The Chemical Constituents and Their Pharmacological Activities of Endemic Medicinal Plants in Korea -Pharmacologically Active Constituents of Fraxinus Species- (한국 특산 생약의 약효성분 및 약리활성 -Fraxinus spp.의 약효 성분-)

  • Three components, fraxetin-8-glucoside, esculetin-6-glucoside and mannitol, were isolated from the stem bark of Fraxinus chiisanensis var. stenophylla, F. japonica var. intermedia and F. densata. The MeOH extract of the cortex of F. densata had the antiinflammatory activity on the carrageenin-induced paw edema in rat. The MeOH extracts of the cortex of all three Fraxinus spp. have the potent analgesic activity on the HOAc-induced writhing syndrome in mouse and the hepatoprotective activity on the $CCl_4-induced$ fatty liver in rat: protection of ballooning formation and inhibition of sGPT and sGOT increased by $CCl_4$.

Study on the Antiinflammatory, Anticoagulative and Analgesic effects of Sambitang in the experimental animal model (삼비탕(三痺湯)의 항염(抗炎), 항응고(抗凝固) 및 진통효과(鎭痛效果)에 대(對)한 연구(硏究))

  • This study was designed to elucidate the antiinflammatory, cardiovascular, antithrombotic, and analgesic effect of Sambitang. The antiinflammatory effects was measured by the method of carrageenin induced edema, protein leakage test using CMC-pouch, and the effect of Sambitang on the cardiovascular system was observed by the change of flow rate of Ringer solution in the vascular system in the ear of rabbit, and the contraction and dilatation of rat tail artery. Death rate, platelet aggregation, plasma coagulation activity was observed for the measurement of the anticoagurative effect of Sambitang, and the analgesic effect was measured by the acetic acid method and hot plate method. The result was as follows: 1. Sambitang administration, edema and protein leakage was significantly decreased. 2. The drug increased the auricular blood flow in rabbit. 3. The drug relaxed the artery contraction by pretreated norepinephrine in rat. 4. The drug inhibited the death rate of mouse which was led to thromboembo- lism by serotonin and collagen. 5. The drug inhibited the platelet aggregation in rat. 6. The drug prolonged the prothrombin time and activated partial thromboplastin time on the test of plasma coagulation factor activity in rat, but was not valuable. 7. The slight anagesic effect of Sambitang extract was confirmed by the observation of writhing syndrome, paw licking time, and escape time.

  • The root of Pseudostellaria heterophylla Pax et Hoffm. (Caryophyllaceae) is a herbal medicine called Taejasam(太子蔘) and used as a good tonic in China that strengthens the functions of the lung and the stomach like Ginseng in Korea. It has been known to have the antitussive activity for tuberculosis, appetizing effect and antifatigue activity, and so on. Especially it has been known to generate body fluids for the severe thirst and the shortage of water in body after a febrile disease for a long time. Recently some components of cyclic peptides, pseudostellarins A,B,C,D,E,F,G, that inhibit the activity of tyrosinase, were separated from this root. In this report several biological activities including acute toxicity, analgesic activity, antifatigue activity, bile juice increasing activity, hepatoprotective activity and antiinflammatory activity were investigated. These results showed as such; $LD_{50}$ of methanol extracts was above 10,000 mg/kg. The analgesic effect was revealed in the writhing test using rats. The BuOH fractions showed a prominent antifatigue effect against immobilized stress, significant hepatoprotective activities against $CCl_4$ intoxication and inhibitory effect on carrageenin-induced edema in rat's paw.

  • The sedative, antipyretic, analgesic and anticonvulsive action, action on the isolated ileum in mice, retricting action on edema, action on blood pressure and respiration of Sipmidojuksan were evaluated. The results were as follows; sedative effects were recognized by the unbalanced effects of spontaneous momentum by wheel cage method, muscle relaxing action by rotor rod method and prolongation of sleeping hours. In mice, a significant antipyretic effect to endotoxin was recognized. Significant anagesic effects by acetic acid and Randall-Sellito method were recognized. Significant anticonvulsive effects to strychnine and picrotoxin were recognized. Spontaneous momentum of isolated ileum in mice was restricted, and relaxing effects on smooth muscle of digestive organ were noted by anti-acetylcholine, anti-barium chloride and anti-histamine effects. Significant antiedemic effects to carrageenin and histamine were recognized. Dilatation of blood vessels and decrease of blood pressure were noted.

