HPLC에 의한 식육조직중의 잔류 설파제 동시 분석 (Simultaneous Determination of Residual Sulfonamides in Meat Tissues by High Performance Liquid Chromatography)

  • Four sulfoanmides ; sulfamerazine ; sulfamethazine, sulfathiazole and sulfadimethoxine from muscle, kindney, liver and heart tissues of pork and chicken by LC. Residual sulfonamides were extracted with dichloromethane and determined on a Sperisorb ODS-1 column(250mm$\times$4.6mm id) with acetonitrile/water/acetic acid (30/70/0.3 v/v) as a mobile phase at 260nm. Recoveries from 4 tissues of pork and chicken samples fortified with 50 and 100 ppb were 71.2~87.2% and 73.7~89.6%, respectively. The detection limit was 0.03 $\mu\textrm{g}$/g in each drug. Sulfamethazine in 5 samples of pork. And sulfadimethoxine in 5 samples and sulfamethazine in 3 samples were also detected from 41 samples of chicken. The order of residue levels of sulfonamides in tissues was kidney>liver>heart>muscle, respectively. The residue levels of sulfonamides from kidney and liver were 0.03~0.15 $\mu\textrm{g}$/g in porks and 0.03~0.10 $\mu\textrm{g}$/g in chickens.

Effective Analysis of Dicyclanil in Lamb and Chicken Muscle using HPLCUV/Vis and LC/MS/MS

  • The authors describe a method for monitoring dicyclanil levels in lamb and chicken muscle tissues. The devised procedure involves dicyclanyl extraction by SPE and its detection HPLC-UV/Vis and LC/MS/MS. The method was found to have LOD and LOQ values of $0.02\;mg\;kg^{-1}$ and $0.05\sim0.06\;mg\;kg^{-1}$, respectively. The intraday precision and an accuracy of spiked samples were found to have 2.3~10.4 RSD% and 80.9~105.7%, respectively.

Differences in the Quality Characteristics between Commercial Korean Native Chickens and Broilers

  • To investigate the differences in the quality characteristics between commercial Korean native chicken (KNC) and broiler (CB), nutritive and quality parameters of the two chicken species were determined. The KNC thigh muscle had a lower content of crude fat and higher crude ash than the CB thigh. In regards to the fatty acid composition, KNC breast muscle had a higher content of arachidonic acid (C20:4) than CB. The level of inosine was higher in the CB thigh muscle than KNC but there was little difference in other nucleotide compounds. The KNC breast had higher amounts of glycine, alanine, and proline than CB, which are closely related to high quality meat flavor. The sensory acceptance was not significantly different between the breast and thigh of KNC and CB. However, KNC had higher cohesiveness, chewiness and gumminess than CB, which are indicative of a unique texture property. Based on these results, commercial KNC may have superior nutritional quality, taste, and unique texture when compared with CB. Thus, the consumer preference for KNC may be partially explained by these distinctive quality characteristics.

Optimizing Carnosine Containing Extract Preparation from Chicken Breast for Anti-glycating agents

  • Optimization of carnosine and anserine extraction from chicken breast was performed using response surface methodology (RSM) to obtain the maximized physiological activities for anti-glycation and anti-oxidation. The optimum extraction conditions were water extraction for 1.6 h in the case of the 20-wk laying hen muscle and water extraction for 2.12 h in the case of 90-wk laying hen muscle. Higher carnosine and anserine contents were measured in the 20-wk laying hen muscle, along with higher physiological activities, which increased in direct proportion with the dipeptide contents. The extracts prepared from the 20-wk laying hen under optimum conditions showed 57% inhibition of advanced glycated end-product formation, 64% inhibition of lipid peroxidation, and 61% of DPPH radical scavenging effects. On the other hand, 52% inhibition of AGE formation, 62% inhibition of lipid peroxidation, and 53% of DPPH radical scavenging effect were demonstrated within the 90-wk laying hen. In addition, the ratio of carnosine was a key indicator for the physiological activities of the extracts.

