Effects of Arctii Fructus on the Atopic dermatitis (우방자(牛蒡子)가 아토피 피부염에 미치는 영향)

  • Objective : Atopic dermatitis has a close relationship with degranulation of mast cell and separation of Histamine. As there's no experiment with herb, using Arctii Fructus, we investigated experimental influence of Arctii Fructus on degranulation of mast cell and separation of histamine in SD rat. Methods : The SD rats are classified into three groups. One group is normal one treated by normal saline before medical treatment. The other is control group prescribed to Compound 48/80 before normal saline treatment. And the third is experimental group prescribed to compound 48/80 after medical treatment of Arctii Fructus. Then, I investigated the experimental results by measuring the degree of degranulation and separation of histamine. The results of investigation on SD rat group showing the degree of inhibitory effect of degranulation of a mast cell are as follow, the normal group treated by normal saline reflecting the degree of degranulation is $6.10{\pm}0.20\;%$, the control group treated by only compound 48/80 is $87.56{\pm}11.00\;%$, the experimental group which treated by compound 48/80 and Arctii Fructus's medical treatment is $16.26{\pm}4.67\;%$. Results : The normal group treated by only normal saline reflecting the degree of degranulation is $6.10{\pm}0.20\;%$, the control group treated by only compound 48/80 is 87.56=11.00 %, the experimental group treated by compound 48/80 and Arctii Fructus's medical treatment is $16.26{\pm}4.67\;%$. This result indicates that the degree of degranulation of mast cell is obviously inhibited (p<0.0l) in the experimental group in comparison with control one. The analysis of data obtained from plasma, which collected from the experimented SDrats' hearts before their death, and the measurement of quantity of histamine secretion show the following results. The quantity of normal group and control one is $25.34{\pm}4.58$ nM, $348.59{\pm}30.77$ nM respectively, and experimental one prescribed to compound 48/80 after medical treatment of Arctii Fructus is $263.56{\pm}21.34$ nM. This result indicates that separation of histamine isobviously inhibited in the experimental group in comparison with control one (p<0.05). Conclusions : Arctii Fructus does obviously inhibit the degree of degranulation of mast cell (p<0.0l) and separation of histamine in the plasma (p<0.05).

  • A membrane reactor concept, which combines the typical characteristics of chemical reaction with separation process, has been analyzed and simulated in this study. The advantages of the use of a membrane reactor include chemical equilibrium shift towards higher reactant conversion and purer product than the traditional reactors. A membrane reactor model which incorporates a catalytic reaction zone and a separation membrane is proposed. The water-gas shift reaction to produce hydrogen was chosen as a model reaction to be investigated. The membrane reactor is divided into smaller parts by number of n and each part (named cell), which contains both reaction and product separation function is modeled. One of the membrane outlet streams is connected to the next cell, which is repeated up to the last cell. The simulation results can be used for various purposes including decision of optimum operating condition and membrane reactor design.

12-Oxoeicosatetraenoic acid, a candidate signal for placenta separation, activates matrix metalloproteinase and induces apoptosis in bovine trophoblast cells

  • Objective: 12-oxo-5Z,8Z,10E,14Z-eicosatetraenoic acid (12-KETE), a metabolite of arachidonic acid, is a strong candidate signal for placenta separation following calf discharge at delivery. In the present study, the effects of 12-KETE on bovine trophoblast cells were investigated to determine its function in the placentome at delivery. Methods: Bovine trophoblast cells derived from blastocysts were used. They were cocultured with or without fibroblasts derived from bovine placentome and/or bovine uterine epithelial cells. 12-KETE was added to the culture medium. Results: Bovine trophoblast cells contained binucleate cells and strongly expressed caudal type homeobox 2 (CDX-2) genes. Addition of 12-KETE to the trophoblast cell colony without feeder cells or that on a fibroblast monolayer induced rapid exfoliation of the colony. After 12-KETE addition, trophoblast cells emitted strong fluorescence caused by the degradation of dye-quenched collagen, indicating that 12-KETE activated matrix metalloproteinase of the trophoblast cells. Exfoliated cell colonies were stained with YOPRO-1, but not propidium iodide (PI). Moreover, DNA fragmentation and Bcl-2 associated X protein (Bax) gene (apoptosis stimulator) upregulation were observed in exfoliated cells, indicating that 12- KETE induced trophoblast cell apoptosis. These results were consistent with previous in vivo observations; however, even a lower concentration of 12-KETE activated trophoblast protease. Meanwhile, fibroblasts derived from the bovine placentome converted arachidonic acid to 12-KETE. Conclusion: These observations indicate that 12-KETE may serve as a signal for placenta separation at delivery.

  • Due to their periodic helical structure, cholesteric liquid crystals (CLC) have a unique ability to selectively reflect light. CLC films reflecting a broad wavelength band were prepared by inducing a pitch gradient in CLC layer through a phase separation. The reflection bandwidth of the CLC cell was broaden as irradiation light intensity decreased and as the amount of the UV absorbing dye increased. Initial reflection bandwidth of 50 nm was broaden to 300 nm Various pitch distributions in the CLC cell was observed using SEM and ATR-IR technique was used to prove that the pitch distributions are induced through the phase separation.

