Nickel Toxicity and Its Interaction with Zinc, Copper and Lead in Growing Chicks (초생추에서의 니켈의 독성과 아연·구리 및 납과의 상호작용)

  • Nickel toxicity and interactions of nickel with zinc, copper, and lead were studied in glowing chicks fed supplemented diet. Feed intake and growth rate of the chick were reduced by 250mg nickel as a sulfate salt per kg of feed. The toxicity of nickel was decreased by zinc or copper supplementation, but not lead. High nickel feed increased nickel level in kidney and decreased zinc levels in tibia and plasma. However, low zinc levels in tibia and in plasma were reversed by zinc supplementation. Hemoglobin, packed cell volume, and aortic elastin content were increased in chicks fed nickel. These results suggest that nickel toxicity is induced by interference with zinc metabolism.

Effects of Some Autonomic Drugs on the Intramural Nervous System of Isolated Jejunum of Chiken Pretreated with Reserpine (몇가지 자율신경성약물(自律神經性藥物)이 Reserpine 처리(處理) 병아리의 척출공장(剔出空腸) 내재성신경(內在性神經)에 미치는 영향(影響))

  • Pharmacological investigation was performed in chickens which were pretreated with reserpine. Transmural stimulations were given to the isolated jejunum of chickens and studied the responses and the effects of dibenamine, propranolol, atropine and tetrodotoxin on them. The results obtained were summerized as follows: 1. Three different patterns of response were obtained from the isolated jejunum of non-treated chickens after giving them transmural stimulation. The first pattern was contraction followed by relaxation, the second pattern was contraction only and the third pattern was relaxation only. 2. The transmural stimulation of the jejunum preparations evoked relaxation while the periarterial stimulation evoked contraction in the reserpinized chick. 3. The relaxation response to the transmural stimulation was not affected by the pretreatment with dibenamine, propranolol and atropine. 4. The relaxation response to the transmural stimulation was abolished by tetrodotoxin. The results obtained in these studies indicate that the relaxation response to the transmural stimulation is due to non-adrenergic inhibitory fibers.

Sole and Combined Usage of Ultra-sonication and Hydrogen Peroxide as Mitigation Techniques of Bio-fouling

  • Mussels are stubborn organism attached to solid substrate by byssus threads and caused operational problems in utility of power generating stations. Sole and combined usage of ultrasonic (28 kHz- and 42 kHz- frequencies) and hydrogen peroxide ($H_2O_2$) has studied for control of blue mussel larvae and adult stage in seawater condition. A theoretical wo rking model using disinfection (Chick and Watson type) approaches is presented based on helpful results of experiments. This study also demonstrate that the combined treatment (ultra-sonication with $H_2O_2$) is overall highly efficient than individual treatment would, but on the basis of exposure time, the ultra-sonication was the most efficient among them. Therefore the development of sole and combined technique might be effective practical mitigation strategy against mussel attachment for water handling facilities.

Identification of a Fusion-associated Protein in the Skeletal Myoblast Using Monoclonal Antibody (단일클론항체를 이용한 배양 계배 근원세포의 융합과 연관된 단백질의 확인)

  • The present study describes the production of monoclonal antibodies against cultured chick myoblast to pursue critical proteins in muscle cell fusion. Among a panel of monoclonal antibodies, three, Mll-3H 13, Mll-3Hl8 and Mll-3H35 were inhibited movblast fusion. A single 101-kDa antigen reactive with monoclonal antibody Mll-3H35 was detected by radioimmu-noprecipitation or by immunoblotting. During the course of myogenesis, the level of the protein remarkably decreased as the cells there differentiated. These results suggest that the protein platys a direct role in the process of myoblast fusion mechanism.

  • As the bromination method with $Br_2^{82}$ for the synthesis of 5-bromouracil-$Br^{82}$ gave products with considerable impurities, e. g. uracil etc. an attempt to produce pure one by isotopic exchange method was performed. Bromide-82 ion such as $NH_4Br^{82}$ or $HBr^{82}$ undergoes no isotopic exchange with 5-bromouracil-Br. However, isotopic exchange between $NH_4Br^{82}$ and $Br_2$, and between $Br_2^{82}$ and 5-bromouracil-Br were too fast to determine the rate. The result indicated that this method can be used in the production of pure 5-bromouracil-$Br^{82}$. It was also found that the use of reducing agent to maintain $Br^{82}$ as bromide form was unnecessary on $NH_4Br^{82}$ production from reactor.

