Effects of Vitamin A on the Antioxidant Systems of the Growing Chicken

  • The present study was conducted to evaluate effects of the increased dietary vitamin A supplementation on the vitamin A, vitamin E and ascorbic acid concentrations in the plasma and liver and activities of some enzymes in the liver of the growing chicken. One hundred and twenty female chickens at 4 weeks of age were divided in 6 equal groups in accordance with their body weight. They were housed in cages and fed on standard wheat-barley-based broiler diet balanced in the major nutrients. Vitamin A was supplemented in the form of retinyl acetate. Control diet was supplemented with 10 IU/g and experimental feeds were supplemented with 50, 100, 500, 1000 and 2000 IU/g. At days 42 and 56 of the development 8 chickens from each group were killed, plasma and liver were collected for vitamin and enzyme analyses. The increased vitamin A supplementation was associated with its increased accumulation in the liver and with a reduction of ${\alpha}-tocopherol$ concentrations in the plasma and liver. The blood plasma was more resistant to vitamin A concentration changes and the retinol level was elevated only when the vitamin A dose exceeded 100 IU/g feed. Ascorbic acid concentration in the liver was elevated when moderately high vitamin A supplementation was used but significantly decreased at the highest vitamin A dose. Similar changes were observed with glycogen concentration in the liver. Activities of hexokinase, glucose-6-phosphatase and lactate dehydrogenase in the chicken liver were also dependent on vitamin A supplementation, decreasing with highest vitamin A doses. Therefore the observations showed that the vitamin A excess compromises antioxidant system of the growing chickens suggesting that prooxidant activity may be responsible for at least part of the toxicity of vitamin A.

The Effect of the Aflatoxin $B_1$ on Liver Tissue of Chick-Embryo (Aflatoxin $B_1$이 계배 간조직의 미세구조에 미치는 영향)

  • To investigate the effect of aflatoxin $B_1$, on survival rate and ultrastructure of liver during chick embryogenesis electron microscopic methods were used. After injection of aflatoxin $B_1$ into the yolk, ultrastructural changes in the liver of chicken embryo were observed. The results were as followed. 1. 12-day old chicken embryos were treated with single injection of aflatoxin $B_1$ with the dose of $0.0005{\mu}g,\;0.005{\mu}g,\;0.05{\mu}g,\;0.5{\mu}g,\;2.5{\mu}g,\;5.0{\mu}g$ each. Chicken embryos treated with the dose of $0.5{\mu}g$ of aflatoxin $B_1$ had survival rate of 22%. The embryos treated with $2.5{\mu}g$ of aflatoxin $B_1$ hardly survived. 2. Chicken embryos treated with $0.05{\mu}g$ of afatoxin $B_1$ had hatched in 30%, but once hatched, they all survived. 3. After administration of $0.05{\mu}g$ of aflatoxin $B_1$ into the 12-day old chicken embryo, the electron microscopic studies were examined during development stages. The nuclei of hapatocytes became irregularly shaped and the structures of endoplasmic reticulum were changed to spherical types at 20-day old chicken embryo. Also, mitochondria became to be dilated and severe fibrosis was induced in the cytoplasm. However, the hepatocytes became almost normal in 30-day old young chicken.

