Optimizing Carnosine Containing Extract Preparation from Chicken Breast for Anti-glycating agents

  • Optimization of carnosine and anserine extraction from chicken breast was performed using response surface methodology (RSM) to obtain the maximized physiological activities for anti-glycation and anti-oxidation. The optimum extraction conditions were water extraction for 1.6 h in the case of the 20-wk laying hen muscle and water extraction for 2.12 h in the case of 90-wk laying hen muscle. Higher carnosine and anserine contents were measured in the 20-wk laying hen muscle, along with higher physiological activities, which increased in direct proportion with the dipeptide contents. The extracts prepared from the 20-wk laying hen under optimum conditions showed 57% inhibition of advanced glycated end-product formation, 64% inhibition of lipid peroxidation, and 61% of DPPH radical scavenging effects. On the other hand, 52% inhibition of AGE formation, 62% inhibition of lipid peroxidation, and 53% of DPPH radical scavenging effect were demonstrated within the 90-wk laying hen. In addition, the ratio of carnosine was a key indicator for the physiological activities of the extracts.

Effects of Surgical Caponization on Growth Performance, Fiber Diameter and Some Physical Properties of Muscles in Taiwan Country Chicken Cockerels

  • An experiment was conducted to determine the effects of caponization on the growth performance, breast and thigh muscles physical properties and fiber diameter of the Pectoralis major and Gastrocnemius pars externa in Taiwan country chicken cockerels. Caponized birds were surgically altered at 10 weeks of age. Birds were fed grower and finisher diets ad libitum during an eighteen-week experimental period. The results indicated that the live weight and feed intake in the capons were significantly (p<0.05) higher and the shank length was significantly (p<0.05) longer than in intact birds. There were no significant (p>0.05) differences in feed conversion and mortality between two treatments at 28 weeks of age. Compared with intact birds, the capons had greater (p<0.05) tenderness in the breast and thigh muscles. Cohesion of the breast muscle in the capons was significantly (p<0.05) better than in the intact birds, but the thigh muscles were not significantly (p>0.05) affected. No treatment differences (p>0.05) were associated with cooking loss, muscle chewiness, and elasticity. The capons had a significantly (p<0.05) smaller fiber diameter in the Pectoralis major, but were not significantly (p>0.05) different in the fiber diameter of the Gastrocnemius pars externa. It is concluded that castration did not depress growth compared with the intact birds, but did improve muscle tenderness. This difference was most pronounced in the thigh muscles.

Underlying mechanisms of phosphodiesterase 10A and glutamate-ammonia ligase genes that regulate inosine monophosphate deposition and thereby affect muscle tenderness in Jingyuan chickens

  • Objective: Inosine monophosphate (IMP) is a key factor that imparts of meat flavor. Differences in the IMP content in the muscles were evaluated to improve chicken meat quality. Methods: For this study, the IMP content was detected by high performance liquid chromatography. The gene expression profiles of Jingyuan chickens with different feeding patterns and different sexes were analyzed by RNA-sequencing (RNA-seq). Results: Breast muscle IMP content in free-range chickens was extremely significantly higher than that of caged chickens (p<0.01). Breast muscle IMP content in hens was also higher than that of cocks, but the difference was not significant. Correlation analysis showed that the breast muscle IMP content in caged hens and cocks was negatively correlated with the shear force, and the breast muscle IMP content in free-range hens was significantly negatively correlated with the shear force (p<0.05). The two key genes associated with IMP synthesis in chickens with different feeding patterns were glutamate-ammonia ligase (GLUL) and phosphodiesterase 10A (PDE10A). Bioinformatics analysis revealed that the GLUL and PDE10A genes are involved in glutamine biosynthesis and purine salvage pathways respectively. In addition, GLUL expression was positively correlated with the IMP content in caged and free-range chickens, and PDE10A expression was significantly positively correlated with the IMP content in caged and free-range chickens (p<0.05). Conclusion: These findings will facilitate the comprehension of the deposition of IMP in the muscles and thereby aid the process of selection and breeding of good quality local chickens.

Relationship of IGF-I mRNA Levels to Tissue Development in Chicken Embryos of Different Strains

  • Insulin-like growth factor-I (IGF-I) mRNA levels in the eyes, heart, liver and breast muscle removed from dwarf egg-type, normal egg-type and normal meat-type chicken embryos at 7, 14 and 20 days of incubation were measured. There was no influence of chicken strain on IGF-I gene expression in the eyes and liver. The IGF-I gene expression in eyes increased significantly along with the incubation period. In the liver, IGF-I gene expression at 20 days of incubation was significantly higher than that at 14 days of incubation. In the muscle, the lowest value for IGF-I gene expression was observed in meat-type chicken embryos. Regression analysis revealed that IGF-I gene expression was significantly correlated to the weights of the eyes and liver, but not the muscle. We conclude that there is little influence of strain on tissue IGF-I gene expression in chicken embryos during incubation but that tissue development in chicken embryos is nevertheless at least partly regulated by the change in IGF-I gene expression.

