Type I allergic reaction and it's related clinical manifestations are known to occur by the effects of various chemical mediators. These chemical mediators are released from circulating basophils and tissue mast cells, which become 'sensitized' through the binding of antigens and antibodies of the IgE type to their cell surface receptors. Efforts to elucidate the mechanism of the release of these mediators, especially that of histamine, have been persued for years. The mechanism is not yet clarified at the present time. Recent reports of hyaluronidase, an enzyme known to be involved in the tissue inflammatory process, as possible participant in type I allergic reaction, initiated this study. Relationships between the hyaluronidase activity and histamine release from the sensitized rat peritoneal mast cells were investigated. Also anti-allergic agents, tranilast and disodium cromoglycate, along with known histamine releasers, morphine and compound 48/80, were used to observe the inhibitory and stimulatory effects of these substances on the hyaluronidase activity as well as histamine release from the rat mast cells. The results obtained are summarized as follows: 1) Hyaluronidase activity and histamine release from sensitiaed rat peritoneal mast cells started to increase on the 4th day of postsensitization. Hyaluronidase activity reached it's peak value on the 7th day of postsensitization and that of histamine release on the 14th day of postsensitization. 2) Hyaluronidase activity and histamine release from sensitized rat peritoneal mast cells, pre-treated with tranilast revealed significant decrease in comparison with those of non-treated cells. 3) Hyaluronidase activity and histamine release from sensitized rat peritoneal mast cells, pre-treated with tranilast, followed by morphine injection, revealed significant increase in comparison with those of tranilast treated cells. 4) In vitro study of hyaluronidase activity and histamine release from un-sensitized rat peritoneal mast cells, using morphine and compound 48/80 as activators, revealed significant increase compared to those of non-activator used cells. 5) In vitro study of hyaluronidase activity and histamine release from un-sensitized rat peritoneal mast cells, pre-treated with tranilast and disodium cromoglycate, using confound 48/80 and morphine as activators revealed significant decrease in comparison with those of tranilast and disodium cromoglycate treated cells. From above results, participation of enzyme hyaluronidase in the process of histamine release from sensitized rat pertioneal mast cells, could be suggested. It was also quite evident that the clinically used anti-allergic agents, tranilast and disodium cromoglycate, have significant inhibitory function on the hyaluronidase activity and histamine release from sensitized rat peritoneal mast cells, while morphine significantly increased the hyaluronidase activity and histamine release from sensitized rat peritoneal mast cells.
The test method for biologics of lot release system is based on 'Test procedure and specification for biological products,' generally, thimerosal content is measured by chemical analysis using O.D. In this study, the comparative analysis was carried out using the gold amalgamation method for thimerosal content was compared to the existing methods, which are described above. The gold amalgamation method, which uses atomic absorption spectrophotometry, was meets all the method validation acceptance criteria. It is considered to be proper as the assay and identification test for thimerosal. In this study, the comparative analysis was performed three times. As a result, gold amalgamation method is more convenient and easy to perform as this assay doesn't have pre-treatment procedure. Also this assay showed good precision and reproducibility compared to the conventional method. Therefore, it is appropriate to alternate the assay method of thimerosal from the conventional chemical analysis to gold amalgamation method to improve the credibility of lot release system and the quality control of biologics, by standardizing test method.
