Purpose: We examined the effects of Dansam (Salvia miltiorrhiza Bunge, SM) and Dansam-eum (DSE) on gastroesophageal reflux disease (GERD) and reflux esophagitis by comparing the inhibitory effects of SM and DSE with the representative treatment of PPI Omeprazole to determine if the effects of the prescription DSE based on Korean medicine are better than those of a single-use of SM. Methods: We performed experiments using both animal models and cancer cells. Results: Comparison of SM and DSE with PPI in the animal model tests revealed that the effects were superior for SM and DSE than for PPI in all categories (8-OHdG, p-IκB, PAR2, COX-1, cathelicidin, p-JNK, Caspase 3, ATP6V1B1, GRPR, serotonin, and NPY). In three categories (COX-1, serotonin, and NPY), SM and DSE showed superior results over the Controls. In the animal model tests, DSE was superior to SM in all categories except for serotonin. The anti-cancer effects observed in cancer cell tests revealed that SM and DSE had meaningful results in terms of cytotoxicity and cell movement rate, as well as in cancer cell apoptosis. Conclusions: We confirmed that SM and DSE can have effects on reflux esophagitis through the regulation of oxidative stress, inflammation, mucosal protection, apoptosis, proton pumping, and the enteroendocrine system in the stomach and esophagus. We also confirmed that SM and DSE have superior effects to those of PPI on all aspects, especially gastric mucosa protection and enteroendocrine system control. We also confirmed that SM and DSE have anti-cancer effects. Above all, we confirmed that DSE has superior effects on almost all aspects compared to using SM alone.
Objectives : The proliferation and migration of human aortic smooth muscle cells (HASMC) in response to activation by various stimuli plays a critical role in the initiation and development of atherosclerosis. This study was conducted to examine the effects of Nelumbo nucifera leaves (NNL) on the proliferation and migration of HASMC. Additionally, the mechanisms involved in any observed effects were also evaluated. Methods : Apoptotic cells were measured by staining with FITC-labeled annexin V, followed by flow cytometric analysis. The expression level of apoptosis related proteins was confirmed by western blot. And MMP-9 activity was measured by gelatin zymography and MMP-9 expression was measured by ELISA Results : NNL completely inhibited the proliferation of HASMC via induction of the expression of apoptotic proteins including annexin V, cleaved poly ADP-ribose polymerase (PARP), and caspase-3 and -8. NNL treatment resulted in the release of cytochrome c into cytosol, a loss of mitochondrial membrane potential, a decrease in Bcl-2 and Bcl-xL and an increase in Bax expression. NNL also blocked HASMC migration via suppression of MMP-9. Conclusions : Taken together, these results indicate that NNL has the potential for use as an anti-artherosclerosis agent.
Purple loosestrife-Lythrum anceps (Koehne) Makino is a herbaceous perennial plant belonging to the Lythraceae family. It has been used for centuries in Korea and other Asian traditional medicine. It has been showed pharmacological effects, including anti-oxidant and anti-microbial effects. However, the mechanisms underlying its anti-cancer mechanisms are not yet understood. In this study, we investigated the mechanism of apoptosis signaling pathways by ethanol extract of Lythrum anceps (Koehne) Makino (ELM) in human leukemia U937 cells. Treatment with ELM significantly inhibited cell growth in a dose-dependent manner by inducing apoptosis, as evidenced by the formation of apoptotic bodies (ApoBDs), DNA fragmentation and increased populations of sub-G1 ratio. Induction of apoptosis by ELM was connected with up-regulation of death receptor (DR) 4 and DR5, pro-apoptotic Bax protein expression and down-regulation of anti-apoptotic Bcl-2 protein, and inhibitor of apoptosis protein (IAP) family proteins (XIAP, cIAP-1, survivin), depending on dosage. This induction was associated with Bid truncation, mitochondrial dysfunction, proteolytic activation of caspases (-3, -8 and -9) and cleavage of poly(ADP-ribose) polymerase protein. Therefore, our data indicate that ELM suppresses U937 cell growth by activating the intrinsic and extrinsic apoptosis pathways, and thus may have applications as a potential source for an anti-leukemic chemotherapeutic agent.