Screening and functional validation of lipid metabolism-related lncRNA-46546 based on the transcriptome analysis of early embryonic muscle tissue in chicken

  • Objective: The study was conducted to screen differentially expressed long noncoding RNA (lncRNA) in chickens by high-throughput sequencing and explore its mechanism of action on intramuscular fat deposition. Methods: Herein, Rose crown and Cbb broiler chicken embryo breast and leg muscle lncRNA and mRNA expression profiles were constructed by RNA sequencing. A total of 96 and 42 differentially expressed lncRNAs were obtained in Rose crown vs Cobb broiler chicken breast and leg muscle, respectively. lncRNA-ENSGALT00000046546, with high interspecific variability and a potential regulatory role in lipid metabolism, and its predicted downstream target gene 1-acylglycerol-3-phosphate-O-acyltransferase 2 (AGPAT2), were selected for further study on the preadipocytes. Results: lncRNA-46546 overexpression in chicken preadipocyte 2 cells significantly increased (p<0.01) the expression levels of AGPAT2 and its downstream genes diacylglycerol acyltransferase 1 and diacylglycerol acyltransferase 2 and those of the fat metabolism-related genes peroxisome proliferator-activated receptor γ, CCAAT/enhancer binding protein α, fatty acid synthase, sterol regulatory element-binding transcription factor 1, and fatty acid binding protein 4. The lipid droplet concentration was higher in the overexpression group than in the control cells, and the triglyceride content in cells and medium was also significantly increased (p<0.01). Conclusion: This study preliminarily concludes that lncRNA-46546 may promote intramuscular fat deposition in chickens, laying a foundation for the study of lncRNAs in chicken early embryonic development and fat deposition.

기능성 어육단백질의 젤화 특성과 산업적 응용-2. 알칼리 공정으로 회수한 어육, 닭고기 가슴살 및 돼지 후지 육 기능성 단백질 젤의 특성과 최적화 (Gelation Properties and Industrial Application of Functional Protein from Fish Muscle-2. Properties of Functional Protein Gel from Fish, Chicken Breast and Pork Leg and Optimum Formulation)

  • 어육, 닭 가슴살 및 돼지 후지 육을 산성 및 알칼리 용액으로 추출하여 등전점 부근에서 회수하고 중성 부근의 pH로 재조절하여 회수한 단백질의 가열 젤 물성과 이들의 혼합에 따른 가열 물성 값의 변화, 최적 물성과 최소비용을 제공하는 혼합 비율을 결정하였다. 갈고등어의 근원섬유단백질은 산 및 알칼리 처리에 의해 가열 젤을 형성하지 못하였으나, 산과 알칼리 처리후 근형질 단백질을 포함한 회수 단백질은 가열 젤을 형성하였다. pH 10.5에서 처리 후 회수한 단백질의 가열 젤의 파괴강도는 갈고등어가 가장 낮았고, 변형 값은 냉동 꼬마민어>닭 가슴살>돼지 후지 육>갈고등어의 순으로 높았으며, 백색도는 냉동 꼬마민어 회수 단백질이 가장 높았다. 갈고등어 회수 단백질의 첨가는 파괴강도, 변형 값, 백색도를 감소시키고 가격을 상승시키는 반면, 닭 가슴살 회수 단백질의 첨가는 파괴 강도와 백 색도를 다소 증가시키고 가격을 현저히 감소시켰다. 냉동 꼬마민어 회수 단백질인 경우, 파괴강도 110 g 이상, 변형 값 4.5 mm 이상 및 회수 단백질의 원료 단가 2000원 이하/kg을 만족하는 최적 혼합 비율은 냉동 꼬마민어 36∼50%, 닭 가슴살 34∼40%, 돼지 후지 육 14∼25%이었다 가열 젤의 구조는 냉동 꼬마민어 회수 단백질이 가장 치밀하였다. 냉동 꼬마민어 회수 단백질을 축으로 닭 가슴살, 돼지 후지 육 회수 단백질의 적절한 혼합 비율의 조절은 물성 값이 다양화한 연제품에 활용 가능할 것으로 판단된다.

  • Slushed ice와 dry chilling chamber에 각각 도계 직후 냉장한 12주생 영계의 가슴 및 다리 근육으로부터 KCl-phosphate buffer를 사용하여 도계후 $1{\sim}25$시간 사이에 함질소물을 추출했다. Extractable nitrogen의 변화는 주로 myosin의 extractability 감소, 그리고 어느정도 actomyosin의 extractability 증가 결과로 일어남이 밝혀졌으며, 한편 stroma. sarcoplasmic protein 및 비단백함질소물(none protein nitrogen)의 변화는 적었다. Myosin extractability는 도계후 처음 3시간 동안에 급격히 감소하여 그후 계속적으로 wet chilling 동안 감소했으나 dry chilling의 경우 myosin extractability의 감소는 가슴근육보다 다리근육에서 더 많았으며 extractability는 도계 17시간 후부터 증가했다. Actomyosin은 wet chilling의 경우 계속적으로 소량밖에 추출되지 않았으나 dry chilling의 경우 도계후 17시간 이후부터 증가했는데, 이런 경향은 breast muscle과 leg muscle이 동일했다.

