In order to evaluate their possible subacute toxicity, groups of 40 male and female ICR mice were given to the diet with chickens irradiated up to 30 kGy for four weeks. During the experimental periods, appearance, behavior, mortality, food and water consumption of mice fed irradiated chickens were not affected compared to the control. In urine analysis, hematological as well as in serum biochemical findings, no significant differences were found between the control (non-irradiated) and the irradiated chickens. Although minor changes in biochemical parameters were observed, they were in the normal range and were not dose dependent. Spotty necrosis was found in the male liver administered with 30 kGy irradiated chicken. However, it seems not related with gamma irradiation, because it lacks either the dose dependency and the secondary changes accompanied. Our results indicate that the adverse effect of gamma irradiated chicken can be observed at more than 30 kGy level.
A total of 100 samples of chicken-viscera were collected from different poultry-slaughtering houses in Austria;(20 samples of each of gizzard, heart, intestine, liver and spleen). Intestine and gizzard were heavy contaminated with moulds than other examined visceral organs($4.4{\times}10^5$ and $2.6{\times}10^4$ colonies/1g of the samples, respectively). Fungal contamination was not detected in all samples of heart and spleen. Eighty-five mould isolates were collected from the examined samples, the majority of isolates belonging to Aspergillus glaucus group(20.0%) and Trichoderma(14.1%). These isolates comprised 15 species belonging to 9 genera. Members of Aspergillus glaucus(telomorph: Eurotium) group and Trichoderma were further confirmed their identification using random amplified polymorphic DNA-polymerase chain reaction(RAPD-PCR) technique.
A great progress in genetic selection, nutrition and management practices has contributed to the improved growth rate of broilers and egg production in laying hens. For the increased productivity of modern poultry, a healthy chicken liver needs to cope with the increased metabolic demands. The liver is the major site of de novo fatty acid synthesis; therefore, hepatic lipogenesis is crucial for producing better quality meat and eggs. When de novo lipogenesis exceeds the capacity of lipid metabolism and secretion, large amounts of lipids accumulate in the liver of broilers, leading to a fatty liver. Upon onset of egg-laying in hens, lipids including free fatty acids, triglycerides, and phospholipids are dramatically increased in blood plasma for the synthesis of yolk precursors in oocytes. Productive hens with fatty liver often have hemorrhagic syndrome and sudden death due to the heavy demands of yolk synthesis, which burdens the liver. Understanding the lipid metabolism and hepatic lipid disorders is a key point in the improvement of the growth and production of chickens. This review focuses on the recent studies on lipid metabolism, the hepatic lipid disorders, and the prevention or reduction of fatty liver in poultry.
The lengths of thick and thin filaments in the sarcomeres of most vertebrate skeletal muscles are precisely regulated and are important structural parameters in understanding muscle contraction. Nebulin is a usually large protein that spans the whole length of thin filaments in the sarcomeres of skeletal muscles. In this paper we used SDS-PAGE and immunoblot to identify nebulin isoform proteins in muscle and non-muscle tissues. We prepared embryonic chicken tissues including skeletal muscle, cardiac muscle, smooth muscle, brain, liver to compare nebulin isoform proteins. The proteins were divided into soluble and insoluble fraction. As a result, we identified tissue specific expression of various nebulin isoform proteins in muscle and non-muscle tissues of chicken. Nebulin was detected in skeletal muscle of adult chicken about 500 kDa. Nebulett was expressed in cardiac muscle of embryonic and adult chicken about 107 kDa. A giant protein with molecular mass of about 380 kDa was identified in brain of non-muscle of chicken. This giant protein was detected in the soluble fraction of chicken embryo. The unequal distribution of the nebulin isoform proteins suggests tissue specific regulation of the isoform expression and indicates a functional specialization of the encoded isoform subtypes.
Background: Chicken anemia virus (CAV) causes chicken infectious anemia, which results in immunosuppression; the virus has spread widely in chicken flocks in China. Objectives: The aim of this study was to understand recent CAV genetic evolution in chicken flocks in Guangxi Province, southern China. Methods: In total, 350 liver samples were collected from eight commercial broiler chicken farms in Guangxi Province in southern China from 2018 to 2020. CAV was detected by conventional PCR, and twenty CAV complete genomes were amplified and used for the phylogenetic analysis and recombination analysis. Results: The overall CAV-positive rate was 17.1%. The genetic analysis revealed that 84 CAVs were distributed in groups A, B, C (subgroups C1-C3) and D. In total, 30 of 47 Chinese CAV sequences from 2005-2020 belong to subgroup C3, including 15 CAVs from this study. There were some specific mutation sites among the intergenotypes in the VP1 protein. The amino acids at position 394Q in the VP1 protein of 20 CAV strains were consistent with the characteristics of a highly pathogenic strain. GX1904B was a putative recombinant. Conclusions: Subgroup C3 was the dominant genotype in Guangxi Province from 2018-2020. The 20 CAV strains in this study might be virulent according to the amino acid residue analysis. These data help improve our understanding of the epidemiological trends of CAV in southern China.
