생체 면역조직에서는 면역세포의 성장, 분화에 있어서 매우 중요한 역할을 수행하는 면역조직 기질세포가 존재하며, 이들은 서로 연결된 3차원적인 그물구조를 형성하면서 그 사이의 공간에 위치한 면역세포와의 상호작용을 통해 다양한 면역반응을 수행한다. 따라서 생체환경을 모사한 면역세포의 배양이 이루어지기 위해서는 면역세포들이 상호작용할 수 있는 3차원적 면역조직 기질세포 뼈대의 구축이 매우 중요한 의의를 지닌다. 특히 면역반응에서 핵심적인 기능을 수행하는 T세포의 생존, 성장 및 분화에 있어서 필수적인 역할을 하는 흉선상피세포에 대한 3차원적 배양은 T세포의 연구에 필수적으로 요구되지만, 아직 이에 관한 연구가 거의 이루어지지 않은 실정이다. 본 연구에서 흉선상피세포는 폴리도파민으로 코팅된 PCL 및 PCL/PLGA 지지체에서 비코팅군에 비해 부착 및 성장이 촉진되었다. 또한 폴리도파민으로 코팅된 지지체에서 흉선상피세포를 배양하였을 때 2차원 배양군에 비해 흉선세포형성촉진인자의 유전자 발현이 더 증가하였다. 따라서 본 연구는 면역조직 기질세포의 3차원 배양 기술의 개발에 크게 기여할 수 있을 것으로 사료된다.

Due to their multipotency, stem cells can differentiate into a variety of specialized cell types, such as chondrocytes, osteoblasts, myoblasts, and nerve cells. As an alternative to mature tissue cells, stem cells are of importance in tissue engineering and regenerative medicine. Since interactions between scaffold and cells play an important role in the tissue development in vitro, synthetic oligopeptides have been immobilized onto polymeric scaffolds to improve specific cell attachment and even to stimulate cell differentiation. In this study, chondrogenic differentiation of stem cells was evaluated using surface-modified PLLA scaffolds, i.e., either hydrophilic acrylic acid (AA)-grafted PLLA or RGD-immobilized one. Porous PLLA scaffolds were prepared using a gas foaming method, followed by plasma treatment and subsequent grafting of AA to introduce a hydrophilicity (PLLA-PAA). This was further processed to fix RGD peptide to make an RGD-immobilized scaffold (PLLA-PAA-RGD). Stem cells were seeded at $1{\times}10^{6}$ cells per scaffold and the cell-PLLA constructs were cultured for up to 4 weeks in the chondrogenic medium. Using these surface-modified scaffolds, adhesion, proliferation, and chondrogenic differentiation of stem cells were evaluated. The surface of PLLA scaffolds turned hydrophilic (water contact angle, 45 degrees) with both plasma treatment and AA grafting. The hydrophilicity of RGD-immobilized surface was not significantly altered. Cell proliferation rate on the either PLLA-PAA or PLLA-PAA-RGD surface was obviously improved, especially with the RGD-immobilized one as compared to the control PLLA one. Chondrogenic differentiation was clearly identified with Safranin O staining of GAG in the AA- or RGD-grafted PLLA substrates. This study demonstrated that modified polymer surfaces may provide better environment for chondrogenesis of stem cells.
PURPOSE. The purpose of this study was to evaluate the properties of a porous zirconia scaffold coated with bioactive materials and compare the in vitro cellular behavior of MC3T3-E1 preosteoblastic cells to titanium and zirconia disks and porous zirconia scaffolds. MATERIALS AND METHODS. Titanium and zirconia disks were prepared. A porous zirconia scaffold was fabricated with an open cell polyurethane disk foam template. The porous zirconia scaffolds were coated with ${\beta}$-TCP, HA and a compound of ${\beta}$-TCP and HA (BCP). The characteristics of the specimens were evaluated using scanning electron microscopy (SEM), energy dispersive x-ray spectrometer (EDX), and x-ray diffractometry (XRD). The dissolution tests were analyzed by an inductively coupled plasma spectrometer (ICP). The osteogenic effect of MC3T3-E1 cells was assessed via cell counting and reverse transcriptase-polymerase chain reaction (RT-PCR). RESULTS. The EDX profiles showed the substrate of zirconia, which was surrounded by the Ca-P layer. In the dissolution test, dissolved $Ca^{2+}$ ions were observed in the following decreasing order; ${\beta}$-TCP > BCP > HA (P<.05). In the cellular experiments, the cell proliferation on titanium disks appeared significantly lower in comparison to the other groups after 5 days (P<.05). The zirconia scaffolds had greater values than the zirconia disks (P<.05). The mRNA level of osteocalcin was highest on the non-coated zirconia scaffolds after 7 days. CONCLUSION. Zirconia had greater osteoblast cell activity than titanium. The interconnecting pores of the zirconia scaffolds showed enhanced proliferation and cell differentiation. The activity of osteoblast was more affected by microstructure than by coating materials.
