토종 '우리맛닭' 부계 및 실용계에서 MC1R 유전자 변이 및 모색과의 연관성 분석 (Genetic Variations of Chicken MC1R Gene and Associations with Feather Color of Korean Native Chicken (KNC) 'Woorimatdag')

  • 본 연구는 '우리맛닭 실용계의 외모 균일화 및 판별 마커개발을 위하여 수탉으로 이용되는 토종닭 순계 H계통 암탉 207수, 수탉 40수와 '우리맛닭' 실용계 60수의 혈액을 채취하고 DNA를 분리하였다. 모색 관련 후보 유전자 MC1R primer를 제작하여 PCR을 수행한 후, direct sequencing을 실시하였다. 개체별 모색 표현형을 조사한 결과, H계통 순계 암탉 207수 중 157수는 흑색 모색, 50수는 흑색+갈색 모색으로 조사되었다. 수탉의 경우, 40수 중 흑색 모색만 있는 개체는 없고, 흑색+갈색의 모색이 다양한 부위에 섞여 있었다. '우리맛닭' 실용계의 경우 흑색 모색 30수와 갈색 모색 30수를 암수 관계없이 샘플링하여 조사하였다. 모색 표현형과 유전형과의 연관성을 알아보기 위하여 모색 관련 후보유전자 MC1R의 sequencing 결과를 분석하여 유전자 변이를 살펴보았지만, 모색 표현형과 유전형과의 유의적인 연관성을 찾을 수 없었다. 이는 토종닭 순계 H계통의 경우, 검정색의 같은 품종 내에서 갈색 이모색을 가진 개체와의 유전적 변이를 탐색한 결과, 그 차이가 분명하게 구별되지 않은 것으로 생각되어진다. '우리맛닭' 실용계에서 haplotype 및 유전자빈도 분석을 통하여 이모색과의 연관성을 보다 명확하게 결정하기 위해서는 부계와 모계의 모색 표현형과 유전특성을 함께 고려해야 할 것으로 판단된다. '우리맛닭'의 모색 균일도 향상을 위한 모색 표현형과 관련 유전자의 연관성분석 및 유전적 특성에 대한 연구는, 향후 이를 이용하여 '우리맛닭'의 불법 유통을 막고 고품질 브랜드화를 위한 분자마커 개발의 기초 자료로 활용될 것으로 생각된다.

Quality Assessment of the Breast Meat from WoorimatdagTM and Broilers

  • The objective of this study was to compare the characteristics that define the quality of Woorimatdag$^{TM}$ (WM, a certified meat-type commercial Korea indigenous chicken breed) and a commercial broiler breed (Ross, CB). Two hundred WM and 200 CB chickens that were 1-d-old and mixed sex were obtained from a commercial hatchery and randomly assigned to floor pens (20 chickens per pen, $3.0{\times}2.0m$) and raised under the same environmental conditions. WM breast meat contained significantly higher crude protein and ash as well as lower crude fat than CB breast meat (p<0.05). WM breast meat had slightly higher alanine, histidine, isoleucine, and glycine as well as lower phenylalanine content than CB breast meat (p<0.05), and the WM breast meat had a low ratio of unsaturated to saturated fatty acid composition (p<0.05). However, arachidonic acid composition was higher in the WM than the CB breast meat. In addition, the inosin-5'-monophosphate content was also higher in the WM compared with the CB breast (p<0.05). The WM breast meat had higher total collagen content compared with CB breast meat. WM soup taste received higher scores with regard to sensory evaluation compared with CB soup (p<0.05). From these results, we conclude that higher amount of protein and flavor precursors and lower amount of fat in the breast meat of WM could be attractive by consumer when compared with CB.

Comparative Study on Various Growth Performances of Korean Oge and White Leghorn Chickens

  • Genetic resource conservation of indigenous species is important to conserve terrestrial, aquatic and avian species throughout the world. The native Korean Oge (KO) chickens are important and protected indigenous avian species in Korea. This KO breed is very popular in Korea because of its external appearances of blackish color. The characterization of morphology and growth performances of KO were performed since 1980s, however the information is still not sufficient for breed selection. In this study, we compared various growth performances including body growth, tibia length, shank length, chest width, chest grith and length of tail feather between male and female chickens of KO and widely known White Leghorn (WL) breeds at 1 week, 2 weeks, 5 weeks, 10 weeks, and 24 weeks. We observed differences on various growth performances at different aged groups between KO and WL chickens. This study may help for the selection of chicken breeds based on age, body growth and meat production.