Inhibition of Angiogenesis by Propolis

  • Propolis, obtained from honeybee hives, has been used in Oriental folk medicine as an anti-inflammatory, anti-carcinogenic, and immunomodulatory agent. There is considerable evidence suggesting that angiogenesis and chronic inflammation are codependent. Blockage of angiogenesis results in an anti-inflammatory effect. Ethanol (EEP) and ether extracts of propolis (REP), and caffeic acid phenethyl ester (CAPE), an active component of propolis, were examined for their anti-angiogenic activities using the chick embryo chorioallantoic membrane (CAM), and the calf pulmonary arterial endothelial (CPAE) cell proliferation, assays. The presence of EEP, REP and CAPE inhibited angiogenesis in the CAM assay and the proliferation of CPAE cells. The results suggest that anti-angiogenic activities of EEP, REP and CAPE are also responsible for their anti-inflammatory effect.

  • In the previous study, we reported that 2'-hydroxy-4'-methxoychalcone, synthetic chalcone inhibited PGE2 production in TPA- stimulated rat peritoneal macrophages by inhibiting the induction of COX-2 protein. The present study was carried out to clarify whether 2'-hydroxy-4'-methoxychalcone inhibit angiogenesis by the experimental methods in vitro and in vivo. 2'-Hydroxy-4'-methoxychalcone decreased angiogenesis of both chick embryos in the chorioallantoic membrane assay and basic fibroblast growth factor-induced vessel formation inthe mouse Martigel plug assay.   • Gardenia jasminoides Ellis has been used in traditional medicine for the treatment of inflammation, jaundice, headache, fever and hypertension. The 70% ethanolic extract of gardenia fruit showed strong anti-angiogenic activity in the chick embryo chorioallantoic membrane (CAM) assay. Among hexane, ethyl acetate, n-butanol and aqueous fractions prepared from the 70% ethanolic extract, the n-butanol fraction was found to be most effective in the CAM assay. (omitted)

  • The growth and development of the metacercariae of F. seoulensis cultivated in vitro or on the chick chorioallantois were assessed by comparison with the optimum process of maturation in albino rats and new born chickens. The process of maturation was divided for convenience into six stages: Stage 1 ; cell multiplication, Stage 2; body shaping, Stage 3; separation of genital anlagen, Stage :1 organogeny, Stage 5; gametogony, and Stage 6: oviposition. In Hank's and Tyrode's .solutions, the metacercariae were alive up to 200 days or more at $4^{\circ}C$ without any development. The in vivo maturation process in rats or chicks was as follows: stage 1 from 6 hours; stage 2 from 24 hours; stage 3 from 48 to 72 hours; stage 4 from 3 to 4 days; stage 5 from 4 to 5 days; and stage 6 from 5 to 8 days. Despite unsuccessful infection of the metacercariae to 12 day old chicks, fully mature worms of stage 5 or 6 were recovered from new born chicks (1 to 2 days old), The metacercariae of F. seoulensis grown in vitro were up to stage 3 and no further maturation was observed. Of various media employed, the medium NCTC 109 (Gibco) or NCTC 135(Gibco) supplemented with 20% egg yolk or 20% whole egg macerate or 0.5% yeast was basically required for the earlier development of the fluke. It took 16.1 days(in average) to reach the stage 3 after cultivation. The metacercariae cultivated on the chorioallantoic membranes of 6∼13 day old chick embryo at 37∼38℃ showed their full development up to stage 5 or 6. However, the worms were in general remarkably retarded, compared with those grown in rats or chickens. In the experiments of worm transplant, although the transfer was failed from in vitro culture to in vivo of rats(Per os), the transplants from in vitro culture to the chorioallantois and from the choriollantois to in vivo of rat host were successful with or without development of the transferred worms. In the present study, it was observed that the metacercariae of F, seoulensis can be maintained in vitro media with poor development as well as fully matured in 1 to 2 day-old chicks or on the chorioallantois at a very low rate.