Telomeres are essential for chromosome stability and are related with cell senescence, apoptosis and cancer. Even though telomere length and telomerase activity have been studied extensively, very little is known to analyze the telomere dynamics in chicken cells. This study was carried out to analyze the telomere distribution and telomerase activity of Korean Native Chicken cells along with aging. The cells were collected from brain, heart, liver, kidney and germinal tissues during physiological stages. Telomere distribution was analyzed by Quantitative-Fluorescence in situ Hybridization (Q-FISH) techniques using the chicken telomeric DNA probe. Telomerase activity was performed by Telomeric Repeat Amplification Protocol (TRAP) assay. In results, the telomeres of chicken were found at the ends of all chromosomes with the interstitial telomeres on chromosomes 1, 2 and 3. The amount of telomeres on chicken cells was decreased along with aging in most tissues. Furthermore, the telomere quantity was significantly different among tissues. The relative amount of telomeres in proliferous cells such as testis cells had much more than those of liver, brain, heart, blood and kidney cells. The telomerase activity was down-regulated in cells of brain, heart and liver tissues. Whereas gonadal cells showed a constitutive activity of telomerase during all stage of life. In conclusions, the telomere quantity and telomerase activity in chicken are closely relate to cell proliferation and tissue specificity during developmental stages and aging. There is also closely correlated between the amounts of telomeric DNA and telomerase activity in chicken tissues.
The response characteristics of a new biosensor, which was developed by co-immobilisation of chicken liver tissue and ferrocene in a carbon paste matrix for the amperometric determination of hydrogen peroxide, was evaluated. In the range of electrode potential observed ( - 0.5∼ + 0.05 V vs Ag/AgCl), the bioelectrode showed the response time ($t_{95%}$) as low as 13 sec and the detection limit of $5.1{\times}10^{-5}$ M, and exhibited a good selectivity in the case of the addition of the possible interferents tested. Also it offered a high biocatalytic stability and opened up the possibility for the construction of a mediatorless biosensor.

Mycotoxins are secondary metabolites produced by molds, such as Aspergillus, Fusarium and Penicillium, that have adverse effects on animals and humans. Aflatoxin, ochratoxin, zearalenone, fumonisin and deoxynivalenol are the mycotoxins of greatest agro-economic importance and cause acute disease called mycotoxicoses. Mycotoxicosis in poultry birds results in decreased meat/egg production, immunosuppressant, and hepatotoxicosis. Some of toxins or their metabolites may be retained in animal or human tissues and induce health problems. This study was designed to develop a sensitive liquid chromatography tandem mass spectrometry (LC-MS/MS) method for the simultaneous detection and quantification of mycotoxins, such as aflatoxin $B_1$, aflatoxin $M_1$, ochratoxin A, zearalenone, fumonisin B and deoxynivalenol, in chicken liver and kidney tissues. The mycotoxins were extracted and purified using modified QUECHERS methods, separated by LC and detected by an electrospray ionisation interface (ESI) and tandem MS. Good precision and linearity were observed for most of six mycotoxins. The recovery test for each mycotoxin in liver and kidney tissues mostly indicated good average recovery rates between 80.94% and 98.10% and the coefficient of variation mostly under 13.78%, except for aflatoxin $M_1$ and fumonisin $B_1$. The limit of detection (LOD) for six mycotoxins was $7.6{\sim}145.79{\mu}g/kg$ in liver tissues and $6.07{\sim}197.20{\mu}g/kg$ in kidney tissues. The quantification limits (LOQ) for 6 mycotoxins were in the range $23.04{\sim}441.78{\mu}g/kg$ in liver tissues and $18.40{\sim}597.59{\mu}g/kg$ in kidney tissues, respectively. The developed multi-mycotoxin method in this study permits simultaneous, simple, and rapid determination of several co-existing mycotoxins in chicken liver and kidney tissues.
In the chickens removed the left thyroid gland or administrated with thiouracil, fatty liver (FL) and fatty liver-hemorrhagic syndrome (FLHS) were induced within five weeks after treatment. FL was observed in all of the thyroidectomized layer and broiler hens, and the occurrence of FLHS was higher in rate at the fifth week than that at the third week after removal. FL and FLHS were observed in the thiouracil-administrated broiler hens, but the rate of incidence was appeared to be lower in the thiouracil-administrated hens than in the thyroidectomized chickens. It was suggested that hypothyoidism could be an important factor inducing FLHS.
The influence of refeeding either protein, carbohydrate or fat on hepatic insulin-like growth factor-I (IGF-I) mRNA level in chicks which had been fasted for 2 days was examined. The hepatic IGF-I mRNA was measured by ribonuclease protection assay. Fasting reduced hepatic IGF-I mRNA levels to less than half of those in the fed control. When chicks were refed either a control, protein or carbohydrate diet, IGF-I mRNA levels significantly increased to those in the fed control until 2 hours of refeeding. Refeeding of fat did not alter hepatic IGF-I mRNA levels. The significant correlation between liver weight and hepatic IGF-I gene expression suggests that when chicks are refed after 2-d fasting, the acute increase in hepatic IGF-I gene expression brought about after refeeding may be partly regulated by the increase in liver protein metabolism.