Determination and comparison of growth performance parameters between two crossbred strains of Korean native chickens with a white semi broiler chicken for 84 days post-hatch

  • The aim of this study was to investigate the growth performance between two different crossbred strains of Korean native chickens denoted as 1E and 2E with a white semibroiler (LL) from hatch to d-84 post-hatch. A total of 450 one-day-old chicks were randomly distributed into cages which included 15 replicate cages each for 1E and 2E with 9 replicate cages for LL. A three phase feeding program was used as a starter (Day 1 - 35; crude protein [CP] 20.3%; metabolizable energy [ME] 3,059 kcal·kg-1), grower (Day 36 - 56; CP 18.6%; ME 3,123 kcal·kg-1) and finisher (Day 57 - 84; CP 16.7%; ME 3,187 kcal·kg-1). The results revealed that LL had a higher (p < 0.05) BW during d-8 to d-84 compared with the other two cross breed groups. A higher (p < 0.05) ADG was observed from the LL during the entire period except from d-1 to d-14 and d-71 to d-84 when compared with 1E and 2E. Shank length did not vary (p > 0.05) between the strains, despite LL having a SL that was 2.86% higher (p < 0.05) when compared with the other strains from d-15 to d-21 post-hatch, and 1E had a SL that was 2.28% (p < 0.05) higher when compared with the other strains during the first week of the experiment.

시중 유통 토종닭의 품종별 품질 및 관능 특성 비교 (Comparison of Meat Quality and Sensory Characteristics of Different Native Chickens in Korean Market)

  • 본 실험에서는 시중에서 주로 유통되고 있는 2종의 토종닭인 HH종과 WD종 닭고기를 비교하기 위해 일반 육계(BR)와 더불어 육질 및 관능 특성을 분석하였다. 토종닭 내에서 HH종의 가슴육만 일반 육계보다 다소 낮은 지방함량을 보였다. 육색에서는 가슴육과 다리육 두 부위 모두에서 HH종의 명도($L^*$)와 적색도($a^*$)가 WD종보다 유의적으로 높게 나타났다(p<0.05). 항산화력 측정을 위한 DPPH 라디칼 소거능과 pH가 WD종이 HH종보다 유의적으로 높았다(p< 0.05). 가슴육에서 C18:1은 일반 육계가 다른 두 품종의 토종닭보다 유의적으로 높았다(p<0.05). 대표적인 n-3 지방산인 C18:3의 경우, WD종이 HH종보다 유의적으로 높았으며(p<0.05), C20:4와 C22:6의 경우, 토종닭이 일반 육계에 비해 약 2.5배이상 높은 조성을 보였다(p<0.05). 관능평가 맛 부분에서는 가슴육에서 WD종이 일반 육계나 다른 HH 토종닭보다 유의적으로 우수한 수치를 보였다(p<0.05). 특히 WD종의 연도가 다른 품종에 비해 매우 우수한 것으로 나타났으며(p<0.05), 전체적인 기호도에서도 WD종이 유의적으로 높은 점수를 보였다(p<0.05). 국내에서 대표적으로 시판되는 토종닭 2품종의 품질 및 관능 특성은 요인별로 유의적 차이를 보이기도 했으며(p<0.05), 이를 바탕으로 소비자 기호에 적합한 새로운 품종의 토종닭 개발을 위한 기초 자료 활용이 가능할 것이다. 그러나 육질 특성은 품종 이외에도 급여한 사료 및 사양 방식에 따라 다른 것이므로, 보다 많은 시료 수를 바탕으로 한 추가적인 연구가 요구된다.

육용계에 있어서 성장단계에 따른 부위별 가식육의 증가양상 추정에 관한 연구 (Studies on the Estimation of Growth Pattern of Meat in the Edible Parts of Broilers in Growing Stages)

