Chick embryos from stage 14 to stage 31 were studied by means of serial section and light microscopy in order to learn the relationship between the settlement sites of the primordial germ cells (PGCs) and the forming genital ridge. The results showed that: when embryo hatched for 53-56 h, the PGCs reached the coelomic epithelial tissue where gonad would be formed, meanwhile the epithelial tissue began thicker before the PGCs reached. Before stage 19, the final region the PGCs arrived was the thickened portion of the coelomic epithelium, the glycogen in the PGCs cytoplasm maintenance remained unchanged. However at the 3.5-5th hatching day, the glycogen in the PGCs cytoplasm reduced gradually. On the 6th hatching day, the gonad of the embryo appeared the feature of ovary, and the glycogen in the PGCs cytoplasm reduced further. On the 7th hatching day, the differentiation of ovary or testis was obvious and the glycogen in the PGCs cytoplasm later disappeared.
본 연구는 기존의 형질전환 닭 생산방법 중의 하나인 1세 포기 수정란에 유전자를 직접 주입하는 유전자 미세주입방법을 개선할 목적으로 리포좀과 외래 표지 발현유전자인 GFP를 사용하여 외래유전자의 핵전이 효율성과 주입된 외래 유전자의 발현의 지속성을 닭의 배자에서 검증하고자 시도하였다 외래유전자는 배반엽 단계 혹은 1세포기 수정란의 세포질에 리포좀과 유전자의 혼합물 혹은 오직 유전자만을 미세주입을 하였다. 연구 결과들은 리포좀을 사용한 경우 naked DNA에 비하여 배반엽 단계와 1세포기 수정란 모두에서 효율적으로 외래 유전자를 핵내로 도입할 수 있음을 배양 3과 4일차 닭의 배자에서 GFP발현 양상을 통하여 확인하였다. 또한 주입된 외래 유전자에 의해 만들어진 GFP는 배자에서 일주일 정도 지속적으로 발현됨이 관찰되었다. 리포좀 방법은naked유전자 주입 방법에 비해 1세포기와 배반엽 단계 수정란 모두에서 효율적으로 외래 유전자를 핵내로 이동시키는 능력을 가지나, 주입된 유전자의 염색체 삽입에는 큰 영향을 미치지 않는 것으로 판단된다. 따라서 닭의 수정란에서 리포좀 방법은 외래유전자 도입에 유용한 수단으로 이용되어질 수 있을 것으로 사료된다.
The purpose of this study was to investigate the teratological effects of enrofloxacin, a veterinary antibiotic, on the embryos and fetus of hatching chicken eggs. A control group and four experimental fertilized egg groups were set up. The four experimental groups were injected with 0.05 ml, 0.1 ml, 0.2 ml, and 0.3 ml enrofloxacin ($50{\mu}g/ml$) respectively. During the hatching period, the weights of total eggs, inside material(yolk and white) and embryos or fetuses, embryo growth, and teratological effects were investigated. In the experimental groups (fertilized eggs injected with 0.05 ml, 0.1 ml, 0.2 ml and 0.3 ml enrofloxacin), the weights of total eggs were decreased, but the yolk and white weights were increased by the higher amount of antibiotics. The weights of embryos (fetuses) in experimental groups were 86.1%, 78.6%, 65.6% and 61.4% of the control, respectively. Retarded growth, deformity and embryo loss were observed in experimental groups. Teratological effects such as undeveloped eyes, wings and legs, and deformed head and bill were also detected. In conclusion, we found that veterinary antibiotics, enrofloxacin have made teratological effects on the embryos and fetus of hatching eggs.
Sphingomyelinase (SMase EC:3.l.4.l2) has been suggested to play important roles in the cell cycle, differentiation, apoptosis, inflammation, and the regulation of eukaryotic stress responses. SMase inhibitors may be a powerful tool to elucidate and regulate these cellular responses in which SMase involves. We first isolated an SMase inhibitor, named sphinin, from a strain of soil actinomycetes, F50970. Sphinin inhibited Mg/sup 2+/ -dependent neutral SMase from chicken embryo at 1.2 ㎍/㎖ of IC/sub 50/ Sphinin also inhibited acidic SMase, but it had no inhibitory activity on PI-PLC and PC-PLC, suggesting that sphinin is a specific inhibitor of SMase. The strain F50970 was identified as a Streptomyces sp. by its spiral spore chain, LL-diaminopimelic acid, menaquinone patterns of MK-9 (H'6) and MK-9 (H'8), FA-2c type of fatty acid pattern, and other morphological, physiological, and cultural characteristics.

We have examined alcohol-induced malformation of chick embryo. Alcohol was considered to induce the malformation of developing embryo and to have bad effects on embryonic stage. We injected alcohol into air sac on day 4 of incubation. Ten % alcohol-treated group showed a little decrease on their body length compared to the untreated group and distilled water-treated group. Thirty % alcohol-treated group showed significant decrease on their body length compared to the untreated group and 10% ethanol treated group. In addition, we have observed malformation of eyeballs and bills. These results indicate that alcohol affects chicken developments and brings on malformation of developing stage.

In case of the TGF-βl and β3 addition group, there was the decrease of EGFR(Epidermal Growth Factor Receptor) immunoreactivity in mesenchymal cells beneath the medial edge epithelium and also in epithelial mesenchymal interface which is between medial edge epithelium and nasal septum in 72 hours, 2, The immunoreactivity of the control group resembles that of normal chicken embryo palate in development, 3. In the view through fluorescence confocal microscopy, there was confluence in TGF-β3 addition group, This shows that the confluence induced by exogenous TGF-β3 is related to EGFR expression in palate of chicken embryo, which is a physiologic cleft palate model.
Insulin-like growth factor-I (IGF-I) mRNA levels in the eyes, heart, liver and breast muscle removed from dwarf egg-type, normal egg-type and normal meat-type chicken embryos at 7, 14 and 20 days of incubation were measured. There was no influence of chicken strain on IGF-I gene expression in the eyes and liver. The IGF-I gene expression in eyes increased significantly along with the incubation period. In the liver, IGF-I gene expression at 20 days of incubation was significantly higher than that at 14 days of incubation. In the muscle, the lowest value for IGF-I gene expression was observed in meat-type chicken embryos. Regression analysis revealed that IGF-I gene expression was significantly correlated to the weights of the eyes and liver, but not the muscle. We conclude that there is little influence of strain on tissue IGF-I gene expression in chicken embryos during incubation but that tissue development in chicken embryos is nevertheless at least partly regulated by the change in IGF-I gene expression.
