One of the most important characteristics in the seventh educational course is an enforcement of differentiated level curriculum. Therefore, development of various different level educational subjects is needed. A new chromatographic experimental kit which is helpful for middle school students to understand chromatographic principles is needed to correct some mistakes of chromatographic part within the 6th educational course's middle school textbook. In this research, a new thin layer chromatography(TLC) experimental kit for middle school students are developed by using acid-base indicator as samples and ethyl alcohol, ethylacetate, hexane as eluents. The kit is applied to two kinds of middle school student groups(general group & genius group) and two kinds of teacher groups(science teacher & non-science teacher) and an efficacy of the new chromatographic kits are evaluated through making up some questions.

A La (III) ion-selective membrane sensor has been fabricated from poly vinyl chloride (PVC) matrix membrane, containing 3-hydroxy-N'-(pyridin-2-ylmethylene)-2-naphthohydrazide (HPMN) as a neutral carrier, potassium tetrakis (p-chlorophenyl) borate (KTpClPB) as an anionic excluder and ortho-nitrophenyloctyl ether (NPOE) as a plasticizing solvent mediator. The effects of membrane composition and pH as well as the influence of the anionic additive on the response properties were investigated. The sensor with 30% PVC, 62% solvent mediator, 6% ionophore and 2% anionic additive, shows the best potentiometric response characteristics. It displays a Nernstian behavior (19.2 mV per decade) across the range of $1.0{\times}10^{-2}-1.0{\times}10^{-7}$ M. The detection limit of the electrode is $7.0{\times}10^{-8}$ M ($\sim$10 ng/mL) and the response time is 15 s from $1.0{\times}10^{-2}$ up to $1.0{\times}10^{-4} $M and 30 s in the range of $1.0 {\times}10^{-5}-1.0{\times}10^{-7}$ M. The sensor can be used in the pH values of 3.0-9.0 for about seven weeks. The membrane sensor was used as an indicator electrode in the potentiometric titration of lanthanum ions with EDTA. It was successfully applied to the lanthanum determination in some mouth wash preparations.
In order to find out a reliable chemical indicator which can be used to tell whether rice, during storage, is invaded by fungi or not, 90 percent milled rice (Tongil) was stored at 26$^{\circ}C$ for 4 weeks and weekly analyzed for changes in several chemical components. The results were as follows : 1) Of several genera of fungi (Aspergillus sp., Penicillium sp., and Rhizopus sp.) observed in stored rice after surface disinfection, Aspergillus sp. were the most frequent fungi. 2) Chitin content in the rice was 25 ug/g at the beginning of the experiment and increased proportionly with moisture content of the storedrice to become 1980 ug/g at 26.9% moisture after 4 weeks. 3) Non-reducing sugartended to decrease with increasing moisture content and or prolonged period. 4) Fat acidity showed a tendency of increase with rise in moisture content of the stared rice. Consequently, changes in chitin content can be mployed as a reliable measure to evaluate the quality of stored rice in connection with fungal invasion.

The 3-[(2-furylmethylene)amino]-2-thioxo-1,3-thiazolidin-4-one (FTT) was used as an excellent ionophore in construction of a $Zn^{2+}$ PVC-based membrane sensor. The best performance was obtained with a membrane composition of 30% poly(vinyl chloride), 62% nitrobenzen (NB), 3% FTT and 5% sodium tetraphenyl borate (TBP). This membrane sensor shows very good selectivity and sensitivity towards $Zn^{2+}$ over a wide variety of cations, including alkali, alkaline earth, transition and heavy metal ions. The membrane sensor revealed a great enhancement in selectivity coefficients for $Zn^{2+}$ ions, in comparison to the previously reported $Zn^{2+}$ membrane sensors. Theoretical studies also showed the selective interaction of TFF and $Zn^{2+}$ ions. The proposed membrane sensor exhibits a Nernstian behavior (with slope of 29.3 ${\pm}$ 0.3 mV per decade) over a wide concentration range (1.0 ${\times}$$10^{-6}$-1.0 ${\times}$$10^{-2}$) with a detection limit of 8.5 ${\times}$$10^{-7}$ M (52 ng mL$^{-1}$). It shows relatively fast response time, in the whole concentration range ($\lt$ 20 s), and can be used for at least 10 weeks in a pH range of 3.0-7.0. The proposed membrane sensor was successfully used in direct determination of $Zn^{2+}$ ions in wastewater of industrial zinc electroplating companies, and also as an indicator electrode in titration with EDTA.

