Determination of ethylene glycol in alkali wastewater by gas chromatography/mass spectrometry (GC/MS를 이용한 알칼리 폐수 중 ethylene glycol 정량)

  • Treatment with aqueous sodium hydroxide solution plays an important role to impart certain desirable properties - feel and luster on the surface of polyester fiber. In this process alkali wastewater contains disodium terephthalate, ethylene glycol (EG) and residual sodium hydroxide. In this paper we report a new method containing the pretreatments of derivatization with benzoyl chloride and solvent extraction using pentane. The calibration curve of EG determined by GC/MS-SIM shows a good linearity in the range of 0.1 to $25{\mu}g/mL$ having the standard deviation of ${\leq}8.7%$. The recovery and the detection limit of this method are 91.9-93.7% and $0.05{\mu}g/mL$ respectively.

  • We have been studing sensitive non-enzymatic chemiluminescence (CL) imaging methods for the detection of DNA. For one of our methods, a unique chemical derivatization reagent. 3'. 4', 5'-trimethoxyphenylglyoxal (TMPG) was utilized. This reagent reacted specifically with guanine bases in nucleic acids to quickly produce a chemiluminescent derivative under mild reaction conditions. (omitted)

Fabrication of Polymer Thin Films on Solid Substrates (고체 기판에 고분자 박막의 고정화)

  • Surface properties are important for determining the functions and uses of materials. So modification of materials with polymer thin films has emerged as an important method to control the physical and chemical properties of the surface layer. We report a simple and effective method to photochemically attach thin polymeric layers to solid surface without chemical derivatization of the substrate and/or the polymer. The system is based on a photoreactive poly(4-vinylpyridine) (P4VP) thin film which is formed on the $SiO_{2}$ surface via spin coating. This substrate is then covered with another polymer film that is reacted with the benzyl radical moieties by UV irradiation. As a result of photochemical reaction, a thin layer of the later polymer is covalently bound to the surface of P4VP. Unbounded polymer is removed by sonication. The thickness of the attached film is a function of the irradiation time and the molecular weight of the polymer. Spatially defined polymer thin films can be fabricated by way of photolithography.

Simultaneous Fluorimetric Determination of On-line Preconcentrated HANs, DCAD and TCAD by Using RPLC with a Postcolumn Derivatization System

  • A simultaneous analytical method has been developed for the fluorimetric determination of haloacetonitriles (HANs) [dichloroacetonitrile (DCAN), trichloroacetonitrile (TCAN), dibromoacetonitrile (DBAN), haloacetamides [dichloroacetamide (DCAD), and trichloroacetamde (TCAD)] in drinking water by using the combined on-line perconcentration/reversed phase liquid chromatography (RPLC)-postcolumn detection system. This on-line perconcentration system was achieved by employing a precolumn packed with a commercial solid phase extraction (SPE) sorbent for the enrichment and purification of the target analytes. The haloacetonitriles and haloacetamides were separated on CN analytical column in a 7.5% methanol-0.02 M phosphate buffered mobile phase at pH 3. The column effluents were reacted with postcolumn reagents of ophthaldialdehyde (OPA) and sulfite ion at pH 11.5, to produce a highly fluorescent isoindole fluorophore, which were measured with a fluorescence detector. Under the optimized conditions for RPLC and the postcolumn derivatization system all of the coefficient of determination of the standard calibration curves for the target analytes were over 0.99 and had a linear range from 5 to 100 ${\mu}g/L$. The detection limits showed 1.6 ${\mu}g/L$ for DCAD, 0.1 ${\mu}g/L$ for TCAD, 0.6 ${\mu}g/L$ for DCAN, 1.6 ${\mu}g/L$ for TCAN and 1 ${\mu}g/L$ for DBAN, and the recoveries were ranged from 64 to 99% except for DCAD with precisions less than 4.9% in distilled water, and from 72(${\pm}4%$) to 116%(${\pm}2%$) in tap water.