Anti-inflammatory and Antinociceptive Activities of 'Coccinia indica W. &A.' Fruit Juice Powder in Animals

  • The fresh fruit juice powder of Coccinia indica W.&A., (Cucurbitaceae, CJP) was studied for the possible activities of antiinflammatory and antinociceptive to rationalize the folkloric use of the plant juice as rasayana. CJP at the doses of 50-200 mg/kg caused a significant (P<0.05 to P<0.001) inhibition of paw edema induced by ${\lambda}$ carrageenin (1%) and histamine $(10^{-3}\;g/ml,\;0.1\;ml)$ in rats. The effect was comparable to the standard cycloxygenase inhibitor brufen at 100 mg/kg and protective percentages were 63.41% and 65.78% respectively. Administration of CJP (50-200 mg/kg) exhibited a moderate increase of the pain threshold on analgesy-meter induced mechanical pain. However CJP significantly prevented the writhing induced by acetic acid in mice and the percentages of inhibitions were 16.98%-35.47%, which is equivalent to 36.67% produced by brufen. These data indicate that the fruit juice of Coccinia indica rationalizes the traditional system of medicine.

Studies on the Concurrent Administration of Medicines(I) -Anti-inflammatory and Analgesic Actions of Galgun-tang and Aspirin- (의약품(醫藥品)의 병용(倂用) 투여(投與) 효과에 관한 연구(硏究)(I) -갈근탕(葛根湯) 엑기스와 Aspirin의 병용(倂用) 투여(投與)가 항염(抗炎) 및 진통작용(鎭痛作用)에 미치는 영향(影響)-)

  • The extract of Galgun-tang at oral doses of 200 and 400 mg/kg showed remarkable inhibition of 42.5% and 45.0%, respectively, on the carrageenin edema in rat paw. In case of combined oral administration of the extract at each dose of 100, 200 and 400mg/kg with 100mg/kg of aspirin, the inhibitions were 70.5%, 83.6% and 80.7% respectively. These results showed the remarkable effects as compared with those of aspirin which showed 50.3% and 69.4% of inhibition at doses of 100 and 200mg/kg, respectively. On the other hand, on the writhing syndrome in mice induced by 0.7% acetic acid solution, the oral administration of the extract showed remarkable inhibition of 18.5 and 47.7% at doses of 200 and 400mg/kg, respectively. In case of combined oral administration of the extract at each dose of 100, 200 and 400mg/kg with 100mg/kg of aspirin, the inhibition percentages of writhing syndromes were 66.6, 72.3 and 80.8, respectively, whereas the inhibition at doses of 100 and 200mg/kg of aspirin showed 59.9 and 69.5%, respectively.

Antiinflammatory, Analgesic and Antihyperuricemic Effects of 'Gyejakjimo-Tang' in Rats (계작지모탕(桂芍知母湯)의 소염.진통작용 및 Alcohol성 고뇨산혈증(高尿酸血症)에 미치는 영향)

  • 'Gyejakjimo-Tang(桂芍知母湯)', which is composed of Cinnamon bark(桂枝), Peony root(芍藥), Anemarrhena root(知母) and other seven herbs, is described as antipyretic, diuretic and analgesic prescription in traditional medical literatures including Geumgwe-Yoryak(金櫃要略). So it is being used in the treatment of gout clinically in oriental region. As the results of the pharmacological and biochemical trials of the prescription, it was found to have antiinflammatory and significant analgesic effects indicated by carrageenin edema, dye permeabilities and writhing behavior. And it potentiated the output of uric acid in blood and the increase of uric acid in urine of alcohol-toxicated rats, which mean the decrease of uric acid level only by excretion procedure. But the prescription didn't show any effects on the activities of adenine/guanine deaminase, purine nucleoside phosphorylase, xanthine oxidase or uricase, which are related with formation and metabolism of uric acid.

