Anthraquinone-2-sulfonate (AQS) release from poly(N-methylpyrrole anthraquinone-2-sulfonate) (PNMP-AQS) was investigated at open circuit and compared with electrochemically stimulated release during potential cycling. It was found that the fast AQS release from PNMP-AQS single layers is substantially retarded and the amounts of spontaneously and electrochemically releasable AQS can be reduced by constructing bilayers, consisting of PNMP-AQS inner layers and PNMP outer layers. PNMP-Cl outer layers exhibited higher effectiveness for entrapping AQS within inner layers than PNMP/poly(styrene slfonate). The effects of outer layer thicknesses on AQS release were also examined with PNMP-AQS:PMP-Cl. The electroactivity enhancement of PNMP-AQS:PNMP-Cl bilayers due to entrapped AQS was confirmed by chronocoulometry.
A method has been developed to estimate the kinetic energy release originating from the reverse critical energy in unimolecular ion dissociation. Contribution from the excess energy was estimated by RRKM theory, the statistical adiabatic model and the modified phase space calculation. This was subtracted from the experimental kinetic energy release distribution (KERD) via deconvolution. The present method has been applied to the KERDs in $H_2$, loss from $C_6H_6^+$ and HF loss from ${CH_2CF_2}^+$. In the present formalism, not only the energy in the reaction coordinate but also the energy in some transitional vibrational degrees of freedom at the transition state is thought to contribute to the experimental kinetic energy release. Details of the methods for treating the transitional modes are found not to be critical to the final outcome. For a reaction with small excess energy and large reverse critical energy. KERD is shown to be mainly governed by the reverse critical energy.
Osmotic pellet system, which is one of the oral drug delivery systems, has been developed to improve manufacturing process, reduce product cost and other problems of osmotic tablet systems. Osmotic pellet is consisted of water swellable seed layer, drug layer, and membrane layer. Among them, the membrane layer plays an important role in a control of the drug release. In this work, we examined the effect of ratio for Eudragit RL and RS on the drug release behavior. Osmotic pellet with nifedipine as a model drug was easily obtained in a good yield by fluidized bed coater. Osmotic pellet showed round morphology with a range of size $1300{\sim}1500\;{\mu}m$. In the experiment of nifedipine release, the release amount increased with the increase of the ratio of Eudragit. This is due to the fact that Eudragit RL contains more hydrophilic quaternary ammonium group than Eudragit RS. Additionally, the release amount was retarded with increasing the membrane thickness. There are no differences in the release amount measured at the different pH 1.2, 6.5, 6.8, and 7.2. In conclusion, it was found that the drug release from osmotic pellets depended on the composition ratio and coating thickness of membrane layer.
Biodegradable poly[(glycine ethyl ester)-(phenylalanine ethyl ester) phosphazene](PGPP) microparticles were fabricated by electrohydrodynamic atomization to apply drug release test. Atomization parameters such as applied voltage, polymer concentration, and molecular weight were investigated to inspect their effects on the size and morphology of microparticles. The average diameter of PGPP microparticles decreased as increasing applied voltage and solution flow rate. Dichloromethane/dioxane mixture shows better results for the preparation of microparticles than single solvent owing to the different PGPP solubility in solvent. Blending PGPP polymers with proper molecular weights not only favored the production of spherical PGPP microparticles via electrohydrodynamic atomization, but also provided a way to adjust drug (rifampicin) release behavior. Drug-loaded biodegradable polyphosphazene microspheres can be fabricated via electrohydrodynamic atomization, which has potential use in biomedical applications.
To graft N-isopropylacrylamide (NIPAAm) onto alginate, varying dosages of ${\gamma}$-rays were irradiated onto alginate films in deionized water and methanol media, which are non-solvents of alginate. We investigated the hydrogels graft ratio, mechanical strength, swelling kinetics and ratio, and behavior with respect to drug release. The graft yield of NIPAAm increased upon increasing the irradiation dose. The use of the aqueous solution increased the graft yield relative to that obtained in methanol. The mechanical strength of the grafted hydrogels increased after grafting with NIPAAm. In a study of the swelling kinetics, we found that all hydrogels reached an equilibrium swollen state within 3 h. The equilibrium swelling ratio of the hydrogels decreased upon increasing the irradiation dose. The swelling ratio of the hydrogels decreased dramatically between 30 and 35$^{\circ}C$ because phase separation of NIPAAm occurred at 32$^{\circ}C$. The swelling process, with respect to the temperature change, was repeatable. An NIPAAm-grafted alginate containing a drug sustained its release rate until 3 h after an initial high drug release caused by a burst effect.