  • Linkage positions in oligosaccharides may be obtained by FAB CAD MS/MS (Fast Atom Bombardment Collision Activated Dissociation Mass Spectrometry/Mass Spectrometry). Acetylated derivatives of the linkage-isomeric trisaccharides exhibited more useful product ion patterns than the free trisaccharides and provided specific fragmentation patterns according to linkage positions. The reason for the useful linkage dependent spectra patterns of acetylated forms is related to the ability of each linkage in the oligosaccharides to absorb different levels of collision energy and rotational freedom of the individual glycosidic linkage.

  • An efficient and novel positive charge-doping on the sidewalls of multi-walled carbon nanotubes has been achieved in the presence of tetrahydrofuran as a dopant and Lewis acidic ionic liquids, [bmim]$Sb_nF_{5n+1}$ (n ${\geq}$ 2; bmim = 1-butyl-3-methylimidazolium), as an activator, leaving air-stable derivatives having positively charged sidewalls and the counter anions, [MWCNT$^{y+}$][SbF$_6^-$]$_y$ (MWCNT = multi-walled carbon nanotube). The derivatization took place very fast in one-pot and under mild reaction conditions. The ionic structure enabled a tunable dissolution of the derivatives in various solvents through anion exchange.

  • The screening of the anti-cancer activity of the chemical library provided 2-thio-4-aminopyrimidine as the initial hit. The confirmation of structure and biological effect of hit was performed by synthesis and biological evaluation. The optimization of hit was performed by derivatization of substituents while keeping the core structure. The evaluation of growth inhibitory activity was carried out using SRB assay against 6 human cancer cell lines and human fibroblast. The growth inhibitory activity of compounds showed substituent dependency and more than 5 compounds showed complete growth inhibition of cancer cell lines at 10 ${\mu}M$ concentration. Chemical library screening followed by synthetic modification provided possibility that 2-thio-4-aminopyrimidine can be used as a new scaffold for the development of anti-cancer agent.

  • Protein phosphorylation plays a key role in signal transduction in cells. Since phosphoproteins are present in low abundance, enrichment methods are required for their purification and analysis. Chemical derivatization strategies have been devised for enriching phosphoproteins and phosphopeptides. In this report, we employed a strategy that replaces the phosphate moieties on serine and threonine residues with a biotin-containing tag via a series of chemical reactions. Ribulose 1,5-bisphosphate carboxylase/oxygenase (RUBISCO)-depleted protein extracts prepared from Arabidopsis seedlings were chemically modified for 'biotin-tagging'. The biotinylated (previously phosphorylated) proteins were then selectively isolated by avidin-biotin affinity chromatography, followed by two-dimensional gel electrophoresis (2-DE) and matrix-assisted laser-desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS). This led to the identification of 31 protein spots, representing 18 different proteins, which are implicated in a variety of cellular processes. Despite its current technical limitations, with further improvements in tools and techniques this strategy may be developed into a useful approach.

  • Co nanoparticles were prepared by the reverse micelle technique (NaBH₄reduction of cobalt chloride in a reversed micelle solution of didodecyldimethylammoniumbromide (DDAB)/toluene). The size and the shape of Co nanoparticles could be easily controlled by changing the water contents and micelle concentrations, and the solubility of Co nanoparticles was systematically tuned by choosing appropriate surface capping organic ligand molecules. Furthermore, a novel nanofabrication process was clearly demonstrated, which generated oxide over-coated Co nanorods from Co nanoparticles in organic solution by slow oxidation with an external magnetic field.

  • A new sensitive fluorimetric method for the determination of N-methylcarbamates, a class of well known insecticides, based on the derivatization with 7-chloro-4-nitrosobenz-2-oxa-1,3-diazole (NBD-Cl) has been developed. Unreacted NBD-Cl was eluted ahead of derivatized carbamates from C-18 bonded column. An argon ion laser was used as an excitation source of chromatographic eluents and its fluorescence signal was monitored with optical multichannel analyzer. The detection limits of various carbamates were about 100 pg range and the working curves were linear to $10^4-10^5$ nanogram ranges.
