Carotenoid Pigments from Suspended and Sinking Particulate Matter in Prydz Bay, Antarctica

  • Suspended and sinking particles were collected during ODP Leg 119 to the Indian Ocean sector of the Antarctic Ocean. Field work was carried out at four sampling sites in Prydz Bay. Two of these sites were located in the Outer Bay, and two in the Inner Bay. At the four locations, a total of ten deployments of a sediment trap array were made. The concentrations of carotenoids both in suspended and sinking particulate matter in Prydz Bay were analyzed using HPLC. Fucoxanthin was the dominant carotenoid pigments both in suspended and sinking particles. The present study also indicates that 19'-hexanoyoxyfucoxanthin-containing prymesiophytes (Phaeocystis spp.) was abundant in the study area. The flux rates of carotenoids were generally highest at 50 m, and approximately double the flux rates at deeper horizons, however, at Inner Bay sites, the mean flux rates of carotenoids were greatest at 200 m, and 3 times greater than that of 50 m. Such anomalous high fluxes at 200 m imply that grazers were locally abundant between 100 m and 200 m at these sites close to land, and this hypothesis is supported by visual evidence of lots of fecal pellets in the 200 m trap. Integrates standing stocks versus sinking pigments data support that particulate material in Prydz Bay was not recycled rapidly.

Carotenoids and total phenolic contents in plant foods commonly consumed in Korea

  • Phytochemicals are reported to provide various biological functions leading to the promotion of health as well as the reduced risk of chronic diseases. Fat-soluble plant pigments, carotenoids, are extensively studied micronutrient phytochemicals for their potential health benefits. It is noteworthy that specific carotenoids may be responsible for different protective effects against certain diseases. In addition, each carotenoid can be obtained from different types of plant foods. Considering the fact that the phytochemical content in foods can vary according to, but not limited to, the varieties and culture conditions, it is important to establish a database of phytochemicals in locally produced plant foods. Currently, information on individual carotenoid content in plant foods commonly consumed in Korea is lacking. As the first step to support the production and consumption of sustainable local plant foods, carotenoids and total phenolic contents of plant foods commonly consumed in Korea are presented and their potential biological functions are discussed in this review.

저장중 호박분말의 카로테노이드 색소 변화 (Changes of Carotenoids of the Pumpkin Powder during Storage)

  • 열풍건조한 호박분말을 2$0^{\circ}C$에서 1개월 저장하였을 때 저장 초기 총 카로테노이드 함량이 14.46mg%이던 것이 저장 1개월후 빛의 유무에 관계없이 약 65~70% 감소하였으며, TLC 분리에서 빛의 영 향에 관계없이 5개 의 반점으로 분리되었으나 저장시일의 경과에 따른 카로테노이드 색소의 차이를 구별하기는 어려웠다. HPLC에서 lutein의 경우 빛을 조사한 분말은 저장 15후 부터 함량이 다소 증가하였고, 빛을 차단한 분말은 1개월간 함량 변화가 일어나지 않았으며, lycopene은 모두 저 장 말기까지 거의 변화가 일어나지 않았다. $\beta$-Carotene 의 경우 빛을 조사한 처리구가 15일경부터 그 함량이 감소한 반면 빛을 차단한 분말은 저장말기에 가서야 감소하였으며, $\alpha$-carotene은 저장 전 기간을 통해 두 처리구 모두 변화를 나타내지 않았다. 색도조사에서 황색 도인 b값은 두처리구 모두 저장 10일 이후부터 감소하 였으나 빛 차단구의 경우가 그 감소폭이 적은 것으로 나타났다.

두유의 광산화 안정성에 영향을 주는 요인 (Factors Affecting the Photooxidative Stability of Soymilk)

  • The effects of chlorophyll, tocopherols($\alpha$-tocopherol, ${\gamma}$-tocopherol and $\delta$-tocopherol), carotenoids ($\beta$-carotene and lutein), light sources, light intensities and strage temperatures on the photooxidative stability of soymilk were studied by measuring TBA value and depleted headspace oxygen(DHO) of soymilk. The samples were stored in the light storage box for 6 days and evaluated for the photooxidative stabilities. As the concentrations of chlorophyll increased, TBA value and DHO of the sample increased significantly(p<0.05), indicating chlorophyll acting as a photosensitizer. However, as the concentrations of tocopherols ($\alpha$-tocopherol, ${\gamma}$-tocopherol and $\delta$-tocopherol) and carotenoids ($\beta$-carotene and lutein) increased, TBA values and DHO of the samples decreased significantly(p<0.05). The light screening effects of carotenoids on DHO in the samples were not significantly different from the control at p>0.05. Therefore, there was no light screening effects of carotenoids on the oxidative stability of soymilk. The results indicate that tocopherols and carotenoids reduce the photooxidative stability of soymilk. $\delta$-Tocopherol was the most effective in photosensitized oxidation followed by ${\gamma}$-and $\alpha$-tocopherols in the order of increasing stability. $\beta$-Carotene was significantly(p<0.05) more effective than lutein in minimizing the chlorophyll-sensitized photooxidation of soymilk. Visible light was more effective than UV light in decreasing the photooxidative stability of soymilk. Therefore, photooxidation of soymilk containing chlorophyll is mainly due to photosensitized oxidation rather than photolysis reaction. As the intensities of fluorescence light increased, TBA values and DHO of the samples increased significantly at P<0.05. However, as the storage temperatures increased, TBA values and DHO of soymilk did not change significantly at p>0.05.

