Objectives : The objective of this study was to examine the effects of P. oleracea into the HCl-ethanol induced gastritis in rats, and to isolate and determine the chemical compounds from P. oleracea. Methods : The rats were orally administered with crude extract or fractions or isolated compounds of P. oleracea 30 mins before the induction of gastric lesion by oral administration of HCl-ethanol. The gastric lesional area was measured using pixel counting software. Then the chemical compounds from P. oleracea was isolated and determined by LC-MS and NMR. Results : The inhibition effect of oral administration of crude extract of P. oleracea at a dose of 500 mg/kg in HCl-ethanol induced gastritis was similar to cimetidine. Then, aqueous fraction at a dose of 240 mg/kg exhibited the effects similar to cimetidine. Then, the aqueous fraction was further separated by MPLC and yielded four sub fractions. Among those sub fractions, agent II at a dose of 40 mg/kg possessed the strongest effect in the HCl-ethanol induced gastritis. The water fraction yielded-Uridine, Adenosine, Guanosine, which were characterized by Mass, 1H-NMR, 13C-NMR. Conclusions : This study suggest that a P. oleracea and its compounds showed potent efficacy on the development of HCl-ethanol induced gastritis. Thus, P. olaracea can be a potential natural resource for the management of gastritis although the mechanism of action involved in the treatment remains to be explored.
The cyutotoxic effects of hot water soluble polysaccharides extract(PS) from the mushroom, Lentinus edodes, and in combinations with vitamin A or vitamin E on life span of ICR mice bearing P388 cancer cells and in vitro against P388 cancer cells were examined. The chemical components of PS and fractions were analyzed and survival time and cell number of P388 treated with extract fractions with and without vatamin A or E supplementation were also measured. The results obtained were summarized as follows; The extract of fraction B was shown to have the highest antitumor activity against P388 implanted in ICR mice. The antitumor fraction B was consisted of 82.0% of polysaccharide and 4.2% of protein. All three fractions seemed to have in vivo antitumor activity against P388, and fraction B showed the highest activity, In vitro P388 cell growht was inhibited 76%, 89%, 54% by the addition of fraction A, B and C respectively. Vitamin A or E did not appear to have any accelerating effects on either in vivo or in vitro cell cytotoxicity when each of them was combined with the PS and fractions. All three fractions contained more than 68% of polysaccharides. The fraction B showed the highest value of 88% in polysaccharides. Monsaccharides of the fraction B were identified as galactose(59.1%), glucose(29.2%), fructose(2.8%) and uronic acid(4.2%). Hydrolysis of protein from the fraction B was didentifed to have 17 kinds of defined and 5 undifined amino acids. The inhibitory effects of the hot water extracts from mushroom against cancer cell growth of P388 were stronger than the control group. And the survival time of ICR mice was shown to be 161% between the control group and the experimental groups.
This study was carried out to investigate the antimicrobial and antioxidant activities for applications of food additives and medicine using Rumex crispus which widely grown in domestic area. Antimicrobial activities of extract fractions were observed in all microbial species except for Pseudomonas areuginosa. Especially, Vibrio vulificus and Saccharomyces cerevisiae presented obvious antimicrobial activity. Antioxidant activities of the fractions of R. crispus extract were determined by DPPH radical scavenger activity. The fraction of ethyl acetate was presented similar antioxidant activities compared with that of BHA and ascorbic acid. Also, ethyl acetate fraction contained higher phenolic compounds than that of others. The antioxidant activity in the fractions of R. crispus extract was closely related with the content of phenolic compounds in that.
