Vinyl Addition Copolymerization of Norbornene/5-Vinyl-2-norbornene with Cationic $\eta^{3_}$-Allyl Palladium Catalyst and Their Post-Reaction (양이온성 $\eta^{3_}$알릴 팔라듐 촉매를 사용한 노보넨/5-비닐-2-노보넨의 비닐 부가 공중합과 이를 이용한 고분자 후반응)

  • Vinyl addition copolymerizations of norbornene (NB) and 5-vinyl-2-norbomene (VNB) were carried out using a cationic η$^3$-allyl palladium catalyst in the various mole ratio of comonomers. The copolymers could be obtained in good yield (65∼85%) with high weight-average molecular weights (M$_{w}$ > 760,000). Depending on increasing VNB contents, the molecular weight and yield of the copolymers decreased. FT-IR analysis confirmed that actual contents of VNB in polymer were proportional to the feeding content of VNB. From $^1$H-NMR spectroscopy, we found that both exo and endo VNB isomer were copolymerized with NB. Thermal stabilities of NB-VNB copolymers were independent on the VNB content and their initial decomposition temperatures were about 300 C. The NB-VNB copolymers were followed by epoxidation by using m-CPBA and hydroxylation by 9-BBN, respectively, and these post-polymers were characterized by FT-IR spectroscopy and $^1$H-NMR analysis..

  • Polydiacetylene (PDA) is made by photopolymerization of self-assembled diacetylene monomers. If diacetylene monomers are arranged systematically and close enough with distance of atoms, 1,4-addition polymerization will occur by the irradiation of 254 nm ultraviolet rays and then PDA will have alternated ene-yne polymer chains at the main structure. Aqueous solutions of diffused PDA is tinged with blue which shows ${\lambda}_{max}$ 640 nm. Visible color changes from blue to red occurs in response to a variety of environmental perturbations, such as temperature, pH, and ligand-receptor interactions. In this study, we synthesized cationic peptides - PCDA(10,12-pentacosadyinoic acid) liposome using a solid phase peptide synthesis (SPPS) method and prepared liposome solutions at various molar ratios using MPEG-PCDA. When mammalian cells were treated with the liposomes, high transfection efficiency and low toxicity were observed.

The Effects of Supplements on the Plasmid Delivery and Expression in the Transfection Using Cationic Liposomes (양이온 리포좀을 이용한 유전자 전달 및 발현서 첨가제의 효과)

  • Cellular transfections with cationic liposomes are widely empolyed for gene and oligonucleotide transfer in vitro because of their safety and ease of use. However, they still suffer from the low transfection efficiency comparing with viral vectors. Substantial effort shave been focused on increasing transfection efficiency by supplementing the liposome/DNA complexes(lipoplex) with various components. In this work, we tired three kinds of supplements, Poly-L-lysine(PLL), transferrin and a mixture of anionic lipids(PS/PE/PC), to study their effects on gene transfer yield and gene expression efficiency. PLL, a polycationic polymer, enhanced gene transfer yield by 3 times but the gene expression efficiency was increased only by 1.5 times. this result implies that PLL can enhance the transfection efficiency mainly by increasing the rate of outermembrane transport of lipoplex into the cells. On the other hand, transferrin which can facilitate the gene transfer via ligand-receptor interaction gave not only increased gene transfer yield but also enhanced gen expression efficiency by 2.8 times. Transferrin seems to contribute to the escape of plasmid from endosomes through ligand-receptor recycle mechanism. When the cells were treated with a mixture of anionic lipids for 3 hours before the transfection, gene transfer yield was slightly decreased but the gene expression efficiency was enhanced by 1.9 times. This is presumably due to the accelerated liposome-plasmid dissociation by the anionic lipids, and the increased delivery of plasmid to the nucleus. According to these results, it is clear that the supplementation to ameliorate transfection efficiency with cationic liposomes should be contrived in the direction of increasing delivery of plasmid.