삼투정 펠렛은 경구를 통한 약물전달 시스템에 폭넓게 사용되고 있으며, 이러한 삼투정 펠렛은 수팽윤성 시드층과 모델약물인 니페디핀을 포함하는 약물층 그리고 약물의 방출을 조절하는 반투막 층으로 구성되어 있다. 이번 연구에서는 모델약물인 니페디핀을 포함한 삼투정 펠렛을 제조하고, 반투막층으로 사용되는 초산셀룰로오스(CA)와 Eudragit RS의 코팅두께에 따른 약물방출 거동과 알긴산 나트륨과 알긴산 나트륨의 가교가 삼투정 펠렛의 약물방출에 미치는 영향에 대하여 확인하고자 하였다. 모델약물인 니페디핀을 포함한 삼투정 펠렛의 제조는 유동층코팅기를 이용하여 제조하였으며, 비교적 높은 코팅 수율로 $1500{\sim}1700{\mu}m$ 내외의 펠렛이 제조됨을 SEM을 통하여 확인하였다. 이렇게 얻어진 펠렛의 반투막 층의 코팅두께에 따른 약물방출 거동을 보면 반투막의 코팅 두께가 증가할수록 약물의 방출이 지연됨을 확인하였다. 알긴산 나트륨을 반투막층 위에 코팅하였을 경우 인공위액(pH 1.2)에서는 약물방출이 거의 일어나지 않았으며, 인공장액(pH 6.8)으로 교체한 후 약물방출이 서서히 증가함을 알 수 있었다. 또한 알긴산 나트륨을 염화칼슘을 이용하여 가교시켰을 경우 약물의 방출이 급격히 감소함을 알 수 있었다. 이번 실험을 통하여 삼투정 펠렛의 약물방출은 반투막충의 코팅두께에 영향을 받으며, 알긴산 나트륨이 삼투정 펠렛의 약물방출에 영향을 끼침을 확인하였다.
약물의 서방화에 있어서 독성이 특히 강하거나 유효 치료영역이 좁은 약물일수록 초기 버스트는 매우 중요하다. 이러한 약물의 전달을 위한 단일층으로 이루어진 나노미립구의 이용은 표면에 존재하는 약물 때문에 초기 버스트가 커서 서방화에 적절치 못하다. 따라서 본 연구에서는 생분해성 고분자인 덱스트란과 락타이드-글리콜라이드 공중합체(PLCA)를 이용한 이중층 나노미립구를 제조하여 서방성 방출 거동을 보이는 약물 전달체 제조에 대한 연구를 수행 하였다. 덱스트란과 PLCA의 나노미립구는 W/O/W법을 이용하여 이중 에멀젼 과정을 통해 제조하였고 계면활성제로는 폴리(비닐 알코올)(PVA)을 사용하였다. 덱스트란의 생체외 방출 거동을 확인하기 위해 동결 건조된 시료를 직경 $3{\times}1mm$ 몰드를 이용하여 웨이퍼를 제조하여 증류수에서 7일간 방출 거동을 확인하였다. 이중층 나노미립구는 각각의 단일고분자로 이루어진 나노미립구에 비해 다른 방출거동을 보였다. 특히 유화제인 PVA농도가 $0.2\%$인 것이 0차 방출에 가까운 결과를 보였다. 본 실험을 통해 대조군인 물리적인 혼합 모델, 덱스트란 또는 PLGA로만 이루어진 웨이퍼 및 단일층 미립구에 비해 이중층 나노미립구의 내부물질인 덱스트란의 방출 거동이 서방형을 보임을 확인할 수 있었으며 PVA의 함량에 따라 방출 거동을 조절할 수 있었다.
유속이 약한 하천이나 호소에 침강된 유기물, 영양염류, 유해화학물질 등의 오염물질은 일단 수중으로부터 제거되어 퇴적물 속에 축적되었다가 확산, 재 부유, 생물교란 등의 물리 화학 생물학적 과정에 의해 다시 수층으로 용출되어 수질 및 수생생태에 직 간접적인 악영향을 미칠 수 있게 된다. 특히 인은 수중생태계의 일차 생산량을 결정하는 중요한 물질중의 하나로 수체 내 물질순환 과정에서 주로 퇴적물에 저장되는 원소로 물리적인 교란 및 생 화학적 작용에 의해 수층으로용출되어 부영양화현상을 가속시키는 것으로 알려져 있다. 본 연구에서는 수층 미생물의 영향을 고려한 경우(호소수)와 미생물의 영향을 배제한(증류수)경우로 구분하여 각각 다른 capping 소재를 적용하여 퇴적물에서의 인 용출 저감 효과를 분석하였다. 실험 결과 화학적소재인 Fe-Gypsum, $SiO_2$-Gypsum로 capping을 할 경우, control보다 인 용출을 약 40% 이상 저감시키는 것을 확인할 수 있었다. 또한 복합소재인 granule gypsum+Sand는 약 50% 이상 용출을 저감 효과가 나타났다. 따라서 화학적 소재인 granule-gypsum과 그 위에 모래와 같은 친환경소재로 capping을 한다면 인 용출 저감에 효과적일 것으로 판단된다. Capping 처리 전후의 퇴적층내의 인 성상변화를 살펴본 결과, gypsum 투입에 의하여 호소 퇴적층에 존재하는 인이 용출되기 어려운 안정한 형태인 apatite-P 변화된 것으로 확인되었다. 이상의 결과를 토대로 capping 기술의 적용을 통하여 하천 및 호소 퇴적물에서 발생되는 인 용출에 대한 저감 효율을 기대할 수 있을 것으로 사료된다.