Talipariti hamabo is a plant growing around salt marshes in the Lava Coast region of Jeju Island, Korea. In this study, the extract of T. hamabo leaves was investigated for the anti-inflammatory, anti-oxidative and anti-bacterial activities and their active constituents were identified. In the anti-inflammatory tests using lipopolysaccharide-stimulated RAW264.7 cells, the ethyl acetate (EtOAc) fraction inhibited the nitric oxide production without causing cell toxicity. Moreover, the EtOAc fraction reduced pro-inflammatory cytokines (tumor necrosis factor-α, interleukin-6) as well as prostaglandin E2. In the anti-oxidative studies with DPPH and ABTS+ radicals, potent scavenging activities were observed in the EtOAc and n-butanol fractions. Upon the anti-bacterial tests using Staphylococcus epidermidis, EtOAc and n-butanol fractions exhibited good activities. Through the phytochemical studies on EtOAc fraction, three components were isolated by repeated column chromatography; oleic acid (1), p-hydroxyphenethyl-trans-ferulate (2), nicotiflorine (3). Based on these results, the extract of T. hamabo leaves can be developed as natural resources for cosmetic applications.

This study was attempted to isolate and identify the anticancer compounds from Eucommia ulmoides leaves using a human colon cancer cell line HCT-116. The petroleum ether extracts with anticancer activity was chromatographed on silica gel TLC and finally anticancer compounds was purified by HPLC. Their chemical structures were roughly elucidate by UV-VIS absorption spectral data HPLC elution pattern and FAM/MS spectroscopy. From this study these compounds were suspected to be pheophytin a formed by the removal of $Mg^{2+}$ from chlorophyll a and pyropheophytina formed by the removal of acetate group from pheophytin a respectively. To confirm the anticancer effects against HCT-116 cancer cell petroleum ether extract fractions of column chromatography and fractions separated on TLC were tested. All samples tested including the extract of petroleum ether fractions of column chromatograph and three bands (0.13,0.19,0.25) of TLC appeared to inhibit the growth of HCT-116 cancer cell however especially 0.19 and 0.25 fractions separated on TLC plate revealed the strongest effect. These results suggest that chlorophyll derivatives in Eucommia ulmoides may be potential anticancer agents against a human colon cancer cell HCT-116.
Studies were conducted on the nature of allelopathic effect of the substances in the waste of ginkgo leaves from pharmaceutic factory. In the first step, to find out whether there was any allelophatic effect, young seedlings of radish and rice were grown in the water (crude) extract of ginkgo leaf waste and in different liquid/lquid partitioned fractions of EtOAc at pH 9, EtOAc at pH 3, and BuOH. As second step, attempts were made to isolate and identify the allelophatic substance in different liquid/liquid partitioned fractions using GC/MS and NMR techniques. The water (crude) extract of ginkgo leaf waste retarded the growth of radish seedlings under 10% concentration. In case of rice seedlings, the water extract of ginkgo leaf extract showed adverse effect on the growth when combined with $3.3{\times}10^{-6}M$ gibberellin A3. All of the liquid/liquid fractions of crude extract showed strong retardation of seedling growth of radish and rice at the concentration of 1%. Allelophatic substance was isolated from the crude extract using liquid/liquid partition, column chromatography and HPLC techniques. The analytical results of isolated componet using GC/MS and NMR proved that the allelophatic substance in the ginkgo leaf wastes is catechol; one of phenol compounds. Based on the experiences current study, a practical method for the testing of allelophatic effect of crude extract of some materials was proposed. In this method, rice seeds were allowed to sprout until the length of coleoptile to reach 0.5 mm. Such seedlings were submerged in the solution containing supposedly allelophatic substance and the length of shoot and root was measured 3 days after treatment.
The chemical properties and physiological activities of the freeze-dried stromata of Cordyceps militaris were examined. A proximate analysis exhibited that the stromata consisted mainly of crude carbohydrate (74.3%), crude protein (11.5%), and moisture content (8.7%), with a low content of crude ash (4.%) and fat (1.3%). The carbohydrate was mostly composed of glucose (88.6%). A large quantity of essential fatty acids, including linolenic acid (33.8%) was also observed. An analysis of the component amino acid showed a relatively high ratio of the essential amino acids, lysine (101.2 mg/g), methionine (62.7 mg/g), and acidic amino acids of glutamic aicd (57.5 mg/g) and aspartic acid (43.9 mg/g), whereas a low of tyrosine content (4.7 mg/g). An examination of the cordycepin content indicated tht the stromata and mycelium-embedded media (silkworm pupae) contained 0.2% and 0.5%, respectively. From the examination of the physiological activities, based on methanol extract (M), ethylacetate extract (EA), and hot-water extract (HW) fractions of the stromata, the hot-water extract (HW) fraction showed the most potent intestinal immune modulating activity, anticoagulant activity, and anticomplementary activity, whereas the ethylacetate extract (EA) fraction exhihited a radical scavenging activity. Therefore, the results from the present study indicate that the stromata of C. militaris contain various healthy chemical ingredients, and that especially boiled water extract of whole body would apprear to provide beneficial physiological activities.