Understanding chondrocyte behavior inside complex, three-dimensional environments with controlled patterning of geometrical factors would provide significant insights into the basic biology of tissue regenerations. One of the fundamental limitations in studying such behavior has been the inability to fabricate controlled 3D structures. To overcome this problem, we have developed a three-dimensional microfabrication system. This system allows fabrication of predesigned internal architectures and pore size by stacking up the photopolymerized materials. Photopolymer SL5180 was used as the material for 3D scaffolds. The results demonstrate that controllable and reproducible inner-architecture can be fabricated. Chondrocytes harvested from human nasal septum were cultured in two kinds of 3D scaffolds to observe cell adhesion behavior. Such 3D scaffolds might provide effective key factors to study cell behavior in complex environments and could eventually lead to optimum design of scaffolds in various tissue regenerations such as cartilage, bone, etc. in a near future.
Electrospinning of Polyvinylalcohol (PVA), Gelatin (GE), and PVA/GE blend solutions in acetic acid were investigated to fabricate biodegradable for tissue engineering. The morphology of the electrospun nanofibers was investigated with a field emission scanning electron microscope. The fibers have average diameters in the range 50-150 nm. The miscibility of PVA/GE blend fibers was examined by differential scanning calorimetry.The PVA and GE were immiscible in the as-spun nanofibrous structure. X-ray diffraction (XRD) determined the crystallinity of the membrane and tensile strength for evaluation physical properties. An in vitro study of PVA/GE blend nanofibers was conducted. To assay the cytocompatibility and cell behavior on the PVA/GE blend nanofibrous scaffolds, cell attachment and spreading of fibroblasts seeded on the scaffolds were studied. Our results indicate that thePVA/GE blend nanofibrous matrix, particularly the one that contained 20% PVA and 80% GE could be a good candidate for tissue engineering scaffolds, because it has an excellent cell attachment and spreading for fibroblast cell.
The achievement of tissue engineering can be highly depending on the capability to generate complicated, cell seeded three dimensional (3D) micro/nano-structures. So, various fabrication techniques that can be used to precisely design the architecture and topography of scaffolding materials will signify a key aspect of multi-functional tissue engineering. Previous methods for obtaining scaffolds based on top-down are often not satisfactory to produce complex micro/nano-structures due to the lack of control on scaffold architecture, porosity, and cellular interactions. However, a bioprinting method can be used to design sophisticated 3D tissue scaffolds that can be engineered to mimic the tissue architecture using computer aided approach. Also, in recent, the method has been modified and optimized to fabricate scaffolds using various natural biopolymers (collagen, alginate, and chitosan etc.). Variation of the topological structure and polymer concentration allowed tailoring the physical and biological properties of the scaffolds. In this presentation, the 3D bioprinting supplemented with a newly designed plasma treatment for attaining highly bioactive and functional scaffolds for tissue engineering applications will be introduced. Moreover, various in vivo and in vitro results will show that the fabricated scaffolds can carry out their structural and biological functionality.

Mesenchymal stem cells(MSCs) have been though to be multipotent cells that can replicate that have the potential to differentiate into lineages of mesenchymal tissue including the bone, cartilage, fat, tendon, muscle, and marrow stroma. Especially, scaffolds to support cell-based tissue engineering are critical determinants of clinical efforts to regenerate and repair the body. Selection of a matrix carrier imvolves consideration of the matrix's role as a scaffold for physical support and host tissue integration as well as its ability to support of synergize the osteoinductive program of the implanted mesenchymal stem cell. The aim of this study is to evaluate the effect of autobone and Bio-$Oss^{(R)}$ to adherent mesenchymal stem cells as scaffolds on sinus augmentation with fibrin glue mixture in a rabbit model. 16 New Zealand White rabbits were divided randomly into 4 groups based on their time of sacrifice(1, 2, 4 and 8 weeks). First, mesenchymal stem cells were isolated from iliac crest marrow of rabbits and expanded in vitro. Cell culture was performed in accordance with the technique described by Tsutsumi et al. In the present study, the animals were sacrificed at 1, 2, 4 and 8 weeks after transplantation, and the bone formation ability of each sides was evaluated clinically, radiologically, histologically and histomorphologically. According to the histological observations, autobone scaffolds group showed integrated graft bone with host bone from sinus wall. At 2 and 4 weeks, it showed active newly formed bone and neovascularization. At 8 weeks, lamellae bone was observed in sinus graft material area. Radiologically, autobone with stem cell showed more radiopaque than Bio-$Oss^{(R)}$ scaffolds group. there were significant differences in bone volume between 4 and 8 weeks(p<0.05).
