in the presence of alkali metal hydroxide, Chitosan itself has been prepared using chitin, one of the most abundant compounds in nature, as a starting material. To elucidate this natural polymer the capacity of adsorbing heavy metal ions, we have performed adsorption experiments using chitosan derivatives of various average molecular weights with different contents of sulfur. The effect of pH, adsorption time and temperature on adsorption efficiency was also studied. The adsorbent derived from chitosan of average molecular weight ranging $5,700{\sim}20,000$ was shown to have the highest capacity of adsorbing heavy metal ions. Adsorbing efficiency was increased as the reaction time was increased and as the reaction temperature range of $25{\sim}45^{\circ}C$. The adsorption capacity at various pH, however, appeared to vary depending on the heavy metal ions studied.

Physical Property Evaluation of Chitosan Mordanted Green Tea Dyed Cellulose - Focusing on the physical property changes upon the repetition of treatment -

  • The UV-protection effect of green-tea dyed fabrics was reported in our previous studies. The chitosan was used as a natural mordant of cellulose fiber for green tea extract because chitosan is a natural bio-polymer. The increase in the UV protection property of summer cellulose fabrics, cotton and linen, upon the repetition of chitosan mordanting and green tea dyeing was observed. However, the physical property change would be followed by this repeated wet processing of the cellulose fabric. Therefore, the physical changes of the chitosan mordanted and green tea dyed cotton and linen fabrics were evaluated by KES-FB system. Tensile, shear, bending, compression, and surface characteristics were tested upon the repetition of mordanting and dyeing treatments. Linearity of tensile force increased in the treated cotton and linen samples. Tensile energy and resilience decreased in all treated fabrics. Shear stiffness increased in the treated cotton and linen in general. Shear hysteresis was increased in all cotton samples and some linen samples. In cotton, the bending rigidity in all treated cottons increased except C3G3. As the chitosan mordanting numbers increased, the bending rigidity tended to decrease. In linen, the bending rigidity and hysteresis increased in all treated samples. Compressional energy and resilience increased as the number of chitosan mordanting increased both in cotton and linen. This could be the result of the increase in thickness upon chitosan mordanting. Surface coefficient of friction increased in the treated cotton and linen in general. Surface roughness tended to increase in cotton.

  • The changes of interfacial properties and microfailure degradation mechanisms of bioabsorbable composites with hydrolysis were investigated using micromechanical test and acoustic emission (AE). As hydrolysis time increased, the tensile strength, the modulus and the elongation of PEA and bioactive glass fibers decreased, whereas those of chitosan fiber changed little. Interfacial shear strength (IFSS) of bioactive glass fiber/poly-L-lactide (PLLA) composite was significantly higher than that two other systems. The decreasing rate of IFSS was the fastest in bioactive glass fiber/PLLA composite, whereas that of chitosan fiber/PLLA composite was the slowest. With increasing hydrolysis time, distribution of AE amplitude was narrow, and AE energy decreased gradually.

  • A interpenetrating polymer network (IPN) hydrogel, composed of chitosan (CS) and poly(diallydimethylammonium chloride) (PDADMAC) was prepared, which exhibited electrical sensitive behavior. The swelling behavior of the CS/PDADMAC SIPN hydrogel was studied by immersion of the gel into various pH buffer solutions, and their stimuli response in electric fields also investigated. In order to clarify the relationship between the equilibrium swelling ratio and bending behavior of the SIPN hydrogels, the state of water in the SIPN hydrogel was also investigated using differential scanning calorimetry (DSC).