Gene Expression of Heart and Adipocyte Fatty Acid-binding Protein in Chickens by FQ-RT-PCR

  • This study was to detect the expression of heart fatty acid-binding protein (H-FABP) and adipocyte fatty acid-binding protein (A-FABP) gene mRNA in different tissues of Rugao and Luyuan chickens at 56 d and 120 d by real-time fluorescence quantitative reverse transcription polymerase-chain reaction (FQ-RT-PCR). The primers were designed according to the sequences of HFABP, A-FABP and GAPDH genes in Gallus gallus, which were used as target genes and internal reference gene, respectively. The levels of H-FABP and A-FABP gene expression were detected by SYBR Green I FQ-RT-PCR. The relative H-FABP and A-FABP gene mRNA expression level was calculated with 2-$^{{\Delta}Ct}$. Melting curve analysis showed a single peak of three genes. Intramuscular fat (IMF) content in breast muscle and leg muscle of the two chicken breeds at 120 d was higher than at 56 d. IMF content in breast muscle and leg muscle at 56 d and 120 d in Luyuan was significantly higher than in Rugao, however, abdominal fat of Luyuan was significantly lower than that of Rugao. The relative H-FABP gene mRNA expression level in cardiac muscle was the highest in both chicken breeds. The relative H-FABP and A-FABP gene expression of different tissues in Luyuan was higher than in Rugao. H-FABP gene mRNA expression had a negative effect on IMF of leg and breast muscles, and was significantly negatively correlated with IMF content. The relative A-FABP gene mRNA level in abdominal fat was higher than in liver. The A-FABP gene mRNA was not expressed in leg, breast and cardiac muscles. A-FABP gene mRNA expression level was significantly positively correlated with abdominal fat and had a significant effect on abdominal fat but not IMF content.

액체크로마토그래피를 이용한 닭고기 시료에서의 마크로라이드계 동시분석법 개발 (Determination of Four Macrolide Antibiotics Residues in Chicken Muscle Using High-Performance Liquid Chromatography)

  • 본 논문에서는 축산물 중 닭고기 내 마크로라이드계(MLs) 항생물질(spiramycin, josamycin, tilmicosin, 그리고 tylosin) 4종을 분석하기 위하여, 액-액 추출 과정을 거쳐서 PDA 검출기가 장착된 액체크로마토그래피를 이용하여 MLs를 효율적으로 동시분석하는 방법을 확립하였다. 컬럼은 Unison UK-C18 ($150mm{\times}3mm$ id, $3{\mu}m$), 이동상 용매는 0.1% TFA와 0.1% TFA in ACN로 기울기 용리를 사용하였으며, 유속은 0.7 mL/min, 그리고, 주입량은 $10{\mu}L$로 설정하여 분석하였다. 확립된 분석조건으로, 표준검정 곡선은 $50-1000{\mu}g/kg$의 농도범위에서 상관계수가 0.9975 이상의 양호한 직선성을 나타내었으며, 회수율은 저, 중, 고농도로 첨가하여 분석한 결과, 80.4-99.1%를 나타내었다. 검출한계는 $8.8-19.6{\mu}g/kg$이었고, 정량한계는 $26.6-59.4{\mu}g/kg$이었으며, 일내(intra-day)와 일간(inter-day) 정밀도(CV%)는 0.9-13.2%, 2.4-13.1%이었다. 따라서, 확립된 분석방법은 축산물 중 닭고기 시료 내 MLs를 효과적으로 분석하는데 이용될 수 있을 것으로 사료된다.

초음파 처리가 가열 계육의 이화학적 성질 및 기호성에 미치는 영향 (Effect of Ultrasonic Treatment on Physicochemical Properties and Palatability of Cooked Chicken Meat)

  • This study was carried out to investigate the effect of ultrasonic treatment on physicochemical and sensory properties of cooked chicken meat. Moisture content of ultrasonic treated breast meat was higher than that of control, fat content of control of breast and leg meat were higher than that of ultrasonic treatment, and protein of control of breast meat was higher than that of ultrasonic treatment. Hunters L(lightness)-and a(redness)-value were not different between cooking methods, L-value of breast meat and a-value of leg meat were higher without regard to cooking method. Hunterb (yellowness)-value was not different among cooking chicken meats. Cooking yield was not different between cooking methods, pH of ultrasonic treated chicken meats were higher than that of control. Water holding capacity and salt soluble protein extractability of ultrasonic treated breast meat were greater than that of control. Water soluble protein extractability of ultrasonic treated leg meat was higher than that of control, and shear force value was not different between cooking methods. Hardness, cohesiveness and gumminess were not different between cooking methods and parts of muscle. Springiness of ultrasonic treated leg meat was greater than that of control and chewiness of breast meat was higher than that of leg meat. Aroma, texture, juiciness and overall acceptability were not different between cooking methods, taste of ultrasonic treated leg meat was higher than that of control. In addition, the sensory scores of parts of muscle were affected greater than that of cooking methods.

