  • The present study was carried out to know the distribution of freshwater mollusk on the chinese liver fluke in the middle of August, 1997 at the upper area of Kyungan stream located in Yongin Kun, Kyungki province. The snails were collected from the shallow and irrigation route in a rural area located along the upper area of Kyungan stream. The snails collected by hand were brought to the laboratory and cercariae naturally shed from the snails, Parafossarulus manchouricus were examined. Parafossarulus manchouricus is medically important intermediated host of the transmission of digenetic trematoda to human. Each snail was placed in aerated tap water for the detection of ceracariae and examined for expelled cercariae. Infection rate of the cercariae of Clonorchis sinensis was 3 (1.5%) among 195 individuals collected in the habitat. The identification of the cercaria of Clonorchis sinensis was performed as described by Komiya and Tajimi.

Prevalence of Haplorchis taichui in Field-Collected Snails: A Molecular Approach

  • The prevalence of the cercarial stage of an intestinal trematode, Haplorchis taichui, in thiarid snails (Gastropoda: Thiaridae) was investigated using light microscope and species-specific PCR procedures. A total of 988 snails were collected from Mae Taeng district, Chiang Mai province, northern Thailand, which comprised of 3 species; Melanoides tuberculata, Tarebia granifera, and Thiara scabra. The overall prevalence of pleurolophocercous cercariae was 21.7% as determined by the morphology. For genetic detection of H. taichui infection in snails, 2 primers Hapt_F (5'-GGCCAACGCAATCGTCATCC-3') and Hapt_R (5'-GCGTCGGGTTTCAGACATGG-3'), were used. The genomic DNA of H. taichui, which was used as a positive control, gave an amplification of the 256 bp fragment. The overall prevalence of H. taichui from specific PCR was 9.7%. The proportion of H. taichui among the pleurolophocercous cercariae in this study was 44.9%.

PREVALENCE OF FASCIOLA CERCARIAE IN LYMNAEID SNAILS IN BANGLADESH

  • A total of 4149 Lymnaea auricularia var rufescens and 401 L. luteola snails were collected and examined in six periods from May, 1989 to April, 1990 at Savar, Dhaka, Bangladesh. Fasciola cercariae (Gymnocephalous cercariae) was found in 13 (0.31%) L. auricularia var rufescens, but was absent in all L. luteola examined. Prevalence of Fasciola cercariae in the snails varied significantly (p < 0.05) in different periods (months) of the year with higher prevalence in July-August (0.77%), followed by September-October (0.52%) and May-June (0.45%). Cercariae did not appear in the snails from November to April.

Morphological Characteristics and Phylogenetic Trends of Trematode Cercariae in Freshwater Snails from Nakhon Nayok Province, Thailand

  • The prevalence of cercarial infection in freshwater snails and their evolutionary trends were studied in Nakhon Nayok province, Thailand. A total of 2,869 individual snails were examined for parasitic infections. The results showed that 12 snail species were found to host larval stages of trematodes with an overall prevalence of 4.7%. The infected specimens included 7 types at the cercarial stage; cercariae, megalurous cercariae, echinostome cercariae, furcocercous cercariae, parapleurolophocercous cercariae, virgulate cercariae, and xiphidiocercariae. Regarding molecular identification, ITS2 sequence data of each larval trematode were analyzed, and a dendrogram was constructed using the neighbor-joining method with 10,000 replicates. The dendrogram was separated into 6 clades (order/family), including Echinostomatida/Echinostomatidae, Echinostomatida/Philophthalmidae, Opisthorchiida/Heterophyidae, Plagiorchiida/Prosthogonimidae, Plagiorchiida/Lecithodendriidae, and Strigeatida/Cyathocotylidae. These findings were used to confirm morphological characteristics and evolutionary trends of each type of cercariae discovered in Nakhon Nayok province. Furthermore, this investigation confirmed that the ITS2 data of cercariae could be used to study on phylogenetic relationships or to determine classification of this species at order and/or family level when possible.