The dense granule protein of Toxoplasma gondii, inhibitor of signal transducer and activator of transcription 1 (IST) is an inhibitor of signal transducer and activator of transcription 1 (STAT1) transcriptional activity that binds to STAT1 and regulates the expression of inflammatory molecules in host cells. A sterile inflammatory liver injury in pathological acute liver failures occurs when excessive innate immune function, such as the massive release of IFN-γ and TNF-α, is activated without infection. In relation to inflammatory liver injury, we hypothesized that Toxoplasma gondii inhibitor of STAT1 transcription (TgIST) can inhibit the inflammatory response induced by activating the STAT1/IRF-1 mechanism in liver inflammation. This study used IFN-γ and TNF-α as inflammatory inducers at the cellular level of murine hepatocytes (Hepa-1c1c7) to determine whether TgIST inhibits the STAT1/IRF-1 axis. In stable cells transfected with TgIST, STAT1 expression decreased with a decrease in interferon regulatory factor (IRF)-1 levels. Furthermore, STAT1 inhibition of TgIST resulted in lower levels of NF-κB and COX2, as well as significantly lower levels of class II transactivator (CIITA), iNOS, and chemokines (CLXCL9/10/11). TgIST also significantly reduced the expression of hepatocyte proapoptotic markers (Caspase3/8/9, P53, and BAX), which are linked to sterile inflammatory liver injury. TgIST also reduced the expression of adhesion (ICAM-1 and VCAM-1) and infiltration markers of programmed death-ligand 1 (PD-L1) induced by hepatocyte and tissue damage. TgIST restored the cell apoptosis induced by IFN-γ/TNF-α stimulation. These results suggest that TgIST can inhibit STAT1-mediated inflammatory and apoptotic responses in hepatocytes stimulated with proinflammatory cytokines.
Eosinophils play critical roles in the maintenance of homeostasis in innate and adaptive immunity. Although primarily known for their roles in parasitic infections and the development of Th2 cell responses, eosinophils also play complex roles in other immune responses ranging from anti-inflammation to defense against viral and bacterial infections. However, the contributions of pattern recognition receptors in general, and NOD-like receptors (NLRs) in particular, to eosinophil involvement in these immune responses remain relatively underappreciated. Our in vivo studies demonstrated that NLRC4 deficient mice had a decreased number of eosinophils and impaired Th2 responses after induction of an allergic airway disease model. Our in vitro data, utilizing human eosinophilic EoL-1 cells, suggested that TLR2 induction markedly induced pro-inflammatory responses and inflammasome forming NLRC4 and NLRP3. Moreover, activation by their specific ligands resulted in caspase-1 cleavage and mature IL-1β secretion. Interestingly, Th2 responses such as secretion of IL-5 and IL-13 decreased after transfection of EoL-1 cells with short interfering RNAs targeting human NLRC4. Specific induction of NLRC4 with PAM3CSK4 and flagellin upregulated the expression of IL-5 receptor and expression of Fc epsilon receptors (FcεR1α, FcεR2). Strikingly, activation of the NLRC4 inflammasome also promoted expression of the costimulatory receptor CD80 as well as expression of immunoregulatory receptors PD-L1 and Siglec-8. Concomitant with NLRC4 upregulation, we found an increase in expression and activation of matrix metalloproteinase (MMP)-9, but not MMP-2. Collectively, our results present new potential roles of NLRC4 in mediating a variety of eosinopilic functions.
Machaerium cuspidatum은 Fabaceae과 legume속에 속하는 캐노피 덩굴식물(canopy liana)로, 열대 우림 지역에 분포하는 식물이다. 본 연구에서는 Machaerium cuspidatum 메탄올 추출물(MEMC)의 항산화능을 확인하고, 항암 활성 및 그 기전을 인체 폐암세포 A549, 인체간암세포 HepG2를 사용하여 분석하였다. 먼저 DPPH를 이용하여 MEMC의 radical 소거능을 분석한 결과, 소거능 50%의 MEMC 농도($IC_{50}$)는 $1.66{\mu}g/ml$l이었으며, MEMC가 추출물인 것을 감안할 때 효과적인 항산화능을 보유하고 있음을 알 수 있었다. 또한 MEMC는 A549, HepG2 및 HT29에 대해 농도의존적으로 세포 사멸 효과를 보였으며, 세포 형태 변화를 유도하였다. A549와 HepG2를 사용하여 세포주기를 분석한 결과, MEMC 처리 농도가 증가할수록 apoptotic 세포를 의미하는 subG1기의 세포가 증가하였다. 따라서 MEMC에 의한 A549 및 HepG2의 apoptosis 유도를 Annexin V/7AAD 염색으로 확인한 결과, A549 및 HepG2에서 MEMC 농도의존적으로 Annexin V 양성 세포의 비율이 증가하였으며, DAPI 염색결과 MEMC 농도의존적으로 A549와 HepG2의 apoptotic body가 증가하였다. 특히 MEMC에 의한 apoptosis는 p53과 Bax의 발현증가 및 Bcl-2의 발현감소와 연관되어 있으며, caspase-3, -8, -9의 활성화와 poly ADP ribose polymerase (PARP)의 단편화를 일으키는 것을 확인하였다. 이러한 결과들로부터 MEMC는 외인성 및 내인성 경로를 통한 apoptosis 유도에 의해 A549와 HepG2의 증식을 억제시키는 것으로 사료된다.