계육의 위생화 및 안전 저장을 위한 감마선 조사 (Sanitizing and Extending of Shelf Life of Chicken Meat by Gamma Irradiation)

  • Microbial populations of total aerobic bacteria and coliforming bacteria, TBA, Hunter's color value, heme pigments, muscle protein solubility, cooking loss and shear force were investigated fro evaluating the shelf life of chicken legs gamma-irradiated at doses of 1, 3, 5 and 10 kGy with air-contained and vacuum-packaged methods. The initial microbial populations decreased with gamma irradiation depending upon the dose, and microorganisms in the vacuum-packaged samples were inhibited more than those in the air-contained samples. Hunter's L and a values of the surface and inside of the legs increased by gamma irradiation, showing a bright red color and the red color was maintained during the storage of both samples. The concentrations of oxymyoglobin among the heme pigments increased by gamma irradiation. Muscle protein solubility slightly increased by increasing the applied dose. There were no significant differences in the cooking loss and shear force values. In conclusion, the combination of gamma irradiation and vacuum-packaging could extend the shelf life of chilled chicken without deterioration of the quality.

Response of Muscle Protein Synthesis to the Infusion of Insulin-like Growth Factor-I and Fasting in Young Chickens

  • In order to elucidate the physiological function of circulating IGF-I on muscle protein synthesis in the chicken under malnutritional conditions, we administrated recombinant chicken IGF-I using a osmotic mini pump to fasted young chickens and measured the rate of muscle protein synthesis and plasma metabolite. The pumps delivered IGF-I at the rate of $22{\mu}g/d\{300{\mu}g{\cdot}(kg\;body\;weight{\cdot}d)^{-1}\}$. Fractional rate of protein synthesis in the muscle was measured using a large dose injection of L-[$2,6-^3H$]phenylalanine. Constant infusion of chicken IGF-I did not affect plasma glucose level. Significant interaction between dietary treatment and IGF-I infusion was observed in plasma NEFA and total cholesterol concentrations. When chicks were fasted, IGF-I infusion decreased plasma NEFA and total cholesterol concentrations. On the other hand, IGF-I administration did not affect plasma levels of both metabolites. Fasting reduced plasma triglyceride concentration significantly. IGF-I infusion also decreased the level of plasma triglyceride. Plasma IGF-I concentration of young chickens was halved by fasting for 1 d. IGF-I infusion using an osmotic minipump for 1 d increased plasma IGF-I concentration in fasted chicks to the level of fed chicks. Fasting decreased body weight and the loss of body weight was significantly ameliorated by IGF-I infusion. There was a significant interaction between dietary treatment and IGF-I infusion in the fractional rate of breast muscle protein synthesis. There was no effect of IGF-I infusion on muscle protein synthesis in fed chicks. Muscle protein synthesis reduced by fasting was ameliorated by IGF-I infusion, but did not reach to the level of fed control. Muscle weight of fasted chicks infused with IGF-I was similar to fasted birds without IGF-I infusion, which suggests that muscle protein degradation would be increased by IGF-I infusion as well as protein synthesis in fasted chicks.

닭 혈청을 포함한 배양액 조성 변화가 QM7 메추리 근육세포의 성장 및 분화에 미치는 영향 분석 (Effect of Culture Medium Containing Chicken Serum on Growth and Differentiation of QM7 Quail Muscle Cells)

  • 메추리 근육세포주인 QM7 세포는 다양한 연구에서 이용되고 있다. 세포배양에 있어서, 배양액의 조건은 세포의 성장과 분열에 많은 영향을 미친다고 알려져 있다. 본 연구는 QM7 근육세포 배양에 좀더 적절한 배양액 조성을 맞추어 나가기 위해 진행되었다. 이를 위해 세포배양 시, 기존에 사용하던 배양액에서 10% tryptose phosphate broth(TPB) 대신 2% chicken serum(CS)를 넣어 만든 배양액을 기존 배양액을 사용하는 경우와 비교하였다. CS를 넣어 만든 배양액에서 QM7 근육세포는 가늘고 뾰족한 모양에서 좀더 넓어지는 모양으로 바뀌었다. 또한, 기존 배양액에서보다 CS를 넣어 만든 배양액에서 더 빠르게 성장하고 분열하였다. 이는 세포 계대 후 2일차부터 증가하기 시작하여 3일차부터는 세포수가 유의적으로 많았다. CS를 넣은 배양액에서 유지한 근육세포는 분화 전에는 그 미분화 상태를 잘 유지하고 있다가, 분화를 유도하면 근관 형성이 잘 일어나 길이가 좀더 길고 일정하게 분화되는 것을 확인하였다. 이상의 결과에 따라, QM7 근육세포를 배양하는데 있어서 TPB를 이용하는 것보다 CS를 이용하면, 세포의 유지 및 분화에 있어 더 좋은 결과를 얻을 수 있을 것으로 사료된다.