Chicken serum uric acid level is regulated by glucose transporter 9

  • Objective: Glucose transporter 9 (GLUT9) is a uric acid transporter that is associated with uric absorption in mice and humans; but it is unknown whether GLUT9 involves in chicken uric acid regulation. This experiment aimed to investigate the chicken GLUT9 expression and serum uric acid (SUA) level. Methods: Sixty chickens were divided into 4 groups (n = 15): a control group (NC); a sulfonamide-treated group (SD) supplemented with sulfamonomethoxine sodium via drinking water (8 mg/L); a fishmeal group (FM) supplemented with 16% fishmeal in diet; and a uric acid-injection group (IU), where uric acid (250 mg/kg) was intraperitoneally injected once a day. The serum was collected weekly to detect the SUA level. Liver, kidney, jejunum, and ileum tissues were collected to detect the GLUT9 mRNA and protein expression. Results: The results showed in the SD and IU groups, the SUA level increased and GLUT9 expression increased in the liver, but decreased in the kidney, jejunum, and ileum. In the FM group, the SUA level decreased slightly and GLUT9 expression increased in the kidney, but decreased in the liver, jejunum, and ileum. Correlation analysis revealed that liver GLUT9 expression correlated positively, and renal GLUT9 expression correlated negatively with the SUA level. Conclusion: These results demonstrate that there may be a feedback regulation of GLUT9 in the chicken liver and kidney to maintain the SUA balance; however, the underlying mechanism needs to be investigated in future studies.

Characterization of Chicken By-products by Mean of Proximate and Nutritional Compositions

  • Though a great amount of chicken by-products are consumed everyday in many countries worldwide, however, no attention has been paid to the investigation of nutritional composition of these by-products. In the present work, the basic information regarding the aspects of nutritional composition of chicken by-products such as; liver, gizzard, heart, lung, crop, small intestines, cecum and duodenum was studied. Our results revealed that the approximate composition range (minimum to maximum) of these by-products was found as such: moisture 76.68-83.23%; fat 0.81-4.53%, protein 10.96-17.70% and calories 983.20-1,426.0 cal/g tissue, in which liver and gizzard had the highest protein content. Liver had higher (p<0.05) vitamin A, B1, B2, B3, B5 and B6 contents in comparison to other remaining byproducts. Total saturated fatty acids (SFA), unsaturated fatty acids (UFA), polyunsaturated fatty acids (PUFA) levels ranged between the by-products from 31.82% to 43.96%, 56.04% to 68.19%, and 18.27% to 32.05%, respectively. Remarkably, all of by-products showed desirable PUFA/SFA ratios. Furthermore, all of chicken by-products, especially liver, contained higher levels of trace elements (e.g., Fe, Mn and Zn) in comparison with those from muscle tissues published in literature. Overall, the study indicated that most of chicken byproducts examined are good sources of essential nutrients and these obtained results will be the useful information to consumers and meat processors.

The Amount of Telomeres and Telomerase Activity on Chicken Embryonic Cells During Developmental Stages (닭의 발생 단계별 세포내 Telomere의 양적 분포양상과 Telomerase 활성도 분석)

  • Telomeres locate at the end of chromosomes and consist of a tandem repeat sequence of $(TIAGGG)^{n}$ and associated proteins. Telomerase is a ribonucleoprotein which act as a template for the synthesis of telomeric DNA. Telomeres are essential for chromosome stability and are related with cell senescence, apoptosis and cancer. This study was carried out to analyze the amount of telomeres and telomerase activity of chicken cells during embryonic and developmental stages. The whole embryos and prenatal tissues such as brain, heart, liver, kidney and testis at different developmental stages were obtained from Korean Native Chicken. The amount of telomeres on embryonic cells was analyzed by quantitative fluorescence in situ hybridization (Q-FISH) techniques using the chicken telomeric DNA probe. Telomerase activity was measured by telomeric repeat amplification protocol (TRAP) assay. Results indicated that the amounts of telomeric DNA on the most embryonic cells were gradually decreased during ontogenesis. Furthermore, the quantity of telomeres was quite different among embryonic tissues according to developmental origin. The relative amount of telomeres has more in regenerative cells such as embryonic disc and testicular cells than in non-regenerative cells such as liver, brain, heart and kidney cells.   • The division and circadian DNA-synthesis rtythm are studied in chick liver immediately after hatching. The vision function is tested by the mitotic and the $^3H-thymidine$ labelling index The DNA synthesis exhibited a cyclic variation of 12 hours immediately after hatching. The rhythmic changes of DNA synthesis was maintained in the liver of meal-fed chickens, but the DNA synthetic activities decreased gradually in the starved chicken liver. From this time, the rhythm of DNA synthesis was greatly afffected by lighting schedule, all these DNA synthesis of chicken liver was accompained by mitosis.