  • The effects of coarsening on microstructure formation in polystyrene-cyclohexane solutions and membranes made from them were studied by scanning electron miccoscopy(SEM). Thermal analysis of the polymer solutions was carried out with a differential scanning calorimeter and the binodal curve was determined from the onset temperature of the heat of demixing peak. Using thermally induced phase separation(TIPS) and a freeze drying technique, it was demonstrated that polymer membrane microstructure can be changed significantly by controlling coarsening time and quench route. For systems undergoing phase separation by spinodal decomposition, resulting in a well interconnecmd, microporous structure with nearly uniform pore sizes, it was found that extending the phase separation time prior m freezing and solvent removal can result in a significant increase in pore or cell size which is highly dependent on both quench depth and coarsening time. Also this study has revealed the important role of polymer concentration in dictating the material continuity of the membranes.

Cluster Cell Separation Algorithm for Automated Cell Tracking (자동 세포 추적을 위한 클러스터 세포 분리 알고리즘)

  • An automated cell tracking system is used to automatically analyze and track the changes in cell behavior in time-lapse cell images acquired using a microscope with a cell culture. Clustering is the partial overlapping of neighboring cells in the process of cell change. Separating clusters into individual cells is very important for cell tracking. In this study, we proposed an algorithm for separating clusters by using ellipse fitting based on a direct least square method. We extracted the contours of clusters, divided them into line segments, and then produced their fitted ellipses using a direct least square method for each line segment. All of the fitted ellipses could be used to separate their corresponding clusters. In experiments, our algorithm separated clusters with average precisions of 91% for two overlapping cells, 84% for three overlapping cells, and about 73% for four overlapping cells.

  • In this study, we investigated the separation behavior of the anticancer agent paclitaxel and its semi-synthetic precursor 10-deacetylpaclitaxel (10-DAP) from plant cell cultures. As a result of sequential separation/purification performed by biomass extraction with solvent, liquid-liquid extraction, adsorbent treatment, hexane precipitation, and fractional precipitation, the adsorbent treatment was found to be the most effective in separating and recovering 10-DAP from paclitaxel. The optimal adsorbent type, crude extract/adsorbent ratio, and adsorbent treatment temperature were sylopute, 1:1.5 (w/w), and $20^{\circ}C$, respectively. The separation/recovery of 10-DAP from paclitaxel was 74.1% in adsorbent treatment process under optimal conditions.

  • Fuel cells have been spotlighted in the world for being highly efficient and environmentally friendly. A hydrogen which is the fuel of fuel cell can be obtained from a number of sources. Hydrogen source for operating the polymer electrolyte membrane fuel cell(PEMFC) in the current underwater environment, such as a submarine and unmanned underwater vehicles are currently from the metal hydride cylinder. However, metal hydride has many limitations for using hydrogen carrier, such as large volume, long charging time, limited storage capacity. To solve these problems, we suggest diesel reformer for hydrogen supply source. Diesel fuel has many advantages, such as high hydrogen storage density, easy to transport and also well-infra structure. However, conventional diesel reforming system for PEMFC requires a large volume and complex CO removal system for lowering the CO level to less than 10 ppm. In addition, because the preferential oxidation(PROX) reaction is the strong exothermic reaction, cooling load is required. By changing this PROX reactor to hydrogen separation membrane, the problem from PROX reactor can be solved. This is because hydrogen separation membranes are small and permeable to pure hydrogen. In this study, we conducted the pressurized diesel reforming and water-gas shift reaction experiment for the hydrogen separation membrane application. Then, the hydrogen permeation experiments were performed using a Pd alloy membrane for the reformate gas.

A Continuous Cell Separator Based on Gravity and Buoyant Forces in Fluids of Dissimilar Density (서로 다른 밀도의 유체 내 바이오 물질이 받는 중력과 부력 차를 이용한 연속적 세포 분리기)

  • We present a continuous cell separator that achieves density-dependent and size-independent cell separation based on the net force of gravity and buoyancy forces on the cells in dissimilar density fluids. Previous cell separators are, based on the size or dielectrophoretic property of the cells and, are suitable for size-dependent and density-independent cell separation. However, these properties can make it difficult to collect the same types of cells with the same density but with size variations. The present separator, however, is capable of collecting the same types of cells based on the cell density in the fluid. Regardless of cell size, the proposed chip isolates low density cells, (white blood cells, or WBCs) at the upper outlet while obtaining high-density cells (red blood cells, or RBCs) from the lower outlet based on density. Efficiency levels for separation of WBCs and RBCs were $90.9{\pm}9.1%$ and $86.4{\pm}1.99%$, respectively. The present separator therefore has the potential for use in the pretreatment of whole blood.