Expanded polytetrafluoroethylene (ePTFE) grafts have been used as vascular access for many patients suffering from end stage renal disease. However, the vascular graft can cause significant clinical problems such as stenosis or thrombosis. For this reason, many studies have been performed to make drug eluting graft, but initial burst is major problem in almost drug eluting systems. Therefore we used biodegradable polymer to reduce initial burst and make sustained drug delivery. The ePTFE grafts were dipped into a paclitaxel-dissolved solution and then PLGA-dissolved solution was passed through the lumen of ePTFE. We analyzed whether the dose of paclitaxel is enough and the loading amount of PLGA on ePTFE graft increases according to the coating solution's concentration. Scanning electron microscope (SEM) images of various concentration of PLGA showed that the porous surface of graft was more packed with PLGA by tetrahydrofuran solution dissolved PLGA. In addition, in vitro release profiles of Ptx-PLGA graft demonstrated that early burst was gradually decreased as increasing the concentration of PLGA. These results suggest that PLGA coating of Ptx loaded graft can retard drug release, it is useful tool to control drug release of medical devices.
$Poly({\varepsilon}-carbobenzoxy\;L-lysine)/poly(ethylene oxide)/poly({\varepsilon}-carbobenzoxy\;L-lysine)$ (LEL) block copolymers containing $poly({\varepsilon}-carbobenzoxy\;L-lysine)$ (PCLL) as the A component and poly(ethylene oxide) (PEO) as the B component were investigated as drug delivery matrix. PCLL homopolymer and LEL block copolymer microspheres containing anticancer drug, cytarabine, were prepared by a solvent evaporation process and the release patterns of cytarabine from the microspheres were investigated in vitro. The size of PCLL homopolymer and LEL block copolymer microspheres was ranged from $0.2\;{\mu}m$ to $1\;{\mu}m$ in diameter and the shape of the microspheres was almost round. The release pattern of cytarabine from the block copolymer microspheres was dependent on the mole % of PEO of the block copolymers.
본 연구는 콘택트렌즈를 중합 시 사용하는 교차결합제의 구조가 콘택트렌즈의 물리적 특성 및 약물 용출에 미치는 영향을 조사하였다. 4종의 교차결합제를 0.3%와 3%를 각각 사용하여 콘택트렌즈를 제작하였으며 약물은 안약에 사용하는 ofloxacin을 사용하였다. 친수성 교차결합제를 사용한 콘택트렌즈는 함수율과 습윤성이 향상되었으며 친수성 작용기가 많을수록 효과가 크게 나타났다. 약물용출 농도는 친수성이 강한 교차결합제 사용 시 가장 높게 나타났으며 교차결합제의 농도가 높은 콘택트렌즈는 용출되는 약물 농도가 낮고 용출시간은 길어졌다. 콘택트렌즈의 교차결합제 구조는 콘택트렌즈의 성능 향상과 약물 방출조절에 영향을 주었다.
This study was carried out to investigate the heat storage/release characteristics of the thermochemical reaction of the calcined dolomite with the packed bed shape experimental apparatus for development of chemical heat pump system. In the present study, it was found that MgO of the calcined dolomite was not hydrated during the hydration process under the experimental conditions. Therefore, the MgO of the calcined dolomite can be regard as an inert material. As a result, it was found that all of CaO packed kept the reaction temperature of about $510^{\circ}C$ through the entire part of the bed. The dehydration reaction was incurred first at the wall side area as the supplied heat was transferred through the wall side into the packed bed. As a result of the temperature and concentration spread, the reaction was completed at the wall side progressed into the center.