Simultaneous Determination of Residual Sulfonamides in Meat Tissues by High Performance Liquid Chromatography (HPLC에 의한 식육조직중의 잔류 설파제 동시 분석)

  • Four sulfoanmides ; sulfamerazine ; sulfamethazine, sulfathiazole and sulfadimethoxine from muscle, kindney, liver and heart tissues of pork and chicken by LC. Residual sulfonamides were extracted with dichloromethane and determined on a Sperisorb ODS-1 column(250mm$\times$4.6mm id) with acetonitrile/water/acetic acid (30/70/0.3 v/v) as a mobile phase at 260nm. Recoveries from 4 tissues of pork and chicken samples fortified with 50 and 100 ppb were 71.2~87.2% and 73.7~89.6%, respectively. The detection limit was 0.03 $\mu\textrm{g}$/g in each drug. Sulfamethazine in 5 samples of pork. And sulfadimethoxine in 5 samples and sulfamethazine in 3 samples were also detected from 41 samples of chicken. The order of residue levels of sulfonamides in tissues was kidney>liver>heart>muscle, respectively. The residue levels of sulfonamides from kidney and liver were 0.03~0.15 $\mu\textrm{g}$/g in porks and 0.03~0.10 $\mu\textrm{g}$/g in chickens.

Survey of Slaughtered Chicken Leucocytozoon Infection Rate in Pusan, Gyeongnam Area (부산, 경남지방 도계육에 대한 Leucocytozoon 감염율 조사)

  • During 9 months (March-November, 1989), observation of Leucocytozoon infection rate to blood and parenchymatous viscera (spleen, liver, heart) in Pusan, Gyeongnam (Kimhae, Yangsan) districts slaughtered chicken and the results obtained were as follow : 1. Among the blood smear sample of 213 heads of the chicken, 43 heads (20.2%) were infected with Leucocytozoon 2nd stage gametocyte and 4 heads (1.9%) were mixed infected with Leucocytozoon 5th stage gametocyte. 2. In histopathological detection of schizont in tissues section of spleen, liver and heart, there were 16.7% in spleen, 5.6% in liver and 9.1% in heart, mean detection rate were 10.6%. Spleen had the highest detection rate of schizont. 3. In seasonal-related Infection rate, summer was higher than spring.

  • The pathologic studies on the cottonseed meal poisoning in chicken were performed by feeding a diet containing 50 per cent cottonseed meal. The results obtained are summarized as follows: The specific effect of cottonseed meal on chicken was apparently limited to the parenchymatous tissue and blood vessels, where its major pathologic manifestations were degenerative changes in acute cases fed for a period of 7 days, while necrosis of portal and central veins of liver and consequent perivascular hemorrhage and coagulative necrosis of liver and intra-glomerular hemorrhage were characteristic lesions in chronic cases fed for a period 20, 40 and 60 days. In addition, specific cottonseed pigment cells were observed in small number in villus of small intestine in acute cases and in large number in villus, liver and spleen in chronic cases.

Identification of Upregulated APOA1 Protein of Chicken Liver in Pullorum Disease (추백리가 감염된 닭의 간에서 발현이 증가하는 APOA1 단백질의 확인)

  • The aim of this study was to investigate differentially expressed proteins between normal chicken liver and chicken liver inffeted by Salmonella pullorum. 2-dimensional electrophoresis (2DE) and mass spectrometry (MS) were used to identify the proteins. More than 300 protein spots were detected on silver stained 2DE gels using pH 3$\~$10 gradients. The most outstanding protein spot was further analyzed by MALDI-TOF MS and protein database using the Mascot search engine. The protein was finally identified as APOAI (Apolipoprotein AI). Based on the known function of the APOAI, this gene acts protective action against the accumulation of platelet thrombin at the site of vascular damage for the pullorum disease. Therefore APOAI protein, identified in this study, can be a valuable biomarker in relation to the pullorum disease in chicken.
