Fumonisin B1-Induced Toxicity Was Not Exacerbated in Glutathione Peroxidase-1/Catalase Double Knock Out Mice

  • Fumonisin B1 (FB1) structurally resembles sphingolipids and interferes with their metabolism leading to sphingolipid dysregulation. We questioned if FB1 could exacerbate liver or kidney toxicities in glutathione peroxidase 1 (Gpx1) and catalase (Cat) knockout mice. While higher serum levels of thiobarbituric acid reactive substances (TBARS) and sphinganine (Sa) were measured in Gpx1/Cat knockout mice (Gpx1/Cat KO) than wild type mice after 5 days of FB1 treatment, serum levels of alanine aminotransferase (ALT), sphingosine-1 phosphate (So-1-P), and sphinganine-1 phosphate (Sa-1-P) were found to be relatively low. Although Sa was highly elevated in Gpx1/Cat KO mice and wild mice, lower levels of So and Sa were found in both the kidney and liver tissues of Gpx/Cat KO mice than wild type mice after FB1 treatment. Paradoxically, FB1-induced cellular apoptosis and necrosis were hastened under oxidative stress in Gpx1/Cat KO mice.

Simultaneous and Sequential Co-Immobilization of Glucose Oxidase and Catalase onto Florisil

  • The co-immobilization of Aspergillus niger glucose oxidase (GOD) with bovine liver catalase (CAT) onto florisil (magnesium silicate-based porous carrier) was investigated to improve the catalytic efficiency of GOD against $H_2O2$ inactivation. The effect of the amount of bound CAT on the GOD activity was also studied for 12 different initial combinations of GOD and CAT, using simultaneous and sequential coupling. The sequentially co-immobilized GOD-CAT showed a higher efficiency than the simultaneously co-immobilized GOD-CAT in terms of the GOD activity and economic costs. The highest activity was shown by the sequentially co-immobilized GOD-CAT when the initial amounts of GOD and CAT were 10 mg and 5 mg per gram of carrier. The optimum pH, buffer concentration, and temperature for GOD activity for the same co-immobilized GOD-CAT sample were then determined as pH 6.5, 50 mM, and $30^{\circ}C$, respectively. When compared with the individually immobilized GOD, the catalytic activity of the co-immobilized GOD-CAT was 70% higher, plus the reusability was more than two-fold. The storage stability of the co-immobilized GOD-CAT was also found to be higher than that of the free form at both $5^{\circ}C\;and\;25^{\circ}C$. The increased GOD activity and reusability resulting from the co-immobilization process may have been due to CAT protecting GOD from inactivation by $H_2O2$ and supplying additional $O_2$ to the reaction system.

The Reduction of Hydrogen Peroxide in Viable Boar Sperm Cryopreserved in the Presence of Catalase (Catalase 첨가에 따른 돼지 정액 동결 및 융해 후 생존 정자에서 Hydrogen Peroxide의 감소)

  • Semen cryopreservation induces the formation of reactive oxygen species (ROS), and the ROS cause sperm damage. We aimed to investigate the effects of the antioxidative enzyme catalase (CAT) on sperm quality and ROS during cryopreservation. Sperm rich fractions collected from five Duroc boars were cryopreserved in freezing extender with (200 or 400 U/mL) or without CAT (control). After thawing, sperm motility, viability, normal morphology, plasma membrane integrity, mitochondrial function and intracellular ROS were evaluated. CAT significantly improved total sperm motility at a concentration of 400 U/mL (P < 0.05), but didn't improve progressive sperm motility, viability, morphological defects, plasma membrane integrity and mitochondrial function in frozen-thawed boar sperm. In evaluation of ROS, CAT had no effect on reduction in ${\cdot}O_2$, but scavenged $H_2O_2$ in viable frozen-thawed boar sperm at concentrations of 200 and 400 U/mL (P < 0.05). In conclusion, CAT was not enough to improve quality of frozen-thawed sperm, but can reduce $H_2O_2$ generation in viable boar sperm during cryopreservation.

Liquid Crystal Droplet Patterns to Monitor Catalase Activity at Femtomolar Levels

  • Catalase (CAT) decomposes hydrogen peroxide that is toxic to the body. In this study, simple and sensitive detector has been developed for observing catalase activity using liquid crystal droplet system. Microscale LC droplet patterns are formed by spreading aldehyde-doped nematic liquid crystal on pre-treated glass slides. When hydrogen peroxide is added, aldehyde is oxidized and amphiphiles are formed. Dodecanoates cause the pattern to transit from bright to dark as they self-assemble to form a carboxyalte monolayer at the interface. When a drop of pre-incubated CAT and hydrogen peroxide mixture is placed onto the pattern, bright fan-shape is observed. This planar optical appearance indicates that catalase has decomposed hydrogen peroxide. Compared to the detectors that have been previously developed, this system is more sensitive with detection limit of 1fM. This research suggests further studies to be on LC droplet patterning to develop highly sensitive and methodologically simple sensors for various chemicals.

