The lack of a clonal renin-secreting cell line has greatly hindered the investigation of the regulatory mechanisms of renin secretion at the cellular, biochemical, and molecular levels. In the present study, we investigated whether it was possible to induce phenotypic switching of the renin-expressing clonal cell line As4.1 from constitutive inactive renin secretion to regulated active renin secretion. When grown to postconfluence for at least two days in media containing fetal bovine serum or insulin-like growth factor-1, the formation of cell-cell contacts via N-cadherin triggered downstream cellular signaling cascades and activated smooth muscle-specific genes, culminating in phenotypic switching to a regulated active renin secretion phenotype, including responding to the key stimuli of active renin secretion. With the use of phenotype-switched As4.1 cells, we provide the first evidence that active renin secretion via exocytosis is regulated by phosphorylation/dephosphorylation of the 20 kDa myosin light chain. The molecular mechanism of phenotypic switching in As4.1 cells described here could serve as a working model for full phenotypic modulation of other secretory cell lines with incomplete phenotypes.
본 연구는 국내 제조기업의 GVC 참여 유형과 거버넌스에 따른 수출성과를 실증분석한 것이다. KOSPI 상장기업의 패널자료를 바탕으로 관계투자, 정보교환, 혁신은 신뢰의 조절역할과 함께 공급망 통합수준과 기업성과에 영향을 미치는 것으로 분석되었다. 또한 공급망 통합 정도가 GSCM의 실적과 수출 실적에 영향을 미치는 것으로 나타났다. GVC 거버넌스 유형의 경우 통합강도가 높을수록 다른 유형에 비해 성과가 높은 것으로 분석되었다. 이러한 결과는 신뢰 기반의 공급망 통합과 지속적인 GVC 투자가 매우 중요함을 보여준다. 이는 높은 통합 강도와 함께 높은 수준의 GVC 거버넌스가 성공적인 GSCM 성과로 이어질 수 있음을 설명한다. GVC 참여기업의 관계역량과 신뢰도에 기반한 혁신적인 투자전략은 글로벌 파트너십 형성 및 통합 정도에 긍정적인 영향을 미칠 수 있다.
${\gamma}-Oryzanol$ is one of the chain breaking antioxidants. Both sterol (triterpene) and phenolic hydroxyl groups in the structure of ${\gamma}-oryzanol$ may be responsible for its antioxidative function. We hypothesize that ${\gamma}-oryzanol$ is more effective in preventing the autoxidation of polyunsaturated fatty acid (PUFA) than the synthetic phenolic compounds in an oil/water (O/W) emulsion system. The antioxidative effectiveness of different concentrations of ${\gamma}-oryzanol$ and synthetic antioxidants was evaluated at different incubation times (0, 4, 8, 16, and 32 h) by measuring both the formation of hydroperoxides and the decomposition product of hydroperoxides (hexanal) in each emulsion system. Overall, the order of effectiveness of various antioxidants for inhibiting the formation of hydroperoxide in the O/W emulsion was: ${\gamma}-oryzanol$> tert-butylhydroquinone (TBHQ)> butylated hydroxytoluene (BHT)> butylated hydroxyanisole (BHA). O/W emulsion with selective lower concentrations of ${\gamma}-oryzanol$ showed better effectiveness than that with higher concentration of synthetic antioxidants. However, the ability of both ${\gamma}-oryzanol$ and synthetic antioxidants to decompose hydroperoxide was similar. ${\gamma}-Oryzanol$ was more effective antioxidant than the synthetic phenolic compounds in preventing the formation of hydroperoxide in the O/W emulsion system.