Protein tyrosine nitration is considered as an important indicator of nitrosative stresses and as one of the main factors for pathogenesis of inflammation and neuronal degeneration. In this study, we investigated various nitrosative modifications of bovine carbonic anhydrase II (CAII) through qualitative and semi-quantitative analysis using the combined strategy of Fourier transformation ion cyclotron resonance mass spectrometry (FT-ICR MS) and ion-trap tandem mass spectrometry (IT-MS/MS). FT-ICR MS and its spectra were used for the search of the pattern of nitrosative modifications. Identification of nitrosatively modified tyrosine sites were executed through IT-MS/MS. In addition, we also tried to infer the reason for the site-specific nitrosative modifications in CAII. In view of the above purpose, we have explored- i) the side chain accessibility, ii) the electrostatic environment originated from the acidic/basic amino acid residues neighboring to the nitrosatively modified site and iii) the existence of competing amino acid residues for nitration.

In general, the parameters of climate change include aerosol chemical compounds, aerosol optical depth, greenhouse gases(carbon dioxide, CFCs, methane, nitrous oxide, tropospheric ozone), ozone distribution, precipitation acidity and chemical compounds, persistent organic pollutants and heavy metals, radioactivity, solar radiation including ultra-violet and standard meteorological parameters. Over the last ten years, the monitoring activities of Korea regarding to the climate change have been progressed within the WMO GAW and ACE-Asia IOP programs centered at the observation sites of Anmyeon and Jeju Gosan islands respectively. The Greenhouse gases were pointed out that standard air quality monitoring techniques are required to enhance data comparability and that data presentation formats need to be harmonized and easily understood. Especially, the impact of atmospheric aerosols on climate depends on their optical properties, which, in turn, are a function of aerosol size distribution and the spectral reflective indices. Aerosol optical depth and single scattering albedo in the visible are used as the two basic parameters in the atmospheric temperature variation studies. The former parameter is an indicator of the attenuation power of aerosols, while the latter represents the relative strength of scattering and absorption by aerosols. For aerosols with weak absorption, surface temperature decreases as the optical depth increases because of the domination of backscattering. For aerosols with strong absorption, however, warming could occur as the optical depth increases. The objective of the study is to characterize the means, variability, and trends of Greenhouse gases and aerosol properties on a regional basis using data from its baseline observatories in Korea peninsula. A further goal is to understand the factors that control radiative forcing of the greenhouse and aerosol.

