Clinical Application of Serum Tumor Associated Material (TAM) from Non-small Cell Lung Cancer Patients

  • Objective: To explore the associations of serum tumor associated material (TAM) with other common tumor markers like carcinoembryonic antigen (CEA), carbohydrate antigen 125 (CA125), carbohydrate antigen19-9 (CA19-9) and its clinical application in non-small cell lung cancer (NSCLC) patients. Methods: A total of 87 patients were enrolled into this study, all with histologically or cytologically confirmed NSCLC. With the method of chemical colorimetry, the level of TAM was determined and compared, while chemiluminescence was used to measure the levels of common tumor markers. Results: The level of TAM decreased after chemotherapy compared with before chemotherapy when CT or MRI scans showed disease control. Furthermore, it increased when disease progessed and there was no statistically significant difference in monitoring of TAM and common tumor markers (P>0.05). Conclusions: Detecting TAM in NSCLC patients has a higher sensitivity and specificity, so it can be used as an indicator for clinical monitoring of lung cancer chemotherapy.

  • A numerical model (SK-EST) for estimating hydraulic conductivity using monitoring data of underground LPG storage cavern was developed. The model calculates hydraulic conductivity from matrix equation which is established from the distribution of hydraulic potential. To verify the applicability of this model, an inverse analysis was performed using the monitoring data of pressure cell of an operating underground LPG storage cavern. And also using the water pressure parker test data which were obtained to look over the operation capability of pressure cell, conductivity variation with depth was estimated using the developed numerical model (SK-EST) and was compared with in situ results.

  • Gambierdiscus toxicus, Adachi et. Fukuyo, is a benthic ciguatoxin-producing armored dinoflagellate, often attached to macroalgae. This organism is the primary causative agent of ciguatera fish poisoning which occurs in tropical and subtropical regions. However, regardless of the fact that the population of Gambierdiscus spp. has expanded to such temperate areas from sub-trophic and trophic areas, monitoring of G. toxicus has been lacking in the Korean coastal waters of temperate areas. This study was performed at the uninhabited Baekdo Islands off the southern coast of Korea and at Seopsom Island in the vicinity of Seogwipo, Jeju Province during April and May, 2011. Cell densities of Gambierdiscus spp. on macroalgae at Baekdo and Jeju Island ranged from zero to 56.4 cells $g^{-1}$. Maximum density was recorded on the brown alga Cladophora japonica at St. 3 of Jeju Island. In particular, the cell densities of Gambierdiscus spp. were influenced by the substrate characteristics of macroalgae. In the future, the continuous monitoring of toxic benthic dinoflagellate is necessary to predict and prevent ciguatera poisoning in Korean coastal waters.

  • According to the "hydrogen economy roadmap" announced recently by the government, fuel cell electric vehicle diffusion and hydrogen refueling station construction are actively being carried out to prepare for the hydrogen economy era. The station will be expanded by introducing various charging station models such as hydrogen complex charging station, package, and mobile. Accordingly, the study on the safety demonstration of the charging station and related regulations should be compromised. The purpose of this study is to collect monitoring data during charging according to the distinct four seasons in Korea, and to use it as safety demonstration data by analyzing the charging status, charging rate and charging time during charging.

Expression of $HpaG_{Xooc}$ Protein in Bacillus subtilis and its Biological Functions

  • $HpaG_{Xooc}$, from rice pathogenic bacterium Xanthomonas oryzae pv. oryzicola, is a member of the harpin group of proteins, eliciting hypersensitive cell death in non-host plants, inducing disease and insect resistance in plants, and enhancing plant growth. To express and secret the $HpaG_{Xooc}$ protein in Bacillus subtilis, we constructed a recombinant expression vector pM43HF with stronger promoter P43 and signal peptide element nprB. The SDS-PAGE and Western blot analysis demonstrated the expression of the protein $HpaG_{Xooc}$ in B. subtilis. The ELISA analysis determined the optimum condition for $HpaG_{Xooc}$ expression in B. subtilis WBHF. The biological function analysis indicated that the protein $HpaG_{Xooc}$ from B. subtilis WBHF elicits hypersensitive response(HR) and enhances the growth of tobacco. The results of RT-PCR analysis revealed that $HpaG_{Xooc}$ induces expression of the pathogenesis-related genes PR-1a and PR-1b in plant defense response.

  • In this paper, we propose a new moving mobile base station (MMBS) scheme for very low power and micro-size RMIMS (radio-interfaced micro information monitoring system) terminals. RMIMS terminals can be used in various application service areas such as pollution monitoring, environment surveillance, traffic monitoring, emergency monitoring (e.g., building, bridge, railroad breakdown), security monitoring (e.g., theft, alarm) and military application. For these applications based on wireless transmission technologies, sensor type RMIMS terminals must satisfy low cost and low power design (e.g., solar power, life limited battery) requirement. In RMIMS terminal design, this low power requirement limits transmission range of uplink or reverse link and means small cell size. Also these applications using RMIMS terminals may have a little bit non real-time traffic characteristic and low scattering density in service area.