Studies on CNS-Depression and Antiinflammatory Action of Ssangwhatang (쌍화탕의 중추억제 및 항염증작용에 관한 연구)

  • Ssangwhatang which has been known to have antipyretic, analgesic and antifatigue effects are composed of Paeoniae Radix, Angelicae gigantis Radix, Astragali Radix, Rehmaniae Radix, Cnidii Rhizoma, Cassiae Cortex, Glycyrrhizae Radix and Puerariae Radix. Effects of some formulations of Ssangwhatang were studied with respect to the analgesic, antipyretic, hypothermic, anticonvulsive and antiinflammatory actions in experimental animals. The results obtained were as follows: 1) Prescription A, B, C and D of Ssangwhatang were showed significant analgesic effect by the acetic acid stimulating method in mice. 2) All Ssangwhatang-prescriptions used in this studies had hypothermic effect in normal mice. 3) Significant antipyretic effect in febrile rabbits were recognized in prescription A and C of Ssangwhatang, while not in prescription B and D of Ssangwhatang. 4) Prescription A, B, C and D of Ssangwhatang showed marked antiinflammatory effect against the edema induced by carrageenin in rats. 5) Prescriptions of Ssangwhatang did not show significant anticonvulsive effect against the convulsion induced by picrotoxin and strychnine in mice. From the above results, analgesic, hypothermic, antipyretic and antiinflammatory effects were recognized in Ssangwhatang. Ssangwhatang A- and C- prescriptions added with Pueraiae Radix showed better effects than prescription B and C of Ssangwhatang.

Studies on the anti-inflammatory activity of aralia continentalis (II)

  • By tracing albumin stabilizing activity an anti-inflammatory component, continentalic acid was isolated from ether-soluble acidic fraction of Aralia continentalis. Continenetalic acid in a concentration of 0.115mg/3ml gave 50% inhibition for heat denaturation of albumin. The protein stabilizing potency of it was approximately three and eleven times that of phenylbutazone and that of salicylic acid, respectively. The anti-inflammatory actions of it and its methylester were investigated employing carrageenin-induced edema in rat paw. Continentalic acid administered s. c. showed an activity of about three times of hydrocortisone. When administered p. o., it was still active, but its methylester was more active than phenylbutazone, suggesting the poor absorption of it in gastorointestinal tract. Its chemical structure was identified by chemical and spectral studies as (-) pimara 8(14), 15-diene-19-oic acid, which was already isolated from A. dordata, but not reported for its biological activity.

Suppressive Effects of Furonaphthoquinone NFD-37 on the Production of Lipopolysaccharide-Inducible Inflammatory Mediators in Macrophages RAW 264.7

  • 2-Methyl-2-(2-methylpropenyl)-2,3-dihydronaphthoquinone[2,3-b]furan-4,9-dione (N FD-37) is a synthetic furonaphthoquinone compound. In this study, we determined that NFD-37 could inhibit the lipopolysaccharide (LPS)-induced production of inflammatory mediators in macrophages RAW 264.7. This compound inhibited LPS-induced nitric oxide (NO) or prostaglandin (PG) $E_{2}$ production in dose-dependent manners, with $IC_{50}$ values of 7.2 ${\mu}M$ and 5.3 ${\mu}m$, respectively. As the positive controls, pyrrolidine dithiocarbamate (30 ${\mu}M$) exhibited a $57{\%}$ inhibition of NO production, and NS-398 ($1{\mu}M$) manifested a $48{\%}$ inhibition of $PGE_2$ production. The inhibitory effects of NFD-37 on NO and $PGE_2$ production were determined to occur in conjunction with the suppression of inducible NO synthase or cyclooxygenase-2 expression. NFD-37 also inhibited the production of LPS-inducible tumor necrosis factor-${\alpha}$, interleukin (IL)-$1{\beta}$ and IL-6, at $IC_{50}$ values of 4.8-8.9 ${\mu}M$. We also determined the anti-inflammatory efficacy of NFD-37 using carrageenin-induced paw edema in experimental mice.