Mifepristone (MIF)와 Tamoxifen (TAM)은 각각 전립선암과 유방암치료제로 오랫동안 사용되고 있다. MIF는 안드로겐수용체(AR) 양성인 세포와 음성이 세포 모두에서 세포사멸을 유도하며, TAM 은, 리간드-수용체작용 기작의 다양한 특성에 의하여 에스트로겐(ER) 양성인 세포뿐 만 아니라 다른 종류의 암세포에서도 세포사멸을 유도하는 것으로 알려져 있다. 본 연구에서는 AR 음성인 DU-145 전립선암세포에 있어서, MIF와 TAM의 세포독성이 세포 내 칼슘농도 변화에 기인된 세포사멸기작에 의한 것임을 보여준다. MIF와 TAM을 처리시 세포성장은 농도와 시간의존적으로 감소하였으며, confocal laser scanning microscopy (CLSM)과 fluorescence-activated cell sorting (FASC)로 세포를 분석한 결과 각각 MIF와 TAM을 2일간 처리한 세포에서 세포사멸이 진행되는 것을 관찰하였다. 세포독성효과를 비교했을 경우, TAM이 MIF 보다 강하게 작용하였다. MIF와 TAM을 처리한 세포 내 칼슘변화 측정 시, 칼슘농도 또한 처리 약물의 농도와 시간 의존적으로 증가하였다. 1.5 mM 칼슘배지와 칼슘제거된 배지에서의 실험결과를 비교한 바, 세포 내 칼슘증가는 외부로부터의 유입에 의한 것으로 생각된다. 세포독성효과와 마찬가지로 칼슘증대 효과 역시 TAM에서 뚜렷하게 나타났다. 수용체 매개 세포사멸기작의 초기에 관여하는 procaspase-8은 MIF 처리 시 뚜렷이 활성화 되었으나, TAM의 경우 활성화가 MIF의 경우에 비해 강하지 못하였다. 그러나, 세포사멸의 중추적인 역할을 하는 caspase-3은 TAM 을 처리한 세포에 있어서 활성 정도가 훨씬 높았다. 세포사멸과정의 중요한 조절 단백질인 Bcl-2 그룹단백질의 발현을 조사해 본 결과, 세포사멸 억제단백질인 Bcl-2의 발현은 MIF, TAM 처리 시 동일하게 감소한 반면, 촉진단백질인 Bax의 발현은 2-3배 가량 증대되었다. 이상의 결과로 보아 MIF와 TAM은 세포 내 칼슘조절을 통하여 세포사멸을 유도하나, 세포사멸의 초기단계는 MIF와 TAM이 서로 다른 경로를 경유할 가능성이 있는 것으로 생각된다.
신장에서 발생한 산화적 스트레스는 조직을 손상시키고 이는 만성신장질환으로 이어질 수 있다. 본 연구에서는 LLC-$PK_1$ 신장세포를 이용하여 산화적 스트레스 개선 효과를 살펴보았다. LLC-$PK_1$ 세포에 무막줄기세포추출물을 처리했을 때 체내 항산화 단백질인 heme-oxygenase-1, thioredoxin reductase 1, 및 NADPH quinine oxidoreductase-1의 발현이 증가함을 확인하였다. LLC-$PK_1$에 산화적 스트레스를 유도하기 위하여 3-morpholinosydnonimine (SIN-1)을 처리한 결과 세포생존율이 감소하여 산화적 스트레스로 인해 세포가 손상됨을 확인하였다. 그러나 무막줄기세포추출물을 처리하였을 때 세포생존율이 증가하였으며, $2.5{\mu}g/mL$에서 세포생존율이 58.84%에서 64.43%까지 증가하였다. 또한 무막줄기세포추출물은 LLC-$PK_1$ 세포에서 SIN-1으로 유도된 염증 및 세포사멸을 조절하였다. 염증 관련 단백질인 inducible nitric oxide synthase와 cyclooxygenase-2는 무막줄기세포 추출물을 처리했을 때 단백질 발현이 감소하였고, 세포사멸과 관련된 B-cell lymphoma-2-associated X protein/B-cell lymphoma-2 비율과 cleaved caspase-3, cleaved-poly (ADP-ribose) polymeras의 단백질 발현이 감소함을 확인하였다. 결과적으로 무막줄기세포출물은 SIN-1을 처리한 LLC-$PK_1$ 세포에서 산화적 스트레스에 대한 보호 효과가 있음을 알 수 있었으며, 이들 결과를 바탕으로 무막줄기세포추출물의 항산화 기능성 소재로서의 활용 가능성을 확인하였다.