Characteristic as a Gene Delivery System of Water Soluble Chitosan Conjugated with Cationic Peptide (양이온 펩타이드가 컨쥬게이트된 수용성 키토산의 유전자 전달체로서의 특성)

  • Recently gene delivery has been designed newly using bioactive biomaterial and applied in the various field by many researchers. In this study, we proposed a new gene delivery system which has the capability of targeting effect in the specific tissue and remarkably enhanced transfection efficiency. We investigated $^1H-NMR$ spectroscopy, particle size analyzer and gel retardation to confirm the correct preparation of gene delivery. Also, we identified the hemo-compatibility of gene delivery by hemolysis assay, non-cytotoxicity by MTT test and transfection efficiency. The uptake mechanism of the gene carrier was confirmed using inhibitor agent such as sodium azide, indomethacin, quercetin, colchicine, and chloropromazine. As a results, it was identified that gene carrier prepared by in this study entered in the cell by the microtubule-dependent, energy-dependent and clathrin-mediated endocytosis pathway.

A Comparative Study on the Conditioning and Dewaterability of Digested Sludge from Intermittent Aerobic Digestion (간헐포기 소화슬러지의 개량과 탈수 특성에 관한 비교 연구)

  • Synthetic organic polyelectrolytes can be used to condition sludges to enhance their dewaterability. When conditioning biological sludges, the charge on the polymer has a significant impact on the effectiveness of the polymer as a conditioner. The objectives of this investigation were to determine the most effective type and dosage of polymer for conditioning digested sludge prior to dewatering, and to investigate the relationship between dewaterability and particle size. 3 types of digested sludge were used under the different digestion processes like anaerobic digestion, aerobic digestion and intermittent aerobic digestion. CST(capillary suction time), TTF(time-to-filter) and SRF(specific resistance to filtration) were tested as a dewaterability index and the number of particle distribution was analyzed using particle size analyzer. The results indicate that cationic polymer appears to be required for effective conditioning of these 3 types digested sludge and the optimal polymer dosage is about 0.6% of SS. CST and TTF are well correlated with mean particle diameter of which the dimension order is 1.7.

  • The present investigation has focused up on the study of the adsorption of three different molecular mass fractions of a polyDiMethylDiAllylAmmoniumChloride (DMDAAC) (8750(LM\ulcorner), 48000(MM\ulcorner) and 1200000(HM\ulcorner)) on bleached chemical fibres. Both kinetics of adsorption and equilibrium adsorption measurements have been conducted and each adsorption has been measured by polyelectrolyte titration. The results show that the LM\ulcorner polymer can reach all of the charges in the fibre wall whereas the MM\ulcorner and HM\ulcorner can only reach the external surfaces of the fibres. It is also shown that the kinetics of adsorption of the LMw polymer is not at all affected by the presence of a saturated layer of HMw polymer on the surface of the fibres. Finally, the results from the investigation show that it is possible to have full coverage of the external surface of the fibres by a high molecular mass polymer and a full coverage of the internal surface of the fibres with a low molecular mass polymer. This is true if the high molecular masspolymer is added first followed by addition of the low molecular masspolymer.

Interaction between Poly(L-lysine) and Poly(N-isopropyl acrylamide-co-acrylic acid) in Aqueous Solution

  • A series of pH/temperature sensitive polymers were synthesized by copolymerizing N-isopro-pyl acrylamide(NIPAAm) and acrylic acid(AAc) . The influence of polyelectrolyte between poly(allyl amine) (PAA) and poly(L-lysine)(PLL) on the lower critical solution temperature(LCST) of pH/temperature sensitive polymer was compared in the range of pH 2∼12. The LCST of PNIPAAm/water in aqueous poly(NIPAAm-co-AAc) solution was determined by cloud point measurements. A polyelectrolyte complex was prepared by mixing poly(NIPAAm-co-AAc) with poly(allyl amine) (PAA) or poly(L-lysine) (PLL) solutions as anionic and cationic polyelectrolytes, respectively. The effect of polyelectrolyte complex formation on the conformation of PLL was studied as a function of temperature by means of circular dichroism(CD). The cloud points of PNIPAAm in the aqueous copolymers solutions were stongly affected by pH, the presence of polyelectrolyte solute, AAc content, and charge density. The polyelectrolyte complex was formed at neutral condition. The influence of more hydrophobic PLL as a polyelectrolyte on the cloud point of PNIPAAm in the aqueous copolymer solution was stronger than that of poly(allyl amine)(PAA). Although polymer-polymer complex was formed between poly(NIPAAm-co-AAc) and PLL, the conformational change of PLL did not occur due to steric hinderance of bulky N-isopropyl groups of PNIPAAm.