Aim of the study: Scaffolds are crucial to tissue engineering/regeneration. Biodegradable polymer/ceramic composite scaffolds can overcome the limitations of conventional ceramic bone substitutes such as brittleness and difficulty in shaping. In this study, poly(L-lactide)/hydroxyapatite(PLLA/HA) composite scaffolds were fabricated for in vivo bone tissue engineering. Material & methods: In this study, PLLA/HA composite microspheres were prepared by double emulsion-solvent evaporation method, and were evaluated in vivo bone tissue engineering. Bone marrow mesenchymal stem cell from rat iliac crest was differentiated to osteoblast by adding osteogenic medium, and was mixed with PLLA/HA composite scaffold in fibrin gel and was injected immediately into rat cranial bone critical size defect(CSD:8mm in diameter). At 1. 2, 4, 8 weeks after implantation, histological analysis by H-E staining, histomorphometric analysis and radiolographic analysis were done. Results: BMP-2 loaded PLLA/HA composite scaffolds in fibrin gel delivered with osteoblasts differentiated from bone marrow mesenchymal stem cells showed rapid and much more bone regeneration in rat cranial bone defects than control group. Conclusion: This results suggest the feasibility and usefulness of this type of scaffold in bone tissue engineering.
The object of this study is to investigate the effects of intermittent cyclic stretching on the smooth muscle cells (SMCs) seeded onto aligned multi-layered fibrous scaffold. To make multi-layered fibrous scaffold, polyurethane (PU) and poly(ethylene oxide) (PEO) were electrospun alternatively, then were immersed into distilled water to extract PEO. Various types of scaffolds were fabricated depending on fiber directions, i.e., aligned or randomly oriented. The direction of stretching was either parallel or vertical to the fiber direction for the aligned scaffolds. The stretching was also applied to the randomly aligned scaffolds. The duration of stretching was 2 min with 15 min resting period. During the stretching, the maximum and minimum strain was adjusted to be 10 and 7%, respectively with the frequency of 1 Hz. The bioactivities of cells on the scaffolds were assessed by quantifying DNA, collagen, and glycosaminoglycan (GAG) levels. And the cell morphology was observed by staining F-actin. SMCs under parallel stretching to the fiber direction responded more positively than those in other conditions. From the results, we could explain the morphological effect of a substrate on cellular activities. In addition the synergistic effects of substrate and mechanical stimuli effects were confirmed.
Poly(lactide-co-glycolic acid)(PLGA)는 좋은 기계적 성질과 생분해성으로 약물전달시스템 또는 조직공학적으로 널리 이용되고 있으나 낮은 세포 부착률을 가지고 있어 피브린을 첨가하여 이를 보완하고자 하였다. 본 연구에서 사용된 지지체는 트롬빈과 피브리노겐, 그리고 세포을 혼합시킨 후 PLGA 지지체 위에 도포시켜 제조하였다. 세포의 부착 및 증식률을 측정하고자 PLGA/피브린 지지체에 늑연골 세포를 파종 후 1, 3일 및 7일 후 SEM과 MTT 분석을 통하여 측정하였으며, 세포외기질 형성에 미치는 피브린의 영향을 확인하고자 세포를 파종 후 누드마우스에 이식하여 GAG 및 콜라겐 합성의 효과를 확인하였다. 따라서 본 연구에서는 피브린이 혼합된 PLGA 지지체가 생체 내 외 환경에서 세포의 부착 및 증식에 미치는 영향을 확인하고자 연구를 진행하였다. 그 결과, PLGA/피브린 지지체가 기존의 PLGA 지지체와 비교하여 탁월한 세포 성장률을 나타내는 것으로 확인하였다.