Survey on Encysted Cercaria of Trematodes from Eresh-water Fishes in Tongjin Riverside Areas in Korea (동진강 유역 담수어에 기생하는 흡충류 피낭유충 조사)

  • In an attempt to clarify the epidemiological feature of distomiasis in Tongjin riverside area, the prevalence of distomiasis in the residents and infection rates of the metacercariae in fresh-water fishes were investigated at the upper, middle and lower reaches of the river from January to April, 1984. The results obtained were summarized as follows: 1. Out of a total of 931 fresh-water fishes which composed of 33 different species, 611 fishes(65.6%) of 31 species were found positive with digenetic trematode metacercariae of 16 different species, and there were some differences in infection rates of the metacercariae among the fishes in the 3 parts of the river; 53.8% in upper, 80.7% in middle, and 61.0% in lower reaches, respectively. 2. Infection rates of the metacercariae of Exorchis oviformis, Metagonimus yokogawai, Echinochasmus japenicus, Metorchis orientalis and Clonorchis sinensis in the fishes were 48%,29%, 115, 7.9% and 6.3oA, respectively. 3. The average number of the encysted larvae of Clonorchis found in fish body/gram showed 4.44 in Pseudorasbera larva, Gnathepegon coreanus (1.2), Microphysogoio yaluensis (0.76), Abbottina springeri (0.4), Acanthorhodeus asmussi (0.21) and Cultriculus eigenmanni (0.17), respectively. 4. The average number of the metacercariae of Metagonimus found in fish body/gram disclosed 34.01 in Zacco platypus, Zacco temmincki (16.46), Carassius carassius (5.35), Moroco oxycephalus (1.54) , Aphyocypris chinensis (1.5) and etc., respectively. 5. Detection rates of the eggs of Clonorchis and Metagonimus among residents were 1.1% and 0.8%, respectively, out of a total 923 Persons.

Complement-mediated tail degradation of Neodiplostomum seoulense cercariae

  • The furcocercus cercariae of Neodiplostomum seoulense (Digenea: Neodiplostomidae) penetrate the skins of tadpoles and shed their tails. The speculated mechanism of this tail loss was physical efforts required to produce a vigorous zigzag motion during skin penetration; no other mechanism has been proposed. We examined the relationship between the host serum and cercarial tail loss. Cercariae of N. seoulense were collected from experimentally infected Segmentina hemisphaerula, and lots of 300 cercariae were cultured in medium 199 contained several types of sera. Cercarial tail degradation was induced in all media, but all the cercariae cultured except those cultured in media containing fetal bovine serum (FBS) died within 48 hr. After 72 hr cultivation in media containing FBS, cercarial tail degradation was induced in 67.0%; in continuous cultivation 13.3% of larvae survived for 7 days. Tail degradation did not occur in the absence of serum and when serum was heat inactivated at $56^{\circ}C$ for 30 min. The addition of 20 mM ethylenediaminetetraacetic acid (EDTA) blocked cercarial tail degradation completely. Moreover, the addition of 20 mM $MgCl_2$ restored tail degradation blocked by EDTA. These results suggest that the alternative complement pathway is related with the N. seoulense cercarial tail degradation induced by serum.

Molecular Differentiation of Schistosoma japonicum and Schistosoma mekongi by Real-Time PCR with High Resolution Melting Analysis

  • Human schistosomiasis caused by Schistosoma japonicum and Schistosoma mekongi is a chronic and debilitating helminthic disease still prevalent in several countries of Asia. Due to morphological similarities of cercariae and eggs of these 2 species, microscopic differentiation is difficult. High resolution melting (HRM) real-time PCR is developed as an alternative tool for the detection and differentiation of these 2 species. A primer pair was designed for targeting the 18S ribosomal RNA gene to generate PCR products of 156 base pairs for both species. The melting points of S. japonicum and S. mekongi PCR products were $84.5{\pm}0.07^{\circ}C$ and $85.7{\pm}0.07^{\circ}C$, respectively. The method permits amplification from a single cercaria or an egg. The HRM real-time PCR is a rapid and simple tool for differentiation of S. japonicum and S. mekongi in the intermediate and final hosts.

Studios on Echinostomu spp. in the Chungju Reservoir and upper streams of the Namhan River (충주호 및 그 상류 지역의 Echinostoma spp.에 대한 연구)