  • Antioxidative activites and anti skin aging effects of radish(Raphanus sativus L. var. sativus) extracts were studied. The DPPH(1,1-Diphenyl-2-picrylhydrazal) inhibitory effects and CAT(Catalase) activities of hydrophilic radish extracts(DPPH 81.0%, CAT 175units/mg) were significantly higher than lipophilic radish extracts(DPPH 50.0%, CAT 45unitg/mg). But, SOD(superoxide dismutase) activities of hydrophilic radish extracts(SOD 81.3%) were lower than lipophilic radish extracts(SOD 92.3%) In the Bovine Achilles Tendon collagen treated with hydrophilic radish extracts and lipophilic radish extracts, the pyridinoline concentration of collagen were decreased, respectively.

The Association between Korean Schizophrenics and Catalase Gene Polymorphism (한국인 정신분열병 환자와 Catalase 유전자 다형성의 연합)

  • Objective:There is increasing evidence that free radical-mediated CNS neuronal dysfunction is involved in the pathophysiology of schizophrenia. This study was performed to examine the relationship between antioxidant defense system and schizophrenia by analyzing polymorphism of catalase gene, an antioxidant enzyme. Method:Genotype and allele frequencies in the promoter region in the catalase gene using restriction fragment length polymorphism were studied, comparing 155 Korean controls with 167 Korean schizophrenics. Results:No difference was found between the schizophrenics and the controls in genotype and allele frequencies of HinfI polymorphism in the catalase gene. Significant difference was found between the female schizophrenics and the female controls in the genotype distribution(${\chi}^2$=11.096, df=2, p=0.004). Conclusions:The results do not support an association between polymorphism of catalase gene and schizophrenia. However, this study suggests that HinfI polymorphism in the catalase gene could be associated with female schizophrenics.

  • This study was conducted to investigate the effects of nickel chloride ($NiCl_2$) exposure on oxidative stress of the abalone Haliotis discus hannai. Experimental groups were composed of one control condition and five nickel chloride exposure conditions (5.9, 8.8, 13.3, 20.0, 30.0 mg/L). Superoxide dismutase (SOD) activity was increased in the foot and hepatopancreas, but decreased in the gill. Catalase (CAT) activity was increased in all exposure groups except 20.0 mg/L and 30.0 mg/L. In the gill, CAT activity was similar to the control group for all exposure groups. In the hepatopancreas, CAT activity was increased compared with the control group. However SOD and CAT activity showed not significant differences (P > 0.05).

  • The cats gene encodes the major catalase in Sreptomyces coelicolor, whose production increases upon H$_2$O$_2$treatment. Besides the previously identified primary promoter (catApl), a minor promoter (catAp2) was newly assigned by S1 nuclease mapping. The catAp2 transcript was observed transiently upon entry into the stationary phase in liquid culture and upon differentiation on solid plates, whereas the level of catApl transcription did not chance significantly during this growth transition. ThecatApl promoter was transcribed by the major vegetative RNA polymerase holoenzyme containing $\sigma$$\^$HrdB/, whereas the catAp2 was transcribed in vitro by the holoenzyme containing $\sigma$$\^$R/ that is activated under oxidative conditions. The cia-element regulating the H$_2$O$_2$-inducibility of catApl was identified within the 23 bp inverted repeat sequence located between -65 and -43 of the catApl promoter. We roamed this sequence HRE (H$_2$O$_2$-responsive Element). The distal half of the inverted repeat was more crucial for H$_2$O$_2$-dependent induction of the catApl transcript than the proximal half. HRE most likely serves as a binding site for the H$_2$O$_2$-responsive repressor CatR.

Antioxidant Effect of Mulberry Leaves and Yacon Tuber Extracts in High-fat Diet-fed Rats

  • The effect of mulberry leaves and yacon tuber extracts (MYE) on antioxidant was tested in this study. The present study investigated the in vivo effects of the anti-oxidative effect of MYE on catalase (CAT), superoxide dismutase (SOD), glutathione S-transferase (GST), glutathione peroxidase (GSH-Px), and thiobarbituric acid reactive substances (TBARS). The seven-day acclimation of the mice was divided into six groups: Normal diet group (NOR), high fat diet group (HFD), high fat diet with 0.5% hydroxycitric acid group diet group for positive group (HHCA), high fat diet with 1% mulberry leaf and 1% yacon diet group (MYE-1), high fat diet with 3% mulberry leaf and 3% yacon group (MYE-3) and high fat diet with 5% mulberry leaf and 5% yacon group (MYE-5). The effect of serum antioxidant in the catalase of MYE-1, MYE-3, and HHCA comparing to HFD by 31.0%, 27.7% and 45.2%, respectively (P<0.05~0.01). The effect on hepatic antioxidant in the catalase of HFD was significantly increased 3.7 (77.3%) times than that of NOR (P<0.01). But, the activities of catalase were decreased significantly in MYE-1, MYE-3, MYE-5 and HHCA by 21.7%, 24.2%, 24.9%, and 28.8% compared to HFD, respectively. GSH-Px was significantly decreased in MYE-1, MYE-3, MYE-5 and HHCA by 15.5%, 37.1%, 23.4%, and 23.7% compared to HFD, respectively (P<0.05). The activities of CAT, SOD, GST, GSH-Px, and TBARS were more significantly decreased in MYE-1 and MYE-3 than those of HFD and HHCA. MYE have shown significant effects on anti-oxidative function against high fat diet.