우렁쉥이 껍질 카로테노이드의 가열분해로 생성되는 휘발성 화합물의 특성 (Volatile Compounds Formed by Thermal Degradation of Ascidian Tunic Carotenoids)

  • 우렁쉥이 껍질로부터 추출한 카로테노이드 색소의 식품첨가물로서의 이용 가능성을 알아보기 위하여 SDE(simultaneous distillation and extraction/concentration) 장치를 이용하여 휘발성 화합물을 추출하였다. 이를 GC 및 GC-MS로 분석하여 총 63성분의 휘발성 화합물을 동정하였고, 대부분의 화합물이 카로테노이드의 가열분해로 생성되어지는 것으로 나타났다. 동정된 화합물중 대표적인 화합물은 1,3,5-trimethylbenzene, 3,5,5-trimethyl-3-cyclohexen-1-ol, 3,5,5-trimethyl-3-cyclohexen-1-one, 1,1,2,3-tetramethyl-2-cyclohexen-5-ol, 1,1,2,3-tetramethyl-2-cyclohexen-5-one, 2,3,4,4-tetramethyl-6-hydroxy-2-cyclohexene-1-one, 1,2,3,8-tetrahydro-3,3,6-trimethyl-1-naphtol, dihydroacetinidolide, ${\beta}-ionone$, 2-(1,1,5-trimethyl-3-hydroxy-5-cyclohexen-6-yl)-1-to-lylethene, 2,6-dimethyl-8-(1,1,5-trimethyl-3-hydroxy-5-cyclohexen-6-yl)-1,3,5-octatriene-7-yne 등 이었다. 이중 몇가지 화합물의 생성기작도 추정해 보았다.

배합사료 내 색소 원료에 따른 비단잉어 홍백C(yprinus carpio) 치어의 성장과 체색 변화 (Effect of Dietary Carotenoids Sources on Growth and Skin Color of Red- and White-colored Fancy Carp Cyprinus carpio var. koi)

  • A feeding trial was conducted to investigate the effects of dietary carotenoids sources on growth and skin color of red- and white-colored fancy carp Cyprinus carpio var. koi. Nine experimental diets (designated as Con, CP, PA, SP, OP, MB, TO, BE and PO) were formulated to contain Carophyll Pink, red paprika, Spirulina, Opuntia, mandarin bark, tomato, beet and Porphyra, respectively. Each experimental diet was fed to two replicate groups of fish (22.9 g/fish) to visual satiation three times a day for 8 weeks. Weight gain and feed efficiency of fish fed the diets containing Spirulina or Opuntia were higher than those of fish fed the control diet (P<0.05). The values of $a^*$, $L^*$ and $b^*$ of fish skin were significantly changed by dietary carotenoids sources (P<0.05). The $a^*$ values of fish fed the diets containing Carophyll Pink, red paprika and Spirulina were higher than those of fish fed other diets (P<0.05). The skin total carotenoids of fish fed the diets containing Spirulina and Opuntia were higher than those of others (P>0.05). Based on the results of this study, it can be concluded that dietary inclusion of red paprika and Spirulina pacifica could increase the skin redness of red- and white-colored fancy carp.

백내장클리닉을 내원한 성인의 렌즈혼탁지수에 따른 항산화 영양소 및 카로티노이드 섭취량 평가 (Intakes of Antioxidant Nutrients and Carotenoids according to Lens Turbidity Level among Adults Visiting Cataract Clinic)

  • This study examined intakes of nutrients, carotenoids and polyphenols according to lens turbidity levels among adults visiting a cataract clinic. A total of 102 males and females aged 40 years or higher agreed to participate in the study, and a face-to-face survey interview was conducted to obtain information on general characteristics, health behaviors, and dietary intakes of each participant. The dietary data were collected by a food frequency questionnaire which was previously validated in a Korean population. In addition to essential nutrients, intakes of a total of 10 carotenoids and polyphenols were calculated based on the tables of food functional composition developed by National Academy of Agricultural Science. The subjects were divided into 3 groups according to turbidity levels (< 16, ${\geq}16$ and < 25, ${\leq}25$), and health behaviors and dietary intakes were compared among the turbidity groups. Data showed seemingly higher energy intake and lower dietary antioxidant (i.e., vitamin A, ${\beta}$-carotene, lutein, zeazanthin, lycopene, cryptozanthin) intakes in the highest turbidity group. However, no statistically significant findings were found in all the comparative analysis on characteristics of health behavior and intakes of nutrients, carotenoids and polyphenols. The current study findings should be cautiously interpreted in consideration of several limitations including a cross-sectional study design, a small sample size, uneven sample size distribution across turbidity groups, and limited generalizability due to using a convenience sample. Therefore we cannot conclude that the risk of cataract is unrelated with dietary antioxidant intakes based solely on the results of this study.