Preparation and Properties of Novel Biodegradable Hydrogel based on Cationic Polyaspartamide Derivative

  • Novel copolymers consisting of poly(2-hydroxyethyl aspartamide-co-N,N'-dimethyl-1,3-propane aspartamide) (PHEA-DPA) were prepared from polysuccinimide (PSI), which is the thermal polycondensation product of aspartic acid, via a ring-opening reaction with N,N'-dimethyl-1,3-propane diamine (DPA) and ethanolamine. The prepared water-soluble copolymer was then crosslinked by reacting it with hexamethylene diisocyanate to provide the corresponding gel. The swelling behavior and morphology of the crosslinked hydrogels were investigated. The degree of swelling decreased with increasing crosslinking reagent due to the higher crosslinking density. It was also confirmed that the swelling property is affected by pH. At low pH (< pH 4), swelling is increased due to the ionization of DPA with a tertiary amine moiety. In addition, a reversible swelling and de-swelling behavior was demonstrated by adjusting the pH of the solution. The prepared hydrogels showed a well-interconnected microporous structure with regular 5-20 $\mu$m sized pores.

Highly Fluorescing Solid DNA-Cationic Polyelectrolyte Complexes Prepared from a Natural DNA and a Poly(fluorenevinylene-alt-phenylene) Bearing Quaternary Ammonium Pendants

  • A fluorescing, copolymer(Q)-bearing, quaternary ammonium pendant was mixed with excess natural salmon sperm DNA with a molecular weight of $1.3{\times}10^6$(2,000 base pairs) to afford highly fluorescing, complex mixtures. The fluorescence life-time of the polymer Q was greatly increased when mixed with DNA: for the mixture of Q:DNA=1:750 the fast and slow decay lifetimes increased from ca. 10 to 100 ps and from 20 ps to ca. 1 ns, respectively. The enhanced fluorescence of the mixtures was ascribed to efficient compartmentalization and reduced conformational relaxation of the polymer Q by complexation with excess DNA.

Synthesis of the Polysaccharide, (1 $\longrightarrow$ 5)-$\alpha$-D-Ribofuranan and Its Catalytic Activities for the Hydrolysis of Phosphates and the Cleavage of Nucleic Acids

  • The polysaccharide, (1\longrightarrow5)-$\alpha$-D-ribofuranan, was synthesized by a cationic ring-opening polymerization of 1,4-anhydro-2,3-di-O-benzyl-$\alpha$-D-ribopyranose with the aid of boron trifluoride etherate and subsequent debenzylation. This polysaccharide catalyzed the hydrolysis of ethyl p-nitrophenyl phosphate, uridylyl(3'\longrightarrow5')uridine ammonium salt, and 4-tert-butylcatechol cyclic phosphate N-methyl pyridinium. The polymer also catalyzed the cleavage of nucleic acids (DNA and RNA). The hydrolysis of ethyl p-nitrophenyl phosphate in the presence of the polymer was accelerated by 1.5 ${\times}$ 10$^3$ times relative to the uncatalyzed reaction. The catalytic activity was attributable to the vic-cis-diols of the riboses being located inside the active center that is formed by polymer chain folding; these diols form hydrogen bonds with two phosphoryl oxygen atoms of the phosphates so as to activate the phosphorus atoms to be attacked by nucleophile ($H_2O$).