Early leaves within the Viola albida complex were investigated by scanning electron microscopy in order to determine the morphological segments during morphogenesis. The early leaf development of V. albida var. albida could be morphologically divided into the eight stages in the following order: I, the initiation of shoot germination; II, the conical growth directionally of the leaf; III, the adaxial and abaxial formation of the leaf; IV, the initiation of the stipule; V, the formation of a transitional zone between the leaf blade and petiole; VI, the expansion of the upper part of the leaf blade; VII, the formation of almost all parts of the early leaf; VIII, the early simple leaf. Viola albida var. takahashii differs from V. albida var. albida by additional stages, i.e., V-1, the initiation of the first lateral lobe at the both lateral parts of the leaf after the stage V and an early lobed leaf. Viola albida var. chaerophylloides is also distinguished from two taxa by two developmental features, V-2, the initiation of a second lateral lobe below of the first lateral lobe, and an early palmately compound leaf. These findings suggest that the Viola albida complex would be in the process of peramorphosis, showing developmental changes in a chain of events, leading to a different leaf shape. These data would also be useful for isolating genes that give rise to different leaf morphogenesis outcomes among the taxa in the Viola albida complex.
Objective: Pluripotent stem cell-derived lymphatic endothelial cells (LECs) show great promise in their therapeutic application in the field of regenerative medicine related to lymphatic vessels. We tested the approach of forced differentiation of mouse embryonal stem cells into LECs using biodegradable poly lactic-co-glycolic acid (PLGA) nanospheres in conjugation with growth factors (vascular endothelial growth factors [VEGF-A and VEGF-C]). Methods: We evaluated the practical use of heparin-conjugated PLGA nanoparticles (molecular weight ~15,000) in conjugation with VEGF-A/C, embryoid body (EB) formation, and LEC differentiation using immunofluorescence staining followed by quantification and quantitative real-time polymerase chain reaction analysis. Results: We showed that formation and differentiation of EB with VEGF-A/C-conjugated PLGA nanospheres, compared to direct supplementation of VEGF-A/C to the EB differentiation media, greatly improved yield of LYVE1(+) LECs. Our analyses revealed that the enhanced potential of LEC differentiation using VEGF-A/C-conjugated PLGA nanospheres was mediated by elevation of expression of the genes that are important for lymphatic vessel formation. Conclusion: Together, we not only established an improved protocol for LEC differentiation using PLGA nanospheres but also provided a platform technology for the mechanistic study of LEC development in mammals.
The phase transformation from cellulose I into cellulose II in woods by way of Na-cellulose I was examined by x-ray diffraction analysis.The formation of Na-cellulose I in woods increased with the increase of treating time in alkali solution. When compression wood was treated with 20% NaOH solution at room temperature for 1 day, the x-ray diagram showed only Na-cellulose I. On the other hand, the x-ray diagram of tension wood showed a mixture of cellulose I and Na-cellulose I. Cellulose I of tension wood could not be transformed completely into Na-cellulose I even after 10-day treatment, but was transformed into Na-cellulose I after 30-day treatment. Na-cellulose I of compression and tension woods was converted to the cellulose I pattern and the mixture of cellulose I and cellulose II, respectively, after washing with water and drying at 20℃. Cellulose I regenerated from Na-cellulose I in wood could not be converted to cellulose II by delignification. Thus, it revealed that the delignification of the alkali-treated wood did not affect their cellulose structures. From the results, therefore, it can be concluded that lignin in woods prevents the formation of the stable Na-cellulose I and the conversion from cellulose I to cellulose II. This means that the conversion of chain polarity of wood cellulose hardly occurs during mercerization because cellulose microfibrils are fixed by lignin which not to be intermingled.
We present a quantitative analysis of the relationship between the gas dynamics and star formation history of DDO 210 which is an irregular dwarf galaxy in the local Universe. We perform profile analysis of an high-resolution neutral hydrogen (HI) data cube of the galaxy taken with the large Very Large Array (VLA) survey, LITTLE THINGS using newly developed algorithm based on a Bayesian Markov Chain Monte Carlo (MCMC) technique. The complex HI structure and kinematics of the galaxy are decomposed into multiple kinematic components in a quantitative way like 1) bulk motions which are most likely to follow the underlying circular rotation of the disk, 2) non-circular motions deviating from the bulk motions, and 3) kinematically cold and warm components with narrower and wider velocity dispersion. The decomposed kinematic components are then spatially correlated with the distribution of stellar populations obtained from the color-magnitude diagram (CMD) fitting method. The cold and warm gas components show negative and positive correlations between their velocity dispersions and the surface star formation rates of the populations with ages of < 40 Myr and 100~400 Myr, respectively. The cold gas is most likely to be associated with the young stellar populations. Then the stellar feedback of the young populations could influence the warm gas. The age difference between the populations which show the correlations indicates the time delay of the stellar feedback.