  • The present study was performed to know the epidemiological statug of echinostomiasis in the Chungju Reservoir and upper streams of Namhan River, together with an experimental study on the life history o( Echinostoma hortense. The stool specimens of 169 inhabitants and 473 junior high school students from 5 different villages revealed 3(0.5%) echinostomatid egg Positive cases. E. hortense adult worms were recovered from one patient after a treatment and purgation. For the other two patients, it was presumed that one had 2. hortense and another E. cinetorchis infection, based on the morphology of eggs. Five kinds of freshwater snails(168 Radix auricularia coreane, 534 Physa arutp, 144 Hippeutis cnntori, 56 Cipcngopaludina chinensis malleata and 125 Semiiulcospira nediila glebus) examined for the cercariae of echinostomes showed negative results. Ten kinds of freshwater fishes examined for E. herten-'emetacercariae revealed positive rates as Misgurnus anguillicaudatus 40.5%, Odentobutis obscura interrupta 20.3%, Moroco onycephalus 3.9%, and Coreoperca hawamebari 2.0%. In the experimental study, the metacercariae of E. horlense were infected to rats, eggs were collected from adult worms and cultivated, and miracidia were obtained. The miracidia were artificially infected to freshwater snails (R. auricularia), and cercarial shedding was studied. It was revealed that, when the snails were kept at a low temperature ($24^{\circ}C$), only 523 cercariae (on average) were produced during 24 hrs, while they were at a high temperature ($30^{\circ}C$), as many as 9, 990 cercariae (on average) were shed during the same time. The experimental infection of E. hortense cercariae to freshwater fishes was successful in 0. obscura interrupta 52.0%, M. anguillicaudatus 30.3%, C. kawamebari 27.0%, Cobitis lutheri 15.0%, M. cxycephazus 7.3%, Pseudogobio esocinus 4.3%, Squalidus cereanus 2.0%, Zccco platypus 1.3%, and Pungtungia herzi 1.3%. However, infection was not successful to snails, C. chinensis. It has been proved that the Chungju Reservoir and upper streams of Namhan riyrr are endemic areas of echinostomiasis, especially of E. horfense, and snails such as R. auritulerie coreana and fishes such as O. obscura interrupts, M. gnguillicgudgtus, and others are taking the role of 6rst and second intermediate hosts, respectively.

The Wormicidal Substance of Fresh Water Fishes on Clonorehis sinensis V. Purification and Chemical Charaeterization of Clonorehicidal Substance from Epidermal Mucus of Cyprinus carpio (간흡충에 대한 살충성 물질에 관한 연구 V. 잉어 체표점액내 살충성물질의 화학적 성상)

  • As a series of studies to clarify clonorchicidal substances in body surface mucus of some freshwater fishes, the substance in the epidermal mucus of Cyprinus carpio was isolated by silica gel column and thin layer chromatography and analysed for its chemical nature. Wormicidal trial was done in vitro, and the results obtained are summarized as follows: The mucus was extracted by ethyl ether and separated into 4 fractions by column chromatography using benzene as solvent. The second fraction with yellowish red colour among them showed the strongest clonorchicidal effect. The yellowish red fraction obtained by column chromatography was then fractionated into 6 spots by thin layer chromatography with petrol/ etherjchloroform (30/70, v/v), and the Rf. 0.714 spot among the 6 spots showed the strongest effect. The Rf. 0.714 spot was further fractionated into 6 spots by thin layer chromatography with benzene/acetone (90/10, vlv), and the Rf. 0.800 spot among the later 6 spots revealed the strongest effect. The Rf. 0.800 spot was chromatographed on column with benzene and 2 fractions were obtained. The second fraction of light brown colour represented the final purified fraction. By these Purification Procedures, clonorchicidal substance was Purified 15-fold with 0.035 yield from the mucus of C. carpio, and 10mg of the final fraction killed the cercaria in 26 min, the metacercaria in 115 min, and the adult in 443 min. Infra red and nuclear magnetic resonance spectrometric analysis of the purified substance revealed that the substance belongs to an ethyl ester of unsaturated fatty acid with 2 double bonds, 15 methylene groups and 1 methyl group.

Experimental Studies on the Second Intermediate Host of Clonorchis sinensis I.   • The present studies were undertaken to determine accurately whether Korean native fresh-water shrimps can serve as the second intermediate host of Clonorchis sinensis. The experimental results obtained were as follows; 1. Metacercaria of Clonorchis sinensis was not discovered in 36 Macrobranchium nipponensis (De Hann), 1992 Palaemon paucidens (De Hann) and Palaemon madestus Heller, and 860 Caridina leucastica Stimpson, collected in Kimhae district, which were digested in artificial gastric juice, using binocular dissecting microscope. 2. Cercariae of the Chinese liver fluke could not approach the body wall of young shrimps due to streaming caused by their motion. 3. Young Palaemon spp. of 247 were divided into two groups and placed in two different room aquariums, one is at high temperature, the other is normal. Then they were contacted with the cercariae. Only 11 degenerated metacercariae were found in 9 Palaemon spp. It was suggested that they had died immediately or within a few days after penetration. Furthermore the temperature did not affect the infective ratio. 4. At the field experimental aquarium, no metacercaria of Clonorchis sinensis was discovered tn 100 Palaemon spp. which had been brought into contact with 5000 Parafossarulus manchouricus two trials. 5. Specific changes in the cercariae mixed with the extract of shrimps meat were not observed through the dissecting microscope. 6. These results suggest that the Korean native shrimps cannot serve as the intermediate host of Clonorchis sinensis.