Uremia is associated with defective protein binding of weakly acidic drugs, whereas the protein binding of basic drugs tends to be normal. The exact chemical nature of compound(s) and mechanism for these changes as yet is unknown, and has not been defined. Organic solvent extraction of pooled normal human urine following hydrolysis by hydrochloric acid produced an extract, which when added to normal human serum, was capable of inducing binding defects similar to those in uremia. Binding defects were observed with the weakly acidic drugs such as nafcillin, salicylate, sulfamethoxazole and phenytoin while the binding of the basic drugs such as trimethoprim and quinidine were unaffected. The binding defects induced by the hydrolyzed urine extract could readily be corrected by same organic solvent extraction of acidified serum and the defects could be transferred to the normal human serum using the organic solvent layer at the physiologic pH (7.4). Followed by reacidification ind extraction of the binding defects induced serum with the same solvent, separated several fractions were obtained on thin-layer chromatography. One of these fractions could reinduce the binding defects and this factor(s) is apparently weakly acidic compounds and tightly bound to serum at physiologic pH, but extractable at acidic pH, and its molecular weight range is approximately 500 or less similar to those seen in uremia. These findings strongly support the hypothesis that the drug binding defect in uremia is due to the accumulation of endogenous metabolic products which arc normally excreted by the kidneys but accumulate in renal failure.
The ameliorating effect of the MeOH extract of the root of Platycodon grandiflorum on the ethanol-induced cognitive dysfunction in mice was investigated. The total MeOH extract was devided into two fractions, saponin rich fraction and non-saponin fraction. The mice with repeated administration of the total MeOH extract and those with saponin rich fraction both showed a markedly prolongεd latency time upon the step-through test (passive avoidance performance) performed after acute ethanol intoxication, while those with non-saponin rich fraction did not. The results suggest that the extracts from the root of Platycodon grandiflorum exert a beneficial effect on ethanol-induced memory impairment in mice and also suggest that these effects of the extracts might be ascribed to the saponin constituents.
We investigated the chemical components of red onion powder dried using the low temperature vacuum method and the inhibitory effects of solvent extracts of the dried red onion powder on the growth of HT-1080 human fibrosarcoma and HT-29 human colon cancer cells and $H_2O_2$-induced oxidative stress. The moisture content of the dried red onion powder was 17.95%, while the vitamin C content was 96 mg/100 g and the total phenols content was 39.1 mg/mL. The inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts of the red onion powder on the growth of HT-1080 and HT-29 cancer cells increased in a dose dependent manner (p<0.05). The inhibitory effect was greater on the growth of HT-29 cells, while the A+M extracts had a higher inhibitory effect than the MeOH extracts. Treatment with the hexane, 85% aq. methanol, butanol and water fractions of the extract led to significant inhibition of the growth of both cancer cell lines (p<0.05). Among the fractions, the hexane and 85% aq. methanol fractions showed a greater inhibitory effect. To determine the protective effect on $H_2O_2$-induced oxidative stress, a DCFH-DA (dichlorodihydrofluorescin diacetate) assay was conducted. All fractions, including the crude extracts of dried red onion, appeared to lead to a significant reduction in the levels of intracellular reactive oxygen species (ROS), and these reductions occurred in a dose dependent fashion (p<0.05). Among the fractions, the 85% methanol fraction showed the greatest protective effect on the production of lipid peroxides.