Modulation of unscheduled DNA synthesis by dehydroepiandrosterone (DHEA) after exposure to various chemical carcinogens was investigated in the primary rat hepatocytes. Unscheduled DNA synthesis was induced by treatment of such direct acting carcinogens as methly methanesulfonate (MMS) and ethyl methanesulfonate (EMS) or procarcinogens including benzo(a)pyrene (BaP) and 7, 12-dimethylbenz(a)anthracene (DMBA). Unscheduled DNA synthesis was determined by measuring [methyl-3H]thymidine radioactivity incorporated into nuclear DNA of hepatocytes treated with carcinogens in the presence or absence of DHEA. Hydroxyurea $(5{\times}10^{-3} M)$was added to growth medium to selectively suppress normal replication. DHEA at concentrations ranging from $(1{\times}10^{-6} M)$ to$(5{\times}10^{-4} M)$ did not significantly inhibit unscheduled DNA synthesis induced by either MMS $(1{\times}10^{-4} M)$ or EMS $(1{\times}10^{-2} M)$. In contrast, DHEA-significantly inhibited unscheduled DNA synthesis induced by BaP $(6.5{\times}10^{-5} M)$ and DMBA.$(2{\times}10^{-5} M)$. DHEA-induced hepatotoxicity in rats was examined using lactate dehydrogenase (LDH) release as an indicator of cytotoxicity. DHEA exhibit no significant increase in LDH release compared with the control at 18 h. These data suggest that nontoxic concentration of DHEA does not affect the DNA excision repair process, but it probably influence the enzymatic system responsible for the metabolic activation of procarcinogens and thereby decreases the amount of the effective DNA adducts formed by the ultimate reactive carcinogenic species.
A novel solid-contact indium(III)-selective sensor based on bis-(1H-benzimidazole-5-methoxy-2-[(4-methoxy-3, 5-dimethyl-1-pyridinyl) 2-methyl]) thiosulfinate, known as an omeprazole dimer (OD) and a neutral ionophore, was constructed, and its performance characteristics were evaluated. The sensor was prepared by applying a membrane cocktail containing the ionophore to a graphite rod pre-coated with polyethylene dioxythiophene (PEDOT) conducting polymer as the ion-to-electron transducer. The membrane contained 3.6% OD, 2.3% oleic acid (OA) and 62% dioctyl phthalate (DOP) as the solvent mediator in PVC and produced a good potentiometric response to indium(III) ions with a Nernstian slope of 19.09 mV/decade. The constructed sensor possessed a linear concentration range from $3{\times}10^{-7}$ to $1{\times}10^{-2}$ M and a lower detection limit (LDL) of $1{\times}10^{-7}$ M indium(III) over a pH range of 4.0-7.0. It also displayed a fast response time and good selectivity for indium(III) over several other ions. The sensor can be used for longer than three months without any considerable divergence in potential. The sensor was utilized for direct and flow injection potentiometric (FIP) determination of indium(III) in alloys. The parameters that control the flow injection method were optimized. Indium(III) was quantitatively recovered, and the results agreed with those obtained using atomic absorption spectrophotometry, as confirmed by the f and t values. The sensor was also utilized as an indicator electrode for the potentiometric titration of fluoride in the presence of chloride, bromide, iodide and thiocyanate ions using indium(III) nitrate as the titrant.
High-voltage dielectric discharges are an emerging technique in environmental pollutant degradation, which are characterized by the production of hydroxyl radicals as the primary degradation species. The initiation and propagation of the electrical discharges depends on several physical, chemical, and electrical parameters such as 1st and 2nd voltage of power, gas supply, conductivity and pH. These parameters also influence the physical and chemical characteristics of the discharges, including the production of reactive species such as OH, $H_2O_2$ and $O_3$. The experimental results showed that the optimum 1st voltage and oxygen flow rate for RNO (N-Dimethyl-4-nitrosoaniline, indicator of the generation of OH radical) degradation were 160 V (2nd voltage of is 15 kV) and 4 L/min, respectively. As the 2nd voltage (4 kV to 15 kV) was increase, RNO degradation was increased and, generated $H_2O_2$ and $O_3$ concentration were increased. The conductivity of the solution was not influencing the RNO degradation, $H_2O_2$ and $O_3$ generation. The pH effect on RNO degradation was not high. However, the lower pH and the conductivity, the higher $H_2O_2$ and $O_3$ generation were observed.
Inhibitory mechanism of colored rice bran against cellular genotoxicity induced by chemical mutagen was studied using organic solvent extracts from a colored rice cultivar termed as Suwon415, and the mutagen, mitomycin C. Inhibitory effects of 70% ethanol extact and chloroform fraction from rice bran of Suwon415 were higher than those from Chuchung used as control. However, antioxidative activities of each fraction from Suwon415 were slightly lower than those from Chuchung, suggesting the involvement of a different inhibitory mechanism not related to antioxidation pathway. Using E. coli as the indicator cell, inhibitory mechanism of rice bran extract from colored rice against mutagenicity induced by mitomycin C was investigated to reveal the possibility that it acts in a desmutagenic manner. Further investigation to quantify the free mitomycin C in reaction mixture following incubation with rice bran extract demonstrated that rice bran extract might inhibit the cellular genotoxicity of mitomycin C by direct adsorption of the mutagen.