Background: The constant consumption of chemical products owing to expanding industrialization has led to an increase in public interest in chemical substances. As the production and disposal processes for these chemical products cause environmental problems, regional information on the hazard level of chemical substances is required considering their effects on humans and in order to ensure environmental safety. Objectives: This study aimed to identify hazard contribution and spatiotemporal characteristics by region and chemical by calculating a hazard-based result score using pollutant release and transfer register (PRTR) data. Methods: This study calculated the chemical discharge and hazard-based result score from the Risk-Screening Environmental Indicators (RSEI) model, analyzed their spatiotemporal patterns, and identified hotspot areas where chemical discharges and high hazard-based scores were concentrated. The amount of chemical discharge and hazard-based risk scores for 250 cities and counties across South Korea were calculated using PRTR data from 2011 to 2018. Results: The chemical discharge (high densities in Incheon, Daegu, and Busan) and hazard-based result scores (high densities in Incheon, Chungcheongnam-do, and some areas of Gyeongsangnam-do Province) showed varying spatial patterns. The chemical discharge (A, B) and hazard-based result score (C, D) hotspots were identified. Additionally, identification of the hazard-based result scores revealed differences in the type of chemicals contributing to the discharge. Ethylbenzene accounted for ≥80% of the discharged chemicals in the discharge hotspots, while chromium accounted for >90% of the discharged chemicals in the hazard-based result score hotspots. Conclusions: The RSEI hazard-based result score is a quantitative indicator that considers the degree of impact on human health as a toxicity-weighted value. It can be used for the management of industries discharging chemical substances as well as local environmental health management.
The Dexamethasone (DEX) loaded chitosan microspheres were prepared by thermal denaturation and chemical cross-linking method using a dierent concentration of glutaraldehyde as chemical cross-linking agent. The prepared microspheres were evaluated for the percentage of Drug Loading (DL), Encapsulation Efficiency (EE) and surface morphology by Scanning Electron Microscopy (SEM). DL and EE were found to be maximum range of 10.0 to 10.79 % and 58.19 to 64.73 % respectively. The SEM Photographs of the resultant microspheres exhibited fairly smooth surfaces and predominantly spherical in appearance. In addition, Fourier Transform Infrared Spectroscopy (FTIR) and Differential Scanning Calorimetry (DSC) shown that there was no interaction between the drug and polymer. In vitro and in vivo release studies revealed that the release of dexamethasone was sustained and extended up to 63 days and effectively controlled by the extent of cross-linking agent. Non-clinical parameters such as paw volume, hematological parameters like Erythrocyte Sedimentation Rate (ESR), Paced Cell Volume (PCV), Total Leucocytes Count (TLC), Hemoglobin (Hb), Differential Cell Count (DCC) were investigated in Fruend's Complete Adjuvant (FCA) induced arthritic rats. Radiology and histopathological studies were also performed in order to evaluate the therapeutic efficacy of the DEX-loaded microspheres in extenuating the rat arthritic model.
Monodispersed microparticles with a poly(D,L-lactide-co-glycolide) (PLGA) core and a poly(ethyl 2-cyanoacrylate) (PE2CA) shell were prepared by Shirasu porous glass (SPG) membrane emulsification to reduce the initial burst release of doxorubicin (DOX). Solution mixtures with different weight ratios of PLGA polymer and E2CA monomer were permeated under pressure through an SPG membrane with $1.9\;{\mu}m$ pore size into a continuous water phase with sodium lauryl sulfate as a surfactant. Core-shell structured microparticles were formed by the mechanism of anionic interfacial polymerization of E2CA and precipitation of both polymers. The average diameter of the resulting microparticles with various PLGA:E2CA ratios ranged from 1.42 to $2.73\;{\mu}m$. The morphology and core-shell structure of the microparticles were observed by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The DOX release profiles revealed that the microparticles with an equivalent PLGA:E2CA weight ratio of 1:1 exhibited the optimal condition to reduce the initial burst of DOX. The initial release rate of DOX was dependent on the PLGA:E2CA ratio, and was minimized at a 1:1 ratio.
Today toxic gas has various uses. If there is a release accident, the gas rapidly disperse into the atmosphere. The extent of damage due to toxic gas accident is very wide and fatal to human being. So, it is necessary for toxic gas facilities which have high risk to construct an emergency response system that prepare to toxic release and make immediate response to be possible at accident appearance. In this study accident scenario were selected and frequency analysis was executed using FTA technique. Dispersion effect of toxic gas release was analyzed using DNV company's PHAST(Ver. 6.2). Finally, an emergency response system was developed using results of quantitative risk analysis.
The objectives of this study were to investigate the influence of gamma irradiation for sterilization on poly(D,L-lactide-co-glycolide) (PLGA) with different molecular weight and the effect of gamma irradiation on the release behavior of 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU, carmustine) from PLGA wafer with various irradiation doses. The effect of gamma irradiation on PLGA was evaluated by gel permeation chromatography (GPC), differential scanning calorimetry (DSC), and electron paramagnetic resonance (EPR). The weight average molecular weight (M$_{w}$) and glass transition temperature (T$_{g}$) of PLGA decreased after gamma irradiation. The extent of M$_{w}$ reduction was dependent on irradiation dose and PLGA molecular weight. Using EPR spectroscopy, we successfully detected gamma irradiation induced free radicals in PLGA. The gamma irradiation increased the release rate of BCNU from PLGA wafer at applied irradiation doses except 2.5 Mrad of irradiation dose in this study.study.