Influence of Caponization on the Carcass Characteristics in Taiwan Country Chicken Cockerels

  • We determined the effect of caponization on the carcass and giblet characteristics, and skin and muscle color in Taiwan country chicken cockerels. Caponized birds were surgically altered at 10 weeks old and were fed growing and finishing diets ad libitum during an eighteen-week experimental period. The results showed that the percentage of dressing, heart, feet, thigh, head and neck were significantly (p<0.05) higher in the intact birds, while the capons had a higher (p<0.05) percentage of abdominal fat, intestine, back, wing and breast. Eviscerated weight, breast width, gizzard, liver and spleen ratios were not affected by the treatments. The breast skin color values for lightness (L*) and yellowness (b*) values in the capons were significantly (p<0.05) higher than in the intact birds, but the thigh and back skin were not significantly(p>0.05)different. Compared with the intact birds, the capons had a significantly (p<0.05) less redness (a*) values in the back skin, but were not significantly (p>0.05) different in the breast and thigh skin. The L* value of the thigh muscle was significantly (p<0.05) greater in the capons than in the intact birds, but were not significantly (p>0.05) different in breast and back muscles. The b* values in the breast, back and thigh muscles of the capons were significantly (p<0.05) greater whereas the intact birds had a higher (p<0.05) a* values in the breast, back, and thigh muscles. Moreover, our findings also indicate that the castration resulted in a significant alteration in dressing percentage, carcass region and organ percentage.

우모분과 pyridoxine에 의한 taurine 강화 계육 생산

  • An experiment was conducted to investigate the effects of dietary supplementation of feather meal (FM its) digests on the performance of broiler chicks and taurine content in broiler meat. A total of 1,000 broiler chickens were assigned to five dietary treatments : Control, FM diet(FM), FM+pyridoxine(FM+Pyridox), H$_2$O$_2$ treated FM diet(H$_2$O$_2$-FM) and enzyme treated FM diet (Enzyme-FM). Treated diets were supplemented with FM or FM digests at the level of 5 % to the control diet. During the stater period, weight gain of chicks fed FM+Pyridox was significantly higher(P<0.05) than those of the other FM or FM digest treatments but was not different from the control. Weight gam of overall period were not significantly different among treatments. Feed intake of the control was greater than that of FM or FM digest treatments. Feed conversion ratio(feed intake/gain) of chicks fed FM and H$_2$O$_2$-FM were significantly higher than those of Enzyme-FM and FM+Pyridox, but were not significantly different from the control. Taurine contents of leg and breast mucle were significantly (P<0.01) different among treatments but those of liver were not significantly different. Taurine content of FM+Pyridox was highest in both leg and breast muscle. It was 85 % higher in leg muscle and 15 % higher in breast muscle than that of the control. Sensory evaluation data showed significant but not consistant responses in various parameters. FM + Pyridox treatment showed highest score in aroma of raw leg muscle of male and in juiciness and tenderness of broiled breast muscle of male chickens. Control group showed highest color score in raw leg muscle of female and lowest overall acceptability score in broiled breast and leg muscle of male chicken. It is concluded that taurine can be enriched especially in broiler leg meat by 5 % FM diet supplemented with pyridoxine.

The Expression Characterization of Chicken Uncoupling Protein Gene

  • The UCPs are members of the mitochondrial inner membrane transporter family, present in the mitochondrial inner membrane. Their main function is increasing the energy expenditure via diminishing the resulting production of ATP from mitochondrial oxidative phosphorylation instead of yielding dissipative heat. They are associated with the metabolism of fat and regulation of energy expenditure. The UCP gene can be viewed as the candidate gene for chicken fatness. In the present study, RT-PCR and Northern Blot methods were developed to investigate the expression of the UCP gene in ten tissues including heart, liver, spleen, lung, kidney, gizzard, intestine, brain, breast muscle and abdominal fat of chicken. The results of both RT-PCR and Northern Blot methods showed that the UCP gene expressed specific in breast muscle. The expression levels of UCP gene in breast muscles from egg-type and meat-type chickens of hatching, 2, 4, 6 and 8 wk of age were detected by RT-PCR assay and results showed that the expression levels of UCP gene were related to breeds. Expression level of UCP gene in layers was higher than that in broilers at various weeks of age except at 6 wk. The UCP gene's expression was higher at 6 wk and had no significant difference among other weeks of age in broilers; in layers the expression level of UCP gene had no significant difference among weeks of age. The experiment results also showed that insulin could increase the expression level of UCP gene by 40% compared with control group.