Objectives : The purpose of this study was to investigate the effect of Dangguibohyultang (DB) and its combination (DB-I; Astragali membraneus BUNGE : Angelica gigas NAKAI=5:1, DB-II; Astragali membraneus BUNGE:Angelica gigas NAKAI=1:1, DB-III; Astragali membraneus BUNGE:Angelica gigas NAKAI=1:5,) on apoptosis in human colorectal adenocarcinoma HCT116 cells. Methods : To study the cytotoxic effect of methanol extract of DB-I, DB-II and DB-III on HCT116 cells, the cell viability was determined by XTT reduction method and ttypan blue exclusion assay. To confirm the induction of apoptosis, the cleavage of poly ADP-ribose polymerase (PARP), a substrate for caspase-3 and a typical sign of apoptosis, and the activation of procaspase-3, -8 and -9 were examined by western blot analysis. Furthermore, DB-induced apoptosis was confirmed by DNA fragmentation. The release of cytochrome C from mitochondria to cytosol, the level of Bcl-2 and Bax, and the expressions of Raf/MEK/ERK were examined by western blot analysis. Results : DB-I and DB-II reduced proliferation of HCT116 cells in a dose-dependent manner. DB-I and DB-II decreased procaspase-3, -8, -9 levels in a dose-dependent manner and induced the clevage of PARP. DB-I and DB-II also triggered the mitochondrial apoptotic signaling by increasing the release of cytochrome C from mitochondria to cytosol, decreasing of anti-apoptotic Bcl-2, and increasing of pro-apoptotic Bax. DB-I and DB-II decreased the activation of Ras/Raf/MEK/ERK cascade in a dose-dependent manner. Conclusion : These results suggest that DB-I and DB-II induce apoptosis via mitochondrial pathway in HCT116 cells. Furthermore, Raf/MEK/ERK cascade is involved in DB-induced apoptosis. These results suggest that DB is potentially useful as a chemotherapeutic agent in human liver cancer.
천궁(C. officinale)은 예로부터 민간처방 약재로 사용되었으며, 항염증, 항산화, 항암 및 신생혈관억제 등의 효능을 가지는 것으로 알려져 있다. 하지만 혈구암세포에서 apoptosis 유발과 관련된 분자생물학적 기전에 대해서는 명확히 밝혀져 있지 않다. 본 연구에서는 인체 혈구암세포인 U937 세포에서 천궁의 열수, 에탄올 및 메탄올 추출물(WECO, EECO 및 MECO)이 유발하는 항암효과 및 항암기전을 조사하였다. 먼저 WECO, EECO 및 MECO가 유발하는 증식억제 정도를 조사한 결과 EECO가 가장 뛰어난 효능을 가진다는 것을 알 수 있었으며, 이러한 현상이 apoptosis 유발에 의한 것임을 annexin-V 염색, apoptotic body 형성, DNA 단편화 및 MMP 소실 등을 통하여 확인하였다. EECO 처리에 의한 apoptosis 유발에는 DR4의 발현 증가와 함께 cIAP-1, Bcl-2 및 total Bid의 발현감소가 관여하였으며, caspases-3, -8 및 -9의 활성화와 함께 caspases-3의 기질 단백질인 PARP, β-catenin 및 PLC γ1의 단편화도 관찰되었다. 또한 EECO는 AMPK signaling pathway를 활성화시키는 것으로 나타났으며, AMPK 억제제인 compound C를 이용하여 AMPK의 활성을 억제하였을 경우 EECO에 의하여 유발되었던 apoptosis가 현저하게 감소되는 것으로 나타났다. 이상의 결과를 살펴볼 때 인체 혈구암세포인 U937 세포에서 EECO에 의하여 유발되는 apoptosis는 AMPK가 중요한 조절자로서 작용하는 것으로 생각된다.

② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