Injectable hydrogels are quite promising materials due to their potential to minimize invasive implantation and this provides versatile fitness irrespective of the damaged regions and facilitates the incorporation of bioactive agents or cells. In situ gel formation through stereocomplex formation is a promising candidate for injectable hydrogels. In this paper, a new series of enantiomeric, four-arm, PEG-PLA block copolymers and their stereocomplexed hydrogels were prepared by bulk ring-opening polymerization of D-lactide and L-lactide, respectively, with stannous octoate as a catalyst. The prepared polymers were characterized by $^1H$ nuclear magnetic resonance (NMR) spectroscopy, Fourier-transform infrared (FT IR) spectroscopy, gel permeation chromatography (GPC) and thermal gravitational analysis (TGA), confirming the tailored structure and chain lengths. The swelling and degradation behavior of the hydrogels formed from a selected copolymer series were observed in different concentrations. The degradation rate decreased with increasing polymer content in the solution. The rheological behavior indicated that the prepared hydrogel underwent in situ gelation and had favorable mechanical strength. In addition, its feasibility as an injectable scaffold was evaluated using a media dependence test for cell culture. A Tris solution was more favorable for in situ gel formation than PBS and DMEM solutions were. These results demonstrated the in situ formation of hydrogel through the construction of a stereocomplex with enantiomeric, 4-arm, PEG-PLA copolymers. Overall, enantiomeric, 4-arm, PEG-PLA copolymers are a new species of stereocomplexed hydrogels that are suitable for further research into injectable hydrogels.
아밀로오스 함량이 다른 옥수수 전분을 121$^{\circ}C$에서 호화시켜 팽윤력과 용해도를 측정했을때, 팽윤력은 찰옥수수 전분인 Amioca가 가장 컸고, PFP, Amaizo, Amylomaize VII 순이었으며 Amaizo의 경우 용출된 겉보기 아밀로오스의 요드 친화력이 가장 높았고, 용해도 역시 높게 나타났다. BrooHield viscometer를 이용하여 0.25 N NaOH 용액으로 측정한 알칼리 호화양상은 Amylomaize VII이 최고점도 l0$\times$$10^3$cp를 나타낸 반면 Amaizo는 70$\times$$10^3$cp로 Amylomaize VII에 비해 높은 값을 나타냈고, PFP는 70$\times$$10^3$cp까지 증가했다가 감소하였다. 전분과 물을 1 : 9로 하여 121$^{\circ}C$에서 가열-냉각 횟수를 4회 반복하며, 각 단계에서 형성된 겔의 성질을 비교하면 Amaizo로 만든 겔의 견고성, 부착성, 탄성과 응집성이 가장 컸으며, 견고성은 가열-냉각처리를 2회 반복한 경우 가장 컸고 그 후 감소하였다. 옥수수전분 겔의 성질은 아밀로오스 함량보다는 사슬길이나 가열 중 용출되는 부분의 성질에 더 상관성이 높았다.
매우 얇고 다공성 비인화성 와이퍼 용지를 아인산 수용액 처리에 의해 준비되었다. H3PO3 처리는 와이퍼 용지의 난연성을 불연성 수준으로 개선시켰다. 열중량 분석 결과 700℃에서 잔량이 50%까지 증가했다. 이것은 본 실험과 같은 아인산 처리가 셀룰로오스 사슬의 탈수를 돕고, 연소중에 차르(char)와 유사한 구조의 형성을 촉진한다는 것을 의미한다. FT-IR과 X선 광전자 분광학에서는 H3PO3의 일부분은 셀룰로오스 사슬의 작용기와 반응하는 것으로 보였다. 반응한 아인산은 셀룰로오스의 체인의 탈수를 촉진하고, 차르와 같은 구조의 형성을 도와 와이퍼 용지의 난연성을 향상시킨다.