  • 육용계에서 가식육량을 효과적으로 추정할 수 있는 방법을 구명코자 White Cornish종과 S. C. W. Leghorn종. 그리고 이 두 품종의 상반교잡종을 각각 120수씩 계 480수를 공시하여 체중. 부위별 가식육 및 체형의 변화, 이들 형질에 발현된 잡종강세와 반성유전효과 및 화학적 조성분함량을 조사하고. 성장단계별 가식육량을 추정한 결과를 요약하면 다음과 같다. 1. W. Cornish종의 8주시 체중 및 사료요구율과 폐사율을 보면 각각 1,818.8$\pm$47.7g와 2.24 및 3.30%였고 S. C. W. Leghorn종에선 각각 667.8$\pm$28.9g와 3.28 및 2.50%였으며 잡종들의 능력은 이 두 품종의 중간정도로 정상발육을 하였다. 2. 체중에 있어서 계종간 차이는 2주령부터 유의성이 인정되었고. 성장곡선의 회귀방정식에서 성장속도를 표시하는 회귀계수률 보면 W. Cornish종( $b_{cc}$ =1,578)은 S. C. W. Leghorn종 ( $b_{LL}$ =1,378)에 비해 유의적으로 크고 교잡종들은 이 두 품종의 중간값( $b_{LC}$ =1,476, $b_{CL}$ =1,470)을 보인다. 가식육량의 경우도 계종간 증가양상이 체중의 경우와 비슷하였으나 육량증가의 회귀방정식에서 회귀계수의 크기가( $b_{LL}$ =1,563, $b_{LC}$ =1,651, b.$_{CL}$ =1,636, $b_{CC}$ =1,766) 체중에서 보다 컷는데 그 원인이 성장속도 차이때문인지를 구명하기 위해 주령(log x)과 체중에 대한 가식육량비율(log y) 간의 선형관계를 추정한 결과 각 계종의 회귀계수( $b_{LL=0.184}$, $b_{LC}$ =0.175, $b_{CL=0.165}$$b_{CC}$ =0.188)는 모두유의성이 인정되어 가식육의 증가속도는 체중의 증가속도보다 현저히 빠른 것으로 판명되었다. 3. 체중구성요소중 주령경과에 따라 그 중량비율이 증가되는 것은 우모, 복강지방, 흉부 및 퇴경부이고, 감소되는 것은 두부, 경부, 가식내장 및 부가식내장이며 방혈량, 각부, 익부 및 배부는 큰 변화를 보이지 않는다.

Association of Insulin-Like Growth Factor-I (IGF-I) Gene Polymorphism with Serum IGF-I Concentration and Body Weight in Korean Native Ogol Chicken

  • IGF-I is involved in the regulation of growth and differentiation in mammals, but its role as a modulator of growth and metabolism in poultry is poorly understood. And, no studies have so far been reported for the comparison between serum IGF-I concentration and body growth in the egg type or the dual purposes (meat and egg type) chicken including the Korean Native Ogol Chicken (KNOC). Therefore, in order to improve the body growth and meat production of the KNOC, this study was conducted for the identification of the polymorphism of IGF-I gene and for its possible association with both body weight and IGF-I concentration. The RFLP patterns for IGF-I gene were identified by the PstI restriction enzyme. The frequencies of +/+, +/-, and -/- genotype were 16.9%, 51.7%, and 31.4%, respectively. Any statistical significance was not observed in all variations except for sex variation (p<0.01) by covariate quadratic model. The significant effect of the IGF-I genotype on body weight by sex indicates that there are different physiological characteristics in gender. Although the body weights of male KNOCs in most ages were not significant, there was a tendency of KNOCs with +/+ IGF-I genotype to be heavier than those with any other genotypes. But all IGF-I genotypes in female did not influence on body weight. The ANOYA revealed no significant effects of IGF-I genotypes on serum IGF-I concentration but sex effect was highly significant on the IGF-I concentration at 20 and 40 weeks (p<0.01). Although the +/+ genotype, in gender, tended to express a higher IGF-I concentration than the other genotypes at all ages in males, a statistical difference among the genotypes was not found except for 60 weeks (p<0.05). Furthermore, since body weight and IGF-I genotypes are associated, it is possible to improve KNOC to a meat type breed if a continuous selection can be made for the body weight and/or IGF-I traits.

닭의 모색 연관 유전자인 MC1R, MITF, TYRP1의 SNP(Single Nucleotide Polymorphism) 규명 (Identification of SNP(Single Nucleotide Polymorphism) from MC1R, MITF and TYRP1 associated with Feather Color in Chicken)

  • 닭을 구분하는데 있어 가장 눈에 띠는 것이 모색이며, 모색 관련 유전자인 MC1R, MITF, TYRP1의 SNP에 따른 유전자형을 확인하고, 각 품종별로 구별이 가능한 SNP 마커를 개발하고자 하는데, 본 연구의 목적이 있다. 마커들을 조합으로 haplotype을 보았을 때, MC1R 유전자의 SNP 조합에서 CGG type일 경우, 재래닭 만을 특별히 구별할 수 있었으며, TAG, TGG, TAA type일 경우에는 아라카나 만을 구별할 수 있었고, CAA type의 경우, 레그혼 만을 특이적으로 구별할 수 있었다. TYRP1 유전자의 SNP 조합에서는 TTTCA, CCTCA type의 경우, 레그혼 만을 구별할 수 있으며, CTTTA type의 경우, 오골계 만을 특이적으로 구별할 수 있었다. MC1R 유전자의 SNP 조합으로 재래닭, 레그혼, 아라카나를 구별할 수 있었고, TYRP1 유전자의 각각의 SNP 및 조합으로 4가지 품종 모두 구별할 수 있었다. 이렇게 품종 간의 유전적 다형성에 대한 연구가 더 많이 진행된다면, 단순 모색만으로 품종을 구별하기보다는 분자생물학적으로 품종 간의 차이를 이해할 수 있게 될 것이라고 생각된다.