아까시재목버섯 유래 카르테노이드 계열 항산화 물질 생산 (Development of carotenoid production process using perenniporia fraxinea)

  • 이 연구에서는 Perenniporia fraxinea를 사용하여 배지가 카르테노이드 생성 및 균사 성장에 미치는 영향을 분석했습니다. 0.1% 미만의 펩톤을 함유한 맥아추출물은 카르테노이드의 생성을 촉진했으며 0.2% 이상의 펩톤이 그 생산을 억제하는 것으로 나타났다. 맥아추출물이 없으면 Perenniporia fraxinea는 소량의 펩톤 첨가에도 불구하고 카르테노이드를 생산하지 못했다. 아카시재목버섯의 배양을 통하여 생산된 균사체를 원심분리로 배양액을 분리하여 상층액을 카르테노이드 용액으로 회수가 가능하였다. 균사체 내부에 축적된 카르테노이드는 에탄올을 사용하여 추출하였다. 배양액에서 분리 또는 추출한 카르테노이드 용액은 우수한 DPPH 라디칼 소거 활성을 나타냈다. 본 연구에서 생산된 항산화 카르테노이드는 천연물 유래 물질로 독성 및 부작용이 없으며 항산화 효과가 우수하여 활성산소에 의해 생성된 산화물을 효과적으로 제거할 수 있다.

FT-IR 스펙트럼 데이터의 다변량 통계분석을 이용한 고기능성 아프리칸 얌 식별 및 기능성 성분 함량 예측 모델링 (Discrimination of African Yams Containing High Functional Compounds Using FT-IR Fingerprinting Combined by Multivariate Analysis and Quantitative Prediction of Functional Compounds by PLS Regression Modeling)

  • 본 연구에서는 UV-VIS spectrophotometer를 이용한 total carotenoids, flavonoids, phenolics 함량 데이터와 FT-IR 스펙트럼 데이터를 다변량통계분석법을 통하여 기능성 성분 함량이 높은 아프리칸 얌 고속 선발 시스템을 구축하였다. 62개 아프리칸 얌의 total carotenoids 함량은 $0.01-0.91{\mu}g{\cdot}g^{-1}$ dry wt 나타냈다. Total flavonoids와 phenolics 함량은 $12.9-229.0{\mu}g{\cdot}g^{-1}$ dry wt와 $0.29-5.2mg{\cdot}g^{-1}$ dry wt로 각각 나타났다. 아프리칸 얌은 FT-IR 스펙트럼상의 1700-1500, 1500-1300, $1,100-950cm^{-1}$, 부위에서 중요한 스펙트럼 변화가 나타났다. 이 부위는 각각 amide I과 II을 포함하는 아미노산 및 단백질계열의 화합물, phosphodiester group을 포함한 핵산 및 인지질 그리고 단당류나 복합 다당류를 포함하는 carbohydrates 계열의 화합물들의 질적, 양적 정보를 반영하는 부위이다. PCA 분석과 PLS-DA 분석에서 62개 아프리칸 얌은 유연성이 높은 종으로 3개의 그룹을 형성하였다. 아프리칸 얌의 FT-IR 스펙트럼 데이터와 UV-VIS spectrophotometer을 이용한 total carotenoids, flavonoids, phenolics 함량 데이터 간에 PLS regression 분석하였다. Total carotenoids, flavonoids, phenolics 함량 성분의 실측 값과 예측 값간에 상관계수($R^2$)가 각각 0.83, 0.86, 0.72로 나타났다. 이 결과, 아프리칸 얌으로부터 FT-IR 스펙트럼을 이용한 total carotenoids, flavonoids, phenolics 함량 예측이 가능하였다. 본 연구에서 확립된 대사체 수준에서 아프리칸 얌의 유용 기능성 성분 함량 예측 모델링을 통해 품종, 계통의 신속한 선발 수단으로 활용이 가능할 것으로 예상된다.

Microalgal Biotechnology: Carotenoid Production by the Green Algae Dunaliella salina

  • Unicellular green algae of the genus Dunaliella thrive in extreme environmental conditions such as high salinity, low pH, high irradiance and subzero temperatures. Species of Dunaliella are well known in the alga biotechnological industry and are employed widely for the production of valuable biochemicals, such as carotenoids. Some strains of Dunaliella are cultivated commercially in large outdoor ponds and are harvested to produce dry algal meals, such as polyunsaturated fatty acids and oils for the health food industry, and coloring agents for the food and cosmetic industries. During the past decade, the advances in molecular biology and biochemistry of microalgae, along with the advances in biotechnology of microalgal mass cultivation, enabled this microalga to become a staple of commercial exploitation. In particular, the advent of molecular biology and mutagenesis in Dunaliella has permitted enhancements in the carotenoids content of this green alga, making it more attractive for biotechnological applications. Accordingly, the present review summarizes the recent developments and advances in biotechnology of carotenoid production in Dunaliella.