THE EFFECTS OF CELL WALL PROTEINS OF STREPTOCOCCUS SPP. ON DNA SYNTHESIS OF L929 CELLS AND THEIR SDS-PAGE PATTERNS (연쇄 구균의 세포벽 단백질이 L929 세포의 DNA합성에 미치는 영향 및 SDS-PAGE 양상에 관한 연구)

  • Bacteria have been regarded as a one of major etiologic factors in root canal infections. In endodontic treatment the effective removal of pathogenic microorganisms in the root canal is the key to successful outcome. Bacterial cell wall components may play an important role in the development of pulpal and periapical disease. The purpose of this study was to evaluate the effect of sonic extracts of Streptococcus spp. on cultured L929 cells and to characterize cell wall protein profiles of Streptococcus spp. Streptococcus spp. were isolated from infected root canals and identified with Vitek Systems(Biomeriux, USA). Five streptococci, namely S. sanguis, S. mitis, S uberis, S. mutans (ATCC 10449) and S. faecalis (ATCC 19433) weere enriched in brain heart infusion broth. Cell pellets were sonicated and cell wall extracts were dialyzed and membrane filtered. Prepared cell wall proteins were applied to cultured L929 cell. The cell reaction were evaluated by monitoring DNA synthesis, cell numbers and the change of cell morphology. The total cell wall protein profiles of microorganisms were characterized by sodium dodecyl sulfate polyacrylamide-gel eledruphoresis(SDS-PAGE). DNA synthesis of L929 cells were reduced by the increasing concentration of sonic extracts. DNA synthesis was significantly suppressed in more than $50{\mu}g$/ml of sonic extract conentration in five streptococci. S. nutans (ATCC 10449) showed stronger suppression on DNA synthesis than remaining four streptococci, which had the similar effect on DNA synthesis. Analysis of DNA synthesis measured by [$^3H$]-thymidine uptake was more sensitvie method than cell counting. Sonic extracts affected the microscopic findings of L929 cells. The protein profiles indicated that all five strains shared two major proteins with molecular masses of 70.8 and 57.5 kD respectively. S. uberis and S. mutans shared common minor proteins of which molecular weights were 147.9 and 112.2 kD respectively. However some minor proteins were unique for S. mitis, S. uberis and S. faecalis.

A new role for the ginsenoside RG3 in antiaging via mitochondria function in ultraviolet-irradiated human dermal fibroblasts

  • Background: The efficacy of ginseng, the representative product of Korea, and its chemical effects have been well investigated. The ginsenoside RG3 has been reported to exhibit apoptotic, anticancer, and antidepressant-like effects. Methods: In this report, the putative effect of RG3 on several cellular function including cell survival, differentiation, development and aging process were evaluated by monitoring each specific marker. Also, mitochondrial morphology and function were investigated in ultraviolet (UV)-irradiated normal human dermal fibroblast cells. Results: RG3 treatment increased the expression of extracellular matrix proteins, growth-associated immediate-early genes, and cell proliferation genes in UV-irradiated normal human dermal fibroblast cells. And, RG3 also resulted in enhanced expression of antioxidant proteins such as nuclear factor erythroid 2-related factor-2 and heme oxygenase-1. In addition, RG3 affects the morphology of UV-induced mitochondria and plays a role in protecting mitochondrial dysfunction. Conclusioin: RG3 restores mitochondrial adenosine triphosphate (ATP) and membrane potential via its antioxidant effects in skin cells damaged by UV irradiation, leading to an increase in proteins linked with the extracellular matrix, cell proliferation, and antioxidant activity.

Production of Glutathione by yeast and Process Monitoring (효모에 의한 글루타치온의 생산과 공정 모니터링)

  • In this work the production of glutathione (GSH) by yeast Saccharomyces cerevisiae and the monitoring of the process were studied. In shaking culture the production of GSH was high at initial pH value of 4 and at temperature of 30$^{\circ}C$. But when L-cysteine was added to the culture medium at the beginning of the cultivation, the productivity of GSH was low. In case 0,5% (v/v) of L-cysteine, glycine and glutamic acid were introduced to the culture medium in the exponential cell growth phase, high concentration of GSH (about 90 mg/L) was produced in the bioreactor. A fed-batch operation with stepwise glucose feeding strategy allowed to produce 102 mg/L of GSH. The cultivation processes were on-line monitored by a 2-dimensional fluorescence sensor. A few off-line data such as cell growth, cystein concentration, phosphate concentration and GSH productivity could be well correlated to the fluorescence intensity of some combinations of excitation and emission wavelengths.

  • FCEV(Fuel Cell Electric Vehicle)는 연료전지스택의 각 셀에서 반응하는 화학에너지를 전기에너지로 변환하여 차량을 구동하는 시스템이다. 이러한 연료전지 셀이 정상적인 발전이 되지 않을 경우 비정상적인 전압이 발전되고 이것을 방치한다면 연료전지 스택의 영구적인 고장을 야기할 수 있다. 이를 방지하기 위해 SVM(Stack Voltage monitor) 제어기는 각 셀의 전압을 측정하고 그 정보를 상위 제어기인 FCU(Fuel cell Control Unit)에 전달한다. 이에 FCU는 연료전지스택의 고장을 운전자에게 알리고 연료전지스택의 발전을 멈추게 한다. 기존에 SVM 제어기는 각 셀마다 분압저항을 통하여 측정하며 이 전압의 차를 이용하여 셀 전압을 계산하는 방식이었다. 이는 상위 셀로 갈수록 오차가 커지는 문제가 있고 다수의 CPU 및 DC/DC 컨버터가 적용이 필요하여 복잡한 구성과 가격이 높은 문제가 있었다. 이러한 문제점을 해결하기 위하여 cell monitoring IC를 적용하였고 좀 더 정밀한 측정과 간단한 인터페이스를 구성할 수 있었다. 본 연구에서는 기존 SVM 제어기보다 안정되고 정밀한 SVM 제어기의 개발에 대해 기술하였다.