Performance differences of Rhode Island Red, Bashang Long-tail Chicken, and their reciprocal crossbreds under natural cold stress

  • Objective: The Bashang Long-tail chicken (BS), an indigenous Chinese breed, is considered cold tolerant. We selected BS, the Rhode Island Red (RIR), and their reciprocal crossbreds for the present study. The objectives were: i) to validate whether BS is cold tolerant and whether egg production and cold tolerance of crossbreds could be improved; and ii) to determine the physiological characteristics that underlie cold tolerance and favorable egg production performance in cold environments. Methods: A total of 916 chickens were reared in warm and natural cold environments (daily mean ambient temperature varied from $7.4^{\circ}C$ to $26.5^{\circ}C$ in the warm environment and from $-17.5^{\circ}C$ to $27.0^{\circ}C$ in the cold environment). To investigate their adaptability to the cold environment, the egg production performance and body weight were monitored and compared between breeds and environments. The cloacal temperature and serum biochemical parameters were monitored to reveal the physiological characteristics underlie cold tolerance and favorable egg production performance in the cold environment. Results: The warm environment experiment showed that RIR had the highest egg production performance, and that the reciprocal crossbreds had a higher egg production performance than BS. While in the cold environment RIR had the lowest egg production performance, and the reciprocal crossbreds had a higher egg production performance than BS. In the cold environment BS and reciprocal crossbreds had higher triiodothyronine, tetraiodothyronine levels than RIR. At 35 and 39 wk of age, when the ambient temperature was extremely low (varied from $-20^{\circ}C$ to $0^{\circ}C$), serum glucose, follicle-stimulating hormone, luteinizing hormone, estradiol of BS and crossbreds were higher than RIR. Conclusion: Bashang Long-tail chicken has a favorable cold tolerance ability. Crossbreeding with RIR and BS is an effective way to develop cold tolerant chickens with improved egg production performance.

육계사료에 항생제 첨가 효과와 경제성에 관한 조사연구 II (the additional effects of antibiotics for the chicken breeding feed and economical efficiency.)

  • The purpose of this experiment is to clarify the additional effect of antibiotics for the chicken breeding feed. This experiment was carried out with 6,560 individuals which were collected from four farms, Dugil, Hyubhoi, Sanggog and Kumsan. The duration of this experiment was 6 months from July to December in 1981. 1. Body weight and body gain. (1) The range of the body gain was 1.91-7.60% The most effective sections was T$_3$, and the effect was T$_2$, T$_4$, T$_1$, T$\sub$5/ in order. (2) The environmental sanitation of the Sanggog farm was not so good. In this farm the body gain was severely occurred in T$_3$ and T$_2$ sections. Mere gain obtained from T$\sub$5/ section. (3) The environmental sanitation was good in Dugil, Hyubhoi and Kumsan. A little body gain was occurred in these farms, but differences were shown according to the place or breeds. 2. Amount of feed intake feed conversion ratio. (1) Amount of feed intake was increased 0.13-3.19%, and feed conversion ratio was improved 1.68-4.62% compared with control. T$_3$ section showed high percent and T$_4$ T$_1$, T$\sub$5/, Is, C in order. (2) The keeping method of the chicken breed in Hyubhoi was floor brooding. From Hyubhoi and Sanggog farms, the efficiency was increased 9.4-9.8%. (3) From Dugil and Kumsan, the rate of improvement was low. 3. Mortality : Mortality was merely decreased in Sanggog and Kumsan farms which the sanitary and fowl caring conditions had been had, by additional antibiotics. But there was no worthy effect. 4. The ratio of carcass and the degree of skin coloration. The additional antibiotics in feed had no effect on the ratio of carcass but had the influence on the yellowish skin coloration. Skin coloring was good in T$\sub$7/ section and T$_1$, T$\sub$6/, T$_4$, C, T$_3$, T$\sub$5/, T$_2$ in order. 5. Economical efficiency (1) The addition of the antibiotics in feed increased 3.76-10.95% of gross earning. The highest earning was occurred in T$_4$ section and T$_3$, T$_1$, T$_2$, T$\sub$5/, C sections in order. (2) The earning rate was influenced by the antibiotic products. (3) In Dugil and Kumsan farms, there was no difference in earning, however, in Hyubhoi and Sanggog farms there was wide difference in earning. Conclusionally, the addition of antibiotics in chicken breeding feed is necessary. Especially in bad condition of environmental sanitation and floor brooding, the effect of additional antibiotics is clear. Selection of antibiotics from various kinds, maker's confidence and quality problems are important.
